5The versatile functions of Sox9 in development,stem cells,and human diseases显示文摘The transcription factor Sox9 was first discovered in patients with campomelic dysplasia,a haploinsufficiency disorder with skeletal deformities caused by dysregulation of Sox9 expression during chondrogenesis.Since then,its role as a cell fate determiner during embryonic development has been well characterized;Sox9 expression differentiates cells derived from all three germ layers into a large variety of specialized tissues and organs.However,recent data has shown that ectoderm-and endoderm-derived tissues continue to express Sox9 in mature organs and stem cell pools,suggesting its role in cell maintenance and specification during adult life.The versatility of Sox9 may be explained by a combination of posttranscriptional modifications,binding partners,and the tissue type in which it is expressed.Considering its importance during both development and adult life,it follows that dysregulation of Sox9 has been implicated in various congenital and acquired diseases,including fibrosis and cancer.This review provides a summary of the various roles of Sox9 in cell fate specification,stem cell biology,and related human diseases.Ultimately,understanding the mechanisms that regulate Sox9 will be crucial for developing effective therapies to treat disease caused by stem cell dysregulation or even reverse organ damage.Alice Jo Sahitya Denduluri Bosi Zhang Zhongliang Wang Liangjun Yin Zhengjian Yan Richard Kang Lewis L.Shi James Mok Michael J.Lee Rex C.Haydon 2014Genes & Diseases2014,1,2:16
6Adjuvant heparanase inhibitor PI-88 therapy for hepatocellular carcinoma recurrence显示文摘AIM: To demonstrate that administering heparanase inhibitor PI-88 at 160 mg/d is safe and promising in reducing hepatocellular carcinoma(HCC) recurrence for up to 3 year following curative resection. METHODS: A total of 143 patients(83.1% of the 172 participants in the phase Ⅱ study) participated in the follow-up study. Of these patients, 50 had received no treatment, 48 had received 160 mg/d PI-88, and 45 had received 250 mg/d PI-88 during the phase Ⅱ trial. Safety parameters and the following efficacy endpoints were investigated:(1) time to recurrence;(2) diseasefree survival; and(3) overall survival. RESULTS: PI-88 at 160 mg/d delayed the onset and frequency of HCC recurrence, and provided a clinically significant survival advantage for up to 3 years after treatment compared with those of the control group:(1) the recurrence-free rate increased from 50% to 63%, and(2) time to recurrence at the 36 th percentile was postponed by 78%. The efficacy of administering PI-88 at 250 mg/d was confounded by a high dropout rate(11 out of 54 patients). Additionally, subgroup analyses of patients with(1) multiple tumors or a single tumor ≥ 2 cm; and(2) hepatitis B or C revealed that administering PI-88 at 160 mg/d conferred the most significant survival advantage(56.8% improvement in disease-free survival, P = 0.045) for patients with both risk factors for recurrence. 8Expression of protein S100A4 is a predictor of recurrence in colorectal cancer显示文摘AIM: To investigate the prognostic significance of S100A4 expression in colorectal cancer and its correlation with expression of E-cadherin and p53. METHODS: A cohort of archival formalin-fixed paraff inembedded specimens was selected from 127 patients with colorectal cancer who underwent surgical resection between April 2000 and March 2004 at the Department of Surgery, Korea University Guro Hospital. The expression of protein S100A4 was evaluated according to the proportion of positively stained cancer cells. In each case, three core biopsies with a diameter of 2 mm were punched out and positioned in a recipient paraff in array block. Four-μm sections of these tissue array blocks were used for immunohistochemical analysis of protein S100A4, E-cadherin, and p53. Clinicopathological data were based on the original histopathologic reports and clinical records of patients. RESULTS: In normal colorectal mucosa, protein S100A4 immunoreactivity was clearly absent in both cytoplasm and nucleus. However, positive immunoreactivity of protein S100A4 was detected in 45 (35.4%) of the tumor cases. There was no significant association between positive immunoreactivity of protein S100A4 and clinicopathological parameters such as tumor differentiation or TNM stage, and also no correlation between the reactivity and E-cadherin or p53 expression. However, positive immunoreactivity of protein S100A4 was found to be associated with tumor recurrence (P = 0.004), and was also associated with significantly worse overall survival in the Kaplan-Meyer survival analysis (P = 0.044). After adjustment for tumor differentiation, tumor depth and nodal status, however, it failed to achieve statistical significance (P = 0.067). 9Green tea and the risk of gastric cancer: Epidemiological evidence显示文摘Gastric cancer (GC) is one of the leading causes of cancer death in the world. Numerous efforts are being made to find chemoprotective agents able to reduce its risk. Amongst these, green tea has been reported to have a protective effect against stomach cancer. This article aims to critically evaluate all epidemiological studies reporting an association between green tea consumption and GC risk. MEDLINE, EBSCOHOST and Google Scholar were used to search for clinical trials of green tea and its correlation to stomach cancer. Studies include cohort and case-control studies. Outcome of interests are inverse association, no association, and positive association. Seventeen epidemiologic studies were reviewed. Eleven studies were conducted in Japan, five in China, and one with Japanese descendent in Hawaii. Ten case-control studies and seven cohortstudies were included. The relative risks or odds ratio of GC for the highest level of green tea consumption was compared. Seven studies suggested no association, eight an inverse association, and one a positive association. One study had shown a significantly lowered GC risk when tea was served warm to cold. Another study also showed a significantly risk with lukewarm tea. All studies that analyzed men and women separately have suggested a reduced risk in women than in men, albeit no significant difference. This review demonstrates that there is insufficient information to support green tea consumption reduces the risk of GC. 13Gravitational wave astrophysics, data analysis and multimessenger astronomy显示文摘This paper reviews gravitational wave sources and their detection. One of the most exciting potential sources of gravitational waves are coalescing binary black hole systems. They can occur on all mass scales and be formed in numerous ways, many of which are not understood. They are generally invisible in electromagnetic waves, and they provide opportunities for deep investigation of Einstein's general theory of relativity. Sect. 1 of this paper considers ways that binary black holes can be created in the universe, and includes the prediction that binary black hole coalescence events are likely to be the first gravitational wave sources to be detected. The next parts of this paper address the detection of chirp waveforms from coalescence events in noisy data.Such analysis is computationally intensive. Sect. 2 reviews a new and powerful method of signal detection based on the GPUimplemented summed parallel infinite impulse response filters. Such filters are intrinsically real time alorithms, that can be used to rapidly detect and localise signals. Sect. 3 of the paper reviews the use of GPU processors for rapid searching for gravitational wave bursts that can arise from black hole births and coalescences. In sect. 4 the use of GPU processors to enable fast efficient statistical significance testing of gravitational wave event candidates is reviewed. 14Multiple malignant extragastrointestinal stromal tumors of the greater omentum and results of immunohistochemistry and mutation analysis:A case report显示文摘To report an extragastrointestinal stromal tumor (EGIST) that occurs outside the gastrointestinal tract and shows unique clinicopathologic and immunohistochemical features. In our case, we experienced multiple soft tissue tumors that originate primarily in the greater omentum, and in immunohistochemical analysis, the tumors showed features that correspond to malignant EGIST. Two large omental masses measured 15 cm x 10 cm and 5 cm × 4 cm sized and several small ovoid fragments were attached to small intestine, mesentery and peritoneum. On histologic findings, the masses were separated from small bowel serosa and had high mitotic count (115/50 HPFs). In the results of immunohistochemical stains, the tumor showed CD117 (c-kit) positive reactivity and high Ki-67 labeling index. On mutation analysis, the c-kit gene mutation was found in the juxtamembrane domain (exon 11) and it was heterozygote. Platelet-derived growth factor receptor (PDGFR) gene mutation was also found in the juxtamemembrane (exon 12) and it was polymorphism. 16Polo-like kinase 1,a new therapeutic target in hepatocellular carcinoma显示文摘AIM:To investigate the role of polo-like kinase 1 (PLK1) as a therapeutic target for hepatocellular carcinoma (HCC).METHODS:PLK1 gene expression was evaluated in HCC tissue and HCC cell lines.Gene knockdown with short-interfering RNA (siRNA) was used to study PLK1 gene and protein expression using real-time reverse transcription polymerase chain reaction (RT-PCR) and Western blotting,and cell proliferation using 3-(4,5-dim ethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2(4-sulf ophenyl)-2H-tetrazolium (MTS) and bromodeoxyuridine (BrdU) assays.Apoptosis was evaluated using the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay,and caspase-inhibition assay.Huh-7 cells were transplanted into nude mice and co-cultured with PLK1 siRNA or control siRNA,and tumor progression was compared with controls.RESULTS:RT-PCR showed that PLK1 was overexpressed 12-fold in tumor samples compared with controls,and also was overexpressed in Huh-7 cells.siRNA against PLK1 showed a reduction in PLK1 gene and protein expression of up to 96% in Huh-7 cells,and a reduction in cell proliferation by 68% and 92% in MTS and BrdU cell proliferation assays,respectively.There was a 3-fold increase in apoptosis events,and TUNEL staining and caspase-3 assays suggested that this was caspase-independent.The pan-caspase inhibitor Z-VAD-FMK was unable to rescue the apoptotic cells.Immnofluorescence co-localized endonuclease-G to fragmented chromosomes,implicating it in apoptosis.Huh-7 cells transplanted subcutaneously into nude mice showed tumor regression in siPLK1-treated mice,but not in controls.CONCLUSION:Knockdown of PLK1 overexpression in HCC was shown to be a potential therapeutic target,leading to apoptosis through the endonuclease-G pathway.Wei Chuen Mok Shanthi Wasser Theresa Tan Seng Gee Lim 2012World Journal of Gastroenterology2012,18,27:4
17Secreted modular calcium-binding proteins in pathophysiological processes and embryonic development显示文摘Objective:Secreted modular calcium-binding proteins (SMOCs) are extracellular glycoproteins of the secreted protein, acidic, and rich in cysteine-related modular calcium-binding protein family and include two isoforms, SMOC1 and SMOC2, in humans. Functionally, SMOCs bind to calcium for various cell functions. In this review, we provided a summary of the most recent advancements and findings of SMOC1 and SMOC2 in development, homeostasis, and disease states.Data sources:All publications in the PubMed database were searched and retrieved (up to July 24, 2019) using various combinations of keywords searching, including SMOC1, SMOC2, and diseases.Study selection:All original studies and review articles of SMOCs in human diseases and embryo development written in English were retrieved and included.Results:SMOC1 and SMOC2 regulate embryonic development, cell homeostasis, and disease pathophysiology. 18The D253N Mutation in the Polymerase Basic 2 Gene in Avian Influenza(H9N2) Virus Contributes to the Pathogenesis of the Virus in Mammalian Hosts显示文摘Mutations in the polymerase basic 2(PB2) gene of avian influenza viruses are important signatures for their adaptation to mammalian hosts. Various adaptive mutations have been identified around the 627 and nuclear localization sequence(NLS) domains of PB2 protein, and these mutations contribute to the replicative ability of avian influenza viruses.However, few studies have focused on adaptive mutations in other regions of PB2. In this study, we investigated the functional roles of the D253N mutation in PB2 in an H9N2 virus. This mutation was found to affect an amino acid residue in the middle domain of the PB2 protein. The virus with the D253N mutation showed higher polymerase activity and transiently increased viral replication in human cells. However, the mutant did not show significant differences in viral replication in the respiratory tract of mice upon infection. 19Decomposition of trifluoromethane in a dielectric barrier discharge non-thermal plasma reactor显示文摘The decomposition of trifluoromethane (CHF3) was carried out using non-thermal plasma generated in a dielectric barrier discharge (DBD) reactor. The effects of reactor temperature, electric power, initial concentration and oxygen content were examined. The DBD reactor was able to completely destroy CHF3 with alumina beads as a packing material. The decomposition efficiency increased with increasing electric power and reactor temperature. The destruction of CHF3 gradually increased with the addition of O2 up to 2%, but further increase in the oxygen content led to a decrease in the decomposition efficiency. The degradation pathways were explained with the identified by-products. The main by-products from CHF3 were found to be COF2, CF4, CO2 and CO although the COF2 and CF4 disappeared when the plasma were combined with alumina catalyst.M. Sanjeeva Gandhi Y. S.
