1Vegetation and soil carbon storage in China显示文摘This study estimated the current vegetation and soil carbon storage in China using a biogeochemical model driven with climate, soil and vegetation data at 0.5°latitude-longitude grid spatial resolution. The results indicate that the total carbon storage in China's vegetation and soils was 13.33 Gt C and 82.65 Gt C respectively, about 3% and 4% of the global total. The nationally mean vegetation and soil carbon densities were 1.47 kg C/m2 and 9.17 kg C/m2, respectively, differing greatly in various regions affected by climate, vegetation, and soil types. They were generally higher in the warm and wet Southeast China and Southwest China than in the arid Northwest China; whereas vegetation carbon density was the highest in the warm Southeast China and Southwest China, soil carbon density was the highest in the cold Northeast China and southeastern fringe of the Qinghai-Tibetan Plateau. 2The IAP family: endogenous caspase inhibitors with multiple biological activities显示文摘IAPs (inhibitors of apoptosis) are a family of proteins containing one or more characteristic BIR domains. These proteins have multiple biological activities that include binding and inhibiting caspases, regulating cell cycle progression, and modulating receptor-mediated signal transduction. Our recent studies found the IAP family members XIAP and c-IAP1 are ubiquitinated and degraded in proteasomes in response to apoptotic stimuli in T cells, and their degradation appears to be important for T cells to commit to death. In addition to three BIR domains, each of these IAPs also contains a RING finger domain.We found this region confers ubiquitin protease ligase (E3) activity to IAPs, and is responsible for the auto-ubiquitination and degradation of IAPs after an apoptotic stimulus. 3Effect of extracorporeal bioartificial liver support system on fulminant hepatic failure rabbits显示文摘AIM To evaluate the possibility of usingcultured human hepatocytes as a bridge betweenbioartificial liver and liver transplantation.METHODS In this experiment,the efficacy ofextracorporeal bioartificial liver support system(EBLSS)consisting of spheriodal human livercells and cultured hepatocytes supernatant wasassessed in vivo using galactosamine inducedrabbit model of fulminant hepatic failure.RESULTS There was no difference of survivalbetween the two groups of rabbits,but in thesupported rabbits serum alanineaminotransferase,total bilirubin and creatininewere significantly lower and hepatocyte necrosiswas markedly milder than those in controlanimals.In addition,a good viability of humanliver cells was noted after the experiment.CONCLUSION EBLSS plays a biologic role inmaintaining and compensating the function ofthe liver.Wang YJ Li MD Wang YM Chen GZ Lu GD Tan ZX 2000World Journal of Gastroenterology2000,6,2:17
4Drug resistance mechanisms and novel drug targets for tuberculosis therapy显示文摘Drug-resistant tuberculosis(TB) poses a significant challenge to the successful treatment and control of TB worldwide.Resistance to anti-TB drugs has existed since the beginning of the chemotherapy era.New insights into the resistant mechanisms of anti-TB drugs have been provided.Better understanding of drug resistance mechanisms helps in the development of new tools for the rapid diagnosis of drugresistant TB.There is also a pressing need in the development of new drugs with novel targets to improve the current treatment of TB and to prevent the emergence of drug resistance in Mycobacterium tuberculosis.This review summarizes the anti-TB drug resistance mechanisms,furnishes some possible novel drug targets in the development of new agents for TB therapy and discusses the usefulness using known targets to develop new anti-TB drugs.Whole genome sequencing is currently an advanced technology to uncover drug resistance mechanisms in M.tuberculosis.However,further research is required to unravel the significance of some newly discovered gene mutations in their contribution to drug resistance.Md Mahmudul Islam H.M. 5Watering techniques and zero-valent iron biochar pH effects on As and Cd concentrations in rice rhizosphere soils,tissues and yield显示文摘Zero-valent iron amended biochar(ZVIB)has been proposed as a promising material in immobilizing heavy metals in paddy fields.In this study,the impacts of p H of ZVIB(p H 6.3 and p H 9.7)and watering management techniques(watering amount in the order of CON(control,5/72)>3/72>1-3/72>3/100>1/72,with 5/72,for example,representing irrigation given to 5 cm above soil surface in 72 hr regular interval)on As and Cd bioavailability for rice and its grain yield(Yield BR)were investigated in a pot experiment.Brown rice As(As BR)content was irrelative to the watering treatments,while significantly decreased(>50%)with the addition of both ZVIB materials.The diminutions of brown rice Cd(Cd BR)content as well as the Yield BR were highly dependent on both the soil amendment materials’p H and watering amount.Among all the watering treatments,3/72 treatment(15%less irrigation water than the CON)with ZVIB 6.3 amendment was the optimum fit for simultaneous reduction of As BR(50%)and Cd BR contents(19%)as well as for significant increment(12%)of the Yield BR.Although high pH(9.7)ZVIB application could also efficiently decrease As and Cd contents in brown rice,it might risk grain yield lost if appropriate(e.g.3/72 in our study)watering management technique was not chosen.Therefore,ZVIB would be an environmentally friendly option as an amendment material with proper selection of watering management technique to utilize As and Cd co-contaminated arable soils safely for paddy cultivation.Md Shafiqul Islam Yali Chen Liping Weng Jie Ma Zulqarnain Haider Khan Zhongbin Liao Abdoul Salam Issiaka Abdoul Magid Yongtao Li 2021Journal of Environmental Sciences2021,33,2:9
6Physicochemical and Sensory Properties of japonica Rice Varied with Production Areas in China显示文摘Northeast of China and Jiangsu Province are major production areas of japonica rice in China.Rice from northeast of China is well-known for its good-eating and appearance quality,and that from Jiangsu Province is viewed as inferior.However,little is known concerning the difference in physicochemical and sensory properties of rice between the major two production areas.Analysis of 16 commercial rice samples showed marked differences in physicochemical properties,including chalky grain rate,contents of amylose and protein and pasting properties between the two main areas.Northeastern rice contained more shortchain amylopectin as compared with Jiangsu rice.However,Jiangsu rice is comparable to northeastern rice in terms of sensory quality including overall acceptability and textural properties of springiness,stickiness and hardness as evaluated by trained panel.Our results indicated the limitation of conventional index of physicochemical properties,and suggested the necessity of identification of new factors controlling rice sensory property.In addition,the taste analyzer from Japan demonstrates limitation in distinguishing the differences between northeastern and Jiangsu rice,and therefore needs localization to fit China.YANG Xiao-yu LIN Zhao-miao LIU Zheng-hui Md A Alim BI Jun-guo LI Gang-hua WANG Qiang-sheng WANG Shao-hua DING Yan-feng 2013Journal of Integrative Agriculture2013,12,10:8
7MicroRNA and mRNA profiling of cerebral cortex in a transgenic mouse model of Alzheimer’s disease by RNA sequencing显示文摘In a previous study,we found that long non-coding genes in Alzheimer’s disease(AD)are a result of endogenous gene disorders caused by the recruitment of microRNA(miRNA)and mRNA,and that miR-200a-3p and other representative miRNAs can mediate cognitive impairment and thus serve as new biomarkers for AD.In this study,we investigated the abnormal expression of miRNA and mRNA and the pathogenesis of AD at the epigenetic level.To this aim,we performed RNA sequencing and an integrative analysis of the cerebral cortex of the widely used amyloid precursor protein and presenilin-1 double transgenic mouse model of AD.Overall,129 mRNAs and 68 miRNAs were aberrantly expressed.Among these,eight down-regulated miRNAs and seven up-regulated miRNAs appeared as promising noninvasive biomarkers and therapeutic targets.The main enriched signaling pathways involved mitogen-activated kinase protein,phosphatidylinositol 3-kinase-protein kinase B,mechanistic target of rapamycin kinase,forkhead box O,and autophagy.An miRNA-mRNA network between dysregulated miRNAs and corresponding target genes connected with AD progression was also constructed.These miRNAs and mRNAs are potential biomarkers and therapeutic targets for new treatment strategies,early diagnosis,and prevention of AD.The present results provide a novel perspective on the role of miRNAs and mRNAs in AD.This study was approved by the Experimental Animal Care and Use Committee of Institute of Medicinal Biotechnology of Beijing,China(approval No.IMB-201909-D6)on September 6,2019.Li Zeng Hai-Lun Jiang Ghulam Md Ashraf Zhuo-Rong Li Rui Liu 2021Neural Regeneration Research2021,16,10:6
8Microarray,SAGE and their applications to cardiovascular diseases显示文摘The wealth of DNA data generated by the human genome project coupling with recently invented high-throughput gene expression profiling techniques has dramatically sped up the process for biomedical researchers on elucidating the role of genes in human diseases. One powerful method to reveal insight into gene functions is the systematic analysis of gene expression. Two popular high-throughput gene expression technologies, microarray and Serial Analysis of Gene Expression (SAGE) are capable of producing large amounts of gene expression data with the potential of providing novel insights into fundamental disease processes, especially complex syndromes such as cardiovascular disease, whose etiologies are due to multiple genetic factors and their interplay with the environment. Microarray and SAGE have already been used to examine gene expression patterns of cell-culture, animal and human tissues models of cardiovascular diseases. In this review, we will first give a brief introduction of microarray and SAGE technologies and point out their limitations. We will then discuss the major discoveries and the new biological insightsthat have emerged from their applications to cardiovascular diseases. 9Putrescine, spermidine, and spermine play distinct roles in rice salt tolerance显示文摘Polyamines(PAs) play diverse roles in plant growth and development,as well as responses to environmental stimuli.In this study,the effects of PAs on rice salt tolerance were investigated.Salt stress resulted in the alteration of endogenous PAs levels in rice roots and leaves,where spermidine(Spd) and spermine(Spm) contents were increased,and putrescine(Put) content was decreased.RT-qPCR analysis revealed that PAs biosynthesis-related genes ADC1,ODC,and Arginase were significantly downregulated by salt treatment;however,SAMDC transcription was significantly upregulated.Exogenous Spm enhanced rice salt tolerance remarkably;however,exogenous Put and Spd undermined rice salt tolerance.Transgenic rice plants overexpressing SAMDC display a higher ratio of Spm/(Put+Spd) and enhanced salt tolerance.Salt stress also increased polyamine oxidase activities in rice,resulting in elevated reactive oxygen species(ROS) production.Our findings revealed that accumulation of Put and Spd substantially reduced salt tolerance in rice,likely by facilitating ROS production;whereas,conversion of Put and Spd to Spm contributes to rice salt tolerance.Md Azizul ISlam PANG Jin-huan MENG Fan-wei LI Ya-wen XU Ning YANG Chao LIU Jun 2020Journal of Integrative Agriculture2020,19,3:5
10Cyber Attack Protection and Control of Microgrids显示文摘Recently, the smart grid has been considered as a next-generation power system to modernize the traditional grid to improve its security, connectivity, efficiency and sustainability.Unfortunately, the smart grid is susceptible to malicious cyber attacks, which can create serious technical, economical, social and control problems in power network operations. In contrast to the traditional cyber attack minimization techniques, this paper proposes a recursive systematic convolutional(RSC) code and Kalman filter(KF) based method in the context of smart grids.Specifically, the proposed RSC code is used to add redundancy in the microgrid states, and the log maximum a-posterior is used to recover the state information, which is affected by random noises and cyber attacks. Once the estimated states are obtained by KF algorithm, a semidefinite programming based optimal feedback controller is proposed to regulate the system states, so that the power system can operate properly. 13鞘内注射吗啡预处理激活大鼠δ、κ、μ受体致心肌保护显示文摘背景鞘内注射小剂量吗啡可产生与静脉注射吗啡相似的心肌保护作用,但鞘内注射吗啡预处理(IT—MPC)与缺血预处理(IPC)作用强度的差别还未知,IT-MPC是否由阿片类受体所介导也未阐明。因此,本实验比较IT—MPC与IPC作用强度差别并探讨阿片类受体的作用。方法80只SD大鼠麻醉后开胸,成功置入鞘内导管后随机分为13组(n=6—7)。IPC组接受3次5分钟缺血-5分钟再灌注的心肌IPC(阻闭左冠状动脉),随之给予30分钟缺血-2小时再灌注诱导心肌缺血再灌注(IR)损伤。IT-MPC组按剂量不同分为4个亚组(分别是0.03、0.3、3及30μg/kg),以3次5分钟注射-5分钟间停的模式平均注入鞘内,随之诱导IR损伤。静脉注射吗啡预处理组(IV-MPC)组经静脉给予吗啡300μg/kg,对照组鞘内给予10μl生理盐水。受体阻滞剂组选择在IT—MPC(3μg/kg)前分别给予选择性δ、κ、μ受体阻滞剂NTD、nor-BNI及CTOP,以评价阿片类受体亚型的作用。通过2,3,5-tripheny ltetrazolium染色,计算心肌梗死面积(IS),即梗死心肌占缺血危险区(AAR)的百分比。结果与对照组相比,鞘内注射0.3~30μg/kg吗啡降低心肌IS,3组IS/AAR分别为33%±10%(0.3μg/kg)、29%±10%(3μg/kg)、29%±16%(30μg/kg),对照组为53%±8%(P〈0.01)。IT-MPC降低心肌IS/AAR程度与IV-MPC(33%±6%,P=0.84)及IPC(22%±4%,P=0.41)相近。注射选择性受体阻滞剂后,由IT-MPC(3μg/kg)产生的心肌预处理效果降低(NTD+IT-MPC,50%±9%:nor-BNI+IT-MPc,43%±6%;CTOP+IT—MPC,53%±9%;与对照组相比,P=0.14)。结论IT—MPC产生与IPC及IV—MPC相近的心肌保护作用,其机制涉及到δ、κ、μ受体。Rui Li Gordon T. 14The α_Mβ_2 integrin and its role in neutrophil function显示文摘Neutrophils are the first cell type to arrive at the in-jury sites and play a critical role in host defense, by virtueof its ability to adhere and transmigrate through endothe-lium, to phagocytose foreign pathogens, and to producefree oxygen radicals and proteolytic enzymes. Yet, inap-propriate neutrophil activation causes tissue damage andvarious inflammatory diseases. These physiological andpathological functions of neutrophils depend on the en-gagement of certain surface receptors, especially αMβ2,the ma jor β2 integrin receptor present on neutrophil sur-face. 16Advances in the development of molecular genetic tools for Mycobacterium tuberculosis显示文摘Mycobacterium tuberculosis, a Gram-positive bacterium of great clinical relevance, is a lethal pathogen owing to its complex physiological characteristics and development of drug resistance. Several molecular genetic tools have been developed in the past few decades to study this microorganism. These tools have been instrumental in understanding how M. tuberculosis became a successful pathogen. Advanced molecular genetic tools have played a significant role in exploring the complex pathways involved in M. tuberculosis pathogenesis. Here, we review various molecular genetic tools used in the study of M. tuberculosis. Further, we discuss the applications of clustered regularly interspaced short palindromic repeat interference(CRISPRi), a novel technology recently applied in M. tuberculosis research to study target gene functions.
