1Component analysis of Chinese medicine and advances in fuming-washing therapy for knee osteoarthritis via unsupervised data mining methods显示文摘OBJECTIVE: To analyze the component law of Chinese medicines in fuming-washing therapy for knee osteoarthritis(KOA), and develop new fuming-washing prescriptions for KOA through unsupervised data mining methods.METHODS: Chinese medicine recipes for fuming-washing therapy for KOA were collected and recorded in a database. The correlation coefficient among herbs, core combinations of herbs, andnew prescriptions were analyzed using modified mutual information, complex system entropy cluster, and unsupervised hierarchical clustering, respectively.RESULTS: Based on analysis of 345 Chinese medicine recipes for fuming-washing therapy, 68 herbs occurred frequently, 33 herb pairs occurred frequently, and 12 core combinations were found.Five new fuming-washing recipes for KOA were developed.CONCLUSION: Chinese medicines for fuming-washing therapy of KOA mainly consist of wind-dampness-dispelling and cold-dispersing herbs, blood-activating and stasis-resolving herbs,and wind-dampness-dispelling and heat-clearing herbs. The treatment of fuming-washing therapy for KOA also includes dispelling wind-dampness and dispersing cold, activating blood and resolving stasis, and dispelling wind-dampness and clearing heat. 2Study on gastric cancer blood plasma based on surface-enhanced Raman spectroscopy combined with multivariate analysis显示文摘A surface-enhanced Raman spectroscopy (SERS) method combined with multivariate analysis was developed for non-invasive gastric cancer detection. SERS measurements were performed on two groups of blood plasma samples: one group from 32 gastric patients and the other group from 33 healthy volunteers. Tentative assignments of the Raman bands in the measured SERS spectra suggest interesting cancer-specific biomolecular changes, including an increase in the relative amounts of nucleic acid, collagen, phospholipids and phenylalanine and a decrease in the percentage of amino acids and saccharide in the blood plasma of gastric cancer patients as compared with those of healthy subjects. Principal components analysis (PCA) and linear discriminant analysis (LDA) were employed to develop effective diagnostic algorithms for classification of SERS spectra between normal and cancer plasma with high sensitivity (79.5%) and specificity (91%). A receiver operating characteristic (ROC) curve was employed to assess the accuracy of diagnostic algorithms based on PCA-LDA. 4Psychological impact of the COVID-19 pandemic on healthcare workers:a cross-sectional study in China显示文摘Background Healthcare workers fighting against the coronavirus disease 2019(COVID-19)pandemic are under tremendous pressure,which puts them at an increased risk of developing psychological problems.Aims This study aimed to investigate the prevalence of psychological problems in different healthcare workers(ie,physicians,medical residents,nurses,technicians and public health professionals)during the COVID-19 pandemic in China and explore factors that are associated with the onset of psychological problems in this population during this public health crisis.Methods A cross-sectional,web-based survey was conducted in February 2020 among healthcare workers during the COVID-19 pandemic.Psychological problems were assessed using the Generalized Anxiety Disorder Scale,Patient Health Questionnaire and Insomnia Severity Index.Logistic regression analyses were used to explore the factors that were associated with psychological problems.Results The prevalence of symptoms of anxiety,depression,insomnia and the overall psychological problems in healthcare workers during the COVID-19 pandemic in China was 46.04%,44.37%,28.75%and 56.59%,respectively.The prevalence of the overall psychological problems in physicians,medical residents,nurses,technicians and public health professionals was 60.35%,50.82%,62.02%,57.54%and 62.40%,respectively.Compared with healthcare workers who did not participate in front-line work,front-line healthcare workers had a higher risk of anxiety,insomnia and overall psychological problems.In addition,attention to negative or neutral information about the pandemic,receiving negative feedback from families and friends who joined front-line work,and unwillingness to join front-line work if given a free choice were three major factors for these psychological problems.Conclusions Psychological problems are pervasive among healthcare workers during the COVID-19 pandemic.Receiving negative information and participating in front-line work appear to be.important risk factors for psychological problems.The psychological health of different healthcare workers should be protected during the COVID-19 pandemic with timely interventions and proper information feedback.Jianyu Que Le Shi Jiahui Deng Jiajia Liu Li Zhang Suying Wu Yimiao Gong Weizhen Huang Kai Yuan Wei Yan Yankun Sun Maosheng Ran Yanping Bao Lin Lu 2020General Psychiatry2020,33,3:8
5Orexin A attenuates the sleep-promoting effect of adenosine in the lateral hypothalamus of rats显示文摘Orexin neurons within the lateral hypothalamus play a crucial role in the promotion and maintenance of arousal. Studies have strongly suggested that orexin neurons are an important target in endogenous adenosine-regulated sleep homeostasis. Orexin A induces a robust increase in the firing activity of orexin neurons, while adenosine has an inhibitory effect. Whether the excitatory action of orexins in the lateral hypothalamus actually promotes wakefulness and reverses the sleep-producing effect of adenosine in vivo is less clear. In this study,electroencephalographic and electromyographic recordings were used to investigate the effects of orexin A and adenosine on sleep and wakefulness in rats. We found that microinjection of orexin A into the lateral hypothalamus increased wakefulness with a concomitant reduction of sleep during the first 3 h of post-injection recording, and this was completely blocked by a selective antagonist for orexin receptor 1,SB 334867. The enhancement of wakefulness also occurred after application of the excitatory neurotransmitter glutamate in the first 3 h post-injection. However, in the presence of the NMDA receptor antagonist APV, orexin A did not induce any change of sleep and wakefulness in the first 3 h.Further, exogenous application of adenosine into the lateral hypothalamus induced a marked increase of sleep in the first 3-h post-injection. No significant change in sleep and wakefulness was detected after adenosine application followed by orexin A administration into the same brain area. 6Effect of a comprehensive therapy plus Gushenyutai plaster administered at Guanyuan(CV 4) on male infertility associated with semen non-liquefaction显示文摘OBJECTIVE: To observe the clinical effect of Gushenyutai plaster administered at the Guanyuan(CV4) acupoint on male infertility associated with semen non-liquefaction.METHODS: Sixty-two male patients with infertility caused by semen non-liquefaction were random-ized into a treatment and control group. The control group received comprehensive therapy, which included oral administration of clarithromycin sustained release tablets, hip bath with Zhongyaoxiaoyan granules, prostate massage, and transurethral microwave treatment. The treatment group was administered Gushenyutai plaster in addition to the comprehensive therapy. Both groups were treated for 8 weeks. 7IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. 8High spatial and temporal resolution synthetic aperture phase microscopy显示文摘A new optical microscopy technique,termed high spatial and temporal resolution synthetic aperture phase microscopy(HISTR-SAPM),is proposed to improve the lateral resolution of wide-field coherent imaging.Under plane wave illumination,the resolution is increased by twofold to around 260 nm,while achieving millisecond-level temporal resolution.In HISTR-SAPM,digital micromirror devices are used to actively change the sample illumination beam angle at high speed with high stability.An off-axis interferometer is used to measure the sample scattered complex fields,which are then processed to reconstruct high-resolution phase images.Using HISTR-SAPM,we are able to map the height profiles of subwavelength photonic structures and resolve the period structures that have 198 nm linewidth and 132 nm gap(i.e.,a full pitch of 330 nm).As the reconstruction averages out laser speckle noise while maintaining high temporal resolution,HISTR-SAPM further enables imaging and quantification of nanoscale dynamics of live cells,such as red blood cell membrane fluctuations and subcellular structure dynamics within nucleated cells.We envision that HISTR-SAPM will broadly benefit research in material science and biology.Cheng Zheng Di Jin Yanping He Hongtao Lin Juejun Hu Zahid Yaqoo Peter T.C.So Renjie Zhou 2020Advanced Photonics2020,2,6:5

11Influence of Tip End-plate on Noise of Small Axial Fan显示文摘In this work, tip end-plate is used to improve the noise performance of small axial fans. Both numerical simulations and experimental methods were adopted to study the fluid flow and noise level of axial fans. Four modified models and the prototype are simulated. Influences of tip end-plate on static characteristics, internal flow field and noise of small axial fans are analyzed. The results show that on basis of the prototype, the model with the tip end-plate of 2 mm width and changed length achieved best noise performance. The overall sound pressure level of the model with the tip end-plate of 2 mm width and changed length is 2.4 dB less than that of the prototype at the monitoring point in specified far field. It is found that the mechanism of noise reduction is due to the decrease of vorticity variation on the surface of blades caused by the tip end-plate. Compared with the prototype, the static pressure of the model with the tip end-plate of 2 mm width and changed length at design flow rate decreases by 2 Pa and the efficiency decreases by 0.8%. 12Developing controllable hypermutable Clostridium cells through manipulating its methyl-directed mismatch repair system显示文摘Development of controllable hypermutable cells can greatly benefit understanding and harnessing microbial evolution.However,there have not been any similar sys-tems developed for Clostridium,an important bacterial genus.Here we report a novel two-step strategy for de-veloping controllable hypermutable cells of Clostridium acetobutylicum,an important and representative indus-trial strain.Firstly,the mutS/L operon essential for methyl-directed mismatch repair(MMR)activity was inactivated from the genome of C.acetobutylicum to generate hy-permutable cells with over 250-fold increased mutation rates.Secondly,a proofreading control system carrying an inducibly expressed mutS/L operon was constructed.The hypermutable cells and the proofreading control system were integrated to form a controllable hypermut-able system SMBMutC,of which the mutation rates can be regulated by the concentration of anhydrotetracycline(aTc).Duplication of the miniPthl-tetR module of the proof-reading control system further signifi cantly expanded the regulatory space of the mutation rates,demonstrating hypermutable Clostridium cells with controllable mutation rates are generated.The developed C.acetobutylicum strain SMBMutC2 showed higher survival capacities than the control strain facing butanol-stress,indicating greatly increased evolvability and adaptability of the controllable hypermutable cells under environmental challenges.Guodong Luan Zhen Cai Fuyu Gong Hongjun Dong Zhao Lin Yanping Zhang Yin Li 2013Protein & Cell2013,4,11:4

14Experimental and theoretical studies of the reaction between cationic vanadium oxide clusters and acetylene显示文摘The time of flight mass spectrometer coupled with a laser ablation/supersonic expansion cluster source and a fast flow reactor was adopted to study the reactivity of cationic vanadium oxide clusters(VmOn+) toward acetylene(C2H2) molecules under gas phase(P,~ 1.14 kPa),under near room temperature(T,~ 350 K) conditions.Association products,VmOnC2H2+(m,n = 2,4;2,6;3,7―8;4,9―11;5,12―13;6,13―16,and 7,17),are observed.The oxidation of C2H2 by(V2O5)+n(n = 1―3) is experimentally identified.The reactivity of(V2O5)+ n decreases as n increases.Density functional theory(DFT) calculations were carried out to interpret the reaction mechanisms.The DFT results indicate that a terminal oxygen atom from V2O5+ can transfer overall barrierlessly to C2H2 at room temperature,which is in agreement with the experimental observation.Other experimental results such as the observation of V2O6C2H+2 and non-observation of V2O7,8C2H+2 in the experiments are also well interpreted based on the DFT calculations.The reactivity of vanadium oxide clusters toward acetylene and other hydrocarbons may be considered in identifying molecular level mechanisms for related heterogeneous catalysis.YIN Shi MA YanPing DU Lin HE ShengGui GE MaoFa 2008Chinese Science Bulletin2008,53,24:3
15Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. In addition, we performed a quantitative analysis of the different pollen grain cells, including sacci, and found that there were 202 connections within a saccus SEM image. 17Regulation of cytoskeleton-associated protein activities: Linking cellular signals to plant cytoskeletal function显示文摘The plant cytoskeleton undergoes dynamic remodeling in response to diverse developmental and environmental cues. Remodeling of the cytoskeleton coordinates growth in plant cells, including trafficking and exocytosis of membrane and wall components during cell expansion, and regulation of hypocotyl elongation in response to light. Cytoskeletal remodeling also has key functions in disease resistance and abiotic stress responses. Many stimuli result in altered activity of cytoskeleton-associatedproteins,microtubuleassociated proteins(MAPs) and actin-binding proteins(ABPs). MAPs and ABPs are the main players determining the spatiotemporally dynamic nature of the cytoskeleton, functioning in a sensory hub that decodes signals to modulate plant cytoskeletal behavior. Moreover, MAP and ABP activities and levels are precisely regulated during development and environmental responses, but our understanding of this process remains limited. In this review, we summarize the evidence linking multiple signaling pathways, MAP and ABP activities and levels, and cytoskeletal rearrangements in plant cells. 19YTHDF2 is essential for spermatogenesis and fertility by mediating a wave of transcriptional transition in spermatogenic cells显示文摘The dynamic and reversible regulation roles of m^(6)A modification and the characterization of m^(6)A readers have provided new insights into spermatogenesis at the post-transcriptional level.YTHDF2,as an m^(6)A reader,has been reported to mediate the m^(6)A-containing transcript decay during the mouse oocyte maturation,embryonic stem cell differentiation,neural development,and zebrafish maternal-to-zygotic transition.However,the roles of YTHDF2 in mammalian spermatogenesis are uncertain.Here,we generated germ cell-specific Ythdf2 mutants(Ythdf2-vKO)at a C57BL/6J background and demonstrated that YTHDF2 is essential for mouse spermatogenesis and fertility.Ythdf2-vKO provides oligoasthenoteratozoospermia phenotype with increased apoptosis in germ cells.High-throughput RNA-seq analysis showed that a group of mRNAs is upregulated in Ythdf2-vKO mouse testis;further analysis and MeRIP-qPCR data showed that most of the upregulated genes in Ythdf2-vKO mouse testis are modified with m^(6)A and are YTHDF2 candidate binding genes.Interestingly,RNA-seq analysis combined with our previous single-cell transcriptomics data of mouse spermatogenesis pointed out the failure of a wave of transcript transition during the spermatogenesis of Ythdf2-vKO mice,which was confirmed by gene expression analysis using qPCR of diplotene spermatocytes and round spermatids obtained through fluorescence-activated cell sorting.Our study demonstrates the fundamental role of YTHDF2 during mouse spermatogenesis and provides a potential candidate for the diagnosis of male infertility with the oligoasthenoteratozoospermia syndrome.Xinxi Zhao Zhen Lin Yong Fan Wenzhi Li Yujie Zhang Fei Li Tong Hong Hua Feng Minghan Tong Ningling Wang Yanping Kuang Qifeng Lyu 2021Acta Biochimica et Biophysica Sinica2021,53,12:2
20Characteristics of bleaching of sandstone in northeast of Ordos Basin and its relationship with natural gas leakage显示文摘Bleaching of sandstone has significant applications to tracing hydrocarbon pathways and evaluating the scale of natural gas seepage. Bleaching of sandstones in the northeast of Ordos Basin is mainly distributed in the Mesozoic Yan'an Formation. Studying on petrology, major elements, REEs and trace elements of bleached sandstones and comparing with adjacent sandstones, combining with geologic-geochemical evidences of gas seepage in the northeast of the basin, the bleached sandstones are formed in the acid environment and reducing fluids. Characteristics of petrology show that the contents of kaolinite are high and the color of margin of ferric oxide minerals is lighter than that of the center. Major elements of sandstone samples show high contents of Al2O3 and low ratio of Fe3+/Fe2+. The TFe2O3 content of the bleached sandstone is lower than that of red rock. REE data show that bleached sandstones have low ∑REE contents and Eu-depleted and slightly Ce-enriched. Trace elements show that the bleached sandstones enrich in Co, deplete in Sr, and slightly enrich in Zr and Hf which are close to the values for the green alteration sandstones, and slightly lower than ore-bearing sandstones. Geochemical characteristics of oil-bearing sandstone in the northern basin suggest that the oil-shows are formed by matured Carboniferous-Permian coal bed methane escaping to the surface, and natural gas in field could migrate to the north margin of the basin. The δ13C (PDB) andδ18O(PDB) values of calcite cement in the study area range from -11.729‰ to -10.210‰ and -14.104‰ to -12.481‰, respectively. Theδ13C (PDB) values less than -10‰ imply the carbon sources part from organic carbon.
