1Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16

5Single-nucleus transcriptomic landscape of primate hippocampal aging显示文摘The hippocampus plays a crucial role in learning and memory,and its progressive deterioration with age is functionally linked to a variety of human neurodegenerative diseases.Yet a systematic profiling of the aging effects on various hippocampal cell types in primates is still missing.Here,we reported a variety of new aging-associated phenotypic changes of the primate hippocampus.These include,in particular,increased DNA damage and heterochromatin erosion with time,alongside loss of proteostasis and elevated inflammation.To understand their cellular and molecular causes,we established the first single-nucleus transcriptomic atlas of primate hippocampal aging.Among the 12 identified cell types,neural transiently amplifying progenitor cell(TAPC)and microglia were most affected by aging.In-depth dissection of gene-expression dynamics revealed impaired TAPC division and compromised neuronal function along the neurogenesis trajectory;additionally elevated pro-inflammatory responses in the aged microglia and oligodendrocyte,as well as dysregulated coagulation pathways in the aged endothelial cells may contribute to a hostile microenvironment for neurogenesis.This rich resource for understanding primate hippocampal aging may provide potential diagnostic biomarkers and therapeutic interventions against age-related neurodegenerative diseases.Hui Zhang Jiaming Li Jie Ren Shuhui Sun Shuai Ma Weiqi Zhang Yang Yu Yusheng Cai Kaowen Yan Wei Li Baoyang Hu Piu Chan Guo-Guang Zhao Juan Carlos Izpisua Belmonte Qi Zhou Jing Qu Si Wang Guang-Hui Liu 2021Protein & Cell2021,12,9:5

7Investigation on experimental load spectrum for high and low cycle combined fatigue test显示文摘As the outfield load spectrum is so complicated that it cannot be used directly for test study in laboratory.This paper presents a method to determine load spectrum for high and low cycle combined fatigue of turbine mortise at elevated temperature through experimental and numerical method.First of all,the low cycle load spectrum with duration time is determined through cumulative damage rule.The rain flow counting method is applied to obtain main cycles and sub cycles,and then the stress cycle is converted into pulsation cycle(stress ratio=0)based on the S-N curve and the Goodman curve;Secondly,three groups of different amplitudes tests are established to determine the high cycle amplitude.Finally,another three groups of the high-low combined cycle fatigue(HLCCF)tests for turbine mortise of a certain type engine are carried out.The results show that the macro and micro failure modes are identical with outfield's,which verifies the accuracy of the conversion method.Rongqiao Wang Jiaming Wei Dianyin Hu Xiuli Shen Jiang Fan 2013Propulsion and Power Research2013,2,4:4
8Effects of adeno-associated virus (AAV) of transforming growth factors β_1 and β_3 (TGFβ_(1,3)) on promoting synthesis of glycosaminoglycan and collagen type II of dedifferentiated nucleus pulposus (NP) cells显示文摘The effects of AAV-TGFβ1 and AAV-TGFβ3 on promoting synthesis of glycosaminoglycan and collagen type II of dedifferentiated rabbit lumbar disc NP cells were studied in this work. The rabbit lumbar disc NP cells were isolated and cultured. The earlier and later dedifferentiated NP cells were established by subculture. The AAV transfection efficiency to dedifferentiated NP cells was analyzed with AAV-EGFP in vitro. After dedifferentiated NP cells were transfected by AAV-TGFβ1 or AAV-TGFβ3, their biological effects on promoting synthesis of glycosaminoglycan or collagen type II were detected and compared by the methods of 35S incorporation or immunoblotting. The experimental results showed that AAV could transfect efficiently the earlier dedifferentiated NP cells, but its transfection rate was shown to be at a low level to the later dedifferentiated NP cells. Both AAV-TGFβ1 and AAV-TGFβ3 could promote the earlier dedifferentiated NP cells to synthesize glycosaminoglycan and collagen type II, and the effect of AAV-TGFβ1 was better than that of AAV-TGFβ3. For the later dedifferentiated NP cells, the AAV-TGFβ3 could promote their synthesis, but AAV-TGFβ1 could slightly inhibit their synthesis. 9A CCAAT-binding factor,SlNFYA10,negatively regulates ascorbate accumulation by modulating the D-mannose/L-galactose pathway in tomato显示文摘Ascorbic acid(AsA),an important antioxidant and growth regulator,and it is essential for plant development and human health.Specifically,humans have to acquire AsA from dietary sources due to their inability to synthesize it.The AsA biosynthesis pathway in plants has been elucidated,but its regulatory mechanism remains largely unknown.In this report,we biochemically identified a CCAAT-box transcription factor(SlNFYA10)that can bind to the promoter of SlGME1,which encodes GDP-Man-3’,5’-epimerase,a pivotal enzyme in the D-mannose/L-galactose pathway.Importantly,SlNFYA10 simultaneously binds to the promoter of SlGGP1,a downstream gene of SlGME1 in the Dmannose/L-galactose pathway.Binding assays in yeast and functional analyses in plants have confirmed that SlNFYA10 exerts a negative effect on the expression of both SlGME1 and SlGGP1.Transgenic tomato lines overexpressing SlNFYA10 show decreased levels of SlGME1 and SlGGP1 abundance and AsA concentration in their leaves and fruits,accompanied by enhanced sensitivity to oxidative stress.Overall,SlNFYA10 is the first CCAAT-binding factor identified to date to negatively regulate the AsA biosynthetic pathway at multiple sites and modulate plant responses to oxidative stress.Weifang Chen Tixu Hu Jie Ye Bing Wang Genzhong Liu Ying Wang Lei Yuan Jiaming Li Fangman Li Zhibiao Ye Yuyang Zhang 2020Horticulture Research2020,7,1:3
10The Variation of Stone Cell Content in 236 Germplasms of Sand Pear(Pyrus pyrifolia)and Identification of Related Candidate Genes显示文摘Stone cells have been described to substantially influence pear fruit quality,as lignin and cellulose are the main components of stone cells.However,there are limited studies on the relationship between the variation and molecular basis of stone cells,lignin and cellulose content among different pear varieties.Here,to reveal the variation of stone cell content within different cultivated species,we collected 236 germplasms of sand pear(Pyrus pyrifolia)at 50 days after flower blooming(DAFB),the key stage of stone cell formation.In our results,we measured the content of stone cells,lignin and cellulose and found that these contents ranged from2.82%to 29.00%,8.84%to 55.30%and 11.52%to 30.55%,respectively.Further analysis showed that the variation coefficient of stone cell,lignin and cellulose content was 39.10%,28.03%and 16.71%,respectively.Additionally,a significant correlation between stone cell,lignin and cellulose content were detected,and the correlation coefficient between the contents of stone cell and lignin(0.912)was higher than between the contents of stone cell and cellulose(0.796).Moreover,the average lignin content(29.73%)was higher than the average cellulose content(18.03%)in stone cells in pear fruits,indicating that lignin is the main component of stone cell in pears.Finally,on the basic of the transcriptome data,we identified 10 transcription factors belonging to bHLH,ERF,MYB,and NAC transcript families,which might be involved in lignin formation in stone cells.qRT-PCR experiments verified coincident trends between expression of candidate genes and stone cell content.This research laid foundation for future studies on genetic variation of stone cells in pear fruits and provided important gene resources for stone cell regulation.Jiaying Zhang Jiaming Li Cheng Xue Runze Wang Mingyue Zhang Kaijie Qi Jing Fan Hongju Hu Shaoling Zhang Jun Wu 2021Horticultural Plant Journal2021,7,2:3
11Effect of Ningmitai capsule plus sertraline on patients with premature ejaculation and enlarged seminal vesicles:A randomized clinical trial显示文摘OBJECTIVE: To investigate the effects of Ningmitai capsule combined with sertraline on patients with premature ejaculation(PE) and an increased anterior-posterior diameter(APD) of the seminal vesicles(SVs).METHODS: Sixty men with acquired PE were enrolled and randomly divided into two groups. The combined group was treated with Ningmitai capsule and sertraline, while the control group was treated with sertraline alone. Main outcomes were measured using the premature ejaculation diagnostic tool(PEDT), APD of SVs, and Clinical Global Impression of Change questionnaire and compared before and after 3 months of treatment.RESULTS: Comparing after treatment with before treatment outcomes within each group, the PEDT score was significantly reduced in the combined group(12.1 ± 2.5 vs 8.6 ± 3.2, P < 0.001, respectively) and control group(12.9 ± 2.6 vs 10.3 ± 1.6, P <0.001, respectively). Furthermore, the PEDT score after treatment was significantly lower in the combined compared with control group(8.6 ± 3.2 vs10.3 ± 1.6, P = 0.011, respectively). The APD of SVs in the combined group was significantly decreased after treatment [(10.8 ± 2.4) vs(12.9 ± 2.2) mm, P =0.001], while the APD of SVs in the control group was equivalent before and after treatment. The treatment response rate was not significantly higher in the combined compared with control group.CONCLUSION: These results indicated that the effect of Ningmitai capsule combined with sertraline was better than that of sertraline alone for the treatment of PE patients exhibiting an increased APD of SVs. 12Accumulation and Elimination Kinetics of 2-Methylisoborneol in Crucian Carp (Carassius carassius)显示文摘2-methylisoborneol(2-MIB) is a common, earthysmelling compound that causes taste and odor issues in aquaculture. To quantify the distribution, absorption, and depuration of 2-MIB in crucian carp(Carassius carassius), we detected and measured 2-MIB it via microwave, headspace solid-phase microextraction(HS-SPME), and gas chromatography before and after exposing fish to two different concentrations of 2-MIB(200 ng/L and 500 ng/L). The results showed that during the exposure time, the fish absorbed 2-MIB rapidly within 4 h. After exposure to 2-MIB for 120 h, the 2-MIB in gill and liver were rapidly removed. The processes of accumulation and elimination of 2-MIB in fish were investigated using the one-compartment kinetic model. The modeling results showed that the range of the uptake rate constant(k1) was 0.49-6.91. The range of the elimination rate constant(k2) was 0.05-0.27 and uptake rates were higher than elimination rates. 13Ground-based Observations of Ultraviolet and Total Solar Radiation in Shenyang,Northeast China显示文摘Objective This work explores the diurnal variation of Solar ultraviolet radiation (UVR) and total solar radiation (TSR) in northeast China,using daily observations of UVR and TSR in Shenyang.Methods UVR and TSR measurements were carried out from March 1st,2006 to December 31st,2009 in Shenyang,Liaoning province,China (41°51' N,123°27' E).Results Both TSR and UVR showed seasonal variation,reaching the highest levels in summer and the lowest in winter.They showed the greatest fluctuation in summer and autumn.The irradiance of TSR and UVR on clear days around the equinoxes and solstices increased substantially compared with the mean seasonal irradiance,especially in autumn.The whole day accumulated dose of UVR in winter was far less than that during the middle part of a summer day (i.e.between 10:00 and 14:00).It was also less than the accumulated summer dose of morning and afternoon (i.e.between 8:00 and 10:00 and 14:00 and 16:00).Conclusion The instant irradiance and daily accumulated amount of UVR are low in Shenyang,especially in autumn and winter.Thus concern about the health effects arising because shortage of UVR in northeast China is warranted.GAO Qian HU LiWen GONG HuiZhi GAO Na YU JiaMing WANG Yang CHU Chun YANG Hui LIU Yang 2011Biomedical and Environmental Sciences2011,24,5:2
14ITO/Cu multilayer electrodes for high-brightness electrochromic displays显示文摘Fabry-Perot(F-P)nanocavity-type electrochromic devices with multicolor tunability have attracted significant interest in the past two years for their potential uses in a wide variety of applications,such as electronic display,military camouflage,and dynamic decoration.However,challenges such as insufficient brightness,lengthy switching times,and poor cycling stability have yet to be overcome.Herein,we demonstrate electrochromic electrodes based on ITO/Cu as both the current collector and the reflective layer,with WO_(3 )as the electrochromic material,forming a unique three-layered structure.The constituted WO_(3 )/ITO/Cu films present a high brightness value of about 84%before coloration,and a decent brightness value of 48%after coloration at−0.8​V.Moreover,a fast switching time between different coloration states(tc​=​2.2​s;tb​=​1​s)is recorded,attributed to the extremely low sheet resistivity of the ITO/Cu current collector.The films also reveal excellent electrochemical cycling stability across 2000 cycles.Xueqing Tang Zishou Hu Zhen Wang Jian Chen Xinyang Mu Ge Song Peiyan Sun Zhengji Wen Jiaming Hao Shan Cong Zhigang Zhao 2022eScience2022,2,6:1

18Unique and complementary suppression of cGAS-STING and RNA sensing- triggered innate immune responses by SARS-CoV-2 proteins显示文摘The emergence of SARS-CoV-2 has resulted in the COVID-19 pandemic,leading to millions of infections and hundreds of thousands of human deaths.The efficient replication and population spread of SARS-CoV-2 indicates an effective evasion of human innate immune responses,although the viral proteins responsible for this immune evasion are not clear.In this study,we identified SARS-CoV-2 structural proteins,accessory proteins,and the main viral protease as potent inhibitors of host innate immune responses of distinct pathways.In particular,the main viral protease was a potent inhibitor of both the RLR and cGAS-STING pathways.Viral accessory protein 0RF3a had the unique ability to inhibit STING,but not the RLR response.On the other hand,structural protein N was a unique RLR inhibitor.0RF3a bound STING in a unique fashion and blocked the nuclear accumulation of p65 to inhibit nuclear factor-KB signaling.3CL of SARS-CoV-2 inhibited K63-ubiquitin modification of STING to disrupt the assembly of the STING functional complex and downstream signaling.Diverse vertebrate STINGs,including those from humans,mice,and chickens,could be inhibited by 0RF3a and 3CL of SARS-CoV-2.The existence of more effective innate immune suppressors in pathogenic coronaviruses may allow them to replicate more efficiently in vivo.Since evasion of host innate immune responses is essential for the survival of all viruses,our study provides insights into the design of therapeutic agents against SARS-CoV-2.Yajuan Rui Jiaming Su Si Shen Ying Hu Dingbo Huang Wenwen Zheng Meng Lou Yifei Shi Meng Wang Shiqi Chen Na Zhao Qi Dong Yong Cai Rongzhen Xu Shu Zheng Xiao-Fang Yu 2021Signal Transduction and Targeted Therapy2021,6,4:1

20Development of Diapirs and Accumulation of Natural Gases in Yinggehai Basin显示文摘Overpressure developed throughout most of the Yinggehai basin. The burial depth to top overpressure varied from about 1 600 m to 4 500 m, with the shallowest top overpressure occurring in the depocenter. The main cause of the overpressure was disequilibrium compaction resulting from rapid sedimentation of fine grained sediments. The overpressure was strengthened by the retention of fluids including gases due to lack of faults before diapir development. The diapirism in the Yinggehai basin was a combined result of the strong overpressure and the tensile stress field caused by the right lateral slip of the boundary fault. The diapirism, a product of the movement of overpressured fluids and plastic shales, shaped the vertical conduits from source to traps that would be absent without overpressured fluid release.
