| 1 | Global kinematics of tectonic plates and subduction zones since the late Paleozoic Era显示文摘Detailed global plate motion models that provide a continuous description of plate boundaries through time are an effective tool for exploring processes both at and below the Earth's surface. A new generation of numerical models of mantle dynamics pre-and post-Pangea timeframes requires global kinematic descriptions with full plate reconstructions extending into the Paleozoic(410 Ma). Current plate models that cover Paleozoic times are characterised by large plate speeds and trench migration rates because they assume that lowermost mantle structures are rigid and fixed through time. When used as a surface boundary constraint in geodynamic models, these plate reconstructions do not accurately reproduce the present-day structure of the lowermost mantle. Building upon previous work, we present a global plate motion model with continuously closing plate boundaries ranging from the early Devonian at 410 Ma to present day.We analyse the model in terms of surface kinematics and predicted lower mantle structure. The magnitude of global plate speeds has been greatly reduced in our reconstruction by modifying the evolution of the synthetic Panthalassa oceanic plates, implementing a Paleozoic reference frame independent of any geodynamic assumptions, and implementing revised models for the Paleozoic evolution of North and South China and the closure of the Rheic Ocean. Paleozoic(410-250 Ma) RMS plate speeds are on average ~8 cm/yr, which is comparable to Mesozoic-Cenozoic rates of ~6 cm/yr on average.Paleozoic global median values of trench migration trend from higher speeds(~2.5 cm/yr) in the late Devonian to rates closer to 0 cm/yr at the end of the Permian(~250 Ma), and during the Mesozoic-Cenozoic(250-0 Ma) generally cluster tightly around ~1.1 cm/yr. Plate motions are best constrained over the past 130 Myr and calculations of global trench convergence rates over this period indicate median rates range between 3.2 cm/yr and 12.4 cm/yr with a present day median rate estimated at~5 cm/yr. For Paleozoic times(410-251 Ma) our model results in median convergence rates largely~5 cm/yr. Globally,~90% of subduction zones modelled in our reconstruction are determined to be in a convergent regime for the period of 120-0 Ma. Over the full span of the model, from 410 Ma to 0 Ma,~93% of subduction zones are calculated to be convergent, and at least 85% of subduction zones are converging for 97% of modelled times. Our changes improve global plate and trench kinematics since the late Paleozoic and our reconstructions of the lowermost mantle structure challenge the proposed fixity of lower mantle structures, suggesting that the eastern margin of the African LLSVP margin has moved by as much as ~1450 km since late Permian times(260 Ma). | 2 | Role of functional imaging in the development and refinement of invasive neuromodulation for psychiatric disorders显示文摘Deep brain stimulation(DBS) is emerging as a pow-erful tool for the alleviation of targeted symptoms in treatment-resistant neuropsychiatric disorders. Despite the expanding use of neuropsychiatric DBS, the mecha-nisms responsible for its effects are only starting to be elucidated. Several modalities such as quantitative elec-troencephalography as well a intraoperative recordings have been utilized to attempt to understand the under-pinnings of this new treatment modality, but functional imaging appears to offer several unique advantages. Functional imaging techniques like positron emission tomography, single photon emission computed tomog-raphy and functional magnetic resonance imaging have been used to examine the effects of focal DBS on activ-ity in a distributed neural network. These investigations are critical for advancing the field of invasive neuro-modulation in a safe and effective manner, particularly in terms of defining the neuroanatomical targets and refining the stimulation protocols. The purpose of this review is to summarize the current functional neuroim-aging findings from neuropsychiatric DBS implantation for three disorders: treatment-resistant depression, obsessive-compulsive disorder, and Tourette syndrome. All of the major targets will be discussed(Nucleus ac-cumbens, anterior limb of internal capsule, subcallosal cingulate, Subthalamic nucleus, Centromedial nucleus of the thalamus-Parafasicular complex, frontal pole, and dorsolateral prefrontal cortex). | 3 | Transcriptomic analysis in Anemone flaccida rhizomes reveals ancillary pathway for triterpene saponins biosynthesis and differential responsiveness to phytohormones显示文摘Anemone flaccida Fr. Schmidt is a perennial medicinal herb that contains pentacyclic triterpenoid saponins as the major bioactive constituents. In China, the rhizomes are used as treatments for a variety of ailments including arthritis. However, yields of the saponins are low, and little is known about the plant's genetic background or phytohormonal responsiveness. Using one-quarter of the 454 pyrosequencing information from the Roche GS FLX Titanium platform, we performed a transcriptomic analysis to identify 157 genes putatively encoding 26 enzymes involved in the synthesis of the bioactive compounds. It was revealed that there are two biosynthetic pathways of triterpene saponins in A. flaccida. One pathway depends on β-amyrin synthase and is similar to that found in other plants. The second, subsidiary('backburner') pathway is catalyzed by camelliol C synthase and yields β-amyrin as minor byproduct. Both pathways used cytochrome P450-dependent monooxygenases(CYPs) and family 1 uridine diphosphate glycosyltransferases(UGTs) to modify the triterpenoid backbone. The expression of CYPs and UGTs were quite different in roots treated with the phytohormones methyl jasmonate, salicylic acid and indole-3-acetic acid. | 4 | Efficacy of Lycium barbarum polysaccharide in adolescents with subthreshold depression: interim analysis of a randomized controlled study显示文摘Subthreshold depression is a highly prevalent condition in adolescents who are at high risk for developing major depressive disorder.In preclinical models of neurological and psychiatric diseases,Lycium barbarum polysaccharide(LBP)extracted from Goji berries had antidepressant effects including but not limited to anti-oxidative and anti-inflammatory properties.However,the effect of LBP on subthreshold depression is unclear.To investigate the clinical efficacy and safety of LBP for treating subthreshold depression in adolescents,we conducted a randomized,double-blind,placebo-controlled trial(RCT)with 29 adolescents with subthreshold depression recruited at The Fifth Affiliated Hospital of Guangzhou Medical University.The participants were randomly assigned to groups where they received either 300 mg LBP(LBP group,n=15,3 boys and 12 girls aged 15.13±2.17 years)or a placebo(placebo group,n=14,2 boys and 12 girls aged 15±1.71 years)for 6 successive weeks.Interim analyses revealed that the LBP group exhibited a greater change in Hamilton Depression Scale(HAMD-24)scores relative to the baseline and a higher remission rate(HAMD-24 total score≤7)at 6 weeks compared with the placebo group.Scores on the Beck Depression Inventory-II(BDI-II),Pittsburgh Sleep Quality Index(PSQI),Kessler Psychological Distress Scale(Kessler),and Screen for Child Anxiety-Related Emotional Disorders(SCARED)were similar between the LBP and placebo groups.No side effects related to the intervention were reported in either group.These results indicate that LBP administration reduced depressive symptoms in adolescents with subthreshold depression.Furthermore,LBP was well tolerated with no treatment-limiting adverse events.Clinical trials involving a larger sample size are needed to further confirm the anti-depressive effects of LBP in adolescents with subthreshold depression.This study was approved by the Medical Ethics Committee of the Fifth Affiliated Hospital of Guangzhou Medical University(Guangzhou,China;approval No.L2019-08)on April 4,2019 and was registered on ClinicalTrials.gov(identifier:NCT04032795)on July 25,2019. | Xiaoyue Li Xuan Mo Tao Liu Robin Shao Kayla Teopiz Roger S.McIntyre Kwok-Fai So Kangguang Lin | 2022 | Neural Regeneration Research2022,17,7: | 6 |

| 6 | Suppression of monosodium urate crystal-induced inflammation by inhibiting TGF-β-activated kinase 1-dependent signaling:role of the ubiquitin proteasome system显示文摘Monosodium urate(MSU)crystals activate inflammatory pathways that overlap with interleukin-1β(IL-1β)signaling.However,the post-translational mechanisms involved and the role of signaling proteins in this activation are unknown.In the present study,we investigated the intracellular signaling mechanisms involved in MSU-induced activation of THP-1 macrophages and human nondiseased synovial fibroblasts(NLSFs)and the in vivo efficacy of an inhibitor of tumor grovArth factor-β(TGF-β)-activated kinase 1(TAK1),5Z-7-oxozeaenoI,in MSU-induced paw inflammation in C57BL76 mice.THP-1 macrophage activation with MSU crystals(25-200 ng/ml)resulted in the rapid and sustained phosphorylation of interleukin-1 receptor-activated kinase 1(IRAK1 Thr^(209))and TAK1(Thr^(184/187))and their association with the E3 ubiquitin ligase TRAF6.At the cellular level,MSU inhibited the deubiquitinases A20 and UCHL2 and increased 20s proteasomal activity,leading to a global decrease in K^(63)-linked ubiquitination and increase in K^(48)-linked ubiquitination in THP-1 macrophages.While MSU did not stimulate cytokine produaion in NLSFs,it significantly amplified IL-1β-induced IL-6,IL-8,and ENA-78/CXCL5 production.Docking studies and MD simulations followed by TAK1 in vitro kinase assays revealed that uric acid molecules are capable of arresting TAK1 in an active-state conformation,resulting in sustained TAK1 kinase activation.Importantly,MSU-induced proinflammatory cytokine production was completely inhibited by 5Z-7-oxozeaenol but not IRAK1/4 or TRAF6 inhibitors.Administration of 5Z-7-oxozeaenol(5 or 15 mg/kg;orally)significantly inhibited MSU-induced paw inflammation in C57BL/6 mice.Our study identifies a novel post-translational mechanism of TAK1 activation by MSU and suggests the therapeutic potential of TAK1 in regulating MSU-induced Inflammation. | Anil K.Singh Mahamudul Haque Kayla O'Sullivan Mukesh Chourasia Madhu M.Ouseph Salahuddin Ahmed | 2021 | Cellular & Molecular Immunology2021,18,1: | 2 |

| 11 | Challenges in the microbiological food safety of fresh produce:Limitations of post-harvest washing and the need for alternative interventions显示文摘Fresh produce(processed fruit and vegetables)continues to be the main source of foodborne illness outbreaks implicating pathogens such as Escherichia coli O157:H7,Salmonella,Listeria monocytogenes and human parasites(e.g.hepatitis A,Cyclospora).Previously,outbreaks were primarily limited to leafy greens,tomatoes,and cantaloupes,but more recently there has been a trend of more diverse produce types(e.g.cucumbers and papayas)being implicated.Although on-farm good agriculture practices(GAP)contribute to preventing pathogens entering the fresh produce chain,it cannot be relied upon completely due to the open nature of farming.As a consequence,there is an identified need for interventions that can remove field-acquired contamination,especially given fresh produce is eaten raw.In the following review,an overview of foodborne illness outbreaks linked to contaminated fresh produce will be described along with potential sources of contamination.Post-harvest washing that was once considered decontamination is now viewed as a high-risk cross-contamination point.The challenges in monitoring the post-harvest wash process will be discussed along with processing factors that need to be considered.A range of alternative,or supplemental,non-aqueous interventions will be described including irradiation,ultraviolet light,high hydrostatic pressure,gas phase(ozone and chlorine dioxide),and hydroxyl radicals generated through advanced oxidative process or gas plasma.All have been proved to be effective at pathogen control on the laboratory scale and are poised to enter commercial application.The current status of these alternative interventions along with challenges of integrating into commercial practice will be described. | Kayla Murray Fan Wu John Shi Sophia Jun Xue Keith Warriner | 2017 | Food Quality and Safety2017,1,4: | 1 |

| 16 | Linking tree growth rate, damage repair, and susceptibility to a genus-specific pest infestation显示文摘Pest preference and subsequent susceptibility of a host individual is likely related to previous growth patterns in that host.Emerald ash borer(Agrilus planipennis Fairmaire)is a pestiferous beetle introduced to North America from Asia.While all species of ash are susceptible to attack,some individual trees appear to survive infestation.We selected ash trees in southeastern Michigan,collected cores and categorized trees as high tolerance to emerald ash borer attack(high overall health,low crown dieback),low tolerance(low overall health,high crown dieback)and intermediate tolerance(in-between the other categories).We artificially wounded trees and measured wound closure after 3 years.Ring width indices were not correlated between high and low tolerance trees.Regression slopes comparing growth and years were significantly different between the three tolerance categories,with high tolerance trees having the steepest slope.Wound closure was greatest in high tolerance trees.High tolerance trees demonstrating more rapid(steeper regression slope),consistent(lower variance),and effective(greater wound closure)growth.Those vigorously growing trees likely had more capacity to repair damage caused by emerald ash borer,leading to healthier trees in our categorization.Linking previous host growth patterns to health may have implications related to identifying individual trees potentially tolerant to attack. | Kayla N.Boyes Kathryn G.Hietala-Henschell Alexander P.Barton Andrew J.Storer Jordan M.Marshall | 2019 | Journal of Forestry Research2019,30,5: | 1 |
