| 1 | Three-dimensional bioprinting of multicell-laden scaffolds containing bone morphogenic protein-4 for promoting M2 macrophage polarization and accelerating bone defect repair in diabetes mellitus显示文摘Critical-sized bone defect repair in patients with diabetes mellitus remains a challenge in clinical treatment because of dysfunction of macrophage polarization and the inflammatory microenvironment in the bone defect region.Three-dimensional(3D)bioprinted scaffolds loaded with live cells and bioactive factors can improve cell viability and the inflammatory microenvironment and further accelerating bone repair.Here,we used modified bioinks comprising gelatin,gelatin methacryloyl(GelMA),and 4-arm poly(ethylene glycol)acrylate(PEG)to fabricate 3D bioprinted scaffolds containing BMSCs,RAW264.7 macrophages,and BMP-4-loaded mesoporous silica nanoparticles(MSNs).Addition of MSNs effectively improved the mechanical strength of GelMA/gelatin/PEG scaffolds.Moreover,MSNs sustainably released BMP-4 for long-term effectiveness.In 3D bioprinted scaffolds,BMP-4 promoted the polarization of RAW264.7 to M2 macrophages,which secrete anti-inflammatory factors and thereby reduce the levels of pro-inflammatory factors.BMP-4 released from MSNs and BMP-2 secreted from M2 macrophages collectively stimulated the osteogenic differentiation of BMSCs in the 3D bioprinted scaffolds.Furthermore,in calvarial critical-size defect models of diabetic rats,3D bioprinted scaffolds loaded with MSNs/BMP-4 induced M2 macrophage polarization and improved the inflammatory microenvironment.And 3D bioprinted scaffolds with MSNs/BMP-4,BMSCs,and RAW264.7 cells significantly accelerated bone repair.In conclusion,our results indicated that implanting 3D bioprinted scaffolds containing MSNs/BMP-4,BMSCs,and RAW264.7 cells in bone defects may be an effective method for improving diabetic bone repair,owing to the direct effects of BMP-4 on promoting osteogenesis of BMSCs and regulating M2 type macrophage polarization to improve the inflammatory microenvironment and secrete BMP-2. | Xin Sun Zhenjiang Ma Xue Zhao Wenjie Jin Chenyu Zhang Jie Ma Lei Qiang Wenhao Wang Qian Deng Han Yang Jinzhong Zhao Qianqian Liang Xiaojun Zhou Tao Li Jinwu Wang | 2021 | Bioactive Materials2021,6,3: | 12 |
| 2 | Responses of Mean and Extreme Precipitation to Different Climate Forcing Under Radiative-Convective Equilibrium显示文摘Understanding the responses of mean and extreme precipitation to climate change is of great importance.Previous studies have mainly focused on the responses to prescribed sea surface warming or warming due to increases of CO2.This study uses a cloud-resolving model under the idealization of radiative-convective equilibrium to examine the responses of mean and extreme precipitation to a variety of climate forcings,including changes in prescribed sea surface temperature,CO2,solar insolation,surface albedo,stratospheric volcanic aerosols,and several tropospheric aerosols.The different responses of mean precipitation are understood by examining the changes in the surface energy budget.It is found that the cancellation between shortwave scattering and longwave radiation leads to a small dependence of the mean precipitation response on forcings.The responses of extreme precipitation are decomposed into three components(thermodynamic,dynamic,and precipitation efficiency).The thermodynamic components for all climate forcings are similar.The dynamic components and the precipitation-efficiency components,which have large spreads among the cases,are negatively correlated,leading to a small dependence of the extreme precipitation response on the forcings. | Chenyu MA Wei YUAN Ji NIE | 2020 | Advances in Atmospheric Sciences2020,37,4: | 2 |

| 5 | Functional analysis of the sbcD (dr1921) gene of the extremely radioresistant bacterium Deinococcus radiodurans显示文摘In eukaryotes, the Mre11-Rad50-Nbs1 (MRN) complex, which resides at the crossroads of DNA repair and checkpoint signaling, rapidly forms prominent foci at damage sites following double-strand break (DSB) induction. This complex carries out the initial processing of the DSB ends. Mutations in the genes that encode components of this complex result in DNA-damage hypersensitivity, genomic in- stability, telomere shortening, and aberrant meiosis. Therefore, the MR proteins are highly conserved during evolution. The bacterial orthologs of Mre11 and Rad50 are the SbcD and SbcC proteins, respec- tively. Deinococcus radiodurans, an extremely radioresistant bacterium, is able to mend hundreds of radiation-induced DSBs. The SbcD and SbcC proteins were identified as the products of the Dr1921 and Dr1922 genes. Disruption of the sbcD gene, by direct reverse-orientation insertional mutagenesis tech- nology, remarkably increases the cells’ sensitivity to various types of DNA damaging agents, such as ionizing radiation, ultraviolet irradiation, hydrogen peroxide, and mitomycin C. We also provide evidence that the drSbcD protein plays an important role in both growth and DNA repair in this organ- ism, especially in repair of DSBs generated after cellular exposure to 6000 Gy of IR. | 6 | Exploring the Evolution of Passenger Flow and Travel Time Reliability with the Expanding Process of Metro System Using Smartcard Data显示文摘Metro system has experienced the global rapid rise over the past decades. However,few studies have paid attention to the evolution in system usage with the network expanding. The paper's main objectives are to analyze passenger flow characteristics and evaluate travel time reliability for the Nanjing Metro network by visualizing the smart card data of April 2014,April 2015 and April 2016. We performed visualization techniques and comparative analyses to examine the changes in system usage between before and after the system expansion. Specifically,workdays,holidays and weekends were specially segmented for analysis.Results showed that workdays had obvious morning and evening peak hours due to daily commuting,while no obvious peak hours existed in weekends and holidays and the daily traffic was evenly distributed. Besides,some metro stations had a serious directional imbalance,especially during the morning and evening peak hours of workdays. Serious unreliability occurred in morning peaks on workdays and the reliability of new lines was relatively low,meanwhile,new stations had negative effects on exiting stations in terms of reliability. | 11 | Impeded degradation of perovskite solar cells via the dual interfacial modification of siloxane显示文摘It is challenging to improve the long-term stability of perovskite solar cells(PSCs) without sacrificing efficiency. The perovskite absorbers degrade from the film surface/interfaces, which follows entangled mechanisms that have not been fully revealed yet.Herein, we decouple and elaborate two distinctive pathways regarding film degradation based on FACsPbI3perovskites.Moreover, a dual interfacial modification strategy has been developed for improving the material’s intrinsic stability, thus leading to the film degrading in a more retardant pathway. The corresponding PSCs achieve a stable power output efficiency of 23.75%.More importantly, the unencapsulated PSCs devices retain over 93% of their initial PCE after the maximum power point(MPP)tracking under the continuous 1-sun illumination and show significantly improved stability after aged under the thermal treatment or stored in ambient atmosphere for over 1500 hours without obvious PCE decay. | 16 | Protective effects and mechanisms of Lizhong decoction(理中汤) against non-alcoholic fatty liver disease in a rat model显示文摘OBJECTIVE: To investigate the protective effects and molecular mechanisms of Lizhong decoction( 理中汤, LZD) against non-alcoholic fatty liver disease(NAFLD). METHODS: Male Wistar rats were fed a high-fat diet for four weeks to induce NAFLD, and were administered LZD by gavage for four weeks. Potential therapeutic targets for NAFLD were analyzed using network pharmacology. Liver pathology was evaluated using Oil Red O and hematoxylin-eosin staining. Furthermore, mitochondrial function, lipid metabolism, oxidative stress, and inflammatory response were examined. RESULTS: Rats with NAFLD exhibited high levels of hepatic damage and cholesterol deposition. Moreover, apoptosis was increased, superoxide dismutase and glutathione content were reduced, malondialdehyde content was increased, and the protein expression of inflammatory cytokines and p-c-Jun N-terminal kinase was increased. The LZD treatment ameliorated mitochondrial dysfunction, reduced liver damage,inhibited oxidative stress and inflammatory response, upregulated peroxisome proliferator-activated receptor(PPAR)-γ expression, and suppressed dipeptidyl peptidase 4(DPP4) expression in the liver. In this paper, the mobility degradation is modeled in combination with the universal NBTI model.The coulomb scattering induced by interface states is revealed to be the dominant component responsible for mobility degradation.
