| 1 | Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro.In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally,once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls.The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time,duration of fever,and adverse events.A total of 197 patients completed chloroquine treatment,and 176patients were included as historical controls.The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation,this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic. | Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan | 2020 | National Science Review2020，7，9： | 20 |
| 2 | DNA image cytometry test for primary screening of esophageal cancer: a population-based multi-center study in high-risk areas in China显示文摘Objective: To evaluate the feasibility of DNA image cytometry(DNA-ICM) as a primary screening method for esophageal squamous cell cancer(ESCC).Methods: A total of 5,382 local residents aged 40–69 years from three high-risk areas in China(Linzhou in Henan province, Feicheng in Shandong province and Cixian in Hebei province) from 2008 to 2011 were recruited in this population-based screening study. And 2,526 subjects declined to receive endoscopic biopsy examination with Lugol's iodine staining, while 9 and 815 subjects were excluded from liquid-based cytology and DNA-ICM test respectively due to slide quality. Finally, 2,856, 5,373 and 4,567 subjects were enrolled in the analysis for endoscopic biopsy examination, liquid-based cytology and DNA-ICM test, respectively. Sensitivity(SE), specificity(SP), negative predictive values(NPV) and positive predictive values(PPV) as well as their 95% confidence intervals(95% CI) for DNA-ICM, liquid-based cytology and the combination of the two methods were calculated. Receiver operating characteristic(ROC) curves were applied to determine the cutoff point of DNA-ICM for esophageal cancer.Results: DNA-ICM results were significantly correlative with esophageal cancer and precancer lesions(χ~2=18.016, P<0.001). The cutoff points were 5,802, 5,803 and 8,002 based on dissimilar pathological types of low grade intraepithelial neoplasia(LGIN), high grade intraepithelial neoplasia(HGIN), and ESCC, respectively, and 5,803 was chosen in this study considering the SE and SP. The SE, SP, PPV, NPV of DNA-ICM test(cutoff point 5,803) combined with liquid-based cytology [threshold atypical squamous cells of undetermined significance(ASCUS)] were separately 72.1%(95% CI: 70.3%-73.9%), 43.3%(95% CI: 41.3%-45.3%), 22.8%(95% CI: 21.1%-24.5%) and 87.0%(95% CI: 85.7%-88.3%) for LGIN, 85.7%(95% CI: 84.3%-87.1%), 41.3%(95% CI: 39.3%-43.3%), 4.6%(95% CI: 3.8%-5.4%) and 98.9%(95% CI: 98.5%-99.3%) for HGIN, and 96.0%(95% CI: 95.2%-96.8%), 40.8%(95% CI: 38.8%-42.8%), 1.7%(95% CI: 1.2%-2.2%) and 99.9%(95% CI: 99.8%-100.0%) for ESCC.Conclusions: It is possible to use DNA-ICM test as a primary screening method before endoscopic screening for esophageal cancer. | Meng Wang Changqing Hao Qing Ma Guohui Song Shanrui Ma Deli Zhao Lin Zhao Xinqing Li Wenqiang Wei | 2016 | Chinese Journal of Cancer Research2016，28，4： | 15 |
| 3 | Distribution and source of microplastics in China's second largest reservoir-Danjiangkou Reservoir显示文摘Fresh water microplastic pollution is of pressing concern globally,but its distribution and sources in reservoirs are poorly documented.Danjiangkou Reservoir is the second largest reservoir in China and is divided into the Han Reservoir and Dan Reservoir.In this work,microplastic abundances and morphological characteristics of the reservoir were investigated.The microplastic abundance of 15 main tributaries of the reservoir was also measured.The vertical distribution(in water column and sediment),horizontal distribution(in Han Reservoir and Dan Reservoir)and source of microplastics were analyzed.Microplastics accumulated in the middle layer of the reservoir,and the size and color of the microplastic particles changed from the surface to the bottom,which implies that surveys of surface water are not enough to determine the microplastic contamination for deep water reservoirs.In the surface water,the microplastic abundance in the Han Reservoir was lower than that in the Dan Reservoir(p<0.05),but microplastic abundance did not differ significantly in the intermediate and bottom water.Tributaries were one of the main sources of microplastics for Han Reservoir but not for Dan Reservoir.Agricultural cultivation in the hydro-fluctuation belt might be an important source of microplastics in the Dan Reservoir,which should be given additional attention.The results of this study can provide valuable information for developing microplastic sampling strategies in deep water reservoirs.Further studies are recommended to investigate the process through which microplastics in the hydro-fluctuation belt enter the reservoir and the sinking behavior of microplastics in the reservoir. | Li Lin Xiong Pan Sheng Zhang Danwen Li Wenliang Zhai Zhen Wang Jingxiang Tao Changqing Mi Qingyun Li John C.Crittenden | 2021 | Journal of Environmental Sciences2021，33，4： | 13 |

| 5 | Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. | 6 | Ectopic expression of fungal EcGDH improves nitrogen assimilation and grain yield in rice显示文摘NADP(H)-dependent glutamate dehydrogenases(GDH) in lower organisms have stronger ammonium affinity than those in higher plants. Here we report that transgenic rice overexpressing the EcGDH from Eurotium cheralieri exhibited significantly enhanced aminating activities.
