2Association of the Late Cornified Envelope-3 Genes with Psoriasis and Psoriatic Arthritis:A Systematic Review显示文摘Psoriasis(Ps)and psoriatic arthritis(Ps A)are genetically complex diseases with strong genetic evidence.Recently,susceptibility genes for Ps and Ps A have been identified within the late cornified envelop(LCE)gene cluster,especially the cluster 3(LCE3)genes.It is noteworthy that the deletion of LCE3B and LCE3C(LCE3C_LCE3B-del)is significantly associated with these two diseases.Gene-gene interactions between LCE3 genes and other genes are associated with Ps and Ps A.LCE3 genes also have pleiotropic effect on some autoimmune diseases,such as rheumatoid arthritis,atopic dermatitis and systemic lupus erythematosus.Further studies need to focus on the potential function of LCE3 genes in the pathogenesis of Ps and Ps A in the future.Changbing Shen Jing Gao Xianyong Yin Yujun Sheng Liangdan Sun Yong Cui Xuejun Zhang 2015Journal of Genetics and Genomics2015,42,2:4
3DNA methylation-based subclassification of psoriasis in the Chinese Han population显示文摘Psoriasis (Ps)is an inflammatory skin disease caused by genetic and environmental factors.Previous studies on DNA methylation (DNAm)found genetic markers that are closely associated with Ps,and evidence has shown that DNAm mediates genetic risk in Ps.In this study,Consensus Clustering was used to analyze DNAm data,and 114Ps patients were divided into three subclassifications.Investigation of the clinical characteristics and copy number variations (CNVs)of DEFB4,IL22,and LCE3C in the three subclassifications revealed no significant differences in gender ratio and in Ps area and severity index (PASI)score.The proportion of late-onset (≥40 years)Ps patients was significantly higher in type I than in types H and III (P =0.035).Type III contained the smallest proportion of smokers and the largest proportion of non-smoking Ps patients (P =0.086).The CNVs of DEFB4and LCE3C showed no significant differences but the CNV oflL22significantly differed among the three subelassifications (P =0.044).This study is the first to profile Ps subclassifications based on DNAm data in the Chinese Han population.These results are useful in the treatment and management of Ps from the molecular and genetic perspectives.Fusheng Zhou Changbing Shen Yi-Hsiang Hsu Jing Gao Jinfa Dou Randy KO Xiaodong Zheng Liangdan Sun Yong Cui Xuejun Zhang 2018Frontiers of Medicine2018,12,6:3

5A rare variant in COL11A1 is strongly associated with adult height in Chinese Han population显示文摘Human height is a highly heritable trait in which multiple genes are involved. Recent genome-wide association studies(GWASs) have identified that COL11A1 is an important susceptibility gene for human height. To determine whether the variants of COL11A1 are associated with adult and children height,we analyzed splicing and coding single-nucleotide variants across COL11A1 through exome-targeted sequencing and two validation stages with a total 20,426 Chinese Han samples. A total of 105 variants were identified by exome-targeted sequencing, of which 30 SNPs were located in coding region. The strongest association signal was chr1_103380393 with P value of 4.8 * 10^(-7). Chr1_103380393 also showed nominal significance in the validation stage(P = 1.21 * 10^(-6)). Combined analysis of 16,738 samples strengthened the original association of chr1_103380393 with adult height(P_(combined)= 3.1 * 10^(-8)), with an increased height of 0.292sd(standard deviation) per G allele(95% CI:0.19-0.40). There was no evidence(P = 0.843) showing that chr1_103380393 altered child height in 3688 child samples. 7Heat-inducible SlWRKY3 confers thermotolerance by activating the SlGRXS1 gene cluster in tomato显示文摘High temperature stress is one of the major environmental factors that affect the growth and development of plants. Although WRKY transcription factors play a critical role in stress responses, there are few studies on the regulation of heat stress by WRKY transcription factors,especially in tomato. Here, we identified a group I WRKY transcription factor, SlWRKY3, involved in thermotolerance in tomato. First, SlWRKY3 was induced and upregulated under heat stress. Accordingly, overexpression of SlWRKY3 led to an increase, whereas knock-out of SlWRKY3 resulted in decreased tolerance to heat stress. Overexpression of SlWRKY3 accumulated less reactive oxygen species(ROS), whereas knock-out of SlWRKY3 accumulated more ROS under heat stress. This indicated that SlWRKY3 positively regulates heat stress in tomato. In addition,SlWRKY3 activated the expression of a range of abiotic stress-responsive genes involved in ROS scavenging, such as a SlGRXS1 gene cluster.Further analysis showed that SlWRKY3 can bind to the promoters of the SlGRXS1 gene cluster and activate their expression. 8Multiple-model GWAS identifies optimal allelic combinations of quantitative trait loci for malic acid in tomato显示文摘Malic acid(MA)is an important flavor acid in fruits and acts as a mediator in a series of metabolic pathways.It is important to understand the factors affecting MA metabolism for fruit flavor improvement and to understand MA-mediated biological processes.However,themetabolic accumulation of MA is controlled by complex heredity and environmental factors,making it difficult to predict and regulate the metabolism of MA.In this study,we carried out a genome-wide association study(GWAS)on MA using eight milestone models with two-environment repeats.A series of associated SNP variations were identified from the GWAS,and 15 high-confidence annotated geneswere further predicted based on linkage disequilibrium and lead SNPs.The transcriptome data of candidate geneswere explored within different tomato organs as well as various fruit tissues,and suggested specific expression patterns in fruit pericarp.Based on the genetic parameters of population differentiation and SNP distribution,tomato MA content has been more influenced by domestication sweeps and less affected by improvement sweeps in the long-term history of tomato breeding.In addition,genotype×environment interaction might contribute to the difference in domestication phenotypic data under different environments.This study provides new genetic insights into how tomato changed its MA content during breeding and makes available function-based markers for breeding by marker-assisted selection.Wenxian Gai Fan Yang Liangdan Yuan Saeed ul Haq Yaru Wang Ying Wang Lele Shang Fangman Li Pingfei Ge Haiqiang Dong Jinbao Tao Fei Wang Xingyu Zhang Yuyang Zhang 2023Horticulture Research2023,10,4:0

10Effect of different plant growth regulators on callus and adventitious shoots induction,polysaccharides accumulation and antioxidant activity of Rhodiola dumulosa显示文摘Objective:As a medicinal plant,the resource of Rhodiola dumulosa is deficient along with the large collection.For the protection and utilization of R.dumulosa,the influence of plant growth regulators(PGRs)on callus induction and adventitious shoots differentiation,polysaccharide production and the antioxidant activity were tested.Methods:Internodes of R.dumulosa were used as explants and cultured on MS medium plus different plant growth regulators(PGRs).The anti-oxidative activities of polysaccharides were evaluated using radical scavenging assays.Results:By response surface plot,0.85 mg/L N6-benzyladenine(BA),0.34 mg/L naphthaleneacetic acid(NAA)and 0.33 mg/L 2,4-dicholorophenoxyacetic acid(2,4-D)were the optimal factors for callus induction(90.03%)from internodes explants on MS medium.The fresh weight of green callus increased 47.26 fold,when callus was inoculated on MS+thidiazuron(TDZ)0.5 mg/L+NAA 2.0 mg/L.Adventitious buds regenerated from callus on the media of MS were fortified with BA 1.0 mg/L plus NAA 0.5 mg/L,and the induction rate was 40.00%.MS plus indole-3-butyric acid(IBA)1.0 mg/L produced the highest rooting rate with 10 to 15 roots in a length of 2–3 cm per shoot.The content of total polysaccharides in callus developed on MS+TDZ 0.5 mg/L+NAA 2.0 mg/L and MS+BA 1.0 mg/L+NAA 0.5 mg/L was as high as 1.72%2.15%.At the dose of 0.5 mg/mL polysaccharides extracted from different callus induced on MS+NAA 2.0 mg/L+TDZ 0.5 mg/L or MS+BA 1.0 mg/L+NAA 0.5 mg/L or MS+BA 0.5 mg/L+2,4-D 0.5 mg/L,the ABTS radical eliminating percentages were 82.78%,80.18%and 68.59%,respectively,much higher than that of wild plant.Conclusion:A rapid micropropagation system for R.dumulosa has been developed.The combination of TDZ and NAA or BA and NAA can increase the yield of the total polysaccharides.The polysaccharides isolated from callus and whole wild plants had stronger free radicals scavenging activities,indicating that polysaccharides from R.dumulosa are the potential pharmaceutical supplements.Xu Lu Liangdan Fei Yi Li Jialin Du Weiwei Ma Hangjun Huang Junli Wang 2023Chinese Herbal Medicines2023,15,2:0
11Clinical efficacy of low-dose emetine for patients with COVID-19:a real-world study显示文摘Objective:Emetine,an isoquinoline alkaloid that is enriched at high concentrations in the lung,has shown potent in vitro activity against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).The aim of this study was to better understand the effectiveness of low-dose emetine for patients with coronavirus disease 2019(COVID-19).Methods:In this real-world study,63 patients with mild or common COVID-19 were recruited from Wuhan Fangcang Shelter Hospital and five COVID-19-designated hospitals in Anhui Province,China from February to March 2020.Thirty-nine patients from Wuhan Fangcang Shelter Hospital were assigned to a pragmatic randomized controlled clinical trial,and 24 patients from the 5 COVID-19-designated hospitals in Anhui Province underwent a real-world study.The medication course of emetine was less than 10 days.The main symptoms and adverse reactions of all patients were observed and recorded.The primary outcome measure was the time required for a negative SARS-CoV-2 RNA result or the negative result rate on day 10.Secondary outcomes included axillary temperature,transcutaneous oxygen saturation,and respiratory frequency recovery.The study was approved by the Ethics Committee of The First Affiliated Hospital of Anhui Medical University on February 20,2019(approval No.PJ2020-03-19)and was registered with the Chinese Clinical Trial Registry on February 20,2019(registration number:ChiCTR2000030022).Results:The oxygen saturation values were higher in the treatment group than in the control group on the first day after enrollment for patients treated at Fangcang Shelter Hospital.The axillary body temperature,respiratory rate,and oxygen saturation among patients in Fangcang Shelter Hospital were related to the time effect but not to the intervention measures.The respiratory rate and oxygen saturation of patients in the Anhui designated hospitals were related to the intervention measures but not to the time effect.The axillary body temperature of patients in Anhui designated hospitals was related to the time effect but not to the intervention measures.Conclusion:Our preliminary study shows that low-dose emetine combined with basic conventional antiviral drugs improves clinical symptoms in patients with mild and common COVID-19 without apparent adverse effects,suggesting that moderately increased doses of emetine may have good potential for treatment and prevention of COVID-19.Song Fan Qi Zhen Cheng Chen Wenjun Wang Qibing Wu Huihui Ma Chengyuan Zhang Li Zhang Baojing Lu Huiyao Ge Liang Yong Bao Li Yafen Yu Weiwei Chen Yiwen Mao Guangbo Qu Li Su Aoli Wang Zhen Ding Haiwen Li Jin Zhang Yonglian Wang Yufeng Gao Xihai Xu Zhongming Zhu Jun Chen Long Zhang Hongqiang Liang Song Wu Meng Huang Quan Xia Ping Li Yehuan Sun Chaozhao Liang Wei Wei Qingsong Liu Liangdan Sun 2021Journal of Bio-X Research2021,4,2:0

13Data-independent acquisition mass spectrometry quantitative proteomic analysis reveals that skin aging-related proteins differ between men and women显示文摘The skin is the largest organ of the human body,and its aging is visible to the naked eye.The aging rate of men and women is slightly different.This study compared the protein expression of skin samples on the curved forearms of 11 healthy women and 9 healthy men.Quantitative proteomics analysis found that the expression of epidermal proteins in men and women of the same age group was different.Compared with female skin,in male skin,20 proteins were upregulated,and 7 proteins were downregulated.These data suggest that men and women have differences in the speed of skin aging.For the first time in this experiment,a non-invasive mass spectrometer was used to detect 27 different-related epidermal proteins between men and women.Compared with women,among the 20 epidermal proteins upregulated in men,their functions can be classified into antioxidants,epidermal lipid metabolism,signal transduction,membrane transport,and cell biological processes;the 7 downregulated proteins are involved in a variety of biological processes and inflammatory reactions.Our experiments have discovered epidermal proteins related to the differences between men and women,enriching the library of epidermal differential proteins between men and women and enriching the mechanism of skin aging between men and women from the perspective of epidermal differential proteins.HUI ZHANG SHIRUI CHEN MENGTING LIU YAOCHI WANG CONG XIN JING MA XIAODONG ZHENG YUANDI HUANG BO ZHANG XUEJUN ZHANG LIANGDAN SUN SEN YANG 2021BIOCELL2021,45,5:0
