1Long-term trend in aerosol optical depth from 1980 to 2001 in north China显示文摘Using the Total Ozone Mapping Spectrometer(TOMS) monthly aerosol optical depth(AOD) at 500 nm data from 1980 to 2001 in north China,the spatial and temporal variations of AOD were examined.Seasonal AODs in Taklimakan Desert were 0.69 and 0.44 in spring and summer,respectively,which were mainly due to frequent occurrences of dust events in this region.Dust activities in spring also led to high aerosol loading in Gobi Desert and in northeast China where spring AODs were 0.33 and 0.29,respectively.Heavily impacted by events such as volcano eruption,forest fires and extraordinary dust storms,AODs showed large inter-annual variations.A decreasing tendency in AOD was observed in north China during 1980-1991,though a reverse tendency was revealed during 1997-2001,especially for spring AOD in northeast China.Further study is required to figure out how much human activities have contributed to the AOD tendency in north China.Jiangxia Xie Xiangao Xia 2008Particuology2008,6,2:5
2s-HBEGF/SIRT1 circuit-dictated crosstalk between vascular endothelial cells and keratinocytes mediates sorafenib-induced hand-foot skin reaction that can be reversed by nicotinamide显示文摘Hand-foot skin reaction(HFSR),among the most significant adverse effects of sorafenib,has been limiting the clinical benefits of this frontline drug in treating various malignant tumors.The mechanism underlying such toxicity remains poorly understood,hence the absence of effective intervention strategies.In the present study,we show that vascular endothelial cells are the primary cellular target of sorafenib-induced HFSR wherein soluble heparin-binding epidermal growth factor(s-HBEGF)mediates the crosstalk between vascular endothelial cells and keratinocytes.Mechanistically,s-HBEGF released from vascular endothelial cells activates the epidermal growth factor receptor(EGFR)on keratinocytes and promotes the phosphorylation of c-Jun N-terminal kinase 2(JNK2),which stabilizes sirtuin 1(SIRT1),an essential keratinization inducer,and ultimately gives rise to HFSR.The administration of s-HBEGF in vivo could sufficiently induce hyper-keratinization without sorafenib treatment.Furthermore,we report that HBEGF neutralization antibody,Sirtl knockdown,and a classic SIRTI inhibitor nicotinamide could all significantly reduce the sorafenib-induced HFSR in the mouse model.It is noteworthy that nicotinic acid,a prodrug of nicotinamide,could substantially reverse the sorafenib-induced HFSR in ten patients in a preliminary clinical study.Collectively,our findings reveal the mechanism of vascular endothelial cell-promoted keratinization in keratinocytes and provide a potentially promising therapeutic strategy for the treatment of sorafenib-induced HFSR.Peihua Luo Hao Yan Xueqin Chen Ying Zhang Ziying Zhao Ji Cao Yi Zhu Jiangxia Du Zhifei Xu Xiaochen Zhang Su Zeng Bo Yang Shenglin Ma Qiaojun He 2020Cell Research2020,30,9:5
3The m6A methylation regulates gonadal sex differentiation in chicken embryo显示文摘Background:As a ubiquitous reversible epigenetic RNA modification,N6-methyladenosine(m6A)plays crucial regulatory roles in multiple biological pathways.However,its functional mechanisms in sex determination and differentiation during gonadal development of chicken embryos are not clear.Therefore,we established a transcriptome-wide m6A map in the female and male chicken left gonads of embryonic day 7(E7)by methylated RNA immunoprecipitation sequencing(MeRIP-seq)to offer insight into the landscape of m6A methylation and investigate the post-transcriptional modification underlying gonadal differentiation.Results:The chicken embryonic gonadal transcriptome was extensively methylated.We found 15,191 and 16,111 m6A peaks in the female and male left gonads,respectively,which were mainly enriched in the coding sequence(CDS)and stop codon.Among these m6A peaks,we identified that 1013 and 751 were hypermethylated in females and males,respectively.These differential peaks covered 281 and 327 genes,such as BMP2,SMAD2,SOX9 and CYP19A1,which were primarily associated with development,morphogenesis and sex differentiation by functional enrichment.Further analysis revealed that the m6A methylation level was positively correlated with gene expression abundance.Furthermore,we found that YTHDC2 could regulate the expression of sex-related genes,especially HEMGN and SOX9,in male mesonephros/gonad mingle cells,which was verified by in vitro experiments,suggesting a regulatory role of m6A methylation in chicken gonad differentiation.Conclusions:This work provided a comprehensive m6A methylation profile of chicken embryonic gonads and revealed YTHDC2 as a key regulator responsible for sex differentiation.Our results contribute to a better understanding of epigenetic factors involved in chicken sex determination and differentiation and to promoting the future development of sex manipulation in poultry industry.Jianbo Li Xiuan Zhang Xiqiong Wang Congjiao Sun Jiangxia Zheng Junying Li Guoqiang Yi Ning Yang 2022Journal of Animal Science and Biotechnology2022,13,6:4
4The influences of SE infection on layers' production performance,egg quality and blood biochemical indicators显示文摘Background:Salmonella enterica serovar Enteritidis(SE),as a major cause of foodbom illness,infects humans mainly through the egg.However,the symptom of laying hens usually is not typical and hard to diagnosis.In the present study,it is studied that the influences of SE infection on layers’ performance,egg quality and blood biochemical indicators.It will help us to improve the strategy to control SE infection in commercial layers.One hundred layers at 20 wk of age were divided into 2 groups,60 hens for experiment and others for control.The experiment group was fed with the dosage of 10~8 CFU SE per hen.The specific PCR was used to detect the deposition of SE.On the 8 d after SE infection,10 hens from the control group and 30 hens from the experimenta group were slaughtered to detect the SE colonization.The production performance,egg quality and blood biochemical indices were also analyzed.Results:The results showed that the colonization rate of SE was highest in caecum contents(55.17%) and lowest in vagina(17.24%).For the eggs the detection rate of SE was highest on the eggshell(80.00%) and lowest in yolk(18.81%).SE infection had no significant influence on production performance and egg qualities(P > 0.05).The difference of laying rate between the experimental and control groups was less than 0.30%,and both were approximately equal to 82.00%.The blood analysis showed that the aspartic aminotransferase(AST) and alanine aminotransferase(ALT) of experimental group was significantly higher than those of control group(P < 0.05).For experimental and control groups AST values were 236.22 U/l and 211.84 U/l respectively,and ALT values were32.19 U/l and 24.55 U/l.All of coefficients were less than 20%.The colonization of SE in organs increases the enzyme activities of AST and ALT in blood.Conclusions:SE in feed could invade the oviduct and infect the forming eggs.It significantly increased the concentration of ALT and AST in blood.However,SE infection was hard to be observed from the appearances of layer and egg.It might be a dangerous risk to human health.Shijie Fan Jiangxia Zheng Zhongyi Duan Ning Yang Guiyun Xu 2014Journal of Animal Science and Biotechnology2014,5,3:2
5Transcriptome responses to heat stress in hypothalamus of a meat-type chicken显示文摘Background:Heat stress has resulted in great losses in poultry production.To address this issue,we systematically analyzed chicken hypothalamus transcriptome responses to thermal stress using a 44 k chicken Agilent microarray,Methods:Hypothalamus samples were collected from a control group reared at 25℃,a heat-stress group treated at 34℃ for 24 h,and a temperature-recovery group reared at 25℃ for 24 h following a heat-stress treatment.We compared the expression profiles between each pair of the three groups using microarray data.Results:A total of 1,967 probe sets were found to be differentially expressed in the three comparisons with P < 0.05 and a fold change(FC) higher than 1.5,and the genes were mainly involved in self-regulation and compensation required to maintain homeostasis.Consistent expression results were found for 11 selected genes by quantitative real-time PCR.Thirty-eight interesting differential expression genes were found from GO term annotation and those genes were related to meat quality,growth,and crucial enzymes.Using these genes for genetic network analysis,we obtained three genetic networks.Moreover,the transcripts of heat-shock protein,including Hsp 40 and Hsp 90,were significantly altered in response to thermal stress.Conclusions:This study provides a broader understanding of molecular mechanisms underlying stress response in chickens and discovery of novel genes that are regulated in a specific thermal-stress manner.Hongyan Sun Runshen Jiang Shengyou Xu Zebin Zhang Guiyun Xu Jiangxia Zheng Lujiang Qu 2015Journal of Animal Science and Biotechnology2015,6,3:2
6Origin and domestication history of Peking ducks deltermined through microsatellite and mitochondrial marker analysis显示文摘In order to elucidate the domestication history of Peking ducks, 190 blood samples from six Chinese indigenous duck breeds were collected with186 individualsgenotyped by 15 microsatellite markers. Both the FST and Nei’s standard genetic distances (Ds) from the microsatellite data indicated high genetic differentiation between Peking duck and other Chinese indigenous breeds. The haplotype network with mtDNA data showed that most of the Peking duck haplotypes were distinctly different from those of other domestic breeds. Although the H01 haplotype was shared by all domesticated duck breeds, Peking ducks displayed 12 specific domestic duck haplotypes, including four similar haplotypes H02, H04, H08 and H22, that formed a single haplogroup (A).
