1Diarrhoea-predominant irritable bowel syndrome distinguishable by 16S rRNA gene phylotype quantification显示文摘AIM:To study whether selected bacterial 16S ribosomal RNA(rRNA)gene phylotypes are capable of disting- uishing irritable bowel syndrome(IBS). METHODS:The faecal microbiota of twenty volunteers with IBS,subdivided into eight diarrhoea-predominant (IBS-D),eight constipation-predominant(IBS-C)and four mixed symptom-subtype(IBS-M)IBS patients,and fifteen control subjects,were analysed at three time-points with a set of fourteen quantitative real-timepolymerase chain reaction assays.All assays targeted 16S rRNA gene phylotypes putatively associated with IBS,based on 16S rRNA gene library sequence analysis. The target phylotypes were affiliated with Actinobac-teria,Bacteroidetes and Firmicutes.Eight of the target phylotypes had less than 95%similarity to cultured bacterial species according to their 16S rRNA gene sequence.The data analyses were made with repeated-measures ANCOVA-type modelling of the data and principle component analysis(PCA)with linear mixed-effects models applied to the principal component scores. RESULTS:Bacterial phylotypes Clostridium cocleatum 88%,Clostridium thermosuccinogenes 85%,Coprobacillus catenaformis 91%,Ruminococcus bromii-like, Ruminococcus torques 91%,and R.torques 93%were detected from all samples analysed.A multivariate analysis of the relative quantities of all 14 bacterial 16S rRNA gene phylotypes suggested that the intestinal microbiota of the IBS-D patients differed from other sample groups.The PCA on the first principal component(PC1),explaining 30.36%of the observed variation in the IBS-D patient group,was significantly altered from all other sample groups(IBS-D vs control, P=0.01;IBS-D vs IBS-M,P=0.00;IBS-D vs IBS-C, P=0.05).Significant differences were also observed in the levels of distinct phylotypes using relative values in proportion to the total amount of bacteria.A phy- lotype with 85%similarity to C.thermosuccinogenes was quantified in significantly different quantities among the IBS-D and control subjects(-4.08±0.90 vs -3.33±1.16,P=0.04)and IBS-D and IBS-M subjects (-4.08±0.90 vs-3.08±1.38,P=0.05).Furthermore,a phylotype with 94%similarity to R.torques was more prevalent in IBS-D patients'intestinal micro- biota than in that of control subjects(-2.43±1.49 vs -4.02±1.63,P=0.01).A phylotype with 93%simi- larity to R.torques was associated with control sam- ples when compared with IBS-M(-2.41±0.53 vs -2.92±0.56,P=0.00).Additionally,a R.bromii-like phylotype was associated with IBS-C patients in com- parison to control subjects(-1.61±1.83 vs-3.69± 2.42,P=0.01).All of the above mentioned phylotype specific alterations were independent of the effect of time. 3Identification of four novel DC-SIGN ligands on Mycobacterium bovis BCG显示文摘Dendritic-cell-specific intercellular adhesion molecule-3-grabbing non-integrin(DC-SIGN;CD209)has an important role in mediating adherence of Mycobacteria species,including M.tuberculosis and M.bovis BCG to human dendritic cells and macrophages,in which these bacteria can survive intracellularly.DC-SIGN is a C-type lectin,and interactions with mycobacterial cells are believed to occur via mannosylated structures on the mycobacterial surface.Recent studies suggest more varied modes of binding to multiple mycobacterial ligands.Here we identify,by affinity chromatography and mass-spectrometry,four novel ligands of M.bovis BCG that bind to DC-SIGN.The novel ligands are chaperone protein DnaK,60 kDa chaperonin-1(Cpn60.1),glyceraldehyde-3 phosphate dehydrogenase(GAPDH)and lipoprotein lprG.Other published work strongly suggests that these are on the cell surface.Of these ligands,lprG appears to bind DC-SIGN via typical proteinglycan interactions,but DnaK and Cpn60.1 binding do not show evidence of carbohydrate-dependent interactions.LprG was also identified as a ligand for DC-SIGNR(L-SIGN;CD299)and the M.tuberculosis orthologue of lprG has been found previously to interact with human toll-like receptor 2.Collectively,these findings offer new targets for combating mycobacterial adhesion and within-host survival,and reinforce the role of DCSIGN as an important host ligand in mycobacterial infection.Maria V.Carroll Robert B.Sim Fabiana Bigi Anne Jäkel Robin Antrobus Daniel A.Mitchell 2010Protein & Cell2010,1,9:7

5Keratinocytes costimulate naive human T cells via CD2: a potential target to prevent the development of proinflammatory Th1 cells in the skin显示文摘The interplay between keratinocytes and immune cells,especially T cells,plays an important role in the pathogenesis of chronic inflammatory skin diseases.During psoriasis,keratinocytes attract T cells by releasing chemokines,while skin-infiltrating selfreactive T cells secrete proinflammatory cytokines,e.g.,IFN γand IL-17A,that cause epidermal hyperplasia.Similarly,in chronic graftversus-host disease,allogenic IFN γ-producing Th1/Tc1 and IL-17-producing Th17/Tc17 cells are recruited by keratinocyte-derived chemokines and accumulate in the skin.However,whether keratinocytes act as nonprofessional antigen-presenting cells to directly activate naive human T cells in the epidermis remains unknown.Here,we demonstrate that under proinflammatory conditions,primary human keratinocytes indeed activate naive human T cells.This activation required cell contact and costimulatory signaling via CD58/CD2 and CD54/LFA-1.Naive T cells costimulated by keratinocytes selectively differentiated into Th1 and Th17 cells.In particular,keratinocyte-initiated Th1 differentiation was dependent on costimulation through CD58/CD2.The latter molecule initiated STAT1 signaling and IFN γproduction in T cells.Costimulation of T cells by keratinocytes resulting in Th1 and Th17 differentiation represents a new explanation for the local enrichment of Th1 and Th17 cells in the skin of patients with a chronic inflammatory skin disease.Consequently,local interference with T cell–keratinocyte interactions may represent a novel strategy for the treatment of Th1 and Th17 cell-driven skin diseases.Christian Orlik Daniel Deibel Johanna Küblbeck Emre Balta Sabina Ganskih Jüri Habicht Beate Niesler Jutta Schröder-Braunstein Knut Schäkel Guido Wabnitz Yvonne Samstag 2020Cellular & Molecular Immunology2020,17,4:4
6Surface-bound myeloperoxidase is a ligand for recognition of late apoptotic neutrophils by human lung surfactant proteins A and D显示文摘Surfactant proteins A(SP-A)and D(SP-D),both members of the collectin family,play a well established role in apoptotic cell recognition and clearance.Recent in vitro data show that SP-A and SP-D interact with apoptotic neutrophils in a distinct manner.SP-A and SP-D bind in a Ca^(2+)-dependent manner to viable and early apoptotic neutrophils whereas the much greater interaction with late apoptotic neutrophils is Ca^(2+)-independent.Cell surface molecules on the apoptotic target cells responsible for these interactions had not been identified and this study was done to find candidate target molecules.Myeloperoxidase(MPO),a specific intracellular defense molecule of neutrophils that becomes exposed on the outside of the cell upon apoptosis,was identified by affinity purification,mass-spectrometry and western blotting as a novel binding molecule for SP-A and SP-D.To confirm its role in recognition,it was shown that purified immobilised MPO binds SP-A and SP-D,and that MPO is surface-exposed on late apoptotic neutrophils.SP-A and SP-D inhibit binding of an anti-MPO monoclonal Ab to late apoptotic cells.Fluorescence microscopy confirmed that anti-MPO mAb and SP-A/SP-D colocalise on late apoptotic neutrophils.Desmoplakin was identified as a further potential ligand for SP-A,and neutrophil defensin as a target for both proteins.Anne Jäkel Howard Clark Kenneth B.M.Reid Robert B.Sim 2010Protein & Cell2010,1,6:2
7Surfactant protein A (SP-A) binds to phosphatidylserine and competes with annexin V binding on late apoptotic cells显示文摘The role of surfactant protein A(SP-A)in the recognition and clearance of apoptotic cells is well established,but to date,it is still not clear which surface molecules of apoptotic cells are involved in the process.Here we present evidence that phosphatidylserine(PS)is a relevant binding molecule for human SP-A.The binding is Ca^(2+)-dependent and is not inhibited by mannose,suggesting that the sugar-binding site of the carbohydrate recognition domain(CRD)of SP-A is not involved.Flow cytometry studies on apoptotic Jurkat cells revealed apparent inhibition of annexin V binding by increasing concentrations of SP-A in late apoptotic but not early apoptotic cells,and this was consistent for Jurkat cells and neutrophils.Supporting these data,confocal microscopy results show a co-localisation of annexin V and SP-A in late apoptotic but not early apoptotic cells.However,we cannot conclude that this inhibition is exclusively due to the binding of SP-A to PS on the cell surface,as annexin V is not wholly specific for PS and SP-A also interacts with other phospholipids that might become exposed on the apoptotic cell surface.Anne Jäkel Kenneth B.M.Reid Howard Clark 2010Protein & Cell2010,1,2:2
8Factors explaining the abundance of rodents in the city of Luang Prabang,Lao PDR,as revealed by field and household surveys显示文摘A field and a household survey, the latter of which included inspections and interviews with the residents of a totalof 1370 properties, were conducted in 2004 in 30 villages of the city of Luang Prabang, Lao PDR, in order to assessthe degree of rodent infestation and to identify potential factors influencing infestations. Roof rats, Rattus rattus,and the Polynesian rat, Rattus exulans, were the only rodents found in the city, and trapping results showed a cleardominance of roof rats (80–90% of all individuals). Measurements of rodent activity using tracking patchescorrelated positively with the trapping data, and revealed a significantly higher degree of rat infestation during therainy season (September) than during the dry season (November). If households in the vicinity of the samplinglocations were considered, villagers’ accounts of indoor rodent infestations recorded during the household surveycorrelated positively with measurements of rodent activity. At least every second household reported indoorinfestations. Using explorative statistical analyses (classification trees, factor analysis) we checked the predictiveor explanatory value of up to 28 variables assessed during household inspections for villagers’ observations onrodent infestation as the dependent variable. Trophic factors such as exposed food (indoors) and garbage (outdoors),and structural features such as open ceilings (indoors) and rat harborage in gardens (outdoors) ranked highest asexplanatory variables. Assessment of a small sample of roof rat droppings collected inside houses revealed thepresence of the potential disease agents Salmonella javiana, Cryptosporidium parvum, Giardia duodenalis and theparasitic nematode Calodium hepaticum (syn. Capillaria hepatica).
