1Squalene epoxidase promotes colorectal cancer cell proliferation through accumulating calcitriol and activating CYP24A1-mediatedMAPK signaling显示文摘Background:Colorectal cancer(CRC)is one of the most malignant tumorswith high incidence,yet its molecular mechanism is not fully understood,hindering the development of targeted therapy.Metabolic abnormalities are a hallmark of cancer.Targeting dysregulated metabolic features has become an important direction for modern anticancer therapy.In this study,we aimed to identify a new metabolic enzyme that promotes proliferation of CRC and to examine the related molecular mechanisms.Methods:We performed RNA sequencing and tissue microarray analyses of human CRC samples to identify new genes involved in CRC.Squalene epoxidase(SQLE)was identified to be highly upregulated in CRC patients.The regulatory function of SQLE in CRC progression and the therapeutic effect of SQLE inhibitors were determined by measuring CRC cell viability,colony and organoid formation,intracellular cholesterol concentration and xenograft tumor growth.Themolecularmechanism of SQLE functionwas explored by combining transcriptome and untargeted metabolomics analysis.Western blotting and realtime PCR were used to assess MAPK signaling activation by SQLE.Results:SQLE-related control of cholesterol biosynthesis was highly upregulated in CRC patients and associated with poor prognosis.SQLE promoted CRC growth in vitro and in vivo.Inhibition of SQLE reduced the levels of calcitriol(active form of vitamin D3)and CYP24A1,followed by an increase in intracellular Ca2+concentration.Subsequently,MAPK signaling was suppressed,resulting in the inhibition of CRC cell growth.Consistently,terbinafine,an SQLE inhibitor,suppressed CRC cell proliferation and organoid and xenograft tumor growth.Conclusions:Our findings demonstrate that SQLE promotes CRC through the accumulation of calcitriol and stimulation of CYP24A1-mediated MAPK signaling,highlighting SQLE as a potential therapeutic target for CRC treatment.Luwei He Huaguang Li Chenyu Pan Yutong Hua Jiayin Peng Zhaocai Zhou Yun Zhao Moubin Lin 2021Cancer Communications2021,41,8:4

4Rhesus monkeys exhibiting spontaneous ritualistic behaviors resembling obsessive-compulsive disorder显示文摘Obsessive-compulsive disorder(OCD)is a chronic and debilitating psychiatric disorder that affects∼2%–3%of the population globally.Studying spontaneous OCD-like behaviors in non-human primates may improve our understanding of the disorder.In large rhesus monkey colonies,we found 10 monkeys spontaneously exhibiting persistent sequential motor behaviors(SMBs)in individual-specific sequences that were repetitive,time-consuming and stable over prolonged periods.Genetic analysis revealed severely damaging mutations in genes associated with OCD risk in humans.Brain imaging showed that monkeys with SMBs had larger gray matter(GM)volumes in the left caudate nucleus and lower fractional anisotropy of the corpus callosum.The GM volume of the left caudate nucleus correlated positively with the daily duration of SMBs.Notably,exposure to a stressor(human presence)significantly increased SMBs.In addition,fluoxetine,a serotonergic medication commonly used for OCD,decreased SMBs in these monkeys.These findings provide a novel foundation for developing better understanding and treatment of OCD.Rongwei Zhai Geya Tong Zheqin Li Weichen Song Yang Hu Sha Xu Qiqi Wei Xiaocheng Zhang Yi Li Bingbing Liao Chenyu Yuan Yinqing Fan Ge Song Yinyin Ouyang Wenxuan Zhang Yaqiu Tang Minghui Jin Yuxian Zhang He Li Zhi Yang Guan Ning Lin Dan JStein Zhi-Qi Xiong Zhen Wang 2023National Science Review2023,10,11:1

6Functional analysis of the sbcD (dr1921) gene of the extremely radioresistant bacterium Deinococcus radiodurans显示文摘In eukaryotes, the Mre11-Rad50-Nbs1 (MRN) complex, which resides at the crossroads of DNA repair and checkpoint signaling, rapidly forms prominent foci at damage sites following double-strand break (DSB) induction. This complex carries out the initial processing of the DSB ends. Mutations in the genes that encode components of this complex result in DNA-damage hypersensitivity, genomic in- stability, telomere shortening, and aberrant meiosis. Therefore, the MR proteins are highly conserved during evolution. The bacterial orthologs of Mre11 and Rad50 are the SbcD and SbcC proteins, respec- tively. Deinococcus radiodurans, an extremely radioresistant bacterium, is able to mend hundreds of radiation-induced DSBs. The SbcD and SbcC proteins were identified as the products of the Dr1921 and Dr1922 genes. Disruption of the sbcD gene, by direct reverse-orientation insertional mutagenesis tech- nology, remarkably increases the cells’ sensitivity to various types of DNA damaging agents, such as ionizing radiation, ultraviolet irradiation, hydrogen peroxide, and mitomycin C. We also provide evidence that the drSbcD protein plays an important role in both growth and DNA repair in this organ- ism, especially in repair of DSBs generated after cellular exposure to 6000 Gy of IR. 8An almost full reversible lithium-rich cathode: Revealing the mechanism of high initial coulombic efficiency显示文摘Low initial Coulombic efficiency (ICE) is an important impediment to practical application of Li-rich layered oxides (LLOs), which is due to the irreversible oxygen release. It is generally considered that surface oxygen vacancies are conducive to the improvement of ICE of LLOs. To reveal the relation of oxygen vacancies and ICE, sample PLO (Li-Mn-Cr-O) and its treated product (TLO) are comprehensive investigated in this work. During the treated process, part of oxygen atoms return to original constructed vacancies. It makes oxygen vacancies in sample TLO much poorer than those in sample PLO, and induces the formation of Li-poor spinel-layered integrated structure. Electrochemical measurement indicates the ICE of sample PLO is only 80.8%, while sample TLO is almost full reversible with the ICE of ~97.1%. In term of high-energy X-ray diffraction, scanning transmission electron microscopy, X-ray photoelectron spectroscopy and synchrotron hard/soft X-ray absorption spectroscopy, we discover that the ICE is difficult to be improved significantly just by building oxygen vacancies. LLOs with high ICE not only have to construct suitable oxygen vacancies, but also require other components with Li-poor structure to stabilize oxygen. 9IMU-based motion capture system for rehabilitation applications: A systematic review显示文摘In recent years,the use of inertial measurement unit(IMU)-based motion capture(Mocap)systems in rehabilitation has grown significantly.This paper aimed to provide an overview of current IMUbased Mocap system designs in the field of rehabilitation,explore the specific applications and implementation of these systems,and discuss potential future developments considering sensor limitations.For this review,a systematic literature search was conducted using Scopus,IEEE Xplore,PubMed,and Web of Science from 2013 to 2022.A total of 65 studies were included and analyzed based on their rehabilitation application,target population,and system deployment and measurement.The proportion of rehabilitation assessment,training,and both were 82%,12%,and 6%respectively.The results showed that primary focus of the studies was stroke that was one of the most commonly studied pathological disease.Additionally,general rehabilitation without targeting a specific pathology was also examined widely,with a particular emphasis on gait analysis.The most common sensor configuration for gait analysis was two IMUs measuring spatiotemporal parameters of the lower limb.However,the lack of training applications and upper limb studies could be attributed to the limited battery life and sensor drift.To address this issue,the use of low-power chips and low-consumption transmission pathways was a potential way to extend usage time for long-term training.Furthermore,we suggest the development of a highly integrated multi-modal system with sensor fusion.Chenyu Gu Weicong Lin Xinyi He Lei Zhang Mingming Zhang 2023Biomimetic Intelligence & Robotics2023,3,2:0
10Model of NBTI combined with mobility degradation显示文摘The mobility degradation induced by negative bias temperature instability(NBTI) is usually ignored in traditional NBTI modeling and simulation, resulting in overestimation of the circuit lifetime, especially after longterm operation. In this paper, the mobility degradation is modeled in combination with the universal NBTI model.The coulomb scattering induced by interface states is revealed to be the dominant component responsible for mobility degradation. 11Hot-exciton effects on exciton diffusion and circular polarization dynamics in a single PbI_(2)nanoflake显示文摘As one of the emerging two-dimensional lead halide materials,lead iodide(PbI_(2))nanosheets have proven to possess strong application potential in the fields of high-energy radiation detection and highly efficient perovskite solar cells.However,the underlying photophysical properties such as hot-exciton-related carrier dynamics remain unclear for PbI_(2)nanosheets.Here,we report the exciton dynamics of a single PbI_(2)nanoflake prepared by an aqueous solution method.Through a three-dimensional(3D)diffusion model,we obtain the exciton annihilation radius and diffusion coefficient of a single PbI_(2)nanoflake under nonresonant and resonant excitation conditions of band-edge exciton state.As initial exciton densities increase,we find the carrier recombination mechanism for a single PbI_(2)nanoflake gradually changes from exciton-exciton annihilation to free-carrier recombination.Finally,we reveal the room-temperature circular polarization of a single PbI_(2)nanoflake is due to free-carrier recombination with a band-edge exciton dissociation time of~120 fs under the resonant excitation condition.XU ChenYu WANG Lei CUI Lin GAO BingRong WANG HaiYu 2024Science China(Technological Sciences)2024,67,1:0

13Low-strain Co-free Li-rich layered cathode with excellent voltage and capacity stability显示文摘Owing to the inherent advantages of low cost and high capacity,cobalt(Co)-free lithium(Li)-rich layered oxides have become one of the most promising cathodes for next-generation high-energy lithium-ion batteries.However,these familial cathodes suffer from serious voltage decay due to many reasons,such as oxygen release and transition metal(TM)migration,which are closely related to nanoscale strain evolution.Here,by combining the synergistic effects of surface integration,bulk doping,and concentration gradient,we successfully construct a Co-free Li-rich layered cathode with a very small volumetric strain(1.05%)between 2.0 and 4.8 V,approaching the critical value of zero strain.Various characterizations indicate that the constructed zero-strain cathode can significantly suppress the TM migration,interfacial reactions,and structural degradation including cracks,lattice defects,phase evolution,and nanovoids,leading to improved voltage stability of Co-free Li-rich layered oxides during the prolonged cycles.This work provides a strategy to eliminate the lattice strain of Li-rich layered cathodes and facilitates the up-scaled application of the as-prepared cathode materials.Zhuo Yao Yong Chen Chenyu Liu Hao Chen Shuxing Wu Dong Luo Zhan Lin Shanqing Zhang 2023Journal of Energy Chemistry2023,,7:0
14Endoscopy demand among county people in southeast China:A cross-sectional study显示文摘Objective The popularization of gastroenteroscopy and the introduction of comfortable medical care have further promoted the growth of people's demand,especially the demand for painless endoscopy.This cross-sectional study aims to investigate the current situation and change in county people's demand for endoscopy to promote the development of endoscopy centers in county hospitals in southeast China.Methods From October to December 2021,patients and their family members who came to the Gastroenterology Department in Suichang County People's Hospital of Zhejiang Province were randomly selected to complete the questionnaire.A total of 838 valid questionnaires were collected.Additionally,the original software data of the Endoscopy Center were sampled and retrieved(from October to December every year from 2018 to 2021)for statistical analysis of real-world data.Those who would choose painless endoscopy the next time in the valid questionnaires were included in the painless endoscopy group,while those who would choose ordinary endoscopy the next time were included in the ordinary endoscopy group.Results The stepwise forward binary logistic regression model analysis showed that,patients with“secondhand smoke exposure”were more willing to choose painless endoscopy(OR=1.459,95%CI:1.050-2.028,p=0.025).However,patients with an education level of“primary and below”and“junior high school”,and patients who are suffering from“currently experiencing abdominal distension”,were more willing to choose ordinary endoscopy(OR=0.270,95%CI:0.149-0.488,p<0.001;OR=0.528,95%CI:0.330-0.845,p=0.008;OR=0.536,95%CI:0.334-0.861,p=0.010).Patients with previous experience in painless endoscopy tended to choose painless endoscopy the next time,while patients with previous experience in ordinary endoscopy tended to choose ordinary endoscopy the next time(χ^(2)=140.97,p<0.001).From 2018 to 2021,the proportion of painless endoscopy has increased yearly(p<0.001).Most patients indicated that they would“regularly review gastroenteroscopy”(477/838,56.9%).Conclusions With Suichang County of Zhejiang Province as the representative,the demand for painless endoscopy for people's gastrointestinal cancer detection in southeast China has been increasing yearly.The development of endoscopy centers in county-level hospitals can basically meet the demand growth.Meanwhile,advanced concepts such as comfortable medical care and regular follow-up are gradually popularized at the grassroots level in southeast China.Huihui Yan Zhenghua Lin Shuangjing Gao Chenyu Fan Mengyue Jiang Liying Que Lanfang Zhou Yingdi Weng Jing Shu Tongyun Zhang Jian Hu Zhiqiang Liu Xi Ye Jianting Cai Guangfa Liao 2023Laparoscopic, Endoscopic and Robotic Surgery2023,6,1:0

16Dual-site Doping to Enhance Oxygen Redox and Structural Stability of Li-rich Layered Oxides显示文摘Cobalt-free Li-rich layered oxides(LLOs)such as Li_(2)MnO_(3) have attracted extensive attention owing to their high specific capacity and low cost.Nonetheless,numerous problems such as continuous voltage fading and capacity decay have become stumbling blocks in its commercial application.In this study,we propose an effective dual-site doping strategy by choosing Mo as the cation and F as the anion to enhance the capacity and cycling performance.The researchdemonstrates that the cycling stability of LLOs enhances with the doping ratio of Mo,and their capacity increases with the doping ratio of F.It is because Mo as a pillar enhances the structural stability and F doping is conducive to the activation of Li;MnO;.What’s more,dual-site doping also promotes the diffusion of Li;and reduces the internal resistance of the electrode.Due to these improvements,the 5F3M sample still maintains a discharge capacity of 190.98 mAh g;after 100 cycles at 200 mA g^(-1),which is much higher than 165.29 mAh g;of the Pristine sample.This discovery provides a new way to develop advanced layered oxide cathodes for both Na-and Li-ion batteries.Zuhao Zhang Xiaoyan Xie Huixian Xie Xiaokai Ding Jiaxiang Cui Chenyu Liu Dong Luo Zhan Lin 2022Chinese Journal of Structural Chemistry2022,41,4:0

19Down-regulation of MeMYB2 leads to anthocyanin accumulation and increases chilling tolerance in cassava(Manihot esculenta Crantz)显示文摘Chilling-induced accumulation of reactive oxygen species(ROS) is harmful to plants,which usually produce anthocyanins to scavenge ROS as protection from chilling stress.As a tropical crop,cassava is hypersensitive to chilling,but the biochemical basis of this hypersensitivity remains unclear.We previously generated Me MYB2-RNAi transgenic cassava with increased chilling tolerance.Here we report that Me MYB2-RNAi transgenic cassava accumulated less ROS but more cyanidin-3-O-glucoside than the wild type under early chilling stress.Under this stress,the anthocyanin biosynthesis pathway was more active in Me MYB2-RNAi lines than in the wild type,and several genes involved in the pathway,including Me TT8,were up-regulated by Me MYB2-RNAi in the transgenic cassava.Me MYB2 bound to the Me TT8 promoter and blocked its expression under both normal and chilling conditions,thereby inhibiting anthocyanin accumulation.Me TT8 was shown to bind to the promoter of Dihydroflavonol 4-reductase(Me DFR-2)and increased Me DFR-2 expression.Me MYB2 appears to act as an inhibitor of chilling-induced anthocyanin accumulation during the rapid response of cassava to chilling stress.Xin Guo Xiaohui Yu Chenyu Lin Pingjuan Zhao Bin Wang Liangping Zou Shuxia Li Xiaoling Yu Yinhua Chen Peng Zhang Ming Peng Mengbin Ruan 2023The Crop Journal2023,11,4:0
