1Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5
2Human embryonic stem cells-derived endothelial cell therapy facilitates kidney regeneration by stimulating renal resident stem cell proliferation in acute kidney injury显示文摘Endothelial cell therapy has been implicated to enhance tissue regeneration and vascularization in ischemic kidney. However, no published study has yet examined direct effects of endothelial cell treatment in kidney recovery. This study investigated the therapeutic efficacy of endothelial cells in a mouse model with acute kidney injury (AKI). Thus, human embryonic stem cells-derived endothelial cells (hESC-ECs) labeled with a reporter system encoding a double fusion reporter gene for firefly luciferase (Fluc) and green fluorescent protein (GFP) were characterized by Fluc imaging and immunofluoresence staining. Cultured hESC-ECs (1×106) were injected into ischemic kidney shortly after AKI. Survival of the transplanted hESC-ECs was monitored in vivo from day 1 to 14 after endothelial cell transplantation and potential impact of hESC-EC treatment on renal regeneration was assessed by histological analyses. We report that a substantial level of bioluminescence activity was detected 24 h after hESC-EC injection followed by a gradual decline from 1 to 14 d. Human ESC-ECs markedly accelerated kidney cell proliferation in response to ischaemia-induced damage, indicated by an elevated number of BrdU+ cells. Co-expression of Sca-1, a kidney stem cell proliferation marker, and BrdU further suggested that the observed stimulation in renal cell regeneration was, at least in part, due to increased proliferation of renal resident stem cells especially within the medullary cords and arteriole. Differentiation of hESC-ECs to smooth muscle cells was also observed at an early stage of kidney recovery. 4Understanding the role of interface in advanced semiconductor nanostructure and its interplay with wave function overlap显示文摘As the proportion of interfaces increases rapidly in nanomaterials,properties and quality of interfaces hugely impact the performance of advanced semiconductors.Here,the effect of interfaces is explored by comparatively studying two InAs/AlSb superlattices with and without the thin InAsSb layers inserted inside each InAs layers.Through strain mapping,it indicates that the addition of interfaces leads to an increase of local strain both near interfaces and inside layers.Meantime,owing to the creation of hole potential wells within the original electron wells,the charge distribution undergoes an extra electron-hole alternating arrangement in the structure with inserted layers than the uninserted counterpart.Such a feature is verified to enhance electron-hole wave function overlap by theoretical simulations,which is a must for better optical performance.Furthermore,with an elaborate design of the inserted layers,the wave function overlap could be boosted without sacrificing other key device performances.Chenyuan Cai Yunhao Zhao Faran Chang Xuebing Zhao Liting Yang Chongyun Liang Guowei Wang Zhichuan Niu Yi Shi Xianhu Liu Yuesheng Li Renchao Che 2020Nano Research2020,13,6:1
5Co-seismic deformation derived from GPS observations during April 20th,2013 Lushan Earthquake,Sichuan,China显示文摘We process the standard 30 s,static GPS data and the 1 s,high-rate GPS(HRGPS)data provided by the Crustal Movement Observation Network of China with GAMIT/GLOBK software package,and obtain the coseismic displacements of near field and far field,and the epoch-by-epoch time series of HRGPS during Lushan earthquake.GPS data from about 20 sites in Sichuan province,which located between 40 and 450 km from the epicenter,are analyzed so as to study the characteristics of the static displacements and the dynamic crustal deformations,with periods ranging from several minutes to over a month.The result shows that:the static displacements caused by Lushan earthquake are limited to several centimeters;the nearest station SCTQ at 43 km from the epicenter has the largest static displacement of about 2 cm,while the other stations generally have insignificant displacements of less than 5 mm.the stations in the east ofSichuan–Yunnan region shifts 5–10 mm toward the southwest,and the stations in the middle-west of Sichuan Basin moves indistinctively 1–2 mm toward the northwest;station SCTQ has the largest kinematic displacement of about 4 and 3 cm peak-to-peak on the north and east component,respectively,and is much greater than the static permanent displacement;for the stations located at a distance greater than 150 km from the epicenter,the kinematic motions are generally insignificant;exceptionally,station SCNC and station SCSN in central Sichuan Basin have significant kinematic motions although they are more than 200 km away from the epicenter.Yujun Du Zemin Wang Shujiang Yang Jiachun An Qiang Liu Guowei Che 2013Earthquake Science2013,26,3:1
6GPR Surveying in the kernel area of Grove Mountains,Antarctica显示文摘The Grove Mountains, located between the Zhongshan Station and Dome A, are a very important area in inland Antarctic research. China has organized five investigations of the Grove Mountains, encompassing the geological structure, ancient climate, meteorites, ice-movement monitoring, basic mapping, meteorological observations, and other multi-disciplinary observational studies. During the 26th Chinese National Antarctic Research Expedition in 2010, the Grove Mountains investigation team applied specialized ground-penetrating radar(GPR) to survey subglacial topography in the eastern kernel area of the Grove Mountains. In this paper, we processed GPS and GPR data gathered in the field and drew, for the first time, two subglacial topographic maps of the Grove Mountains kernel area using professional graphics software. The preliminary results reveal the mystery of the nunatak landform of this area, give an exploratory sense of the real bedrock landforms, and indicate a possible sedimentary basin under the Pliocene epoch fossil ice in the Grove Mountains area. 9Development of the Relationship between Angiogenesis and Tumor Dormancy显示文摘Tumor dormancy, a complex and still poorly understood phenomenon, has been defined by the long-term persistence of occult can- cer cells during tumor progression. Recurrence and metastasis may occur just because of an activation of a small portion of the tumor cells. In our view, sustained angiogenesis is considered essential in triggering invasive tumor growth. Here we analyze the correlation between angiogenesis and tumor dormancy, the establishment of tumor dormancy models, the imaging strategies and the new biomarkers for dececting microscopic tumors before or during the angiogenic switch. 10Lithium-site substituted argyrodite-type Li_(6)PS_(5)I solid electrolytes with enhanced ionic conduction for all-solid-state batteries显示文摘Argyrodites,Li_(6)PS_(5)X(X=Cl,Br,I),have piqued the interest of researchers by offering promising lithium ionic conductivity for their application in all-solid-state batteries(ASSBs).However,other than Li_(6)PS_(5)Cl(651Cl)and Li_(6)PS_(5)Br(651Br),Li_(6)PS_(5)I(651I)shows poor ionic conductivity(10^(-7)S cm^(-1)at 298 K).Herein,we present Al-doped 651I with I^(-)/S^(2-)site disordering to lower activation energy(Ea)and improve ionic conductivity.They formed argyrodite-type solid solutions with a composition of(Li_(6–3x)Al_(x))PS_(5)I in 0≤x≤0.10,and structural analysis revealed that Al^(3+)is located at Li sites.Also,the Al-doped samples contained anion I^(-)/S^(2-)site disorders in the crystal structures and smaller lattice parameters than the non-doped samples.Impedance spectroscopy measurements indicated that Al-doping reduced the ionic diffusion barrier,Ea,and increased the ionic conductivity to 10^(-5)S cm^(-1);the(Li5.7Al0.1)PS5I had the highest ionic conductivity in the studied system,at 2.6×10^(-5)S cm^(-1).In a lab-scale ASSB,with(Li_(5.7)Al_(0.1))PS_(5)I functioned as a solid electrolyte,demonstrating the characteristics of a pure ionic conductor with negligible electronic conductivity.The evaluated ionic conduction is due to decreased Li+content and I^(-)/S^(2-)disorder formation.Li-site cation doping enables an in-depth understanding of the structure and provides an additional approach to designing betterperforming SEs in the argyrodite system.GAO Ling XIE YuLin TONG Yan XU Miao YOU JiaLe WEI HuiPing YU XiangXiang XU SiQi ZHANG Yi CHE Yong TANG Ya SUZUKI Kota KANNO Ryoji ZHAO GuoWei 2023Science China(Technological Sciences)2023,66,7:0
