| 1 | Magnetogenetics: remote non-invasive magnetic activation of neuronal activity with a magnetoreceptor显示文摘Current neuromodulation techniques such as optogenetics and deep-brain stimulation are transforming basic and translational neuroscience. These two neuromodulation approaches are, however, invasive since surgical implantation of an optical fiber or wire electrode is required. Here, we have invented a non-invasive magnetogenetics that combines the genetic targeting of a magnetoreceptor with remote magnetic stimulation. The noninvasive activation of neurons was achieved by neuronal expression of an exogenous magnetoreceptor, an iron-sulfur cluster assembly protein 1(Isca1). In HEK-293 cells and cultured hippocampal neurons expressing this magnetoreceptor, application of an external magnetic field resulted in membrane depolarization and calcium influx in a reproducible and reversible manner, as indicated by the ultrasensitive fluorescent calcium indicator GCa MP6 s.Moreover, the magnetogenetic control of neuronal activity might be dependent on the direction of the magnetic field and exhibits on-response and off-response patterns for the external magnetic field applied. The activation of this magnetoreceptor can depolarize neurons and elicit trains of action potentials, which can be triggered repetitively with a remote magnetic field in whole-cell patch-clamp recording.In transgenic Caenorhabditis elegans expressing this magnetoreceptor in myo-3-specific muscle cells or mec-4-specific neurons, application of the external magnetic field triggered muscle contraction and withdrawal behavior of the worms, indicative of magnet-dependent activation of muscle cells and touch receptor neurons, respectively. The advantages of magnetogenetics over optogenetics are its exclusive non-invasive, deep penetration, long-term continuous dosing, unlimited accessibility, spatial uniformity and relative safety. Like optogenetics that has gone through decade-long improvements, magnetogenetics, with continuous modification and maturation, will reshape the current landscape of neuromodulation toolboxes and will have a broad range of applications to basic and translational neuroscience as well as other biological sciences. | 4 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. In the peripheral lymphoid system, mature B cells are activated by self or/and foreign antigens andsignals from helper T cells for differentiating into either memory B cells or antibody-producing plasma cells.Accumulating evidence has shown that epigenetic regulations modulate somatic hypermutation and class switchDNA recombination during B-cell activation and differentiation. Any abnormalities in these complex regulatoryprocesses may contribute to aberrant antibody production, resulting in autoimmune pathogenesis such as systemiclupus erythematosus. Newly generated knowledge from advanced modern technologies such as next-generationsequencing, single-cell sequencing and DNA methylation sequencing has enabled us to better understand B-cellbiology and its role in autoimmune development. | 6 | A new method to predict fatigue crack growth rate of materials based on average cyclic plasticity strain damage accumulation显示文摘By introducing a fatigue blunting factor, the cyclic elasto-plastic Hutchinson-Rice-Rosengren (HRR) field near the crack tip under the cyclic loading is modified. And, an average damage per loading-cycle in the cyclic plastic deformation region is defined due to Manson-Coffin law. Then, according to the linear damage accumulation theory-Miner law, a new model for predicting the fatigue crack growth (FCG) of the opening mode crack based on the low cycle fatigue (LCF) damage is set up. The step length of crack propagation is assumed to be the size of cyclic plastic zone. It is clear that every parameter of the new model has clearly physical meaning which does not need any human debugging. Based on the LCF test data, the FCG predictions given by the new model are consistent with the FCG test results of Cr2Ni2MoV and X12CrMoWVNbN 10-1-1. | 7 | Cystathionine-γ-lyase ameliorates the histone demethylase JMJD3-mediated autoimmune response in rheumatoid arthritis显示文摘Cystathionine-γ-lyase(CSE),an enzyme associated with hydrogen sulfide(H2S)production,is an important endogenous regulator of inflammation.Jumonji domain-containing protein 3(JMJD3)is implicated in the immune response and inflammation.Here,we investigated the potential contribution of JMJD3 to endogenous CSE-mediated inflammation in rheumatoid arthritis(RA).Upregulated CSE and JMJD3 were identified in synovial fibroblasts(SFs)from RA patients as well as in the joints of arthritic mice.Knocking down CSE augmented inflammation in IL-1β-induced SFs by increasing JMJD3 expression.In addition,CSE−/−mice with collagen-induced arthritis(CIA)developed severe joint inflammation and bone erosion.Conversely,overexpressing CSE inhibited JMJD3 expression by the transcription factor Sp-1 and was accompanied by reduced inflammation in IL-1β-treated SFs.Furthermore,JMJD3 silencing or the administration of the JMJD3 inhibitor GSK-J4 significantly decreased the inflammatory response in IL-1β-treated SFs,mainly by controlling the methylation status of H3K27me3 at the promoter of its target genes.GSK-J4 markedly attenuated the severity of arthritis in CIA mice.In conclusion,suppressing JMJD3 expression by the transcription factor Sp-1 is likely responsible for the ability of CSE to negatively modulate the inflammatory response and reduce the progression of RA. | Weijun Wu Ming Qin Wanwan Jia Zheng Huang Zhongzheng Li Di Yang Mengwei Huang Chenxi Xiao Fen Long Jianchun Mao Philip K.Moore Xinhua Liu Yi Zhun Zhu | 2019 | Cellular & Molecular Immunology2019,16,8: | 9 |
| 8 | Impact of an innovative tuberculosis financing and payment model on health service utilization by tuberculosis patients in China:do the poor fare better than the rich?显示文摘Background:Tuberculosis(TB)prevalence is closely associated with poverty in China,and poor patients face more barriers to treatment.Using an insurance-based approach,the China-Gates TB program Phase II was implemented between 2012 and 2014 in three cities in China to improve access to TB care and reduce the financial burden on patients,particularly among the poor.This study aims to assess the program effects on service use,and its equity impact across different income groups.Methods:Data from 788 and 775 patients at baseline and final evaluation were available for analysis respectively.Inpatient and outpatient service utilization,treatment adherence,and patient satisfaction were assessed before and after the program,across different income groups(extreme poverty,moderate poverty and non-poverty),and in various program cities,using descriptive statistics and multi-variate regression models.Key stakeholder interviews were conducted to qualitatively evaluate program implementation and impacts.Results:After program implementation,the hospital admission rate increased more for the extreme poverty group(48.5 to 70.7%)and moderate poverty group(45.0 to 68.1%),compared to the non-poverty group(52.9 to 643%).The largest increase in the number of outpatient visits was also for the extreme poverty group(4.6 to 5.7).The proportion of patients with good medication adherence increased by 15 percentage points in the extreme poverty group and by ten percentage points in the other groups.Satisfaction rates were high in all groups.Qualitative feedback from stakeholders also suggested that increased reimbursement rates,easier reimbursement procedures,and allowance improved patients'service utilization.Implementation of case-based payment made service provision more compliant to clinical pathways.Conclusion:Patients in extreme or moderate poverty benefited more from the program compared to a non-poverty group,indicating improved equity in TB service access.The pro-poor design of the program provides important丨essons to other TB programs in China and other countries to better address TB care for the poor. | Di Dong Wei-Xi Jiang Qian Long Fei Huang Hui Zhang Jia-Ying Chen Li Xiang Qiang Li Sheng-Lan Tang Henry Lucas | 2019 | Infectious Diseases of Poverty2019,8,3: | 8 |

| 10 | Effect of continuous negative pressure water supply on the growth, development and physiological mechanism of Capsicum annuum L.显示文摘Effects of continuous negative pressure water supply on water consumption, growth and development, as well as physiological mechanism and quality of Capsicum annuum L. were investigated in this paper. Meanwhile, the optimal negative pressure water supply conditions for growth of C. annuum L. were screened out to achieve the goals of water conservation, high yield and high quality, thus providing theoretical foundation for its field production. The pot experiment within the greenhouse was utilized; the continuous negative pressure water supply was adopted; the four treatments, artificial watering(CK), –5 k Pa(T1), –10 k Pa(T2), and –15 k Pa(T3) were set; and the daily water consumption, yield, as well as the biomass, nitrate reductase, root activity, vitamin C, capsaicin, and nutrient uptakes of nitrogen(N), phosphorus(P) and potassium(K) during various stages of its growth were determined. Compared with CK, when the water supply pressure was controlled at –5 to –15 k Pa in the experiment, the total water consumption of C. annuum L. reduced by 53.42 to 67.75%, the total water consumption intensity reduced by 54.29 to 67.14%, and the water use efficiency increased by 12.66 to 124.67%. The N accumulation in a single strain of C. annuum L. from the color turning stage to the red ripe stage increased by 15.99 to 100.55%, respectively, compared with that of CK; the P accumulation increased by 20.47 to 154.00% relative to that of CK, and the K accumulation increased by 64.92 to 144.9% compared with that of CK. Compared with CK, C. annuum L. yield was remarkably improved by 13.79% at T1, and contents of vitamin C, capsaicin as well as carotenoids at all growth stages were enhanced by 13.42–147.01%, 11.54–71.01%, and 41.1–568.06%, respectively. Nitrate reductase activity, root activity and chlorophyll(a+b) were markedly increased by 335.78–500%, 79.6–140.68% and 114.95–676.19%, respectively, from immature stage to full ripe stage. | 12 | IFN-γ increases efficiency of DNA vaccine in protecting ducks against infection显示文摘AIM: To detect the effects of DNA vaccines in combination with duck IFN-γ gene on the protection of ducks against duck hepatitis B virus (DHBV) infection.METHODS: DuIFN-γ cDNA was cloned and expressed in COS-7 cells, and the antiviral activity of DuIFN-γ was detected and neutralized by specific antibodies. Ducks were vaccinated with DHBpreS/S DNA alone or coimmunized with plasmid expressing DuIFN-γ. DuIFN-γmRNA in peripheral blood mononuclear cells (PBMCs) from immunized ducks was detected by semi-quantitative competitive RT-PCR. Anti-DHBpreS was titrated by enzyme-linked immunosorbent assay (ELISA). DHBV DNA in sera and liver was detected by Southern blot hybridization, after ducks were challenged with high doses of DHBV.RESULTS: DuIFN-γ expressed by COS-7 was able to protect duck fibroblasts against vesicular stomatitis virus (VSV) infection in a dose-dependent fashion, and antiDuIFN-γ antibodies neutralized the antiviral effects. DuIFN-γin the supernatant also inhibited the release of DHBV DNA from LMH-D2 cells. When ducks were co-immunized with DNA vaccine expressing DHBpreS/S and DuIFN-γ gene as an adjuvant, the level of DuIFN-γ mRNA in PBMCs was higher than that in ducks vaccinated with DHBpreS/S DNA alone. However, the titer of anti-DHBpreS elicited by DHBpreS/S DNA alone was higher than that co-immunized with DuIFN-γ gene and DHBpreS/S DNA. | 13 | Validation and application of water levels derived from Sentinel-3A for the Brahmaputra River显示文摘For the first time ever, this study aims at applying Sentinel-3 A to the Great Brahmaputra River(GBR) and validating water levels derived from this newly-launched altimetry satellite mission. The GBR is divided into three primary parts:(1) a large section of the Yarlung Zangbo River in China, also termed the Upper Brahmaputra River(UBR) in this study, featured by high elevation,complex terrain, narrow river widths(from less than 100 to 400 m), and limited in situ measurements;(2) the Middle Brahmaputra River(MBR) with widths varying from ~400 m to ~1 km;(3) the Lower Brahmaputra River(LBR), dominated by braided channels with wide river channels(up to several kilometers). For the altimetry data, both waveform retracking and hooking effect correction were applied to mitigate the influence caused by land contamination and to improve the accuracy of water level retrievals. Water levels were derived from 41 virtual stations(VSs) across the GBR and different retracking algorithms were compared with in situ data from two gauging stations in the UBR. Time series of altimetry-based water levels were categorized into three types based on the quality: no data(type 1), degraded(type 2), and good(type 3). Results showed that the VSs(type 1) only existed in the mountainous regions, accounting for ~ a half of the total in the UBR. Validation with the gauged water levels showed that the TIC algorithm performed best among all of the retrackers applied, followed by the Ice-1 algorithm. The standard deviation of the difference between the gauged and TIC-derived water levels ranged from 0.41 to 0.76 m among four different VSs(type 3). In addition, the quality of VSs in the LBR was best, followed by the MBR. | 14 | An improved modeling of precipitation phase and snow in the Lancang River Basin in Southwest China显示文摘Precipitation phase(e.g., rainfall and snowfall) and snow(e.g., snowpack and snowmelt runoff) in high-mountain regions may largely affect runoff generation, which is critical to water supply, hydropower generation, agricultural irrigation, and ecosystems downstream. Accurately modeling precipitation phase and snow is therefore fundamental to developing a better understanding of hydrological processes for high-mountain regions and their lower reaches. The Lancang River(LR, or the Upper Mekong River)in China, among the most important transboundary rivers originating from the Tibetan Plateau, features active dam construction and complex water resources allocation of various stakeholders in Southeast Asian countries under climate change. This study aims to improve precipitation phase and snow modeling for the LR basin with a hydrological model and multisource remotely sensed data. Results show that joint use of the Moderate Resolution Imaging Spectroradiometer(MODIS) land surface temperature product with high spatial resolution(1 km×1 km) and an air temperature product can more precisely distinguish precipitation phase than air and wet-bulb temperature products in the LR basin. Snowfall and snowmelt were found to be controlled primarily by rainfall and snowfall temperature thresholds in snow modeling. The rainfall and snowfall temperature thresholds derived from the hydrological model through calibration with remotely sensed snowpack at basin scales were considerably lower than those derived from in situ observations. Rainfall and snowfall temperature thresholds derived from in situ observations could lead to the overestimation of snowmelt runoff due mostly to the lack of representation of point-based measurements at basin scales. | 15 | Optimization of cantilevered piezoelectric energy harvester with a fixed resonance frequency显示文摘Piezoelectric energy harvesting is widely used to scavenge vibration energy in the environment.For some vibration sources with fixed frequency,cantilevered harvester can generate the energy effectively,so the optimization theory for cantilevered harvester in such an application is needed.In this article,we present the theoretical and experimental studies of the cantilevered piezoelectric energy harvester with a fixed resonance frequency.An analytical model based on energy method is used to estimate the open-circuit voltage and generated energy.Considering that the harvester may be subjected to the static force or steady-state sinusoidal vibration excitation,static and dynamic analysis is performed for device structure to achieve efficient energy.In the analysis,the effects of geometrical dimension on the energy harvesting performance are discussed comprehensively.Eventually,a prototype is designed and fabricated using(1-x)Pb(Mg1/3Nb2/3)O3-xPbTiO3(PMN-PT)single crystal with ultrahigh piezoelectric properties and coupling factor.Performances of the cantilever with different clamped length are evaluated under sinusoidal vibration excitation,proving the good consistency between experimental results and theoretical prediction.The established analysis can provide useful guidelines for the structure design of cantilevered piezoelectric energy harvester with a fixed resonance frequency. | LIANG Zhu XU ChunDong REN Bo DI WenNing LI Long LUO HaoSu CHEN Zhen SU Jun | 2014 | Science China(Technological Sciences)2014,57,6: | 6 |
| 16 | Impact of an innovative financing and payment model on tuberculosis patients'financial burden:is tuberculosis care more affordable for the poor?显示文摘Background:In response to the high financial burden of health services facing tuberculosis(TB)patients in China,the China-Gates TB project,PhaseⅡ,has implemented a new financing and payment model as an important component of the overall project in three cities in eastern,central and western China.The model focuses on increasing the reimbursement rate for TB patients and reforming provider payment methods by replacing fee-for-service with a case-based payment approach.This study investigated changes in out-of-pocket(OOP)health expenditure and the financial burden on TB patients before and after the interventions,with a focus on potential differential impacts on patients from different income groups.Methods:Three sample counties in each of the three prefectures:Zhenjiang,Yichang and Hanzhong were chosen as study sites.TB patients who started and completed treatment before,and during the intervention period,were randomly sampled and surveyed at the baseline in 2013 and final evaluation in 2015 respectively.OOP health expenditure and percentage of patients incurring catastrophic health expenditure(CHE)were calculated for different income groups.OLS regression and Iogit regression were conducted to explore the intervention's impacts on patient OOP health expenditure and financial burden after adjusting for other covariates.Key-informant interviews and focus group discussions were conducted to understand the reasons for any observed changes.Results:Data from 738(baseline)and 735(evaluation)patients were available for analysis.Patient mean OOP health expenditure increased from RMB 3576 to RMB 5791,and the percentage of patients incurring CHE also increased after intervention.The percentage increase in OOP health expenditure and the likelihood of incurring CHE were significantly lower for patients from the highest income group as compared to the lowest.Qualitative findings indicated that increased use of health services not covered by the standard package of the model was likely to have caused the increase in financial burden.Conclusions:The implementation of the new financing and payment model did not protect patients,especially those from the lowest income group,from financial difficulty,due partly to their increased use of health service.More financial resources should be mobilized to increase financial protection,particularly for poor patients,while cost containment strategies need to be developed and effectively implemented to improve the effective coverage of essential healthcare in China. | Wei-Xi Jiang Qian Long Henry Lucas Di Dong Jia-Ying Chen Li Xiang Qiang Li Fei Huang Hong Wang Chris Elbers Frank Cobelens Sheng-Lan Tang | 2019 | Infectious Diseases of Poverty2019,8,2: | 4 |
| 17 | Characteristic model based all-coefficient adaptive control of an AMB suspended energy storage flywheel test rig显示文摘Feedback control of active magnetic bearing(AMB) suspended energy storage flywheel systems is critical in the operation of the systems and has been well studied. Both the classical proportional-integralderivative(PID) control design method and modern control theory, such as H∞ control and μ-synthesis,have been explored. PID control is easy to implement but is not effective in handling complex rotordynamics. Modern control design methods usually require a plant model and an accurate characterization of the uncertainties. In each case, few experimental validation results on the closed-loop performance are available because of the costs and the technical challenges associated with the construction of experimental test rigs.In this paper, we apply the characteristic model based all-coefficient adaptive control(ACAC) design method for the stabilization of an AMB suspended flywheel test rig we recently constructed. | 18 | Clinical Analysis of 10 AIDS Patients with Malignant Lymphoma显示文摘Objective This work summarizes the clinical features and treatment of 10 AIDS patients with malignant lymphoma. Methods A total of 10 AIDS patients with malignant lymphoma seen in Beijing Ditan Hospital since 2009 were enrolled.Clinical manifestations,pathological examinations,immunity levels,Epstein-Barr virus antibody examinations,complications,treatments,and outcomes were retrospectively analyzed. Results The main clinical manifestations of these patients included intermittent fever in 2 cases,neck masses and fever in 3 ases,auxiliary lymph node enlargement in 2 cases,and abdominal pain and bloating with fever in 3 cases.Up to 7 patients were pathologically diagnosed with diffuse large B cell lymphoma(DLBCL),and 3 patients were pathologically diagnosed with Burkitt’s lymphoma.Up to 8 patients had CD4 cell counts below 200/uL,and 2 patients had a level of more than 200/μL.Up to 7 patients were negative for EBV-IgM antibodies and 3 patients were not examined.Six patients underwent different chemotherapy and their prognoses were different.One patient with Burkitt’s lymphoma alternatively took CODOXM and IVAC for 3 turns after VP chemotherapy;1 patient with liver metastasis took R-CHOP 5 times,then changed therapy regimen to R-MINE and MINE. One patient with adrenal DLBCL took CHOP 6 times.Three patients with DLBCL took CHOP 1 or 2 times.Four patients gave up treatment.Various infections and side effects occurred,including bone marrow suppression,gastrointestinal bleeding,and renal (?)stunction during chemotherapy.Six patients took HAART,and 4 did not.Six patients died,whereas 3 patients got improved;and 1 (?)atient was discharged. | 20 | BMI and BMI Changes to All-cause Mortality among the Elderly in Beijing: a 20-year Cohort Study显示文摘Objective To explore the association between body mass index(BMI)and all-cause mortality among the elderly in Beijing.Methods This analysis was based on the Beijing multidimensional longitudinal study of aging(BLSA),which included 2,090 subjects over 55 years old and was followed-up from 1992 to 2012.BMI-mortality curves were drawn to find the optimal BMI range with the lowest mortality.Cox proportional hazard models were used to obtain the hazard ratios(HRs)for BMI and BMI changes in the overall population and in specific stratified populations.Results During follow-up,1,164 deaths were recorded;BMI-mortality curve was U-shaped,with the lowest mortality at a BMI of approximately 25 kg/m^2.After adjusting for gender,age,smoking,drinking and some pre-existing diseases,HRs for underweight,overweight and obesity compared with normal weight were 1.372(95%CI:1.154-1.631),0.767(95%CI:0.666-0.884)and 0.871(95%CI:0.830-1.246),respectively.HR for BMI drop was 3.245(95%CI:0.824-12.772)in the underweight group and 1.892(95%CI:0.830-1.246)in the normal weight group,HR for BMI rise was 1.795(95%CI:1.243-2.591)in normal weight group and 1.962(95%CI:1.202-3.203)in the overweight group.Conclusion Keeping BMI in an overweight status and stable is related to a reduced mortality. | WANG Yun Feng TANG Zhe GUO Jin TAO Li Xin LIU Long LI Hai Bin LI Di Tian GUO Xiu Hua YANG Xing Hua | 2017 | Biomedical and Environmental Sciences2017,30,2: | 3 |
