| 3 | Application of computational fluid dynamic to model the hydraulic performance of subsurface flow wetlands显示文摘A subsurface flow wetland(SSFW)was simulated using a commercial computational fluid dynamic(CFD)code.The constructed media was simulated using porous media and the liquid resident time distribution(RTD)in the SSFW was obtained using the particle trajectory model.The effect of wetland configuration and operating conditions on the hydraulic performance of the SSFW were investigated.The results indicated that the hydraulic performance of the SSFW was predominantly affected by the wetland configuration.The hydraulic effciency of the SSFW with an inlet at the middle edge of the upper media was 0.584 and the best among the SSFWs with an inlet at the top,the middle,and the bottom edge of the upper media.The constructed media affected the hydraulic performance by the ratio(K)of the upper and lower media resistance.The selection of appropriate media resistance in the protection layer can improve the hydraulic effciency.When the viscous resistance coeffcient of the media in the protection layer changed from 2.315×105 to 1.200×108,the hydraulic effciency of the SSFW increased from 0.301 to 0.751.However,the effect of operating conditions on the hydraulic effciency of the SSFW was slight. | Liwei FAN Hai Reti Wenxing WANG Zexiang LU Zhiming YANG | 2008 | Journal of Environmental Sciences2008,20,12: | 17 |
| 4 | Effect of Mach number on transonic flow past a circular cylinder显示文摘The effect of Mach number on transonic flow past a circular cylinder is investigated numerically for the free-stream Mach number M∞ from 0.85 to 0.98 and the Reynolds number 2×105 based on the diameter of the cylinder. The work provides an insight into several salient features, including unsteady and quasi-steady flow state, formation of local supersonic zone, and evolution of turbulent shear layer. Results show that there exist two flow states dependent of a critical Mach number Mcr around 0.9. One is an unsteady flow state characterized by moving shock waves interacting with the turbulent flow in the near region of the cylinder for M∞Mcr, suppressing vortex shedding from the cylin-der. Some flow behaviors in the unsteady and quasi-steady flow states are revealed. From time evolu-tion of flow structures, local supersonic zones are identified in the wake and generated by two typical processes, i.e. reverse flow behind the cylinder and shed vortices in the near wake. | 5 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. | 6 | Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. | 7 | IL-10-producing regulatory B cells restrain the T follicular helper cell response in primary Sjögren’s syndrome显示文摘Increased numbers of T follicular helper(Tfh)cells have been implicated in the development of autoimmune diseases including primary Sjögren’s syndrome(pSS),but how the Tfh cell response is regulated during autoimmune pathogenesis remains largely unclear.Here,we first found negative correlations between IL-10^(+)regulatory B(Breg)cell numbers and Tfh cell responses and disease activity in patients with pSS and mice with experimental Sjögren’s syndrome(ESS).Moreover,we detected high expression of IL-10 receptor on Tfh cells and their precursors in both humans and mice.In culture,IL-10 suppressed human and murine Tfh cell differentiation by promoting STAT5 phosphorylation.By using an adoptive transfer approach and two-photon live imaging,we found significantly increased numbers of Tfh cells with enhanced T cell homing into B cell follicles in the draining cervical lymph nodes of RAG-2−/−mice transferred with IL-10-deficient B cells during ESS development compared with those of RAG-2−/−mice transferred with wild-type B cells.In ESS mice,CD19^(+)CD1d^(hi)CD5^(+)Breg cells with decreased IL-10 production exhibited severely impaired suppressive effects on T cell proliferation.Consistently,CD19^(+)CD24^(+)CD38^(hi) Breg cells from pSS patients showed significantly reduced IL-10 production with defective inhibitory function in the suppression of autologous Tfh cell expansion.Furthermore,the adoptive transfer of IL-10-producing Breg cells markedly suppressed the Tfh cell response and ameliorated ESS progression in ESS mice.Together,these findings demonstrate a critical role for IL-10-producing Breg cells in restraining the effector Tfh cell response during pSS development. | Xiang Lin Xiaohui Wang Fan Xiao Kongyang Ma Lixiong Liu Xiaoqi Wang Dong Xu Fei Wang Xiaofei Shi Dongzhou Liu Yan Zhao Liwei Lu | 2019 | Cellular & Molecular Immunology2019,16,12: | 13 |

| 9 | Olfactory ecto-mesenchymal stem cell-derived exosomes ameliorate murine Sjögren’s syndrome by modulating the function of myeloid-derived suppressor cells显示文摘Sjögren’s syndrome(SS)is a systemic autoimmune disease characterized by progressive inflammation and tissue damage in salivary glands and lacrimal glands.Our previous studies showed that myeloid-derived suppressor cells(MDSCs)exhibited impaired immunosuppressive function during disease progression in patients with SS and mice with experimental Sjögren’s syndrome(ESS),but it remains unclear whether restoring the function of MDSCs can effectively ameliorate the development of ESS.In this study,we found that murine olfactory ecto-mesenchymal stem cell-derived exosomes(OE-MSC-Exos)significantly enhanced the suppressive function of MDSCs by upregulating arginase expression and increasing ROS and NO levels.Moreover,treatment with OE-MSC-Exos via intravenous injection markedly attenuated disease progression and restored MDSC function in ESS mice.Mechanistically,OE-MSC-Exo-secreted IL-6 activated the Jak2/Stat3 pathway in MDSCs.In addition,the abundant S100A4 in OE-MSC-Exos acted as a key factor in mediating the endogenous production of IL-6 by MDSCs via TLR4 signaling,indicating an autocrine pathway of MDSC functional modulation by IL-6.Taken together,our results demonstrated that OE-MSC-Exos possess therapeutic potential to attenuate ESS progression by enhancing the immunosuppressive function of MDSCs,possibly constituting a new strategy for the treatment of Sjögren’s syndrome and other autoimmune diseases. | Ke Rui Yue Hong Qiugang Zhu Xiaofei Shi Fan Xiao Hailong Fu Qing Yin Yida Xing Xinfeng Wu Xiaodan Kong Huaxi Xu Jie Tian Shengjun Wang Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,2: | 12 |
| 10 | Diagnostic value of 5 serum biomarkers for hepatocellular carcinoma with different epidemiological backgrounds:A large-scale,retrospective study显示文摘Objective:Hepatocellular carcinoma(HCC)is a lethal global disease that requires an accurate diagnosis.We assessed the potential of 5 serum biomarkers(AFP,AFU,GGT-II,GPC3,and HGF)in the diagnosis of HCC.Methods:In this retrospective study,we measured the serum levels of each biomarker using ELISAs in 921 participants,including 298 patients with HCC,154 patients with chronic hepatitis(CH),122 patients with liver cirrhosis(LC),and 347 healthy controls from 3 hospitals.Patients negative for hepatitis B surface antigen and hepatitis C antibody(called'NBNC-HCC')and patients positive for the above indices(called'HBV-HCC and HCV-HCC')were enrolled.The selected diagnostic model was constructed using a training cohort(n=468),and a validation cohort(n=453)was used to validate our results.Receiver operating characteristic analysis was used to evaluate the diagnostic accuracy.Results:Theα-L-fucosidase(AFU)/α-fetoprotein(AFP)combination was best able to distinguish NBNC-HCC[area under the curve:0.986(95%confidence interval:0.958–0.997),sensitivity:92.6%,specificity:98.9%]from healthy controls in the test cohort.For screening populations at risk of developing HCC(CH and LC),the AFP/AFU combination improved the diagnostic specificity for early-stage HCC[area under the curve:0.776(0.712–0.831),sensitivity:52.5%,specificity:91.6%in the test group].In all-stage HBV-HCC and HCV-HCC,AFU was also the best candidate biomarker combined with AFP[area under the curve:0.835(0.784–0.877),sensitivity 69.1%,specificity:87.4%in the test group].All results were verified in the validation group.Conclusions:The AFP/AFU combination could be used to identify NBNC-HCC from healthy controls and hepatitis-related HCC from at-risk patients. | Dongming Liu Yi Luo Lu Chen Liwei Chen Duo Zuo Yueguo Li Xiaofang Zhang Jing Wu Qing Xi Guangtao Li Lisha Qi Xiaofen Yue Xiehua Zhang Zhuoyu Sun Ning Zhang Tianqiang Song Wei Lu Hua Guo | 2021 | Cancer Biology & Medicine2021,18,1: | 10 |

In the peripheral lymphoid system, mature B cells are activated by self or/and foreign antigens andsignals from helper T cells for differentiating into either memory B cells or antibody-producing plasma cells.Accumulating evidence has shown that epigenetic regulations modulate somatic hypermutation and class switchDNA recombination during B-cell activation and differentiation. Any abnormalities in these complex regulatoryprocesses may contribute to aberrant antibody production, resulting in autoimmune pathogenesis such as systemiclupus erythematosus. Newly generated knowledge from advanced modern technologies such as next-generationsequencing, single-cell sequencing and DNA methylation sequencing has enabled us to better understand B-cellbiology and its role in autoimmune development. | 14 | IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE. | Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,7: | 6 |
| 15 | Recent advances in biological detection with magnetic nanoparticles as a useful tool显示文摘Magnetic nanoparticles have emerged as a powerful tool for magnetic resonance imaging, biodetection, drug delivery, and hyperthermia. This review focuses on the biological detection of magnetic nanoparticles as well as their physicochemical properties. Substantial progress in the sensitivity of detection has been made by developing variety of methods. Five applications of magnetic nanoparticles in biological detection are discussed in this review: magnetic separation, magnetic sensing, magnetic manipulation, magnetic catalysis, and signal enhancer for surface plasmon resonance(SPR). | 16 | Proteasome inhibition suppresses Th17 cell generation and ameliorates autoimmune development in experimental Sjögren’s syndrome显示文摘Immunoproteasome activation in immune cells is involved in the modulation of immune responses.Increasing evidence indicates that proteasome inhibitors show beneficial effects in treating autoimmune diseases,but it remains unclear whether proteasome inhibition is an effective approach for suppressing autoimmune development in Sjögren’s syndrome(SS).Our previous work has demonstrated a critical role for Th17 cells in the development of experimental SS(ESS)in mice.In this study,we detected high levels of low-molecular-weight protein 7(LMP7),a key subunit of the immunoproteasome,in Th17 cells from ESS mice.Moreover,treatment with bortezomib(BTZ),a proteasome inhibitor,markedly suppressed Th17 differentiation in both murine and human naive T cells in culture.Furthermore,ESS mice treated with BTZ displayed significantly higher saliva flow rates and a reduction in tissue destruction in the salivary glands compared with vehicle-treated ESS mice.Notably,BTZ-treated ESS mice showed markedly decreased Th17 cells,germinal center B cells and plasma cells in the peripheral lymphoid organs.In addition,adoptively transferred wild type naive CD4+T cells rapidly differentiated into Th17 cells and induced salivary dysfunction in IL-17-deficient mice immunized for ESS induction.However,BTZ treatment profoundly suppressed the donor T-cell-derived Th17 response and ameliorated the reduction in salivary secretion in IL-17-deficient recipient mice.Taken together,our findings demonstrate that proteasome inhibition can effectively ameliorate ESS by suppressing the Th17 response,which may contribute to the development of a novel therapeutic strategy for the treatment of SS. | Fan Xiao Xiang Lin Jie Tian Xiaohui Wang Qian Chen Ke Rui Jie Ma Shengjun Wang Qingwen Wang Xiaoqi Wang Dongzhou Liu Lingyun Sun Liwei Lu | 2017 | Cellular & Molecular Immunology2017,14,11: | 6 |

| 18 | Hot deformation characterization of as-homogenized Al-Cu-Li X2A66 alloy through processing maps and microstructural evolution显示文摘Uniaxial compression tests were carried out on an Al-Cu-Li alloy at a temperature range of 300–500℃ and a strain rate range of 0.001–10 s^-1. Four representative instability maps based on Gegel, AlexanderMalas(A-M), Kumar-Prasad(K-P) and Murty-Rao(M-R) criteria were constructed. Through formula deduction and microstructural observation, it can be concluded that M-R criterion is more accurate than K-P criterion, and the first two criteria are better than Gegel and A-M criteria. From a power dissipation map and a M-R instability map, the optimized processing parameters are 480–500℃/0.001–0.1 s^-1 and 420–480℃/0.1-1 s^-1. The corresponding microstructural analysis shows that dynamic recovery and partial dynamic recrystallization are main dynamic softening mechanisms. Transmission electron microscopy observation indicated that a large number of primary coarse T1(Al2 Cu Li) particles precipitated in the homogenized specimen.
