First observation of tropospheric nitrogen dioxide from the Environmental Trace Gases Monitoring Instrument onboard the GaoFen-5 satellite显示文摘The Environmental Trace Gases Monitoring Instrument(EMI)is the first Chinese satellite-borne UV–Vis spectrometer aiming to measure the distribution of atmospheric trace gases on a global scale. The EMI instrument onboard the GaoFen-5 satellite was launched on 9 May 2018.In this paper,we present the tropospheric nitrogen dioxide(NO2)vertical column density(VCD)retrieval algorithm dedicated to EMI measurement. We report the first successful retrieval of tropospheric NO_(2) VCD from the EMI instrument. Our retrieval improved the original EMI NO_(2) prototype algorithm by modifying the settings of the spectral fit and air mass factor calculations to account for the on-orbit instrumental performance changes. The retrieved EMI NO_(2) VCDs generally show good spatiotemporal agreement with the satellite-borne Ozone Monitoring Instrument and TROPOspheric Monitoring Instrument(correlation coefficient R of ~0.9,bias<50%).A comparison with ground-based MAX-DOAS(Multi-Axis Differential Optical Absorption Spectroscopy)observations also shows good correlation with an R of 0.82.The results indicate that the EMI NO_(2) retrieval algorithm derives reliable and precise results, and this algorithm can feasibly produce stable operational products that can contribute to global air pollution monitoring.Chengxin Zhang Cheng Liu Ka Lok Chan Qihou Hu Haoran Liu Bo Li Chengzhi Xing Wei Tan Haijin Zhou Fuqi Si Jianguo Liu 2020Light(Science & Applications)2020,9,1:11

Cell cycle-related kinase reprograms the liver immune microenvironment to promote cancer metastasis显示文摘The liver is an immunologically tolerant organ and a common metastatic site of multiple cancer types.Although a role for cancer cell invasion programs has been well characterized, whether and how liver-intrinsic factors drive metastatic spread is incompletely understood. Here,we show that aberrantly activated hepatocyte-intrinsic cell cycle-related kinase(CCRK)signaling in chronic liver diseases is critical for cancer metastasis by reprogramming an immunosuppressive microenvironment.Using an inducible liverspecific transgenic model,we found that CCRK overexpression dramatically increased both B16F10 melanoma and MC38 colorectal cancer(CRC)metastasis to the liver, which was highly infiltrated by polymorphonuclear-myeloid-derived suppressor cells(PMNMDSCs)and lacking natural killer T(NKT)cells.Depletion of PMN-MDSCs in CCRK transgenic mice restored NKT cell levels and their interferon gamma production and reduced liver metastasis to 2.7% and 0.7%(metastatic tumor weights)in the melanoma and CRC models, respectively.Mechanistically,CCRK activated nuclear factor-kappa B(NF-κB)signaling to increase the PMN-MDSC trafficking chemokine C-X-C motif ligand 1(CXCL1),which was positively correlated with liver-infiltrating PMN-MDSC levels in CCRK transgenic mice.Accordingly,CRC liver metastasis patients exhibited hyperaaivation of hepatic CCRK/NF-κB/CXCL1 signaling, which was associated with accumulation of PMN-MDSCs and paucity of NKT cells compared to healthy liver transplantation donors. In summary, this study demonstrates that immunosuppressive reprogramming by hepatic CCRK signaling undermines antimetastatic immunosurveillance. Our findings offer new mechanistic insights and therapeutic targets for liver metastasis intervention.Xuezhen Zeng Jingying Zhou Zhewen Xiong Hanyong Sun Weiqin Yang Myth T.S.Mok Jing Wang Jingqing Li Man Liu Wenshu Tang Yu Feng Hector Kwong-Sang W ang Shun-Wa Tsang King-Lau Chow Philip Chun Yeung John Wong Paul Bo-San Lai Anthony Wing-Hung Chan Ka Fai To Stephen Lam Chan Qiang Xia Jing Xue Xiao Chen Jun Yu Sui Peng Joseph Jao-Yiu Sung Ming Kuang Alfred Sze-Lok Cheng 2021Cellular & Molecular Immunology2021,18,4:5
Oral microbiome and pancreatic cancer显示文摘BACKGROUND Microbiota profiles differ between patients with pancreatic cancer and healthy people, and understanding these differences may help in early detection of pancreatic cancer.Saliva sampling is an easy and cost-effective way to determine microbiota profiles compared to fecal and tissue sample collection.AIM To investigate the saliva microbiome distribution in patients with pancreatic adenocarcinoma(PDAC)and the role of oral microbiota profiles in detection and risk prediction of pancreatic cancer.METHODS We conducted a prospective study of patients with pancreatic cancer(n=41)and healthy individuals(n=69).Bacterial taxa were identified by 16S ribosomal ribonucleic acid gene sequencing, and a linear discriminant analysis effect size algorithm was used to identify differences in taxa.Operational taxonomic unit values of all selected taxa were converted into a normalized Z-score,and logistic regressions were used to calculate risk prediction of pancreatic cancer.RESULTS Compared with the healthy control group, carriage of Streptococcus and Leptotrichina(z-score)was associated with a higher risk of PDAC[odds ratio(OR)=5.344,95%confidence interval(CI):1.282-22.282,P=0.021 and OR=6.886,95%CI:1.423-33.337,P=0.016,respectively].Veillonella and Neisseria(z-score)were considered a protective microbe that decreased the risk of PDAC(OR=0.187,95%CI:0.055-0.631,P=0.007 and OR=0.309,95%CI:0.100-0.952,P=0.041,respectively).Among the patients with PDAC,patients reporting bloating have a higher abundance of Porphyromonas(P=0.039),Fusobacterium(P=0.024),and Alloprevotella(P=0.041);while patients reporting jaundice had a higher amount of Prevotella(P=0.008);patients reporting dark brown urine had a higher amount of Veillonella(P=0.035).Patients reporting diarrhea had a lower amount of Neisseria and Campylobacter(P=0.024 and P=0.034),and patients reporting vomiting had decreased Alloprevotella(P=0.036).CONCLUSION Saliva microbiome was able to distinguish patients with pancreatic cancer and healthy individuals.Leptotrichia may be specific for patients living in Sichuan Province, southwest China.Symptomatic patients had different bacteria profiles than asymptomatic patients.Combined symptom and microbiome evaluation may help in the early detection of pancreatic cancer.Ai-Lin Wei Mao Li Guo-Qing Li Xuan Wang Wei-Ming Hu Zhen-Lu Li Jue Yuan Hong-Ying Liu Li-Li Zhou Ka Li Ang Li Mei Rosemary Fu 2020World Journal of Gastroenterology2020,26,48:4
Basal internode elongation of rice as affected by light intensity and leaf area显示文摘Short basal internodes are important for lodging resistance of rice(Oryza sativa L.).Several canopy indices affect the elongation of basal internodes, but uncertainty as to the key factors determining elongation of basal internodes persists. The objectives of this study were(1)to identify key factors affecting the elongation of basal internodes and(2)to establish a quantitative relationship between basal internode length and canopy indices. An inbred rice cultivar,Yinjingruanzhan, was grown in two split-plot field experiments with three N rates(0,75,and 150 kg N ha−1 in early season and 0,90,and 180 kg N ha−1 in late season)as main plots, three seedling densities(16.7,75.0,and 187.5 seedlings m−2)as subplots, and three replications in the 2015 early and late seasons in Guangzhou,China.Light intensity at base of canopy(Lb),light quality as determined from red/far-red light ratio(R/FR),light transmission ratio(LTR),leaf area index(LAI),leaf N concentration(NLV)and final length of second internode(counted from soil surface upward)(FIL)were recorded.Higher N rate and seedling density resulted in significantly longer FIL.FIL was negatively correlated with Lb,LTR,and R/FR(P<0.01)and positively correlated with LAI(P<0.01),but not correlated with NLV(P>0.05).Stepwise linear regression analysis showed that FIL was strongly associated with Lb and LAI(R2=0.82).Heavy N application to pot-grown rice at the beginning of first internode elongation did not change FIL.We conclude that FIL is determined mainly by Lb and LAI at jointing stage.NLV has no direct effect on the elongation of basal internodes.N application indirectly affects FIL by changing LAI and light conditions in the rice canopy.Reducing LAI and improving canopy light transmission at jointing stage can shorten the basal internodes and increase the lodging resistance of rice.Xuhua Zhong Kaiming Liang Bilin Peng Ka Tian Xiaojuan Li Nongrong Huang Yanzhuo Liu Junfeng Pan 2020The Crop Journal2020,8,1:3
Aberrant cholesterol metabolic signaling impairs antitumor immunosurveillance through natural killer T cell dysfunction in obese liver显示文摘Obesity is a major risk factor for cancers including hepatocellular carcinoma(HCC)that develops from a background of non-alcoholic fatty liver disease(NAFLD).Hypercholesterolemia is a common comorbidity of obesity.Although cholesterol biosynthesis mainly occurs in the liver, its role in HCC development of obese people remains obscure.Using high-fat high-carbohydrate diet-associated orthotopic and spontaneous NAFLD-HCC mouse models,we found that hepatic cholesterol accumulation in obesity selectively suppressed natural killer T(NKT)cell-mediated antitumor immunosurveillance.Transcriptome analysis of human liver revealed aberrant cholesterol metabolism and NKT cell dysfunction in NAFLD patients.Notably,cholesterol-lowering rosuvastatin restored NKT expansion and cytotoxicity to prevent obesogenic diet-promoted HCC development. Moreover, suppression of hepatic cholesterol biosynthesis by a mammalian target of rapamycin(mTOR)inhibitor vistusertib preceded tumor regression, which was abolished by NKT inactivation but not CD8^(+)T cell depletion.Mechanistically, sterol regulatory element-binding protein 2(SREBP2)-driven excessive cholesterol production from hepatocytes induced lipid peroxide accumulation and deficient cytotoxicity in NKT cells, which were supported by findings in people with obesity,NAFLD and NAFLD-HCC.This study highlights mTORC1/SREBP2/cholesterol-mediated NKT dysfunction in the tumor-promoting NAFLD liver microenvironment, providing intervention strategies that invigorating NKT cells to control HCC in the obesity epidemic.Wenshu Tang Jingying Zhou Weiqin Yang Yu Feng Haoran Wu Myth T.S.Mok Lingyun Zhang Zhixian Liang Xiaoyu Liu Zhewen Xiong Xuezhen Zeng Jing Wang Jiahuan Lu Jingqing Li Hanyong Sun Xiaoyu Tian Philip Chun Yeung Yong Hou Heung Man Lee Candice C.H.Lam Howard H.W.Leung Anthony W.H.Chan Ka Fai To John Wong Paul B.S.Lai Kelvin K.C.Ng Simon K.H.Wong Vincent W.S.Wong Alice P.S.Kong Joseph J.Y.Sung Alfred S.L.Cheng 2022Cellular & Molecular Immunology2022,19,7:3

Identification of Sheep Ovary Genes Potentially Associated with Off-season Reproduction显示文摘Off-season reproduction is a favorable economic trait for sheep industry.Hu sheep,an indigenous Chinese sheep breed, demonstrates a higher productivity of lambs and displays year-around oestrous behavior under proper nutrition and environment. The genetic basis behind these traits, however,is not well understood. In order to identify genes associated with the off-season reproduction,we constructed a suppression subtractive hybridization(SSH) cDNA library using pooled ovary mRNAs of 6 oestrous Hu females as a tester and the pooled ovary mRNAs of 6 non-oestrous Chinese Merino females as a driver.A total of 382 resulting positive clones were obtained after the SSH.We identified 114 differentially up-regulated genes in oestrous Hu sheep by using subsequent screening and DNA sequencing,of which 8 were previously known,93 were reported for the first time in sheep, and 13 were novel with no significant homology to any sequence in the DNA databases.Functions of the genes identified are related to cell division, signal transduction, structure, metabolism,or cell defense. To validate the results of SSH,6 genes(Ntrk2,Ppap2b,Htra1,Nid1,Serpine2 and Foxola) were selected for conformational analysis using quantitative real-time PCR(qRT-PCR),and two of them(Htral and Foxola) were verified by Northern blot. All of the 6 genes were differentially up-regulated in the ovary of oestrous Hu.It is obvious that off-season reproduction is a complex trait involving multiple genes in multiple organs. This study helps to provide a foundation for the final identification of functional genes involved in the sheep ovary.Lei Chen Ka Liu Zongsheng Zhao Hugh T.Blair Peng Zhang Daquan Li Runlin Z.Ma 2012Journal of Genetics and Genomics2012,39,4:3

A rare case of Staphylococcus lugdunensis septicemia associated with myocarditis and atrioventricular block显示文摘Myocarditis is a relatively rare, possibly life-threatening disease characterized by the inflammation of the myocardium.111 The disease pathogenesis is primarily initiated by acute injury and necrosis of cardiomyocytes, leading to an inflammatory response mediated by the immune system that can potentially cause further aggravation of myocardial damage and organ dysfunction.Prognosis in patients with myocarditis depends on the clinical presentation, which ranges from an asymptomatic disease course to the concomitant development of cardiac arrhythmias, heart failure, cardiogenic shock and even the occurrence of death in extreme cases[1].Alex Tsz Lai Nganh Sharen Lee Tong LIU Mark Tam Ka Hou Christien Li Michelle Vangi Wong Michael Huen Sum Lam Gary Tse Ishan Lakhani 2019Journal of Geriatric Cardiology2019,16,1:2
