1Effects of Tai Ji Quan training on gait kinematics in older Chinese women with knee osteoarthritis: A randomized controlled trial显示文摘Background: Although Tai Ji Quan has been shown to relieve pain and improve functional mobility in people with knee osteoarthritis(OA), little is known about its potential benefits on gait characteristics among older Chinese women who have a high prevalence of both radiographic and symptomatic knee OA. This study aims to assess the efficacy of a tailored Tai Ji Quan intervention on gait kinematics for older Chinese women with knee OA.Methods: A randomized controlled trial involving 46 older women in Shanghai, China, with clinically diagnosed knee OA. Randomized(1:1)participants received either a 60 min Tai Ji Quan session(n = 23) 3 times weekly or a 60 min bi-weekly educational session(n = 23) for 24 weeks.Primary outcomes were changes in gait kinematic measures from baseline to 24 weeks. Secondary outcomes included changes in scores on the Western Ontario and Mc Master University Osteoarthritis Index(WOMAC) and Short Physical Performance Battery(SPPB).Results: After 24 weeks the Tai Ji Quan group demonstrated better performance in gait velocity(mean difference, 8.40 cm/s, p = 0.01), step length(mean difference, 3.52 cm, p = 0.004), initial contact angle(mean difference, 2.19°, p = 0.01), and maximal angle(mean difference, 2.61°,p = 0.003) of flexed knees during stance phase compared to the control group. In addition, the Tai Ji Quan group showed significant improvement in WOMAC scores(p < 0.01)(mean difference,-4.22 points in pain, p = 0.002;-2.41 points in stiffness, p < 0.001;-11.04 points in physical function, p = 0.006) and SPPB scores(mean difference, 1.22 points, p < 0.001).Conclusion: Among older Chinese women with knee OA, a tailored Tai Ji Quan intervention improved gait outcomes. The intervention also improved overall function as indexed by the WOMAC and SPPB. 2Prognostic value of interim ^(18)F-FDG PET/CT in diffuse large B-cell lymphoma显示文摘Objective: Diffuse large B-cell lymphoma (DLBCL) is a heterogeneous disease. The prognostic factor currently used is not accurate enough to predict the outcomes of patients with DLBCL. The prognostic significance of interim PET/CT in DLBCL remains controversial. The aim of this study is to determine the predictive value of interim 18F-FDG PET/CT after first-line treatment in patients with DLBCL. Methods: Thirty-two patients with DLBCL underwent baseline, interim and post-treatment 18F-FDG PET/CT scans. Imaging results were analyzed for the survival of patients via software SPSS 13.0, retrospectively. Results: Thirty-one of the 32 patients were treated with R-CHOP regimen, and interim 18F-FDG PET/CT of 24 patients was performed after 2 cycles of treatment. After a median follow-up period of 16.7 months, the 2-year progression-free survival (PFS) rates were significantly different between the groups above and below SUV max cut-off value of 2.5 (P=0.039). No significant differences were found in the 2-year PFS rates if SUV max cut-off values were set as 2.0 and 3.0, respectively (P=0.360; P=0.113). 3Prognostic value of pre-and post-transplantation 18F-fluorodeoxyglucose positron emission tomography results in non-Hodgkin lymphoma patients receiving autologous stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous stem cell transplantation(ASCT) is the standard of care in the upfront or relapsed/refractory setting in some patients with non-Hodgkin lymphoma(NHL).However,a proportion of patients do not respond to ASCT.^(18)F-fluorodeoxyglucose(FDG) positron emission tomography(PET)/computed tomography(CT) has been widely used for staging,response evaluation,and prognosis prediction.Here,we investigated the prognostic role of PET/CT in NHL patients before and after ASCT.Methods: A retrospective study was conducted at Peking University Cancer Hospital.All NHL patients who underwent ASCT between March 2010 and July 2016 were identified.Patients who had PET/CT scan before and after ASCT were included.Deauville criteria(5-point scale) were used to interpret PET scans.Univariate and multivariate survival analyses were performed using Cox regression.The predictive value of PET scanning was estimated by comparing the area under the receiver operating characteristic(ROC) curve.Results: In total,79 patients were enrolled in this study.In univariate analysis,pre-and post-ASCT PET result was identified as prognostic factors for 3-year progression-free survival(PFS) and overall survival(OS).Patients with negative pre-ASCT PET result demonstrated significantly better PFS(84.2% vs.54.2%) and OS(89.2% vs.63.6%) than patients with positive pre-ASCT PET result.PFS(91.6% vs.25.3%) and OS(96.5% vs.36.8%) were also significantly different between patients with negative and positive post-ASCT PET result.Multivariate analysis also showed a significant association between survival and post-ASCT PET result.ROC analysis revealed that the predictive value of post-ASCT PET result was superior to that of pre-ASCT PET result alone.Combined pre-and post-ASCT PET result is better for predicting outcomes in patients with NHL receiving transplantation.Deauville criteria score >3 was identified as the best cutoff value for post-ASCT PET.Conclusions: Post-ASCT PET result was more important than pre-ASCT PET result in predicting outcomes for NHL patients who underwent ASCT.The prognostic significance can be improved when combining preASCT PET result with post-ASCT PET result.Deauville criteria can be used for interpreting PET scans in this scenario.Zhitao Ying Lan Mi Xuejuan Wang Yuewei Zhang Zhi Yang Yuqin Song Xiaopei Wang WenZheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Chen Zhang Weiping Liu LijuanDeng Jun Zhu 2017Chinese Journal of Cancer Research2017,29,6:3
4The traditional Chinese medicines treat chronic heart failure and their main bioactive constituents and mechanisms显示文摘Chronic heart failure(CHF)is a severe public health problem with increasing morbidity and mortality,any treatment targeting a single session is insufficient to tackle this.CHF is characterized by reduced cardiac output resulting from neurohumoral dysregulation and cardiac remodeling,which might be related to oxidative stress,inflammation,endoplasmic reticulum stress,apoptosis,autophagy,mitochondrial function,and angiogenesis.These molecular mechanisms interact with each other through crosstalk.Historically,Chinese medicinal herbs have been widely applied in the treatment of CHF,and therapeutic effects of Chinese medicinal herbs and their ingredients have been scientifically confirmed over the past decades.Traditional Chinese medicine(TCM)with multiple components can confront the different pathogenesis of CHF through multiple targets.This review analyzes commonly used TCM patent drugs and TCM decoctions that are applicable to different stages of CHF based on clinical trials.Diverse bioactive ingredients in Chinese medicinal herbs have been found to treat CHF via multiple molecular mechanisms.This review comprehensively covers the key works on the effects and underlying mechanisms of TCM,herbal ingredients and synergistic effects of constituent compatibility in treating CHF,providing additional ideas to address this threat.Jie Chen Xiaohong Wei Qian Zhang Yuzhuo Wu Guiyang Xia Huan Xia Lingyan Wang Hongcai Shang Sheng Lin 2023Acta Pharmaceutica Sinica B2023,13,5:2
5PEG-asparaginase in BFM-90 regimen improves outcomes in adults with newly diagnosed lymphoblastic lymphoma显示文摘Objective: Although L-asparaginase(L-ASP) is a standard treatment for lymphoblastic lymphoma(LBL),hypersensitivity reactions by some patients limit its application. Polyethylene glycol-conjugated asparaginase(PEGASP) has a lower immunogenicity and is a standard treatment in all pediatric acute lymphoblastic leukemia(ALL).In this study, we investigated the efficacy and toxicity of PEG-ASP instead of L-ASP as used in the BFM-90regimen(PEG-ASP-BFM-90) for adult LBL.Methods: Between June 2012 and July 2015, we treated 30 adult patients with newly diagnosed LBL, using PEGASP-BFM-90 in a prospective, multicenter and single-arm clinical study at 5 participating institutions in China.Results: All the 30 patients, including 19 males and 11 females with a median age of 30(range: 18–62) years,completed 128 times of the PEG-ASP, with the median of 4(range: 2–6) times. Patients did not receive radiotherapy at this time. The overall response rate was 86.7%(26/30), with 50.0%(15/30) complete response and36.7%(11/30) partial response. The 3-year overall survival was 46.0% [95% confidence interval(95% CI),28.2%–64.8%], and the 3-year progression-free survival was 43.0%(95% CI, 25.7%–62.0%). Major adverse events were myelosuppression, reduced fibrinogen, liver dysfunction and digestive tract toxicities. No allergic reaction and no treatment-related mortality or severe complications were recorded.Conclusions: Our clinical data and observed outcomes indicate that 1 dose of PEG-ASP can replace multiple doses of native L-ASP in BFM-90, with predominantly grade 3–4 neutropenia for adult LBL, and no therapyrelated deaths. The effect is similar to previous reports of PEG-ASP-containing regimens for adult ALL. 6A durable 4-1BB-based CD19 CAR-T cell for treatment of relapsed or refractory non-Hodgkin lymphoma显示文摘Objective:Previous studies reported that 4-1BB-based CD19 chimeric antigen receptor(CAR)-T cells were more beneficial for the clinical outcomes than CD28-based CAR-T cells,especially the lower incidence rate of severe adverse events.However,the median progression-free survival(mPFS)of 4-1BB-based product Kymriah was shorter than that of CD28-based Yescarta(2.9 months vs.5.9 months),suggesting that Kymriah was limited in the long-term efficacy.Thus,a safe and durable 4-1BB-based CD19 CAR-T needs to be developed.Methods:We designed a CD19-targeted CAR-T(named as IM19)which consisted of an FMC63 scFv,4-1BB and CD3ζintracellular domain and was manufactured into a memory T-enriched formulation.A phase I/II clinical trial was launched to evaluate the clinical outcomes of IM19 in relapsed or refractory(r/r)B cell non-Hodgkin lymphoma(B-NHL).Dose-escalation investigation(at a dose of 5×10^(5)/kg,1×10^(6)/kg and 3×106/kg)was performed in 22 r/r B-NHL patients.All patients received a single infusion of IM19 after 3-day conditional regimen.Results:At month 3,the overall response rate(ORR)was 59.1%,the complete response rate(CRR)was 50.0%.The mPFS was 6 months and the 1-year overall survival rate was 77.8%.Cytokine release syndrome(CRS)occurred in 13 patients(59.1%),with 54.5%of grade 1−2 CRS.Only one patient(4.5%)experienced grade 3 CRS and grade 3 neurotoxicity.Conclusions:These results demonstrated the safety and durable efficacy of a 4-1BB-based CD19 CAR-T,IM19,which is promising for further development and clinical investigation.Zhitao Ying Ting He Shanzhao Jin Xiaopei Wang Wen Zheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Weiping Liu Lijuan Deng Yanping Ding Xuelian Hu Bing Bu Xin’an Lu Yuqin Song Jun Zhu 2022Chinese Journal of Cancer Research2022,34,1:2

8Prognostic value of ^(18)F-fluorodeoxyglucose positron emission tomography using Deauville criteria in diffuse large B cell lymphoma treated with autologous hematopoietic stem cell transplantation显示文摘Objective: High-dose chemotherapy(HDC) followed by autologous hematopoietic stem cell transplantation(auto-HSCT) plays an important role in improving outcomes of diffuse large B cell lymphoma(DLBCL) patients.18 F-fluorodeoxyglucose(18 F-FDG) positron emission tomography(PET)/computed tomography(CT) has been widely accepted in response assessment and prediction of prognosis in DLBCL. Here, we report the value of 18 FFDG PET/CT pre-and post-HSCT in predicting outcomes of patients with DLBCL.Methods: DLBCL patients who had PET/CT scan before and after HSCT were included. PET results were interpreted based upon Deauville criteria. The prognostic value of 18 F-FDG PET/CT in auto-HSCT was evaluated.Results: Eighty-four patients were enrolled. In univariate analysis, pre-and post-HSCT PET findings were correlated with 3-year progression-free survival(PFS) [hazard ratio(HR)=4.391, P=0.001; HR=7.607, P<0.001] and overall survival(OS)(HR=4.792, P=0.008; HR=26.138, P<0.001). Patients receiving upfront auto-HSCT after firstline treatment had better outcomes than relapsed/refractory DLBCL patients(3-year PFS, P<0.001; 3-year OS,P<0.001). In the relapsed/refractory patients, pre-and post-HSCT PET findings were also associated with 3-year PFS(P=0.003 vs. P<0.001) and OS(P=0.027 vs. P<0.001). A significant correlation was observed between clinical response to chemotherapy before auto-HSCT and outcomes of patients in the entire cohort(3-year PFS, P<0.001;3-year OS, P<0.001) and in the subgroup of 21 patients with positive pre-HSCT PET(3-year PFS, P=0.084; 3-year OS, P=0.240). A significant association between survival and post-HSCT PET findings was observed in multivariate analysis(HR=5.168, P<0.001).Conclusions: PET results before and after HSCT are useful prognostic factors for DLBCL patients receiving HSCT. 9Effect of Pingan Fang, a Traditional Chinese Medicine compound, on behavioral sensitization and conditioned place preference induced by ethanol in mice显示文摘OBJECTIVE:To observe the effect of Pingan Fang(PG)on behavioral sensitization and conditioned place preference(CPP)induced by ethanol in mice,and to determine the intervention mechanism of PG on alcohol addiction.METHODS:A behavioral sensitization mouse model induced by ethanol was established to observe the effect of PG on the development and expression of behavioral sensitization induced by ethanol by recording the spontaneous activity of mice.The resident time of mice in a white box was measured to evaluate the effect of PG on developing CPP induced by ethanol.Concentrations of dopamine(DA),Glutamate(Glu),and?-aminobutyric acid(GABA)in the corresponding mesolimbic region of mice were determined by enzyme-linked immunosorbent assay.RESULTS:Although PG did not alter spontaneous activity in mice,it reduced the growth of spontaneous activity stimulated by ethanol.The residence time in the white box after-ethanol-training of mice in CPP experiments was decreased.CONCLUSION:Our data suggested that PG blocked the development and expression of behavioral sensitization induced by ethanol and the development of CPP in mice.The mechanism might be related to the decreased content of DA and Glu and increased content of GABA in the mesolimbic dopamine system.This suggests that PG might be useful for the prevention and treatment of alcohol addiction.Jia Lingyan Li Yibei Hu Xiaoyu Lin Wu Wen Dachao Zhong Sen 2016Journal of Traditional Chinese Medicine2016,36,2:1

14(+)/(-)-Yanhusuosines A and B, two dimeric benzylisoquinoline-protoberberine alkaloid atropo-enantiomers featuring polycyclic skeletons from Corydalis yanhusuo显示文摘(+)/(-)-Yanhusuosines A(1) and B(2), two pairs of trace benzylisoquinoline-protoberberine atropoenantiomeric homodimers featuring an unprecedented 6/7/6/6/6/6 hexacyclic skeleton, were isolated from the tubers of Corydalis yanhusuo. The structures of(+)/(-)-1 and(+)/(-)-2 were elucidated using spectroscopic and quantum-chemical calculation approaches.(+)/(-)-Yanhusuosines A(1) and B(2)represent a new class of alkaloid dimers biogenetically constructed by a molecule of benzylisoquinoline with a unit of protoberberine via an intermolecular [4 + 3] cycloaddition. 17Study on Extraction Process of Sipunculus nudus Polysaccharide and Its Antioxidant Activity显示文摘[Objectives] This study was conducted to optimize the extraction process of Sipunculus nudus polysaccharide. [Methods]S. nudus polysaccharide was extracted by ultrasonic water extraction,with the marine S. nudus as a raw material from Sanya City,Hainan Province,by single factor variable method,to find the optimal extraction conditions including ultrasonic power,solid-liquid ratio,ultrasonic extraction temperature,ultrasonic extraction time,ultrasonic extraction times and lye concentration. The absorbance of the polysaccharide was determined by an ultraviolet spectrophotometer,and polysaccharide yield were calculated. The antioxidant activity of scavenging hydroxyl and DPPH radicals was measured at different concentrations. [Results]The optimal extraction conditions for S. nudus polysaccharide were as follows: solid-liquid ratio at 1∶11 mg/ml,extraction temperature at 60 ℃,three times of ultrasonic extraction,ultrasonic power of 960 W,extraction time of 1. 5 h,and lye concentration of 5%,under which the polysaccharide yield was 3. 21%. The scavenging rates of hydroxyl radical and DPPH radical were 12. 58% at the concentration of 0. 20 mg/ml. Moreover,the scavenging rates increased significantly with the increase of concentration. 18(+)/(-)-Yanhusamides A-C,three pairs of unprecedented benzylisoquinoline-pyrrole hetero-dimeric alkaloid enantiomers from Corydalis yanhusuo显示文摘A chemical investigation on the aqueous extract of Corydalis yanhusuo tubers led to the isolation and structural elucidation of three pairs of trace enantiomeric hetero-dimeric alkaloids,(+)/(-)-yanhusamides A-C(1-3),featuring an unprecedented 3,8-diazatricylco[5.2.2.0^(2,6)]undecane-8,10-diene bridged system.Their structures were exhaustively characterized by X-ray diffraction,comprehensive spectroscopic data analysis,and computational methods.Guided by the hypothetical biosynthetic pathway for 1-3,a gram-scale biomimetic synthesis of(±)-1 was achieved in 3 steps using photoenolization/Diels-Alder(PEDA)[4+2]cycloaddition.Compounds 1-3 exhibited potent inhibition of NO production induced by LPS in RAW264.7 macrophages.The in vivo assay showed that oral administration of 30 mg/kg of(±)-1 attenuated the severity of rat adjuvant-induced arthritis(AIA).Additionally,(±)-1 induced a dose-dependent antinociceptive effect in the acetic acid-induced mice writhing assay.Lingyan Wang Guiyang Xia Huan Xia Xiaohong Wei Yanan Wang Sheng Lin 2023Acta Pharmaceutica Sinica B2023,13,2:0
19TiO_(2)/Cu_(2)O heterostructure enabling selective and uniform lithium deposition towards stable lithium metal anodes显示文摘Lithium(Li)metal is the ultimate anode choice for next generation high energy density batteries.However,the high nucleation energy barrier and nonuniform electric field distribution,as well as huge volume expansion,lead to the uncontrollable growth of Li dendrites and poor utilization of Li metal,which hinders its practical application.Herein,titanium dioxide/cuprous oxide(TiO_(2)/Cu_(2)O)heterostructure is constructed on the rimous skeleton of Cu mesh,and the heterostructure decorated rimous Cu mesh(H-CM)can act as both current collector and host for dendrite-free Li metal anode.The TiO_(2)/Cu_(2)O heterostructure realizes selective Li nucleation by nano TiO_(2)and then induces fast and uniform Li conduction with the aid of heterostructure interface and nano Cu_(2)O contributing to dendrite-free Li deposition.While the internal and external space of rimous skeletons in H-CM is used to accommodate the deposited Li and buffer its volume change.Therefore,the cycling reversibility of the derived Li metal anode in H-CM is improved to a high Coulombic efficiency of 98.8%for more than 350 cycles at a current density of 1 mA·cm−2,and 1,000 h(equals to 500 cycles)stable repeated Li plating/stripping can be operated in a symmetric cell.Furthermore,full cells with limited Li anode and high loading LiFePO4 cathode present excellent cycling and rate performances.Lingyan Ruan Xianying Qin Kui Lin Zijin Yang Qiuchan Cai Tong Li Fangting Wu Feiyu Kang Baohua Li 2023Nano Research2023,16,4:0
20Magnetorheological elastomer and smartphone enable microfluidic biosensing of foodborne pathogen显示文摘Rapid detection of foodborne pathogens is crucial to prevent the outbreaks of foodborne diseases.In this work,we proposed a novel microfluidic biosensor based on magnetorheological elastomer(MRE)and smartphone.First,micropump and microvalves were constructed by deforming the MRE under magnetic actuation and integrated into the microfluidic biosensor for fluidic control.Then,the micropump was used to deliver immune porous gold@platinum nanocatalysts(Au@PtNCs),bacterial sample,and immunomagnetic nanoparticles(MNPs)into a micromixer,where they were mixed,incubated and magnetically separated to obtain the Au@PtNC-bacteria-MNP complexes.After 3,3',5,5'-tetramethylbenzidine and hydrogen peroxide were injected and catalyzed by the Au@PtNCs,smartphone was used to measure the color of the catalysate for quantitative analysis of target bacteria.Under optimal conditions,this biosensor could detect Salmonella typhimurium quantitatively and automatically in 1 h with a linear detection range of 8.0×10^(1) CFU/mL to 8.0×10^(4) CFU/mL and a detection limit of 62 CFU/mL.The microfluidic biosensor was compact in size,simple to use,and efficient for detection,and might be used for in-field screening of foodborne pathogens to prevent food poisoning.Gaozhe Cai Yuhe Wang Yingchao Zhang Lingyan Zheng Jianhan Lin 2023Chinese Chemical Letters2023,34,8:0
