Predictive factors associated with gefitinib response in patients with advanced non-small-cell lung cancer(NSCLC)显示文摘Purpose: A number of different clinical characteristics have been reported to singly correlate with therapeutic activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in advanced non-small-cell lung cancer(NSCLC). This study aimed to identify predictive factors associated with prognostic benefits of gefitinib.Patients and methods: EGFR gene typing in 33 advanced NSCLC patients received gefitinib(250 mg/day) were analyzed with mutant-enriched PCR assay. Gefitinib response was evaluated with potential predictive factors retrospectively. Results: The overall objective response rate(ORR) and median progression-free survival(PFS) in the 33 patients treated by gefitinib were 45.5% and 3.0(2.0-4.0) months. The ORR and median PFS in EGFR gene mutation patients were significantly higher/longer than those in EGFR gene wild-type patients(P<0.01). Similarly, the ORR and median PFS in non-smoker patients were significantly higher/longer than those in smoker patients(P<0.05, P<0.01, respectively). However, no difference for ORR and median PFS occurred between male and female patients. Logistic multivariate analysis showed that only EGFR mutated gene was significantly associated with the ORR(P<0.01). Both EGFR mutated gene and non-smoker were the major factors that contributed to PFS(P<0.05). Conclusions: EGFR mutated gene and non-smoker status are potential predictors for gefitinib response in NSCLC patients. | Lian Chen Rui Chen Zhe Zhu Yichen Zhang Zhengwei Wen Yun Li Xiaoming Li Yuwen Luo Liyu Ma Shuguang Lin Xin Chen | 2014 | Chinese Journal of Cancer Research2014,26,4: | 7
Facile one-step synthesis of graphitic carbon nitride-modified biochar for the removal of reactive red 120 through adsorption and photocatalytic degradation显示文摘Converting waste biomass into value-added biochar has been considered as a green and sustainable strategy for resource management and pollution control. In this study, graphitic carbon nitride(g-C_(3)N_(4))modified biochars(BCs)were produced through one-pot pyrolysis of urea and hickory chips in differential ratios at 520℃.The resulting BC/g-C_(3)N_(4)composites were evaluated in laboratory for their physicochemical, adsorptive, and photocatalytic properties. The characterization tests showed the successful synthesis of the BC/g-C_(3)N_(4)composites that introduced g-C_(3)N_(4)structure,N-containing surface func-tional groups, reduced surface area, and better thermal stability to the biochar. After modification, the BC/g-C_(3)N_(4)composites showed better adsorption ability to reactive red 120(RR120)than the pristine BC,due to the strong electrostatic attrition between N-containing functional groups of g-C_(3)N_(4)on biochar surface and anionic RR120.The BC/g-C_(3)N_(4)composites also inherited g-C_(3)N_(4)’s photocatalytic activity, which is visible light responsive to generate free radicals for RR120 degradation. In addition, the composites with higher urea modification ratios were more effective in the degradation of RR120.Overall,this study demonstrates the feasibility and promising potential of combining biochar and photocatalyst for the removal of aqueous dye.Because of the synergistic adsorption and photodegradation ability,BC/g-C_(3)N_(4)composites present a novel and cost-effective solution for the removal of aqueous dye and other photodegradable contaminants under natural conditions. | Yulin Zheng Yichen Yang Yue Zhang Weixin Zou Yidan Luo Lin Dong Bin Gao | 2019 | Biochar2019,1,1: | 5
SLC1A1-mediated cellular and mitochondrial influx of R-2-hydroxyglutarate in vascular endothelial cells promotes tumor angiogenesis in IDH1-mutant solid tumors显示文摘Muta nt isocitrate dehydrog en ase 1(mlDH1)drives tumorigenesis via produci ng on cometabolite R-2-hydroxyglutarate(R-2-HG)across various tumor types. However,mlDHl in hibitors appear only effective in hematological tumors. The therapeutic ben efit in solid tumors remains elusive, likely due to the complex tumor microenvironment. In this study,we discover that R-2-HG produced by IDH1-mutant tumor cells is preferentially imported into vascular endothelial cells and remodels mitochondrial respiration to promote tumor angiogenesis, conferring a therapeutic vulnerability in IDH1-mutant solid tumors.Mechanistically,SLC1 Alz a Na'-depe ndent glutamate tran sporter that is prefere ntially expressed in en dothelial cells, facilitates the in flux of R-2-HG from the tumor microenvironment into the endothelial cells as well as the intracellular trafficking of R-2-HG from cytoplasm to mitochondria.R-2-HG hijacks SLC1A1 to promote mitochondrial Na^(+)/Ca^(2+)exchange,which activates the mitochondrial respiratory chain and fuels vascular en dothelial cell migratio n in tumor an giogenesis.SLC1A1 deficiency in mice abolishes mlDHl-promoted tumor an giogenesis as well as the therapeutic benefit of mlDHl in hibitor in solid tumors. Moreover,we report that HH2301,a newly discovered mlDHl inhibitor, shows promising efficacy in treating IDH1-mutant cholangiocarcinoma in preclinical models.Together,we identify a new role of SLC1A1 as a gatekeeper of R-2-HG-mediated crosstalk between IDH1-mutant tumor cells and vascular en dothelial cells, and dem on strate the therapeutic potential of mlDHl in hibitors in treating IDH1-muta nt solid tumors via disrupting R-2-HG-promoted tumor angiogenesis. | Xiaomin Wang Ziqi Chen Jun Xu Shuai Tang Nan An Lei Jiang Yixiang Zhang Shaoying Zhang Qingli Zhang Yanyan Shen Shijie Chen Xiaojing Lan Ting Wang Linhui Zhai Siyuwei Cao Siqi Guo Yingluo Liu Aiwei Bi Yuehong Chen Xiameng Gai Yichen Duan Ying Zheng Yixian Fu Yize Li Liang Yuan Linjiang Tong Kun Mo Mingcheng Wang Shu-Hai Lin Minjia Tan Cheng Luo Yi Chen Jia Liu Qiansen Zhang Leping Li Min Huang | 2022 | Cell Research2022,32,7: | 2

Spatio-temporal Variation of PM_(2.5) in China from 1998 to 2016显示文摘With the rapid development of China's economy, people's demand for a healthy living environment is increasing, and air quality has gradually been widely concerned by all sectors of society.Using the big data of air quality monitoring from 1998 to 2016,based on the exploratory spatio-temporal analysis method, this paper explored the spatio-temporal evolution of PM_(2.5) at the national scale, and drew the following conclusions:①PM_(2.5) heavy pollution is mainly in central and eastern China, north and south China, and the pollution degree is relatively light in northwest and northeast China.Meanwhile,PM_(2.5) concentration in heavily polluted areas increased significantly over time, while PM_(2.5) concentration in low-polluted areas showed a long-term stable trend.②The number and area of cities with moderate and high PM_(2.5) pollution levels showed an inverted U-shaped curve from 1998 to 2016,and 2007 was the inflection point.③The spatial autocorrelation coefficient of PM_(2.5) is high over the years, and the spatial neighbor effect of PM_(2.5) is significant. The high-pollution clusters are mainly concentrated in the Beijing-Tianjin-Hebei region, the Yangtze River Delta and the Pearl River Delta, and the pollution concentration in these three regions has increased rapidly in recent years. It is necessary to focus on joint prevention and control. | Jianghui WANG Peng ZHOU Yichen LIN Longhao ZHANG Mengyu WU | 2022 | Meteorological and Environmental Research2022,13,1: | 1
Maternal genetic history of ancient Tibetans over the past 4000 years显示文摘The settlement of the Tibetan Plateau epitomizes human adaptation to a high-altitude environment that poses great challenges to human activity. Here,we reconstruct a 4000-year maternal genetic history of Tibetans using 128 ancient mitochondrial genome data from 37 sites in Tibet. The phylogeny of haplotypes M9a1a,M9a1b,D4g2,G2a’c,and D4i show that ancient Tibetans share the most recent common ancestor with ancient Middle and Upper Yellow River populations around the Early and Middle Holocene. In addition, the connections between Tibetans and Northeastern Asians vary over the past 4000 years, with a stronger matrilineal connection between the two during 4000 BPe3000 BP,and a weakened connection after 3000 BP,that are coincident with climate change, followed by a reinforced connection after the Tubo period(1400 BPe1100 BP).Besides,an over 4000-year matrilineal continuity is observed in some of the maternal lineages. We also find the maternal genetic structure of ancient Tibetans is correlated to the geography and interactions between ancient Tibetans and ancient Nepal and Pakistan populations.Overall, the maternal genetic history of Tibetans can be characterized as a long-term matrilineal continuity with frequent internal and external population interactions that are dynamically shaped by geography, climate changes,as well as historical events. | Ganyu Zhang Can Cui Shargan Wangdue Hongliang Lu Honghai Chen Lin Xi Wei He Haibing Yuan Tinley Tsring Zujun Chen Feng Yang Tashi Tsering Shuai Li Norbu Tashi Tsho Yang Yan Tong Xiaohong Wu Linhui Li Yuanhong He Peng Cao Qingyan Dai Feng Liu Xiaotian Feng Tianyi Wang Ruowei Yang Wanjing Ping Ming Zhang Xing Gao Yichen Liu Wenjun Wang Qiaomei Fu | 2023 | Journal of Genetics and Genomics2023,50,10: | 1

An integrative protocol for one‑step PCR amplicon library construction and accurate demultiplexing of pooled sequencing data显示文摘High-throughput sequencing of amplicons has been widely used to precisely and efficiently identify species compositions and analyze community structures, greatly promoting biological studies involving large amounts of complex samples, especially those involving environmental and pathogen-monitoring ones.Commercial library preparation kits for amplicon sequencing, which generally require multiple steps, including adapter ligation and indexing, are expensive and time-consuming,especially for applications at a large scale. To overcome these limitations,a“one-step PCR approach”has been previously proposed for constructions of amplicon libraries using long fusion primers. However, efficient amplifications of target genes and accurate demultiplexing of pooled sequencing data remain to be addressed. To tackle these,we present an integrative protocol for one-step PCR amplicon library construction(OSPALC).High-quality reads have been generated by this approach to reliably identify species compositions of mock bacterial communities and environmental samples. With this protocol, the amplicon library is constructed through one regular PCR with long primers, and the total cost per DNA/cDNA sample decreases to just 7%of the typical cost via the multi-step PCR approach.Empirically tested primers and optimized PCR conditions to construct OSPALC libraries for 16S rDNA V4 regions are demonstrated as a case study.Tools to design primers targeting at any genomic regions are also presented. In principle,OSPALC can be readily applied to construct amplicon libraries of any target genes using DNA or RNA samples, and will facilitate research in numerous fields. | Jiahao Ni Jiao Pan Yaohai Wang Tianhao Chen Xinshi Feng Yichen Li Tongtong Lin Michael Lynch Hongan Long Weiyi Li | 2023 | Marine Life Science & Technology2023,5,4: | 1
Carbon deficiency introduced plasticity of rock-salt-structured transition metal carbides显示文摘High-hardness rock-salt structured transitional metal carbides(TMC)are attracting substantial interest as potential next-generation thermal protection materials. However, the intrinsic brittleness of TMC ceramics impedes their performance in aerodynamically harsh environments. In this work,a promising strategy is proposed to introduce plasticity in TaC–HfC solid solutions by manipulating carbon deficiency. The approach combines density-functional theory(DFT)with experiments and takes Pugh's ratio(k)as the criteria.Depletion of carbon atoms in TaC–HfC solid solutions results in the de-localizing of valence electrons, deviation of spatial modulus along different crystal plane directions, and leading to significant elastic anisotropy. The carbon deficient Ta_(0.8)Hf_(0.2)C_(0.8) is predicted to be a‘softer phase’with reduced modulus and Pugh's ratio(k=0.58).A series of Ta1–xHfxCy(x=0.2 and 0.8,y=0.8,0.9,and 1.0)bulk ceramics are experimentally fabricated by an excessive metal alloying method.Trigonal and hexagonal close-packed structured carbides are derived when the carbon deficiency y decreased to 0.7.The indentation modulus drops from 641.8±14.8 GPa for Ta_(0.8)Hf_(0.2)C_(1.0) to 555.8±9.9 GPa for Ta0.8Hf0.2C0.8.The specific stoichiometric composition of Ta_(0.8)Hf_(0.2)C_(0.8) is experimentally verified to possess both plasticity(k=0.41)and ultra-high nanohardness(41.3±1.3 GPa). | Buhao Zhang Youwei Wang Jie Yin Yichen Wang Hangfeng Zhang Tamás Csanádi Ján Dusza Michael J.Reece Nan Lin Xiao Yang Xuejian Liu Zhengren Huang Dongliang Jiang | 2023 | Journal of Materials Science & Technology2023,,33: | 0
对比分析黑斑息肉综合征合并大肠癌组织肿瘤相关基因(英文)显示文摘Objective:The aim of the study was to screen the differentially expressed genes of Peutz-Jeghers syndrome (PJS) and colorectal carcinoma (CRC).Methods:This study used cDNA microarray to comparatively analyze the gene expression profiles of 4 cases of PJS combined with colorectal adenocarcinoma vs.normal mucosae. The cDNA microarray contained 8064 human genes, and then using RT-PCR to test three genes of all. Results:The experimental data showed that fourteen genes were differentially expressed, which were up-regulated in PJS.Fifty-one genes expressions were altered in CRCs,of which 32 were up-regulated,as compared to the normal mucosae. In addition,5 genes were similarly altered in both PJS and CRCs.RT-PCR analyses confirmed the cDNA microarray data for three of those genes:LATS2,APC and MADH4.Conclusion:LCN2,USP4,GRO3,HYAL1 and APC-these differentially expressed genes likely represent biomarkers for early detection of CRC and may be potential therapeutic targets. | Xiaosan Zhu Yichen Dai Qingzhen Nan Zhangxin Chen Junpei Xie Wei Zeng Yuka Fu Yuanyuan Lin Qingna Lian Qiaofang Sang Xiaojuan Zhan | 2011 | The Chinese-German Journal of Clinical Oncology2011,10,8: | 0
TNFR2 is a potent prognostic biomarker for post-transplant lung metastasis in patients with hepatocellular carcinoma显示文摘Objective:Lung metastasis is a common and fatal complication of liver transplantation for hepatocellular carcinoma(HCC).The precise prediction of post-transplant lung metastasis in the early phase is of great value. Methods:The mRNA profiles of primary and paired lung metastatic lesions were analyzed to determine key signaling pathways. We enrolled 241 HCC patients who underwent liver transplantation from three centers.Tissue microarrays were used to evaluate the prognostic capacity of tumor necrosis factor(TNF),tumor necrosis factor receptor 1(TNFR1),and TNFR2,particularly for post-transplant lung metastasis. Results:Comparison of primary and lung metastatic lesions revealed that the TNF-dependent signaling pathway was related to lung metastasis of HCC.The expression of TNF was degraded in comparison to that in para-tumor tissues(P<0.001).The expression of key receptors in the TNF-dependent signaling pathway,TNFR1 and TNFR2,was higher in HCC tissues than in para-tumor tissues(P<0.001).TNF and TNFR1 showed no relationship with patients’outcomes, whereas elevated TNFR2 in tumor tissue was significantly associated with worse overall survival(OS)and increased recurrence risk(5-year OS rate:31.9%vs.62.5%,P<0.001).Notably,elevated TNFR2 levels were also associated with an increased risk of post-transplant lung metastasis(hazard ratio:1.146;P<0.001).Cox regression analysis revealed that TNFR2,Hangzhou criteria, age, and hepatitis B surface antigen were independent risk factors for post-transplant lung metastasis, and a novel nomogram was established accordingly. The nomogram achieved excellent prognostic efficiency(area under time-dependent receiver operating characteristic=0.755,concordance-index=0.779)and was superior to conventional models, such as the Milan criteria.Conclusions:TNFR2 is a potent prognostic biomarker for predicting post-transplant lung metastasis in patients with HCC.A nomogram incorporating TNFR2 deserves to be a helpful prognostic tool in liver transplantation for HCC. | Huigang Li Zuyuan Lin Jianyong Zhuo Modan Yang Wei Shen Zhihang Hu Yichen Ding Hao Chen Chiyu He Xinyu Yang Siyi Dong Xuyong Wei Beicheng Sun Shusen Zheng Ren Lang Di Lu Xiao Xu | 2023 | Chinese Journal of Cancer Research2023,35,1: | 0
Virtual Power Plants for Grid Resilience: A Concise Overview of Research and Applications显示文摘The power grid is undergoing a transformation from synchronous generators(SGs) toward inverter-based resources(IBRs). The stochasticity, asynchronicity, and limited-inertia characteristics of IBRs bring about challenges to grid resilience. Virtual power plants(VPPs) are emerging technologies to improve the grid resilience and advance the transformation. By judiciously aggregating geographically distributed energy resources(DERs) as individual electrical entities, VPPs can provide capacity and ancillary services to grid operations and participate in electricity wholesale markets. This paper aims to provide a concise overview of the concept and development of VPPs and the latest progresses in VPP operation, with the focus on VPP scheduling and control. Based on this overview, we identify a few potential challenges in VPP operation and discuss the opportunities of integrating the multi-agent system(MAS)-based strategy into the VPP operation to enhance its scalability, performance and resilience. | Yijing Xie Yichen Zhang Wei-Jen Lee Zongli Lin Yacov A.Shamash | 2024 | IEEE/CAA Journal of Automatica Sinica2024,11,2: | 0
Ambipolar tribotronic transistor of MoTe_(2)显示文摘Two-dimensional(2D)tribotronic devices have been successfully involved in electromechanical modulation for channel conductance and applied in intelligent sensing system, touch screen, and logic gates.Ambipolar transistors and corresponding complementary inverters based on one type of semiconductors are highly promising due to the facile fabrication process and readily tunable polarity. Here,we demonstrate an ambipolar tribotronic transistor of molybdenum ditelluride(MoTe_(2)),which shows typical ambipolar transport properties modulated by triboelectric potential. It is comprised of a MoTe_(2)transistor and a lateral sliding triboelectric nanogenerator(TENG).The induced triboelectric potential by Maxwell’s displacement current(a driving force for TENG)can readily modulate the transport properties of both electrons and holes in MoTe_(2)channel and effectively drive the transistor.High performance tribotronic properties have been achieved, including low cutoff current below 1 pA·μm^(−1)and high current on/off ratio of~103 for holes and electrons dominated transports. The working mechanism on how to achieve tribotronic ambipolarity is discussed in detail.A complementary tribotronic inverter based on single flake of MoTe_(2)is also demonstrated with low power consumption and high stability. This work presents an active approach to efficiently modulate semiconductor devices and logic circuits based on 2D materials through external mechanical signal, which has great potential in human–machine interaction, intelligent sensor, and other wearable devices. | Yonghai Li Jinran Yu Yichen Wei Yifei Wang Liuqi Cheng Zhenyu Feng Ya Yang Zhong Lin Wang Qijun Sun | 2023 | Nano Research2023,16,9: | 0
Integration of biocompatible organic resistive memory and photoresistor for wearable image sensing application显示文摘The integration of multiple functional devices to achieve complex functions has become an essential requirement for future wearable biomedical electronic devices and systems. In this paper, we present a flexible multi-functional device composed of a biocompatible organic polymer resistive random-access memory(RRAM) and a photoresistor for wearable image sensing application. The resistive layer of organic polymer RRAM is composed by polychloro-para-xylylene(parylene-C), which is a flexible, transparent, biocompatibility and chemical stability polymer material. What is more, parylene-C is quite safe to be used within human body as it is a Food and Drug Administration(FDA)-approved material. This organic RRAM shows stable switching characteristics, low operation voltages(3.25 V for set voltage and-0.55 V for reset voltage), low static power consumption, high storage window and good retention properties(>10~4 s).A multi-functional device that can detect the light intensity of incident light and simultaneously store the information in the memory devices for wearable image sensing application was proposed and fabricated by integrating the organic resistive memory and a photoresistor. The threshold of incident light intensity can be easily adjust by changing the external voltage. | 19 | Altered gut microbiome in FUT2 loss-of-function mutants in support of personalized medicine for inflammatory bowel diseases显示文摘The FUT2 loss-of-function mutations are highly prevalent and are associated with inflammatory bowel disease(IBD).To investigate the impact of FUT2 loss-of-function mutation on the gut microbiota in patients with IBD,81 endoscopically confirmed IBD patients were genotyped and divided into 3 groups:homozygous for functional FUT2 genes(SeSe),with one copy of non-functional FUT2 gene(Sese),or homozygous for non-functional FUT2 genes(sese).Escherichia, which attaches to fucosylated glycoconjugates, was the only abundant genus exhibiting decreased abundance in sese patients.Compared with SeSe or Sese patients, sese patients exhibited higher abundance in CD8+inducing Alistipe and Phascolarctobacterium and Th17 inducing Erysipelotrichaceae UCG-003.Counter-intuitively,butyrate-producing bacteria were more abundant in sese patients.Consistently, metabolomics analysis found higher levels of butyrate in sese patients. Our data support the hypothesis that FUT2 loss-of-function mutation participates in the IBD pathogenesis by decreasing binding sites for adherent bacteria and thus altering the gut microbiota.Decreased abundances of adherent bacteria may allow the overgrowth of bacteria that induce inflammatory T cells, leading to intestinal inflammation. As FUT2 loss-of-function mutations are highly prevalent, the identification of T cell inducing bacteria in sese patients could be valuable for the development of personalized microbial intervention for IBD. | Sijing Cheng Jun Hu Xianrui Wu Ji-An Pan Na Jiao Yichen Li Yibo Huang Xutao Lin Yifeng Zou Yuan Chen Lixin Zhu Min Zhi Ping Lan | 2021 | Journal of Genetics and Genomics2021,48,9: | 0
