1Syntheses and characterization of anti-thrombotic and anti-oxidative Gastrodin-modified polyurethane for vascular tissue engineering显示文摘Vascular grafts must avoid negative inflammatory responses and thrombogenesis to prohibit fibrotic deposition immediately upon implantation and promote the regeneration of small diameter blood vessels(<6 mm inner diameter).Here,polyurethane(PU)elastomers incorporating anti-coagulative and anti-inflammatory Gastrodin were fabricated.The films had inter-connected pores with porosities equal to or greater than 86%and pore sizes ranging from 250 to 400μm.Incorporation of Gastrodin into PU films resulted in desirable mechanical properties,hydrophilicity,swelling ratios and degradation rates without collapse.The released Gastrodin maintained bioactivity over 21 days as assessed by its anti-oxidative capability.The Gastrodin/PU had better anti-coagulation response(less observable BSA,fibrinogen and platelet adhesion/activation and suppressed clotting in whole blood).Red blood cell compatibility,measured by hemolysis,was greatly improved with 2Gastrodin/PU compared to other Gastrodin/PU groups.Notably,Gastrodin/PU upregulated anti-oxidant factors Nrf2 and HO-1 expression in H2O2 treated HUVECs,correlated with decreasing pro-inflammatory cytokines TNF-αand IL-1β in RAW 264.7 cells.Upon implantation in a subcutaneous pocket,PU was encapsulated by an obvious fibrous capsule,concurrent with a large amount of inflammatory cell infiltration,while Gastrodin/PU induced a thinner fibrous capsule,especially 2Gastrodin/PU.Further,enhanced adhesion and proliferation of HUVECs seeded onto films in vitro demonstrated that 2Gastrodin/PU could help cell recruitment,as evidenced by rapid host cell infiltration and substantial blood vessel formation in vivo.These results indicate that 2Gastrodin/PU has the potential to facilitate blood vessel regeneration,thus providing new insight into the development of clinically effective vascular grafts.Meng Zheng Jiazhi Guo Qing Li Jian Yang Yi Han Hongcai Yang Mali Yu Lianmei Zhong Di Lu Limei Li Lin Sun 2021Bioactive Materials2021,6,2:7
2Preparation and in vitro-in vivo evaluation of intestinal retention pellets of Berberine chloride to enhance hypoglycemic and lipid-lowing efficacy显示文摘Berberine chloride(BBR) is a pharmacokinetic profile of drug with poor bioavailability but good therapeutic efficacy,which is closely related to the discovery of BBR intestinal target.The major aim of this paper is to develop BBR intestinal retention type sustained-release pellets and evaluate their in vivo and in vitro behaviors base on the aspect of local action on intestinal tract. Here,wet milling technology is used to improve dissolution and dissolution rate of BBR by decreasing the particle size and increasing the wettability. The pellets are prepared by liquid layer deposition technology,and then the core pellets are coated with Eudragit~?L30 D-55 and Eudragit~?NE30 D aqueous dispersion. The prepared pellets show high drug loading capacity,and the drug loading up to 93%. Meanwhile,it possesses significant sustained drug release effect in purified water which is expected to improve the pharmacokinetic behavior of BBR. The pharmacokinetics results demonstrate that the halflife of BBR was increased significantly from 24 h to 36 h and the inter-and intra-subject variability are decreased compared to commercial BBR tablets. The retention test results indicate that the pellet size and Eudragit~?NE30 D plays an important role in retention time of the pellet,and it is found that the pellets with small particle size and high Eudragit~?NE30 D coating content can stay longer in the intestine than the pellets with large particle size. 3MS1 is essential for male fertility by regulating the microsporocyte cell plate expansion in soybean显示文摘Male sterility is an essential trait in hybrid seed production,especially for monoclinous and autogamous food crops.Soybean male-sterile ms1 mutant has been known for more than 50 years and could be instrumental in making hybrid seeds.However,the gene responsible for the male-sterile phenotype has remained unknown.Here,we report the map-based cloning and characterization of the MS1 gene in soybean.MS1 encodes a kinesin protein and localizes to the nucleus,where it is required for the male meiotic cytokinesis after telophase Ⅱ.We further substantiated that MS1 colocalizes with microtubules and is essential for cell plate formation in soybean male gametogenesis through immunostaining.Both ms1 and CRISPR/Cas9 knockout mutants show complete male sterility but are otherwise phenotypically normal,making them perfect tools for producing hybrid seeds.The identification of MS1 has the practical potential for assembling the sterility system and speeding up hybrid soybean breeding.Xiaolong Fang Xiaoyuan Sun Xiangdong Yang Qing Li Chunjing Lin Jie Xu Wenjun Gong Yifan Wang Lu Liu Limei Zhao Baohui Liu Jun Qin Mengchen Zhang Chunbao Zhang Fanjiang Kong Meina Li 2021Science China(Life Sciences)2021,64,9:4

5The RALF1-FERONIA complex interacts with and activates TOR signaling in response to low nutrients显示文摘Target of rapamycin(TOR)kinase is an evolutionarily conserved major regulator of nutrient metabolism and organismal growth in eukaryotes.In plants,nutrients are remobilized and reallocated between shoots and roots under low-nutrient conditions,and nitrogen and nitrogen-related nutrients(e.g.,amino acids)are key upstream signals leading to TOR activation in shoots under low-nutrient conditions.However,how these forms of nitrogen can be sensed to activate TOR in plants is still poorly understood.Here we report that the Arabidopsis receptor kinase FERON IA(FER)interacts with the TOR pathway to regulate nutrient(nitrogen and amino acid)signaling under low-nutrient conditions and exerts similar metabolic effects in response to nitrogen deficiency.We found that FER and its partner,RPM1-induced protein kinase(RIPK),interact with the TOR/RAPTOR complex to positively modulate TOR signaling activity.During this process,the receptor complex FER/RIPK phosphorylates the TOR complex component RAPTOR1B.The RALF1 peptide,a ligand of the FER/RIPK receptor complex,increases TOR activation in the young leaf by enhancing FER-TOR interactions,leading to promotion of true leaf growth in Arabidopsis under lownutrient conditions.Furthermore,we showed that specific amino acids(e.g.,Gin,Asp,and Gly)promote true leaf growth under nitrogen-deficient conditions via the FER-TOR axis.Collectively,our study reveals a mechanism by which the RALF1-FER pathway activates TOR in the plant adaptive response to low nutrients and suggests that plants prioritize nutritional stress response over RALF1-mediated inhibition of cell growth under low-nutrient conditions.Limei Song Guoyun Xu Tingting Li Huina Zhou Qinlu Lin Jia Chen Long Wang Dousheng Wu Xiaoxu Li Lifeng Wang Sirui Zhu Feng Yu 2022Molecular Plant2022,15,7:2
6Characterization of a Novel Weak Allele of RGA1/D1 and Its Potential Application in Rice Breeding显示文摘Semi-dwarfing improves the lodging resistance and yield of rice,and the vast majority of modern rice varieties harbor the sd1 allele to decrease plant height,resulting in reduced genetic diversity and negative agronomic traits.Thus,exploring alternative sources of dwarfism is imperative for rice breeding.Here,we identified a novel RGA1 allele,d1-w,from a local indica variety Xiaolixiang(XLX)using a map-based cloning approach.Compared with other rice varieties,RGA1 in XLX contained a unique single nucleotide polymorphism that resulted in an additional transcript and reduced functional RGA1 transcript level.The RGA1 from Nipponbare was introduced into XLX to estimate the value of d1-w in rice breeding.Compared with transgenic XLX plants(XLX^(D1)),XLX exhibited reduced plant height,increased stem strength,lower reactive oxygen species accumulation,delayed senescence,stronger photosynthesis,higher grain yield and quality(including external,milling and nutritional qualities),and enhanced resistance to drought and Rhizoctonia solani.Therefore,we proposed that the d1-w allele has potential as an excellent dwarfism resource for rice breeding.LIU Yantong LI Ting JIANG Zhishu ZENG Chuihai HE Rong QIU Jiao LIN Xiaoli PENG Limei SONG Yongping ZHOU Dahu CAI Yicong ZHU Changlan FU Junru HE Haohua XU Jie 2022Rice science2022,29,6:2
7Neuro-regenerative imidazole-functionalized GelMA hydrogel loaded with hAMSC and SDF-1α promote stem cell differentiation and repair focal brain injury显示文摘Brain tissues that are severely damaged by traumatic brain injury(TBI)is hardly regenerated,which leads to a cavity or a repair with glial scarring.Stem-cell therapy is one viable option to treat TBI-caused brain tissue damage,whose use is,whereas,limited by the low survival rate and differentiation efficiency of stem cells.To approach this problem,we developed an injectable hydrogel using imidazole groups-modified gelatin methacrylate(GelMA-imid).In addition,polydopamine(PDA)nanoparticles were used as carrier for stromal-cell derived factor-1(SDF-1α).GelMA-imid hydrogel loaded with PDA@SDF-1αnanoparticles and human amniotic mesenchymal stromal cells(hAMSCs)were injected into the damaged area in an in-vivo cryogenic injury model in rats.The hydrogel had low module and its average pore size was 204.61±41.41 nm,which were suitable for the migration,proliferation and differentiation of stem cells.In-vitro cell scratch and differentiation assays showed that the imidazole groups and SDF-1αcould promote the migration of hAMSCs to injury site and their differentiation into nerve cells.The highest amount of nissl body was detected in the group of GelMA-imid/SDF-1α/hAMSCs hydrogel in the in-vivo model.Additionally,histological analysis showed that GelMA-imid/SDF-1α/hAMSCs hydrogel could facilitate the regeneration of regenerate endogenous nerve cells.In summary,the GelMA-imid/SDF-1α/hAMSCs hydrogel promoted homing and differentiation of hAMSCs into nerve cells,and showed great application potential for the physiological recovery of TBI.Yantao Zheng Gang Wu Limei Chen Ying Zhang Yuwei Luo Yong Zheng Fengjun Hu Tymor Forouzanfar Haiyan Lin Bin Liu 2021Bioactive Materials2021,6,3:2

12MALE STERILITY 3 encodes a plant homeodomain-finger protein for male fertility in soybean显示文摘Male-sterile plants are used in hybrid breeding to improve yield in soybean(Glycine max(L.)Merr.).Developing the capability to alter fertility under different environmental conditions could broaden germplasm resources and simplify hybrid production.However,molecular mechanisms potentially underlying such a system in soybean were unclear.Here,using positional cloning,we identified a gene,MALE STERILITY 3(MS3),which encodes a nuclear-localized protein containing a plant homeodomain(PHD)-finger domain.A spontaneous mutation in ms3 causing premature termination of MS3 translation and partial loss of the PHD-finger.Transgenetic analysis indicated that MS3 knockout resulted in nonfunctional pollen and no selfpollinated pods,and RNA-seq analysis revealed that MS3 affects the expression of genes associated with carbohydrate metabolism.Strikingly,the fertility of mutant ms3 can restore under longd conditions.The mutant could thus be used to create a new,more stable photoperiod-sensitive genic male sterility line for two-line hybrid seed production,with significant impact on hybrid breeding and production.Jingjing Hou Weiwei Fan Ruirui Ma Bing Li Zhihui Yuan Wenxuan Huang Yueying Wu Quan Hu Chunjing Lin Xingqi Zhao Bao Peng Limei Zhao Chunbao Zhang Lianjun Sun 2022Journal of Integrative Plant Biology2022,64,5:1

20Synthesis, crystal structure and photocatalytic performance of polyoxometalate K_(13)[Gd(GeW_(11)O_(39))_2]·34H_2O显示文摘A polyoxometalate K_(13)[Gd(GeW_(11)O_(39))_2]·34 H_2 O was synthesized and characterized by elemental analysis,IR spectroscopy, UV spectra, XRD and thermal gravimetric analysis. X-ray single-crystal structural analysis indicates that the title compound crystallizes in the triclinic crystal system. The heteropolyanion of [Gd(GeW_(11)O_(39))_2]^(13) consists of two [GeW_(11)O_(39)]^(8-) vacant Keggin moieties linked via Gd^(3+). Photocatalytic performance of K_(13)[Gd(GeW_(11)O_(39))_2]·34 H_2 O in photodegradation of X-3 B was also studied.When the reaction time is 180 min, the decoloring rate reaches 71.19%.
