5Clinical effects of Xingnao Kaiqiao acupuncture on neurological impairment following cerebral infarction显示文摘BACKGROUND: Although the curative effects of acupuncture have been confirmed by various treatments of cerebral infarction, few studies have investigated when acupuncture can attain the best clinical effect. OBJECTIVE: Four different time points were selected for acupuncture treatment of cerebral infarction to evaluate the appropriate time course for Xingnao Kaiqiao therapy in terms of improved neurological function. DESIGN: Controlled observation. SETTING: Department of Traditional Chinese Medicine Rehabilitation and Physiotherapy of the Affiliated Hospital of Medical College of Chinese Armed Police Forces. PARTICIPANTS: A total of 120 inpatients with cerebral infarction of different stages, including 75 males and 45 females, aged 41-75 years, were selected from November 2005 to December 2006 at the Department of Traditional Chinese Medicine Rehabilitation and Physiotherapy in Affiliated Hospital of Medical College of Chinese Armed Police Forces. Diagnostic criteria: in accordance with 'main points of diagnosis on different cerebrovascular disease' secondly revised in the Second Cerebrovascular Disease Academic Meeting of Chinese Medicine Association in 1986. All accepted subjects provided confirmed consent, and the experiment received ethical permission from the hospital’s ethics committee. METHODS: ① Experiment grouping: All inpatients were divided into four groups with non-stochastic concurrent control method according to the disease course: Group I (onset within 7 hours), group Ⅱ (onset from 7 hours to 3 days), group Ⅲ (onset within 4-7 days), and group Ⅳ (onset within 21-180 days). On the basis of symptomatic treatment with western medicine, each group received Xingnao Kaiqiao therapy after onset within 7 hours, 7 hours to 3 days, 4 to 7days, and 21 to 180 days. ② The principal acupoints were Neiguan, Renzhong, and Sanyinjiao. ③ The auxiliary acupoints were Jiquan, Chize, and Weizhong. ④Acupuncture manipulations: initially, Neiguan (PC6, bilateral) was needled at 0.5-1.0 cun vertically with a reducing technique achieved by rotating-swirling and lifting-inserting for 1 minute. Subsequently, Renzhong (GV26) was needled at 0.3-0.5 cun obliquely towards the nasal septum, with a heavy bird-pecking method until eyeballs were moist or shed tears. Sanyinjiao (SP6, affected limb) was needled at 1.0-1.5 cun obliquely along the medial border of the tibia with a 45° angle between the needle and skin. The technique of reinforcing was achieved by a lifting-inserting method until the affected lower limb twitched three times. The auxiliary acupoint, Jiquan, was selected when the patient extended the affected upper limb abduction. Jiquan was along the downward channel, 1 cun from the quondam Jiquan (HT1) acupoint; the armpit hair was avoided. Jiquan (affected limb) was needled vertically at 1.0-1.5 cun. The technique of reduction was achieved by lifting-inserting until the affected upper limb twitched three times. Chize (LU5) was selected when the patient flexed the elbow at 120°. Chize was vertically needled at 1 cun with a lifting-inserting reducing technique until the affected forearm and fingers twitched three times. Weizhong (BL40) was selected when the patient’s leg was raised straight in a supine position. Weizhong was vertically needled at 0.5-1.0 cun with a lifting-inserting reducing technique until the affected lower limb twitched three times. ⑤ Acupuncture manipulations were achieved by the doctor who is an associate chief physician and doctor of acupuncture and moxibustion and has aptitude for the post’s specification. ⑥One course of treatment lasted for 10 days and each group was treated for two courses of treatment. ⑦Experiment evaluation: To apply neurological impairment scores for evaluating clinical curative effects before and after treatment with Xingnao Kaiqiao therapy on day 7, 14, and 21(range of 0-45; decreasing score indicates functional improvement).To evaluate clinical curative effect by clinical effective power:Basic recovery: equal to ormore than 81%; significant improvement: 56% to 81% (including 56%); Improvement: between 36% to 56% (including 36%); Slight Improvement: 11% to 36% (including 11%); No change: less than 11%; Aggravation (including death): negative value. Total effective rate was computed at the same time. MAIN OUTCOME MEASURES: The total effective rate and amelioration of neurological impairment of the patients after treatment. RESULTS: All 120 patients with cerebral infarction were included in the final analysis, without deletion. ① Comparison of the total effective rate: the total effective rate of group Ⅰ, group Ⅱ, group Ⅲ, and group Ⅳ was 97%, 93%, 93%, and 67%, respectively. There were significant differences of interclass synthetic curative effects (χ2 = 36.351 7, P < 0.01). ② Amelioration of neurological impairment: A decreased neurological impairment score was observed in group I on day 7, 14, and 21 after Xingnao Kaiqiao therapy. The differences between group IV and the remaining three groups were significant (P < 0.05). 8Effects of extracts from Chuanwu(Aconitum Carmichaelii) and Banxia(Rhizoma Pinelliae) on excisional wound healing in a rat's model显示文摘OBJEVTIVE: To investigate the effects of extracts from Chuanwu(Aconitum Carmichaelii) and Banxia(Rhizoma Pinelliae) on the excisional wound healing in a rat's model.METHODS: Rats were performed a surgical lesion with a 2.0 cm resected tissue in the dorsal fascia.Following, animals were randomly divided into model group, YNB group and APE group those were respectively treated with saline, 1 mg/mL of Yunnan Baiyao and 1 mg/mL of Chuanwu(Aconitum Carmichaelii) and Banxia(Rhizoma Pinelliae) extracts. Wound contractions in days 0, 3, 7, 14 and 21 were calculated by an image analyzer. Histological analysis was analyzed using hematoxilin and eosin.Levels of tumor necrosis factor-2(IL-2), IL-4, IL-10, transformαin(TNF-g growα), interleukinth factor-1(TGF-nd basic fibroblast growth factor(bFββ1) a-GF) were determined by real-time quantitative PCR.RESULTS: Compared with that of YNB group and APE gtoup, the skin of rats showed poor re-modeling and re-epithelization characterized by a significant decrease of neovascularization, epithelialization and fibroblast in the model group. 9Application of a neural network system combined with genetic algorithm to rank coalbed methane reservoirs in the order of exploitation priority显示文摘A new method based on the combination of a neural network and a genetic algorithm was proposed to rank the order of exploitation priority of coalbed methane reservoirs.The neural network was used to acquire the weights of reservoir parameters through sample training and genetic algorithm was used to optimize the initial connection weights of nerve cells in case the neural network fell into a local minimum.Additionally,subordinate functions of each parameter were established to normalize the actual values of parameters of coalbed methane reservoirs in the range between zero and unity.Eventually, evaluation values of all coalbed methane reservoirs could be obtained by using the comprehensive evaluation method,which is the basis to rank the coalbed methane reservoirs in the order of exploitation priority.The greater the evaluation value,the higher the exploitation priority.The ranking method was verified in this paper by ten exploited coalbed methane reservoirs in China.The evaluation results are in agreement with the actual exploitation cases.The method can ensure the truthfulness and credibility of the weights of parameters and avoid the subjectivity caused by experts.Furthermore,the probability of falling into local minima is reduced,because genetic the algorithm is used to optimize the neural network system.Li Weichao Wu Xiaodong Shi Junfeng 2008Petroleum Science2008,5,4:4

11Orchestration of energy metabolism and osteogenesis by Mg^(2+)facilitates low-dose BMP-2-driven regeneration显示文摘The clinical application of bone morphogenetic protein-2(BMP-2)is limited by several factors,including ineffectiveness at low doses and severe adverse effects at high doses.To address these efficacy and safety limitations,we explored whether orchestration of energy metabolism and osteogenesis by magnesium ion(Mg^(2+))could reduce the dose and thereby improve the safety of BMP-2.Our results demonstrated that rapid metabolic activation triggered by BMP-2 was indispensable for subsequent osteogenesis.Moreover,inadequate metabolic stimulation was shown to be responsible for the ineffectiveness of low-dose BMP-2.Next,we identified that Mg^(2+),as an'energy propellant',substantially increased cellular bioenergetic levels to support the osteogenesis via the Akt-glycolysis-Mrs2-mitochondrial axis,and consequently enhanced the osteoinductivity of BMP-2.Based on the mechanistic discovery,microgel composite hydrogels were fabricated as low-dose BMP-2/Mg^(2+)codelivery system through microfluidic and 3D printing technologies.An in vivo study further confirmed that rapid and robust bone regeneration was induced by the codelivery system.Collectively,these results suggest that this bioenergetic-driven,cost-effective,low-dose BMP-2-based strategy has substantial potential for bone repair.Sihan Lin Shi Yin Junfeng Shi Guangzheng Yang Xutao Wen Wenjie Zhang Mingliang Zhou Xinquan Jiang 2022Bioactive Materials2022,7,12:2
12Structure-guided analysis of Arabidopsis JASMONATE-INDUCED OXYGENASE(JOX)2 reveals key residues for recognition of jasmonic acid substrate by plant JOXs显示文摘The jasmonic acid(JA)signaling pathway is used by plants to control wound responses.The persistent accumulation of JA inhibits plant growth,and the hydroxylation of JA to 12-hydroxy-JA by JASMONATE-INDUCED OXYGENASEs(JOXs,also named jasmonic acid oxidases)is therefore vital for plant growth,while structural details of JA recognition by JOXs are unknown.Here,we present the 2.65Åresolution X-ray crystal structure of Arabidopsis JOX2 in complex with its substrate JA and its co-substrates 2-oxoglutarate and Fe(Ⅱ).JOX2 contains a distorted double-stranded p helix(DSBH)core flanked by a helices and loops.JA is bound in the narrow substrate pocket by hydrogen bonds with the arginine triad R225,R350,and R354 and by hydrophobic interactions mainly with the phenylalanine triad F157,F317,and F346.The most critical residues for JA binding are F157 and R225,both from the DSBH core,which interact with the cyclopentane ring of JA.The spatial distribution of critical residues for JA binding and the shape of the substrate-binding pocket together define the substrate selectivity of the JOXs.Sequence alignment shows that these critical residues are conserved among JOXs from higher plants.Collectively,our study provides insights into the mechanism by which higher plants hydroxylate the hormone JA.Xin Zhang Dongli Wang Joyce Elberse Linlu Qi Wei Shi You-Liang Peng Robert C.Schuurink Guido Van den Ackerveken Junfeng Liu 2021Molecular Plant2021,14,5:2

14On-chip multiplexed single-cell patterning and controllable intracellular delivery显示文摘Conventional electroporation approaches show limitations in the delivery of macromolecules in vitro and in vivo.These limitations include low efficiency,noticeable cell damage and nonuniform delivery of cells.Here,we present a simple 3D electroporation platform that enables massively parallel single-cell manipulation and the intracellular delivery of macromolecules and small molecules.A pyramid pit micropore array chip was fabricated based on a silicon wet-etching method.A controllable vacuum system was adopted to trap a single cell on each micropore.Using this chip,safe single-cell electroporation was performed at low voltage.Cargoes of various sizes ranging from oligonucleotides(molecular beacons,22 bp)to plasmid DNA(CRISPR-Cas9 expression vectors,>9 kb)were delivered into targeted cells with a significantly higher transfection efficiency than that of multiple benchmark methods(e.g.,commercial electroporation devices and Lipofectamine).The delivered dose of the chemotherapeutic drug could be controlled by adjusting the applied voltage.By using CRISPR-Cas9 transfection with this system,the p62 gene and CXCR7 gene were knocked out in tumor cells,which effectively inhibited their cellular activity.Overall,this vacuum-assisted micropore array platform provides a simple,efficient,high-throughput intracellular delivery method that may facilitate on-chip cell manipulation,intracellular investigation and cancer therapy.Zaizai Dong Yanli Jiao Bingteng Xie Yongcun Hao Pan Wang Yuanyuan Liu Junfeng Shi Chandani Chitrakar Stephen Black Yu-Chieh Wang LJames Lee Mo Li Yubo Fan Lingqian Chang 2020Microsystems & Nanoengineering2020,6,1:2

Then, we assessed the effects of varying doses of CGZBT on an established chronic unpredictable mild stress(CUMS) model in rats. Whether animals were depressed was evaluated by sucrose preference test, open field test and forced swimming test. Neurotransmitters of hippocampus were detected by liquid chromatography-mass spectrometry. Serum levels of tumor necrosis factor-alpha(TNF-α), interleukin(IL)-1β, IL-6, IL-4, and IL-10 were measured by enzyme-linked immunosorbent assay. Expressions of toll-like receptor 4(TLR4), myeloid differentiation factor 88(My D88), phospho-nuclear factorkappa B(p-NF-κB), cyclooxygenase-2(COX-2), ionized calcium binding adapter molecule-1(IBA-1) were assessed by immunohistochemical staining and western blotting. RESULTS: Eight compounds were identified from CGZBT, moreover, our results showed that CGZBT effectively reversed the CUMS-induced decrease in sucrose preference, shortened the movement distance and prolonged immobility time. CGZBT significantly increased levels of 5-hydroxytryptamine, dopamine, norepinephrine, 5-hydroxyindoleacetic acid levels, and reduced the expression of TNF-α, IL-1β, IL-6, yet increased IL-4 and IL-10. Furthermore, the expressions of TLR4, My D88, COX-2, p-NF-κB and IBA-1 in hippocampus were effectively reversed after treatment with CGZBT. 18NGS-metabarcoding revealing novel foraminiferal diversity in the Western Pacific Magellan Seamount sediments显示文摘Seamount is a unique deep-sea ecosystem widely distributed in the world.Its biodiversity is vibrant due to its specific geographical and hydrological conditions.However,the diversity and features of foraminifera in such an environment have rarely been studied.We extracted environmental DNA(eDNA)in sedime nts and amplified the partial small subunit ribo somal DNA(SSU rDNA)of fo raminife ra to understand the foraminiferal diversity from four sites in Magellan Seamount(Western Pacific Ocean).Partial S SU rDNA sequencing was conducted and 912979 foraminiferal reads were obtained and gathered into 266 operational taxonomic units(OTUs).In the available dataset,a high proportion of rare OTUs and low identity OTUs in each studied sample showed that the Magellan Seamount foraminiferal community might have a high genetic novelty.The relative abundance of foraminifera varied between replicates probably due to the existence of bias in amplification process and patchiness of the deep-sea floor.It showed that the Magellan Seamount has a relatively high benthic foraminiferal diversity characterized mainly by monothalamiids(76.37%of total reads)in association with rotaliids(19.03%of total reads),including planktic foraminiferal sequences(38.58%of rotaliids;7.36%of total reads).The remaining reads were assigned to miliolids(0.83%of total reads)and textulariids(0.66%of total reads),and 3.11%of total reads are unassigned to a specific family.The co mparative analysis with foraminiferal assemblage s from coastal and deep-sea environme nts indicated that seamounts could aggregate species from a nearby deep-sea.Junfeng SHI Yanli LEI Haotian LI Tiegang LI 2021Journal of Oceanology and Limnology2021,39,5:1

20Strontium-incorporated bioceramic scaffolds for enhanced osteoporosis bone regeneration显示文摘The restoration of bone defects caused by osteoporosis remains a challenge for surgeons.Strontium ranelate has been applied in preventative treatment approaches due to the biological functions of the trace element strontium(Sr).In this study,we aimed to fabricate bioactive scaffolds through Sr incorporation based on our previously developed modified amino-functional mesoporous bioactive glass(MBG)and to systematically investigate the bioactivity of the resulting scaffold in vitro and in vivo in an osteoporotic rat model.The results suggested that Sr-incorporated amino-functional MBG scaffolds possessed favorable biocompatibility.Moreover,with the incorporation of Sr,osteogenic and angiogenic capacities were upregulated in vitro.The in vivo results showed that the Sr-incorporated amino-functional MBG scaffolds achieved better bone regeneration and vessel formation.Furthermore,bioinformatics analysis indicated that the Sr-incorporated amino-functional MBG scaffolds could reduce reactive oxygen species levels in bone marrow mesenchymal stem cells in the osteoporotic model by activating the cAMP/PKA signaling pathway,thus playing an anti-osteoporosis role while promoting osteogenesis.This study demonstrated the feasibility of incorporating trace elements into scaffolds and provided new insights into biomaterial design for facilitating bone regeneration in the treatment of osteoporosis.Qianju Wu Longwei Hu Ran Yan Junfeng Shi Hao Gu Yuwei Deng Ruixue Jiang Jin Wen Xinquan Jiang 2022Bone Research2022,10,4:1
