| 1 | Health risks associated with heavy metals in the drinking water of Swat,northern Pakistan显示文摘The concentrations of heavy metals such as Cd, Cr, Cu, Mn, Ni, Pb and Zn were investigated in drinking water sources(surface and groundwater) collected from Swat valley, Khyber Pakhtunkhwa, Pakistan. The potential health risks of heavy metals to the local population and their possible source apportionment were also studied. Heavy metal concentrations were analysed using atomic absorption spectrometer and compared with permissible limits set by Pakistan Environmental Protection Agency and World Health Organization. The concentrations of Cd, Cr, Ni and Pb were higher than their respective permissible limits, while Cu, Mn and Zn concentrations were observed within their respective limits. Health risk indicators such as chronic daily intake(CDI) and health risk index(HRI) were calculated for adults and children separately. CDIs and HRIs of heavy metals were found in the order of Cr > Mn >Ni > Zn > Cd > Cu > Pb and Cd > Ni > Mn > Cr > Cu > Pb > Zn, respectively. HRIs of selected heavy metals in the drinking water were less than 1, indicating no health risk to the local people. | 2 | Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism. | Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu | 2019 | National Science Review2019,6,6: | 9 |

| 5 | Behavioral interventions to eliminate fear responses显示文摘Fear memory underlies anxiety-related disorders, including posttraumatic stress disorder(PTSD). PTSD is a fear-based disorder,characterized by difficulties in extinguishing the learned fear response and maintaining extinction. Currently, the first-line treatment for PTSD is exposure therapy, which forms an extinction memory to compete with the original fear memory. However,the extinguished fear often returns under numerous circumstances, suggesting that novel methods are needed to eliminate fear memory or facilitate extinction memory. This review discusses research that targeted extinction and reconsolidation to manipulate fear memory. Recent studies indicate that sleep is an active state that can regulate memory processes. We also discuss the influence of sleep on fear memory. For each manipulation, we briefly summarize the neural mechanisms that have been identified in human studies. | 6 | Climate change induced eutrophication of cold-water lake in an ecologically fragile nature reserve显示文摘Aquatic ecosystem sustainability around the globe is facing crucial challenges because of increasing anthropogenic and natural disturbances. In this study, the Tianchi Lake, a typical cold-water lake and a UNESCO/MAB(Man and Biosphere) nature reserve located in high latitude and elevation with the relatively low intensity of human activity was chosen as a system to examine the linkages between climate change and eutrophication. As a part of the UNESCO Bogda Man and Biosphere Reserve, Tianchi Lake has been well preserved for prevention from human intervention, but why has it been infected with eutrophication recent years? Our results show that climate change played a significant role in the eutrophication in the Tianchi Lake. Increased temperature, changed precipitation pattern and wind-induced hydrodynamic fluctuations in the summer season were suggested to make a major contribution to the accelerated eutrophication. The results also showed that the local temperature and precipitation changes were closely linked to the large-scale atmospheric circulation, which opens the door for the method to be applied in other regions without local climatic information. | 7 | Role of digestive protease enzymes and related genes in host plant adaptation of a polyphagous pest,Spodoptera frugiperda显示文摘The evolutionary success of phytophagous insects depends on their ability toefficiently exploit plants as a source of energy for survival.Herbivorous insects largelydepend on the efficiency,flexibility,and diversity of their digestive physiology and sophistication of their detoxification system to use chemically diverse host plants as foodsources.The fall armyworm,Spodoptera frugiperda(J.E.Smith),is a polyphagous pest ofmany commercially important crops.To elucidate the ability of this insect pest to adaptto host plant mechanisms,we evaluated the impact of primary(corn)and alternate(rice)host plants after 1l generations on gut digestive enzymatic activity and expression profiles of related genes.Results indicated that the total protease and classspecific trypsinand chymotrypsinlike protease activity of S.frugiperda significantly differed among hostplant treatments.The classspecifiq protease profiles greatly differed in S.frugiperdamidguts upon larval exposure to different treatments with inhibitors compared with treatments without inhibitors.Similarly,the single and cumulative effects of the enzymespecific inhibitors TLCK,TPCK,and E64 significantly increased larval mortality and reduced larval growth/mass across different plant treatments.Furthermore,the quantitativereverse transcription polymerase chain reaction results revealed increased transcription oftwo trypsin(SfTry3,SfTry7)and one chymotrypsin gene(Sfchym9),which indicatedthat they have roles in host plant adaptation.Knockdown of these genes resulted in significantly reduced mRNA expression levels of the trypsin genes.This was related to theincreased mortality observed in treatments compared with the dsRED control.This resultindicates possible roles of S.frugiperda gut digestive enzymes and related genes in hostplant adaptation. | Muhammad Hafeez Xiao-Wei Li Jin-Ming Zhang Zhi-Jun Zhang Jun Huang Li-Kun Wang Muhammad Musa Khan Sakhawat Shah G.Mandela Fernandez-Grandon Yao-Bin Lu | 2021 | Insect Science2021,28,3: | 2 |
