| 4 | The effects of temperature and resource availability on denitrification and relative N_(2)O production in boreal lake sediments显示文摘Anthropogenic environmental stressors(like atmospheric deposition,land use change,and climate warming) are predicted to increase inorganic nitrogen and organic carbon loading to northern boreal lakes,with potential consequences for denitrification in lakes.However,our ability to predict effects of these changes is currently limited as northern boreal lakes have been largely neglected in denitrification studies.The aim of this study was therefore to assess how maximum potential denitrification and N_2O production rates,and the relationship between the two(relative N_2O production),is controlled by availability of nitrate(NO_3^-),carbon(C),phosphorus(P),and temperature.Experiments were performed using the acetylene inhibition technique on sediments from a small,nutrient poor boreal lake in northern Sweden in 2014.Maximum potential denitrification and N_2O production rates at 4°C were reached already at NO_3^-additions of 106–120 μg NO_3^-–N/L,and remained unchanged with higher NO_3 amendments.Higher incubation temperatures increased maximum potential denitrification and N_2O production rates,and Q10 was somewhat higher for N_2O production(1.77) than for denitrification(1.69).The relative N_2O production ranged between 13% and 64%,and was not related to NO_3^-concentration,but the ratio increased when incubations were amended with C and P(from a median of 16% to 27%).Combined,our results suggests that unproductive northern boreal lakes currently have low potential for denitrification but are susceptible to small changes in NO_3 loading especially if these are accompanied by enhanced C and P availability,likely promoting higher N_2O production relative to N_2. | Maria Myrstener Anders Jonsson Ann-Kristin Bergstrom | 2016 | Journal of Environmental Sciences2016，28，9： | 5 |
| 5 | Bone remodeling effect of a chitosan and calcium phosphate-based composite显示文摘Chitosan is a biocompatible polymer that has been widely studied for tissue engineering purposes.The aim of this research was to assess bone regenerative properties of an injectable chitosan and calcium phosphate-based composite and identify optimal degree of deacetylation(%DDA)of the chitosan polymer.Drill holes were generated on the left side of a mandible in Sprague-Dawley rats,and the hole was either left empty or filled with the implant.The animals were sacrificed at several time points after surgery(7–22 days)and bone was investigated using micro-CT and histology.No significant new bone formation was observed in the implants themselves at any time points.However,substantial new bone formation was observed in the rat mandible further away from the drill hole.Morphological changes indicating bone formation were found in specimens explanted on Day 7 in animals that received implant.Similar bone formation pattern was seen in control animals with an empty drill hole at later time points but not to the same extent.A second experiment was performed to examine if the%DDA of the chitosan polymer influenced the bone remodeling response.The results suggest that chitosan polymers with%DDA between 50 and 70%enhance the natural bone remodeling mechanism. | Lilja Kjalarsdottir Arna Dyrfjord Atli Dagbjartsson Elin H.Laxdal Gissur Orlygsson Johannes Gislason Jon M.Einarsson Chuen-How Ng Halldor Jonsson Jr | 2019 | Regenerative Biomaterials2019，6，4： | 3 |
| 6 | Macrophage secretory products induce an inflammatory phenotype in hepatocytes显示文摘AIM:To investigate the influence of macrophages on hepatocyte phenotype and function.METHODS:Macrophages were differentiated from THP-1 monocytes via phorbol myristate acetate stimulation and the effects of monocyte or macrophageconditioned medium on HepG2 mRNA and protein expression determined.The in vivo relevance of these findings was confirmed using liver biopsies from 147 patients with hepatitis C virus(HCV)infection.RESULTS:Conditioned media from macrophages,but not monocytes,induced a transient morphological change in hepatocytes associated with upregulation of vimentin(7.8±2.5-fold,P=0.045)and transforming growth factor(TGF)-β1(2.6±0.2-fold,P<0.001)and downregulation of epithelial cadherin(1.7±0.02-fold,P=0.017)mRNA expression.Microarray analysis revealed significant upregulation of lipocalin-2(17-fold,P <0.001)and pathways associated with inflammation,and substantial downregulation of pathways related to hepatocyte function.In patients with chronic HCV,realtime polymerase chain reaction and immunohistochemistry confirmed an increase in lipocalin-2 mRNA(F0 1.0 ±0.3,F1 2.2±0.2,F2 3.0±9.3,F3/4 4.0±0.8,P= 0.003)and protein expression(F1 1.0±0.5,F2 1.3± 0.4,F3/4 3.6±0.4,P=0.014)with increasing liver injury.High performance liquid chromatography-tandem mass spectrometry analysis identified elevated levels of matrix metalloproteinase(MMP)-9 in macrophageconditioned medium,and a chemical inhibitor of MMP-9 attenuated the change in morphology and mRNA expression of TGF-β1(2.9±0.2 vs 1.04±0.1,P<0.001) in macrophage-conditioned media treated HepG2 cells.In patients with chronic HCV infection,hepatic mRNA expression of CD163(F0 1.0±0.2,F1/2 2.8±0.3,F3/4 5.3±1.0,P=0.001)and MMP-9(F0 1.0±0.4,F1/2 2.8±0.3,F3/4 4.1±0.8,P=0.011)was significantly associated with increasing stage of fibrosis.CONCLUSION:Secreted macrophage products alter the phenotype and function of hepatocytes,with increased expression of inflammatory mediators,suggesting that hepatocytes actively participate in liver injury. | Michelle Melino Victoria L Gadd Gene V Walker Richard Skoien Helen D Barrie Dinesh Jothimani Leigh Horsfall Alun Jones Matthew J Sweet Gethin P Thomas Andrew D Clouston Julie R Jonsson Elizabeth E Powell | 2012 | World Journal of Gastroenterology2012，18，15： | 3 |

| 14 | Irritable bowel syndrome: Physicians' awareness and patients' experience显示文摘AIM: To study if and how physicians use the irritable bowel syndrome (IBS) diagnostic criteria and to assess treatment strategies in IBS patients. METHODS: A questionnaire was sent to 191 physicians regarding IBS criteria, diagnostic methods and treatment. Furthermore, 94 patients who were diagnosed with IBS underwent telephone interview. RESULTS: A total of 80/191 (41.9%) physicians responded to the survey. Overall, 13 patients were diag-nosed monthly with IBS by specialists in gastroenterology (SGs) and 2.5 patients by general practitioners (GPs). All the SGs knew of the criteria to diagnose IBS, as did 46/70 (65.7%) GPs. Seventy-nine percent used the patient's history, 38% used a physical examination, and 38% exclusion of other diseases to diagnose IBS. Only 18/80 (22.5%) physicians used specific IBS criteria. Of the patients interviewed, 59/94 (62.8%) knew they had experienced IBS. Two out of five patients knew IBS and had seen a physician because of IBS symptoms. Half of those received a diagnosis of IBS. A total of 13% were satisfied with treatment. IBS affected daily activities in 43% of cases. CONCLUSION: Half of the patients with IBS who consulted a physician received a diagnosis.
