| 2 | Kinetics of the accumulation of group 2 innate lymphoid cells in IL-33-induced and IL-25-induced murine models of asthma:a potential role for the chemokine CXCL16显示文摘ILC2s are implicated in asthma pathogenesis, but little is known about the mechanisms underlying their accumulation in airways.We investigated the time course of ILC2 accumulation in different tissues in murine models of asthma induced by a serial per-nasalchallenge with ovalbumin (OVA), house dust mice (HDM), IL-25 and IL-33 and explored the potential roles of ILC2-attractingchemokines in this phenomenon. Flow cytometry was used to enumerate ILC2s at various time points. The effects of cytokines andchemokines on ILC2 migration were measured in vitro using a chemotaxis assay and in vivo using small animal imaging. Comparedwith saline and OVA challenge, both IL-25 and IL-33 challenge alone induced significant accumulation of ILC2s in the mediastinallymph nodes, lung tissue and bronchoalveolar lavage fluid of challenged animals, but with a distinct potency and kinetics. In vitro,IL-33 and CXCL16, but not IL-25 or CCL25, directly induced ILC2 migration. Small animal in vivo imaging further confirmed that asingle intranasal provocation with IL-33 or CXCL16 was sufficient to induce the accumulation of ILC2s in the lungs followinginjection via the tail vein. Moreover, IL-33-induced ILC2 migration involved the activation of ERK1/2, p38, Akt, JNK and NF-κB, whileCXCL16-induced ILC2 migration involved the activation of ERK1/2, p38 and Akt. These data support the hypothesis that epitheliumderived IL-25 and IL-33 induce lung accumulation of ILC2s, while IL-33 exerts a direct chemotactic effect in this process. | 3 | Increasing yield potential through manipulating of an ARE1 ortholog related to nitrogen use efficiency in wheat by CRISPR/Cas9显示文摘Wheat(Triticum aestivum L.)is a staple food crop consumed by more than 30%of world population.Nitrogen(N)fertilizer has been applied broadly in agriculture practice to improve wheat yield to meet the growing demands for food production.However,undue N fertilizer application and the low N use efficiency(NUE)of modern wheat varieties are aggravating environmental pollution and ecological deterioration.Under nitrogen-limiting conditions,the rice(Oryza sativa)abnormal cytokinin response1 repressor1(are1)mutant exhibits increased NUE,delayed senescence and consequently,increased grain yield.However,the function of ARE1 ortholog in wheat remains unknown.Here,we isolated and characterized three TaARE1 homoeologs from the elite Chinese winter wheat cultivar ZhengMai 7698.We then used CRISPR/Cas9-mediated targeted mutagenesis to generate a series of transgene-free mutant lines either with partial or triple-null taare1 alleles.All transgene-free mutant lines showed enhanced tolerance to N starvation,and showed delayed senescence and increased grain yield in field conditions.In particular,the AABBdd and aabbDD mutant lines exhibited delayed senescence and significantly increased grain yield without growth defects compared to the wild-type control.Together,our results underscore the potential to manipulate ARE1 orthologs through gene editing for breeding of high-yield wheat as well as other cereal crops with improved NUE. | Jiahui Zhang Huating Zhang Shaoya Li Jingying Li Lei Yan Lanqin Xia | 2021 | Journal of Integrative Plant Biology2021,63,9: | 13 |
| 4 | Myofiber necroptosis promotes muscle stem cell proliferation via releasing Tenascin-C during regeneration显示文摘Necroptosis,a form of programmed cell death,is characterized by the loss of membrane integrity and release of intracellular contents,the execution of which depends on the membrane-disrupting activity of the Mixed Lineage Kinase Domain-Like protein(MLKL)upon its phosphorylation.Here we found myofibers committed MLKL-dependent necroptosis after muscle injury.Either pharmacological inhibition of the necroptosis upstream kinase Receptor Interacting Protein Kinases 1(RIPK1)or genetic ablation of MLKL expression in myofibers led to significant muscle regeneration defects.By releasing factors into the muscle stem cell(MuSC)microenvironment,necroptotic myofibers facilitated muscle regeneration.Tenascin-C(TNC),released by necroptotic myofibers,was found to be critical for MuSC proliferation.The temporary expression of TNC in myofibers is tightly controlled by necroptosis;the extracellular release of TNC depends on necroptotic membrane rupture.TNC directly activated EGF receptor(EGFR)signaling pathway in MuSCs through its N-terminus assembly domain together with the EGF-like domain.These findings indicate that necroptosis plays a key role in promoting MuSC proliferation to facilitate muscle regeneration. | Shen'ao Zhou Wei Zhang Gaihong Cai Yingzhe Ding Caixia Wei Sheng Li Yu Yang Jie Qin Dan Liu Hao Zhang Xiexiang Shao Jianhua Wang Hongye Wang Wenjun Yang Huating Wang She Chen Ping Hu Liming Sun | 2020 | Cell Research2020,30,12: | 4 |

| 6 | Fibroblast growth factor 21:a novel metabolic regulator from pharmacology to physiology显示文摘Fibroblast growth factor 21(FGF21)is a member of the fibroblast growth factor family.It actually functions as endocrine hormones but does not regulate cell growth and differentiation.It is demonstrated that FGF21 acts on multiple tissue to coordinate carbohydrate and lipid metabolism,including enhancing insulin sensitivity,decreasing triglyceride concentrations,causing weight loss,ameliorating obesity-associated hyperglycemia and hyperlipidemia.Moreover,FGF21 also plays important roles in some physiological processes,such as fasting and feeding,growth hormone axis and thermogenic function of brown adipose tissue.Clinical relevance of FGF21 in humans is still unclear,and the basis and consequences of increased FGF21 in metabolic disease remain to be determined.Both the pharmacological actions and physiological roles make FGF21 attractive drug candidates for treating metabolic disease,but some questions remain to be answered.This article concentrates on recent advances in our understanding of FGF21. | Huating Li Jing Zhang Weiping Jia | 2013 | Frontiers of Medicine2013,7,1: | 2 |
