1Combined genomic, transcriptomic, and metabolomic an alyses provide in sights into chayote (Sechium edule) evolution and fruit development显示文摘Chayote(Sechium edule)is an agricultural crop in the Cucurbitaceae family that is rich in bioactive components.To enhance genetic research on chayote,we used Nanopore third-generation sequencing combined with Hi-C data to assemble a draft chayote genome.A chromosome-level assembly anchored on 14 chromosomes(N50 contig and scaffold sizes of 8.40 and 46.56 Mb,respectively)estimated the genome size as 606.42 Mb,which is large for the Cucurbitaceae,with 65.94%(401.08 Mb)ofthe genome comprising repetitive sequences;28,237 protein-coding genes were predicted.Comparative genome analysis indicated that chayote and snake gourd diverged from sponge gourd and that a whole-genome duplication(WGD)event occurred in chayote at 25±4 Mya.Transcriptional and metabolic analysis revealed genes involved in fruit texture,pigment,fl avor,fl avonoids,antioxidants,and plant hormones during chayote fruit development.The analysis of the genome,transcriptome,and metabolome provides insights into chayote evolution and lays the groundwork for future research on fruit and tuber development and genetic improvements in chayote.Anzhen Fu Qing Wang Jianlou Mu Lili Ma Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuechuan Zhang Xuewen Zhang Fengling Wang Donald Grierson Jinhua Zuo 2021Horticulture Research2021,8,1:6
2Enhancement of T Follicular Helper Cell-Mediated Humoral Immunity Reponses During Development of Experimental Autoimmune Myasthenia Gravis显示文摘Myasthenia gravis(MG) is a prototypical antibody-mediated neurological autoimmune disease with the involvement of humoral immune responses in its pathogenesis. T follicular helper(Tfh) cells have been implicated in many autoimmune diseases. However, whether and how Tfh cells are involved in MG remain unclear.Here, we established and studied a widely-used and approved animal model of human MG, the rat model with acetylcholine receptor alpha(AChRa) subunit(RAChR97–116)-induced experimental autoimmune myasthenia gravis(EAMG). This model presented mild bodyweight loss 10 days after the first immunization(representing the early stage of disease) and more obvious clinical manifestations and body-weight loss 7 days after the second immunization(representing the late stage of disease). AChR-specific pre-Tfh cells and mature Tfh cells were detected in these two stages, respectively. In cocultures of Tfh cells and B cells, the number of IgG2 bsecreting B cells and the level of anti-AChR antibodies in the supernatant were higher in the cultures containing EAMG-derived Tfh cells. In immunohistochemistry and immunofluorescence assays, a substantial number of CD4^+/Bcl-6^+ T cells and a greater number of larger germinal centers were observed in lymph node tissues resected from EAMG rats. 4The genome and transcriptome analysis of snake gourd provide insights into its evolution and fruit development and ripening显示文摘Snake gourd(Trichosanthes anguina L.),which belongs to the Cucurbitaceae family,is a popular ornamental and food crop species with medicinal value and is grown in many parts of the world.Although progress has been made in its genetic improvement,the organization,composition,and evolution of the snake gourd genome remain largely unknown.Here,we report a high-quality genome assembly for snake gourd,comprising 202 contigs,with a total size of 919.8 Mb and an N50 size of 20.1 Mb.These findings indicate that snake gourd has one of the largest genomes of Cucurbitaceae species sequenced to date.The snake gourd genome assembly harbors 22,874 protein-coding genes and 80.0%of the genome consists of repetitive sequences.Phylogenetic analysis reveals that snake gourd is closely related to sponge gourd but diverged from their common ancestor~33–47 million years ago.The genome sequence reported here serves as a valuable resource for snake gourd genetic research and comparative genomic studies in Cucurbitaceae and other plant species.In addition,fruit transcriptome analysis reveals the candidate genes related to quality traits during snake gourd fruit development and provides a basis for future research on snake gourd fruit development and ripening at the transcript level.Lili Ma Qing Wang Jianlou Mu Anzhen Fu Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuewen Zhang Xuechuan Zhang Zhangjun Fei Donald Grierson Jinhua Zuo 2020Horticulture Research2020,7,1:2
5Long non-coding RNA LncKdm2b regulates cortical neuronal differentiation by cis-activating Kdm2b显示文摘The mechanisms underlying spatial and temporal control of cortical neurogenesis of the brain are largely elusive.Long non-coding RNAs(lncRNAs)have emerged as essential cell fate regulators.Here we found LncKdm2b(also known as Kancr),a lncRNA divergently transcribed from a bidirectional promoter of Kdm2b,is transiently expressed during early differentiation of cortical projection neurons.Interestingly,Kdm2b’s transcription is positively regulated in cis by LncKdm2b,which has intrinsic-activating function and facilitates a permissive chromatin environment at the Kdm2b’s promoter by associating with hnRNPAB.Lineage tracing experiments and phenotypic analyses indicated LncKdm2b and Kdm2b are crucial in proper differentiation and migration of cortical projection neurons.These observations unveiled a lncRNA-dependent machinery in regulating cortical neuronal differentiation.Wei Li Wenchen Shen Bo Zhang Kuan Tian Yamu Li Lili Mu Zhiyuan Luo Xiaoling Zhong Xudong Wu Ying Liu Yan Zhou 2020Protein & Cell2020,11,3:2
6NUMB enhances Notch signaling by repressing ubiquitination of NOTCH1 intracellular domain显示文摘The release and nuclear translocation of the intracellular domain of Notch receptor(NICD)is the prerequisite for Notch signaling-mediated transcriptional activation.NICD is subjected to various posttranslational modifications including ubiquitination.Here,we surprisingly found that NUMB proteins stabilize the intracellular domain of NOTCH1 receptor(N1ICD)by regulating the ubiquitin–proteasome machinery,which is independent of NUMB’s role in modulating endocytosis.BAP1,a deubiquitinating enzyme(DUB),was further identified as a positive N1ICD regulator,and NUMB facilitates the association between N1ICD and BAP1 to stabilize N1ICD.Intriguingly,BAP1 stabilizes N1ICD independent of its DUB activity but relying on the BRCA1-inhibiting function.BAP1 strengthens Notch signaling and maintains stem-like properties of cortical neural progenitor cells.Thus,NUMB enhances Notch signaling by regulating the ubiquitinating activity of the BAP1–BRCA1 complex.Zhiyuan Luo Lili Mu Yue Zheng Wenchen Shen Jiali Li Lichao Xu Bo Zhong Ying Liu Yan Zhou 2020Journal of Molecular Cell Biology2020,12,5:2

20Upconversion fluorescence modulation of CaTiO_(3):Yb^(3+)/Er^(3+)nanocubes via Zn^(2+)introduction显示文摘To investigate the effect of Zn^(2+)ions on the luminescence properties of rare earth(RE)doped calcium titanate materials,CaTiO_(3):Yb^(3+)/Er^(3+)/Zn^(2+)nanocubes with uniform size were prepared by solvothermal method in this paper.They were respectively characterized by X-ray diffractometer(XRD),scanning electron microscope(SEM),photoluminescence(PL)spectroscopy and their fluorescence lifetimes.The results show that the average size of the nanocubes is about 550 nm×650 nm×850 nm with good upconversion luminescence(UCL)properties.Under 980 nm laser excitation,the effects of the ratio between activator Er^(3+)and sensitizer Yb^(3+)and Zn^(2+)doping on the upconversion fluorescence properties were investigated,and the optimal ion ratio was obtained.The results of steady-state spectra show that the strongest fluorescence intensity of CaTiO_(3):Yb^(3+)/Er^(3+)was obtained with the addition of 10 mol%Zn^(2+)at a Yb^(3+)/Er^(3+)molar doping ratio of 3:0.3,which was attributed to the crystal field asymmetry generated by the introduction of Zn^(2+)ions.The energy transfer and upconversion mechanism between Yb^(3+)and Er^(3+)ions in CaTiO_(3) nanocubes were investigated by analyzing the upconversion fluorescence kinetics.MU Jiajia LIU Jinyu GAO Lili 2022Optoelectronics Letters2022,18,3:0
