| 1 | Apoptosis and Proinflammatory Cytokine Responses of Primary Mouse Microglia and Astrocytes Induced by Human H1N1 and Avian H5N1 Influenza Viruses显示文摘Patients with an influenza virus infection can be complicated by acute encephalopathy and encephalitis. To investigate the immune reactions involved in the neurocomplication,mouse microglia and astrocytes were isolated,infected with human H1N1 and avian H5N1 influenza viruses,and examined for their immune responses. We observed homogeneously distributed viral receptors,sialic acid (SA)-α2,3-Galactose (Gal) and SA-α2,6-Gal,on microglia and astrocytes. Both viruses were replicative and productive in microglia and astrocytes. Virus-induced apoptosis and cytopathy in infected cells were observed at 24 h post-infection (p.i.). Expression of IL-1β,IL-6 and TNF-α mRNA examined at 6 h and 24 h p.i. was up-regulated,and their expression levels were considerably higher in H5N1 infection. The amounts of secreted proinflammatory IL-1β,IL-6 and TNF-α at 6 h and 24 h p.i. were also induced,with greater induction by H5N1 infection. This study is the first demonstration that both human H1N1 and avian H5N1 influenza viruses can infect mouse microglia and astrocytes and induce apoptosis,cytopathy,and proinflammatory cytokine production in them in vitro. | 2 | Design of heat exchanger network based on entransy theory显示文摘The heat exchanger network(HEN) synthesis problem based on entransy theory is analyzed. According to the characteristics of entransy representation of thermal potential energy, the entransy dissipation represents the irreversibility of the heat transfer process, the temperature difference determines the entransy dissipation, and four HEN design steps based on entransy theory are put forward. The present study shows how it is possible to set energy targets based on entransy and achieve them with a network of heat exchangers by an example of heat exchanger network design for four streams. In order to verify the correctness of the heat exchanger networks design method based on entransy theory, the synthesis of the HEN for the diesel hydrogenation unit is studied. Using the heat exchange networks design method based on entransy theory, the HEN obtained is consistent with energy targets. | 4 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |

| 10 | Barriers and facilitators in Pre-exposure Prophylaxis(PrEP)use intention among Chinese homosexual men显示文摘Background:Despite strengthened efforts on human immunodeficiency virus(HIV)prevention and control,new HIV infections continue to increase among men who have sex with men(MSM)in China.Pre-exposure prophylaxis(PrEP),a highly effective HIV-prevention tool,has recently been included in China’s Action Plan of HIV Prevention and Control.To promote future PrEP implementation,this study aims to identify the barriers and facilitators in PrEP use intention among MSM in China.Methods:In 2018,a cross-sectional survey was conducted among 300 MSM in Nanjing,Jiangsu Province.Questions on demographics,sexual behavior(including a seven-item high-risk behavior index),PrEP use intention,PrEP-related awareness and accessibility,and a seven-item public HIV stigma scale were included in the questionnaire.Bivariate and multivariate logistic regression analyses were conducted to identify factors associated with PrEP use intention.Results:Overall,44.67%of the participants had more than two male sexual partners and 5.56%had HIV-positive sexual partners.Only 57.00%had heard of PrEP and only four(1.33%)participants had used PrEP.However,75.34%expressed the willingness to use oral PrEP if its efficacy was assured.The beliefs that“PrEP can prevent HIV if taken as prescribed”(adjusted odds ratio(AOR)=4.84,P<0.001)and“PrEP can be scaled up in the community”(AOR=3.24,P<0.001)were positively associated with oral PrEP use intention.Concerns of side effects were negatively associated with oral PrEP use intention(AOR=0.32,P=0.006).Further,77.00%of the participants would choose injectable or implanted PrEP instead of oral PrEP.One of the PrEP stigma items,“Not certain if doctors can prescribe PrEP if I go to the clinic”was positively associated with the intention to use injectable or implanted PrEP(AOR=3.03).The items“Heard of PrEP”(AOR=2.74)and“PrEP can prevent HIV if taken as prescribed”(AOR=2.65)were also positively related to the intention to use injectable or implanted PrEP.The most common concerns regarding PrEP use were efficacy(44.67%)and side effects(38.67%).The most common reasons for using injectable and implanted PrEP were adherence(81.94%and 77.86%,respectively)and privacy concerns(56.48%and 55.00%,respectively).Affordable price and coverage by health insurance were the driving factors for PrEP use.Conclusion:Given the low level of awareness of PrEP-related information in China,it’s necessary to provide interventions for high-risk individuals and communities,in order to increase their awareness and knowledge of PrEP.Furthermore,additional alternatives to HIV prevention,such as long-acting injectable or implanted PrEP,should be investigated to reduce the risk of HIV infection in at-risk MSM.Even if this programme could be approved by the National Medical Products Administration of China and implemented,a supportive social environment for MSM is essential during its implementing. | Mingyu Si Xiaoyou Su Li Yan Yu Jiang Yuanli Liu Chongyi Wei Hongjing Yan | 2020 | Global Health Journal2020,4,3: | 1 |
| 11 | New interpretation on the provenance changes of the upper Pinghu–lower Huagang Formation within Xihu Depression,East China Sea Shelf Basin显示文摘Both Pinghu and Huagang formations are important hydrocarbon reservoirs of the Xihu Depression in the East China Sea Shelf Basin.Clarifying the source suppliers and restoring source-to-sink transport routes are of great significance to the future petroleum and gas undertakings.Previous researchers were largely confined by either limitation of geological records,highly dependence on a singular method or low-precision dating techniques.Our study integrated heavy mineral assemblages,geochemical analyses and detrital zircon U-Pb dating to reconstruct multiple source-to-sink pathways,and to provide a better understanding on the provenance evolution for the upper Pinghu–lower Huagang depositions of the Xihu Depression.At least three major provenances have been confirmed and systematically investigated for their separate compositional features.The Hupijiao Uplift(or even farther northern area)was dominated by a major Paleoproterozoic population peaked at ca.1830 Ma along with minor Mesozoic clusters.The Haijiao Uplift to the west and the Yushan Low Uplift to the southwest,on the other hand,generate opposite U-Pb age spectra with apparently larger peaks of Indosinian and Yanshanian-aged zircons.To be noted,both Indosinian and Paleoproterozoic peaks are almost identical in proportion for the Haijiao Uplift.The overall sedimentary pattern of late Eocene-early Oligocene was featured by both spatial and temporal distinction.The Hupijiao Uplift was likely to cast limited impact during the late Eocene,whereas the broad southern Xihu Depression was transported by a large abundance of materials from the nearby Haijiao and Yushan Low Uplifts.The northern source substantially extended its influence to the farther south during the early Oligocene by delivering plentiful sediments of higher-degree metamorphic parent rocks.Combined with the proximal western and southwestern suppliers,the overall Xihu Depression was under control from both distant and local provenances. | Jinshui Liu Shuai Li Kaifei Liao Yuchi Cui Lei Shao Peijun Qiao Yi Lu Yuanli Hou Thian Lai Goh Yongjian Yao | 2023 | Acta Oceanologica Sinica2023,42,3: | 1 |

| 16 | Comparison of the Pipeline embolisation device alone or combined with coiling for treatment of different sizes of intracranial aneurysms显示文摘Objectives The aim of this study was to compare complications and outcomes between intracranial aneurysms treated with the Pipeline embolisation device(PED)alone or with PED combined with coiling for different-sized aneurysms.Method Patients with aneurysms treated by PED were collected from the PED in China postmarket multicentre registry study.We performed a propensity match analysis to compare the efficacy and safety between PED alone and PED combined with coiling treatment,and then aneurysms were organised into three groups based on their size:small(≤7 mm),medium(≤15 mm to>7 mm)and large/giant(>15 mm).Complications and aneurysm occlusion rates in the aneurysm size groups were compared between PED alone and PED combined with coiling patients.Result A total of 1171 patients with 1322 aneurysms were included.All patients received clinical follow-up,while angiographic follow-up was available in 967 aneurysms.For small aneurysms,there was no difference in the aneurysm occlusion rate between two groups(79.1%vs 88.4%,respectively),while there was a significant increase in the ischaemic complication rate(8.3%vs 19.3%,respectively,p=0.0001).For medium and large/giant saccular aneurysms,PED combined with coiling significantly improved the occlusion rate(medium aneurysms:74.7%vs 88.8%,respectively,p<0.0001;large/giant saccular aneurysms:72.9%vs 86.9%,respectively,p=0.018),while there were no differences in the total complication rate.For large/giant non-saccular aneurysms,two groups showed no differences.Conclusion Use of the PED with adjunctive coils can significantly improve the occlusion rate of medium aneurysms,without increasing the total complication rate. | Chao Wang Bin Luo Tianxiao Li Aisha Maimaitili Guohua Mao Donglei Song Yunyan Wang Wenfeng Feng Yang Wang Huaizhang Shi Jieqing Wan Jianmin Liu Sheng Guan Yuanli Zhao Hongqi Zhang | 2022 | Stroke & Vascular Neurology2022,7,4: | 1 |

| 18 | In vitro genotoxicity evaluation and metabolic study of residual glutaraldehyde in animal-derived biomaterials显示文摘Glutaraldehyde(GA)is an important additive that is mainly used in animal-derived biomaterials to improve their mechanical and antimicrobial capacities.However,GA chemical toxicity and the metabolic mechanism remain relatively unknown.Therefore,residual GA has always been a major health risk consideration for animal-derived medical devices.In this study,extracts of three biopatches were tested via the GA determination test and mouse lymphoma assay(MLA).The results showed that dissolved GA was a potential mutagen,which could induce significant cytotoxic and mutagenic effects in mouse lymphoma cells.These toxic reactions were relieved by the S9 metabolic activation(MA)system.Furthermore,we confirmed that GA concentration decreased and glutaric acid was generated during the catalytic process.We revealed GA could be oxidized via cytochrome P450 which was the main metabolic factor of S9.We found that even though GA was possibly responsible for positive reactions of animal-derived biomaterials’biocompatibility evaluation,it may not represent the real situation occurring in human bodies,owing to the presence of various detoxification mechanisms including the S9 system.Overall,in order to achieve a general balance between risk management and practical application,rational decisions based on comprehensive analyses must be considered. | Jianfeng Shi Huan Lian Yuanli Huang Danmei Zhao Han Wang Chunren Wang Jingli Li Linnan Ke | 2020 | Regenerative Biomaterials2020,7,6: | 1 |
