1A rare variant in COL11A1 is strongly associated with adult height in Chinese Han population显示文摘Human height is a highly heritable trait in which multiple genes are involved. Recent genome-wide association studies(GWASs) have identified that COL11A1 is an important susceptibility gene for human height. To determine whether the variants of COL11A1 are associated with adult and children height,we analyzed splicing and coding single-nucleotide variants across COL11A1 through exome-targeted sequencing and two validation stages with a total 20,426 Chinese Han samples. A total of 105 variants were identified by exome-targeted sequencing, of which 30 SNPs were located in coding region. The strongest association signal was chr1_103380393 with P value of 4.8 * 10^(-7). Chr1_103380393 also showed nominal significance in the validation stage(P = 1.21 * 10^(-6)). Combined analysis of 16,738 samples strengthened the original association of chr1_103380393 with adult height(P_(combined)= 3.1 * 10^(-8)), with an increased height of 0.292sd(standard deviation) per G allele(95% CI:0.19-0.40). There was no evidence(P = 0.843) showing that chr1_103380393 altered child height in 3688 child samples. 2Enhancing tylosin production by combinatorial overexpression of efflux,SAM biosynthesis,and regulatory genes in hyperproducing Streptomyces xinghaiensis strain显示文摘Tylosin is a 16-membered macrolide antibiotic widely used in veterinary medicine to control infections caused by Gram-positive pathogens and mycoplasmas.To improve the fermentation titer of tylosin in the hyperproducing Streptomyces xinghaiensis strain TL01,we sequenced its whole genome and identified the biosynthetic gene cluster therein.Overexpression of the tylosin efflux gene tlrC,the cluster-situated S-adenosyl methionine(SAM)synthetase gene metK_(cs),the SAM biosynthetic genes adoK_(cs)-metFcs,or the pathway-specific activator gene tylR enhanced tylosin production by 18%,12%,11%,and 11%in the respective engineered strains TLPH08-2,TLPH09,TLPH10,and TLPH12.Co-overexpression of metK_(cs)and adoK_(cs)-metFcs as two transcripts increased tylosin production by 22%in the resultant strain TLPH11 compared to that in TL01.Furthermore,combinational overexpression of tlrC,metK_(cs),adoK_(cs)-metFcs,and tylR as four transcripts increased tylosin production by 23%(10.93g/L)in the resultant strain TLPH17 compared to that in TL01.However,a negligible additive effect was displayed upon combinational overexpression in TLPH17 as suggested by the limited increment of fermentation titer compared to that in TLPH08-2.Transcription analyses indicated that the expression of tlrC and three SAM biosynthetic genes in TLPH17 was considerably lower than that of TLPH08-2 and TLPH11.Based on this observation,the five genes were rearranged into one or two operons to coordinate their overexpression,yielding two engineered strains TLPH23 and TLPH24,and leading to further enhancement of tylosin production over TLPH17.In particular,the production of TLPH23 reached 11.35 g/L.These findings indicated that the combinatorial strategy is a promising approach for enhancing tylosin production in high-yielding industrial strains.Penghui Dai Yuyao Qin Luyuan Li Haidi Li Lihuo Lv Danying Xu Yuqing Song Tingting Huang Shuangjun Lin Zixin Deng Meifeng Tao 2023Synthetic and Systems Biotechnology2023,8,3:0
3Influence and evaluation of saturated soil shield construction on existing highway显示文摘In order to study the shield construction under an existing highway,the initial displacement and the excess pore water pressure solution from Biot consolidation equation were used to derive the analytical solutions of the vertical deformation and pore water pressure of saturated soil caused by the frontal friction and side friction of the cutterhead.In addition,by introducing the layered method and combining it with other theoretical analytical equations,the expressions of total vertical deformation and total pore water pressure caused by tunnel excavation with different overlying materials were obtained.The simulation results on an engineering project showed that the angleαbetween the direction of the road and the propulsion axis of the shield had a significant influence on the surface settlement of the road.When the angleαwas increased,the settlement curve had higher variation;but the variation would not exceed the maximum settlement value above the shield axis.When the road was perpendicular to the shield axis,a critical point of the road bulging and settlement was formed above the incision.Due to the grinding resistance of the cutterhead,the pore water pressure under the roadbed was distributed asymmetrically on both sides of the shield axis.Liuqi Xu Chongfu Wu Liwei Wang Shuangjun Xu 2021Transportation Safety and Environment2021,3,1:0
