| 2 | Release of VEGF and BMP9 from injectable alginate based composite hydrogel for treatment of myocardial infarction显示文摘Myocardial infarction(MI)is one of cardiovascular diseases that pose a serious threat to human health.The pathophysiology of MI is complex and contains several sequential phases including blockage of a coronary artery,necrosis of myocardial cells,inflammation,and myocardial fibrosis.Aiming at the treatment of different stages of MI,in this work,an injectable alginate based composite hydrogel is developed to load vascular endothelial active factor(VEGF)and silk fibroin(SF)microspheres containing bone morphogenetic protein 9(BMP9)for releasing VEGF and BMP9 to realize their respective functions.The results of in vitro experiments indicate a rapid initial release of VEGF during the first few days and a relatively slow and sustained release of BMP9 for days,facilitating the formation of blood vessels in the early stage and inhibiting myocardial fibrosis in the long-term stage,respectively.Intramyocardial injection of such composite hydrogel into the infarct border zone of mice MI model via multiple points promotes angiogenesis and reduces the infarction size.Taken together,these results indicate that the dual-release of VEGF and BMP9 from the composite hydrogel results in a collaborative effect on the treatment of MI and improvement of heart function,showing a promising potential for cardiac clinical application. | Yong Wu Tianqi Chang Weiqian Chen Xiaoyu Wang Jingjing Li Yueqiu Chen You Yu Zhenya Shen Qian Yu Yanxia Zhang | 2021 | Bioactive Materials2021,6,2: | 7 |

| 5 | Application of regional citrate anticoagulation in patients at high risk of bleeding during intermittent hemodialysis:a prospective multicenter randomized controlled trial显示文摘Objective:Safe and effective anticoagulation is essential for hemodialysis patients who are at high risk of bleeding.The purpose of this trial is to evaluate the effectiveness and safety of two-stage regional citrate anticoagulation(RCA)combined with sequential anticoagulation and standard calcium-containing dialysate in intermittent hemodialysis(IHD)treatment.Methods:Patients at high risk of bleeding who underwent IHD from September 2019 to May 2021 were prospectively enrolled in 13 blood purification centers of nephrology departments,and were randomly divided into RCA group and saline flushing group.In the RCA group,0.04 g/mL sodium citrate was infused from the start of the dialysis line during blood draining and at the venous expansion chamber.The sodium citrate was stopped after 3 h of dialysis,which was changed to sequential dialysis without anticoagulant.The hazard ratios for coagulation were according to baseline.Results:A total of 159 patients and 208 sessions were enrolled,including RCA group(80 patients,110 sessions)and saline flushing group(79 patients,98 sessions).The incidence of severe coagulation events of extracorporeal circulation in the RCA group was significantly lower than that in the saline flushing group(3.64%vs.20.41%,P<0.001).The survival time of the filter pipeline in the RCA group was significantly longer than that in the saline flushing group((238.34±9.33)min vs.(221.73±34.10)min,P<0.001).The urea clearance index(Kt/V)in the RCA group was similar to that in the saline flushing group with no statistically significant difference(1.12±0.34 vs.1.08±0.34,P=0.41).Conclusions:Compared with saline flushing,the two-stage RCA combined with a sequential anticoagulation strategy significantly reduced extracorporeal circulation clotting events and prolonged the dialysis time without serious adverse events. | Xiaoyan TANG Dezheng CHEN Ling ZHANG Ping FU Yanxia CHEN Zhou XIAO Xiangcheng XIAO Weisheng PENG Li CHENG Yanmin ZHANG Hongbo LI Kehui LI Bizhen GOU Xin WU Qian YU Lijun JIAN Zaizhi ZHU Yu WEN Cheng LIU Hen XUE Hongyu ZHANG Xin HE Bin YAN Liping ZHONG Bin HUANG Mingying MAO | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,11: | 4 |

| 8 | Safety,tolerability,and pharmacokinetics of BAT8001 in patients with HER2-positive breast cancer:An open-label,dose-escalation,phase I study显示文摘Background:The introductions of anti-human epidermal growth factor receptor-2(HER2)agents have significantly improved the treatment outcome of patients with HER2-positive breast cancer.BAT8001 is a novel antibodydrug conjugate targeting human epidermal growth factor receptor-2(HER2)-expressing cells composed of a trastuzumab biosimilar linked to the drug-linker Batansine.This dose-escalation,phase I study was designed to assess the safety,tolerability,pharmacokinetics,and preliminary anti-tumor activity of BAT8001 in patients with HER2-positive locally advanced or metastatic breast cancer.Methods:This trial was conducted in subjects with histologically confirmed HER2-positive breast cancer(having evaluable lesions and an Eastern Cooperative Oncology Group performance status of 0 or 1)using a 3+3 design of escalating BAT8001 doses.Patients received BAT8001 intravenously in a 21-day cycle,with dose escalation in 5 cohorts:1.2,2.4,3.6,4.8,and 6.0 mg/kg.The primary objective was to evaluate the safety and tolerability of BAT8001.Preliminary activity of BAT8001 was also assessed as a secondary objective.Results:Between March 2017 to May 2018,29 HER2-positive breast cancer patients were enrolled.The observed dose-limiting toxicities were grade 4 thrombocytopenia and grade 3 elevated transaminase.The maximum tolerated dose was determined to be 3.6 mg/kg.Grade 3 or greater adverse events(AEs)occurred in 14(48.3%)of 29 patients,including thrombocytopenia in 12(41.4%)patients,aspartate aminotransferase increased in 4(13.8%)patients,γ-glutamyl transferase increased in 2(6.9%)patients,alanine aminotransferase increased in 2(6.9%)patients,diarrhea in 2(6.9%)patients.Objective response was observed in 12(41.4%,95%confidence interval[CI]=23.5%-61.1%)and disease control(including patients achieving objective response and stable disease)was observed in 24(82.8%,95%CI=64.2%-94.2%)patients.Conclusions:BAT8001 demonstrated favorable safety profiles,with promising anti-tumor activity in patients with HER2-positive locally advanced or metastatic breast cancer.BAT8001 has the potential to provide a new therapeutic option in patients with metastatic HER2-positive breast cancer. | Ruoxi Hong Wen Xia Liye Wang Kaping Lee Qianyi Lu Kuikui Jiang Shengfeng Li Jinquan Yu Jin Wei Weijia Tang Danyang Zhou Xin An Jiajia Huang Cong Xue Xiwen Bi Yanxia Shi Zhongyu Yuan Fei Xu Shusen Wang | 2021 | Cancer Communications2021,41,2: | 2 |
| 9 | Reprogramming of phytopathogen transcriptome by a non-bactericidal pesticide residue alleviates its virulence in rice显示文摘Bacteria equipped with virulence systems based on highly bioactive small molecules can circumvent their host's defense mechanisms.Pathogens employing this strategy are currently threatening global rice production.In the present study,variations in the virulence of the highly destructive Barkholderiaplantarii were observed in different rice-producing regions.The environment-linked variation was not attributable to any known host-related or external factors.Co-occurrence analyses indicated a connection between reduced virulence and 5-Amino-l,3,4-thiadiazole-2-thiol(ATT),a non-bactericidal organic compound.ATT,which accumulates in rice plants during metabolization of specific agrochemicals,was found to reduce virulence factor secretion by B.plantarii up to 88.8%and inhibit pathogen virulence by hijacking an upstream signaling cascade.Detailed assessment of the newly discovered virulence inhibitor resulted in mechanistic insights into positive effects of ATT accumulation in plant tissues.Mechanisms of virulence alleviation were deciphered by integrating high-throughput data,gene knockout mutants,and molecular interaction assays.TroK,a histidine protein kinase in a two-component system that regulates virulence factor secretion,is likely the molecular target antagonized by ATT.Our findings provide novel insights into virulence modulation in an important plant-pathogen system that relies on the host's metabolic activity and subsequent signaling interference. | Haruna Matsumoto Yuan Qian Xiaoyan Fan Sunlu Chen Yanxia Nie Kun Qiao Dandan Xiang Xinzhong Zhang Meng Li Bo Guo Peilin Shen Qiangwei Wang Yunlong Yu Tomislav Cemava Mengcen Wang | 2022 | Fundamental Research2022,2,2: | 2 |

| 19 | Core pluripotency factors promote glycolysis of human embryonic stem cells by activating GLUT1 enhancer显示文摘Human embryonic stem cells (hESCs) depend on glycolysis for energy and substrates for biosynthesis. To understand the mechanisms governing the metabolism of hESCs, we investigated the transcriptional regulation of glucose transporter 1 (GLUT1, SLC2A1), a key glycolytic gene to maintain pluripotency. By combining the genome-wide data of bin ding sites of the core pluripotency factors (SOX2, OCT4, NANOG, denoted SON), chromosomal interaction and histone modification in hESCs, we identified a potential enhancer of the GLUT1 gene in hESCs, de noted GLUT1 enhancer (GE) element GE interacts with the promoter of GLUT1, and the deletion of GE significantly reduces the expression of GLUT1, glucose uptake and glycolysis of hESCs, confirming that GE is an enhancer of GLUT1 in hESCs. In addition, the mutation of SON binding motifs within GE reduced the expression of GLUT1 as well as the interaction between GE and GLUT1 promoter, indicating that the binding of SON to GE is important for its activity.
