1Expression of vascular endothelial growth factor C and chemokine receptor CCR7 in gastric carcinoma and their values in predicting lymph node metastasis显示文摘AIM:To study the expression of vascular endothelial growth factor C (VEGF-C) and chemokine receptor CCR7 in gastric carcinoma and to investigate their associations with lymph node metastasis of gastric carcinoma and their values in predicting lymph node metastasis.METHODS:The expression of VEGF-C and CCR7 in gastric carcinoma tissues obtained from 118 patients who underwent curative gastrectomy was examined by immunohistochemisty.Among these patients,39 patients underwent multi-slice spiral CT (MSCT) examination.RESULTS:VEGF-C and CCR7 were positively expressed in 52.5 and 53.4% of patients. VEGF-C expression was more frequently found in tumors with lymph node metastasis than those without it (P<0.001).VEGF-C expression was also closely related to lymphatic invasion (P<0.001), vascular invasion (P<0.01),and TNM stage (P<0.001). However,there was no significant correlation between VEGF-C expression and age at surgery, gender, tumor size, tumor location,Lauren classification,and depth of invasion. CCR7 expression was significantly higher in patients with lymph node metastasis compared with those without lymph node metastasis (P<0.001) and was also associated with tumorsize (P<0.01), depth of invasion (P<0.001), lymphatic invasion (P<0.001),and TNM stage (P<0.001).However,the presence of CCR7 had no correlation to age at surgery,gender, tumor location, Lauren classification,and vascular invasion. Among the 39 patients who underwent MSCT examination,only CCR7 expression was related to lymph node metastasis determined by MSCT (P<0.05).In the current retrospective study,the sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of VEGF-C and CCR7 expression in the diagnosis of lymph node metastasis for patients with gastric carcinoma were 73.8%, 70.2%,72.6%,71.4% and 72.0%,and 82.0%,77.2%,79.4%,80.0% and 79.7%,respectively.After subdivision according to the combination of VEGF-C and CCR7 expression, receiver operating characteristic (ROC) analysis showed that the accuracy of the combined examination of VEGF-C and CCR7 expression in predicting lymph node metastasis was relatively high (area under ROC curve [Az]=0.83).CONCLUSION:The expression of VEGF-C and CCR7 is related to lymph node metastasis of gastric carcinoma and both of them may become new targets for the treatment of gastric carcinoma.Furthermore,the combined examination of VEGF-C and CCR7 expression in endoscopic biopsy specimens may be useful in predicting lymph node metastasis of gastric carcinoma and deciding the extent of surgical lymph node resection.ChaoYan Zheng-GangZhu Ying-YanYu JunJi YiZhang Yu-BaoJi MinYan JunChen Bing-YaLin 2004World Journal of Gastroenterology2004,10,6:54

3m^6A Regulates Neurogenesis and Neuronal Development by Modulating Histone Methyltransferase Ezh2显示文摘N6-methyladenosine (m6A),catalyzed by the methyltransferase complex consisting of Mettl3 and Mettl14,is the most abundant RNA modification in mRNAs and participates in diverse biological processes. However,the roles and precise mechanisms of m6A modification in regulating neuronal development and adult neurogenesis remain unclear. Here,we examined the function of Mettl3,the key component of the complex,in neuronal development and adult neurogenesis of mice. We found that the depletion of Mettl3 significantly reduced m6A levels in adult neural stem cells (aNSCs) and inhibited the proliferation of aNSCs. Mettl3 depletion not only inhibited neu-ronal development and skewed the differentiation of aNSCs more toward glial lineage,but also affected the morphological maturation of newborn neurons in the adult brain. m6A immunoprecip-itation combined with deep sequencing (MeRIP-seq) revealed that m6A was predominantly enriched in transcripts related to neurogenesis and neuronal development. Mechanistically,m6A was present on the transcripts of histone methyltransferase Ezh2,and its reduction upon Mettl3 knockdown decreased both Ezh2 protein expression and consequent H3K27me3 levels. The defects of neurogenesis and neuronal development induced by Mettl3 depletion could be rescued by Ezh2 overexpression. 4Linkage between permafrost distribution and river runoff changes across the Arctic and the Tibetan Plateau显示文摘River runoff in the Arctic and the Tibetan Plateau(TP) change significantly in recent decades. However, the mechanisms of the physical processes of permafrost river runoff change remain uncertain across large scale. This study investigated the mainstreams and tributaries of main Arctic and TP rivers dominated by permafrost and assessed the linkage between hydrological regime change and permafrost. The results show that the effects of permafrost on river runoff are highly dependent on the permafrost coverage of a watershed. For the past decades, the majority of the Arctic and TP basins showed increased discharge, while all of the studied basins showed increased baseflow, with faster increasing speed than total discharge.Both total discharge and baseflow annual change rate(ΔQ and ΔBF) increased with permafrost coverage, indicating the increments of streamflow are enhanced with high permafrost coverage. Meanwhile, the annual change of precipitation showed weak connection with total discharge and baseflow change. The high permafrost coverage basins showed high annual maximum/minimum discharge ratio(Qmax/Qmin), while the Qmax/Qminchanged slightly in low permafrost cover basins. Our results highlight the importance of permafrost coverage on streamflow regime change for permafrost basins across the northern hemisphere. 5Transfection of apoptosis related gene Fas ligand in human hepatocellular carcinoma cells and its significance in apoptosis显示文摘AIM: To evaluate the expression of apoptosis related gene Fas ligand (FasL) in human hepatocellular carcinoma (HCC) cells HepG2 and its significance in apoptosis. METHODS: Levels of soluble Fas ligand (sFasL) in a group of patients with hepatitis B virus (HBV)-induced chronichepatitis, HBV-positive liver cirrhosis and HCC were evaluated. In a further study, the recombinant eukaryotic expression plasmid pcDNA3.1hisB-FasL was transfected into HCC cells HepG2 by lipofection, and then soluble FasL was examined in the supernatant of culture cells by EIA, FasL expression in HepG2 cells was detected by immuohistochemistry. After being stained by annexin V and propidium iodine, cells were passed through a flow cytometer and examined by a fluorescence microscope and a laser scanning microscope.RESULTS: The sFasL levels were significantly lower in patients with HCC when compared to the patients with hepatitis or liver cirrhosis. In comparison with untransfected cells, the soluble FasL could be detected in the supernatant of transfected cells. FasL was expressed on the membranes and cytoplasm of transfected cells. The apoptotic cell rate was 36.30% in transfected cells, and was 11.53% in untransfected cells. 6Transparently curved metamaterial with broadband millimeter wave absorption显示文摘We present a conformal metamaterial with simultaneous optical transparency and broadband millimeter-wave absorption for a curved surface. By tailoring the reflection response of meta-atoms at oblique angles, it is possible to achieve excellent absorption performance from 26.5 to 40.0 GHz within a wide angular range from 0° to 60°for transverse-electric and transverse-magnetic waves. In the meantime, by employing transparent substrates,including polyvinyl chloride and polyethylene terephthalate, good optical transmittance(80.1%) and flexibility are obtained simultaneously. The reflectivity of a curved metallic surface coated with the proposed curved metamaterial is simulated and measured experimentally. 8花岗岩动态断裂能各向异性试验研究显示文摘由于微裂纹的存在以及长期构造应力下矿物晶粒排列所造成的自然各向异性,岩石的力学性质大多表现出各向异性的特点。研究选取Barre花岗岩和Standstead花岗岩,利用改装的分离式霍普金森压杆系统和国际岩石力学学会推荐的直切槽半圆盘弯曲试样,进行了花岗岩动态断裂能各向异性的试验。试验中根据纵波波速所确定的主方向制备岩石试样,每种岩石制备三组试样。试验采用波形整形技术实现动态力平衡,并利用激光测量裂缝表面张开位移。利用这些测量结果并结合能量分析,确定2种岩石样品的动态断裂能。研究结果表明,加载率在0~200 GPa·m1/2/s范围,2种岩石三个方向的动态断裂能均随着加载率的增加而增加。在相同的加载率下,2种岩石的动态断裂参数具有明显的各向异性。2种岩石X方向(波速最小)的动态断裂能最小,Z方向(波速最大)的动态断裂能最大。此外,随着加载率的提高,2种岩石动态断裂能的各向异性差异越来越明显,这主要是由于高加载率下扩展过程中损伤区域的增大造成的。徐颖 ZHANG Junchen 姚伟 夏开文 2018岩石力学与工程学报2018,37,A01:6
9Large-scale blow spinning of heat-resistant nanofibrous air filters显示文摘Particulate matter(PM)pollution has become a serious problem worldwide and various kinds of nanofibrous filters aiming to solve the problem have been developed.It is urgent to remove PM from high-temperature pollution sources,such as industrial emissions,coal furnaces,and automobile exhaust gases.However,filtration at pollution sources remains challenging because most existing air filters are not resistant to high temperature.Herein,heat-resistant polyimide(PI)nanofibrous air filters are fabricated via a simple and scalable solution blow-spinning method.These air filters show excellent thermal stability at high temperature up to 420℃.They exhibit a filtration efficiency as high as 99.73%at ambient temperature and over 97%at 300℃.In addition,a field test shows that the filters remove>97%of PM from the car exhaust fumes.Hence,the blow-spun PI nanofibrous membranes combined with the facile preparation strategy have great potential in high temperature air filtration fields and other similar applications such as water purification and protein separation.Ziwei Li Jianan Song Yuanzheng Long Chao Jia Zhenglian Liu Lei Li Cheng Yang Junchen Liu Sen Lin Haiyang Wang Yibo Liu Minghao Fang Hui Wu 2020Nano Research2020,13,3:6
10A Consortium Blockchain Paradigm on Hyperledger-Based Peer-to-Peer Lending System显示文摘This paper focuses on how to use consortium blockchain to improve the regulation of peer-to-peer(P2 P) lending market. The partial decentralized consortium blockchain with limited pre-set nodes can well improve transparency and security, which is suitable for financial regulation. Considering irregularities of the P2P lending market, the Hyperledger-based Peer-to-Peer Lending System(HyperP2PLS) is proposed. First elaborate the application scenario and business logic of the system, where a national P2P Lending Trading Center will be established to integrate all transactions and information of P2P lending market. Then construct the system architecture consisting of the blockchain network, HTTP server, and applications. The algorithm of implementation process and the web application for users have been well illustrated. 14Drop casting based superhydrophobic and electrically conductive coating for high performance strain sensing显示文摘Conductive polymer composites(CPCs)strain sensors exhibit promising applications in flexible electronics,people’s health monitoring,etc.It remains a big challenge to develop a simple and cost-effective method to prepare CPCs with high conductivity,corrosion resistance,strong interfacial adhesion and high sensitivity.Here,we propose a facile“drop-casting and fluorination”strategy to fabricate superhydrophobic and highly electrically conductive coating by Ag precursor adsorption onto a commercially available elastic tape,subsequent chemical reduction and final fluorination.The Ag nanoparticles could not only construct the electrically conductive network but also greatly enhance the surface roughness.The contact angle and electrical conductivity of the coating can reach as high as 156°and 126 S/cm,respectively.When used for strain sensing,the superhydrophobic and conductive coating shows a high gauger factor(up to 7631 with the strain from 44%to 50%)and outstanding recyclability.The strain sensor could monitor different body joint motions with the stable and reliable sensing signals even after long time treatment in a corrosive solution.Ling Wang Lisheng Wu Yuqing Wang Junchen Luo Huaiguo Xue Jiefeng Gao 2022Nano Materials Science2022,4,2:2

17Comparison of dimension reduction-based logistic regression models for case-control genome-wide association study:principal components analysis vs.partial least squares显示文摘With recent advances in biotechnology, genome-wide association study(GWAS) has been widely used to identify genetic variants that underlie human complex diseases and traits. In case-control GWAS, typical statistical strategy is traditional logistical regression(LR) based on single-locus analysis. However, such a single-locus analysis leads to the well-known multiplicity problem, with a risk of inflating type I error and reducing power. Dimension reduction-based techniques, such as principal component-based logistic regression(PC-LR), partial least squares-based logistic regression(PLS-LR), have recently gained much attention in the analysis of high dimensional genomic data. However, the perfor?mance of these methods is still not clear, especially in GWAS. We conducted simulations and real data application to compare the type I error and power of PC-LR, PLS-LR and LR applicable to GWAS within a defined single nucleotide polymorphism(SNP) set region. We found that PC-LR and PLS can reasonably control type I error under null hypothesis.On contrast, LR, which is corrected by Bonferroni method, was more conserved in all simulation settings. In particular, we found that PC-LR and PLS-LR had comparable power and they both outperformed LR, especially when the causal SNP was in high linkage disequilibrium with genotyped ones and with a small effective size in simulation. 18Construction of recombinant eukaryotic expression plasmid containing murine CD40 ligand gene and its expression in H22 cells显示文摘AIM: To construct a recombinant murine CD40 ligand (mCD40L) eukaryotic expression vector for gene therapy and target therapy of hepatocellular carcinoma (HCC).METHODS: mCD40L cDNA was synthesized by RT-PCR with the specific primers and directly cloned into T vector to generate middle recombinant. After digestion with restriction endonuclease, the target fragment was subcloned into the multi-clone sites of the eukaryotic vector. The constructed vector was verified by enzyme digestion and sequencing,and the product expressed was detected by RT-PCR and immunofluorescence methods.RESULTS: The full-length mCD40L-cDNA was successfully cloned into the eukaryotic vector through electrophoresis,and mCD40L gene was integrated into the genome of infected H22 cells by RT-PCR. Murine CD40L antigen molecule was observed in the plasma of mCD40L-H22 by indirect immuno-fluorescence staining.CONCLUSION: The recombined mCD40L eukaryotic expression vector can be expressed in H22 cell line. 19Joint formation mechanism of high depth-to-width ratio friction stir welding显示文摘High depth-to-width ratio friction stir welding is an attractive method for the joining demands of alum inum profiles, which is sparked with its extrem ely low heat input and high mechanical performance. In this study, the joint form ation mechanism was studied by a num erical model of plastic flow combined with experim ental approaches. A fluid-solid-interaction algorithm was proposed to establish the coupling model, and the material to be welded was treated as non-New tonian fluid. The thread structure and the milling facets on tool pin prom oted drastic turbulence of material. The thread structure converged the plasticized material by its inclined plane, and then drove the attached material to refill the welds. The milling facets brought about the periodic dynamic material flow. The thread structure and the milling facets increased the strain rate greatly under the extremely low heat input, which avoided the welding defects. The condition of the peak tem perature of 648 K and the strain rate of 151 s^-1 attributed to the lowest coarsening degree of precipitate. The tensile strength of the joint reached 265 MPa, equivalent to 86% of base material.
