1Four kinds of ENU-induced white spot mice and chromosome locations of the mutant genes显示文摘Phenotype-driven is the name for an ap-proach used to study gene functions through mutagenesis, location and cloning of the mutant gene. In this study, 150 male C57BL/6J(B6) mice were treated with ENU and repro-duced a total offspring of 3860. Of these descendants, 210 exhibited mutation phenotypes by screening, and more than 10 of them are hereditable. Four kinds of mutant mice, named Wbct, W-1Bao, W-2Bao, and W-3Bao, showed dominant hereditary white spot mutation with partial albinism on their belly, distal limbs and tail terminal. To map these mutant genes, 39 microsatellites, equally distributed on the mouse genome and with difference between B6 and DBA/2J (D2), were selected to scan the genome after discrimination of the white spots in the F2 mice [(B6×D2)×D2]. It is found that, the log odds score (LODS) between W-1Bao and D5Mit168 is 0.56, and the LODS of W-1Bao and D5Mit352 is 4.47. With the gradual application of microsatellites D5Mit290, D5Mit312, D5Mit308 and D5Mit356 that are close to the mutant gene, and the number of F2 mice going up to 537, the mutant W-1Bao is located between D5Mit356 and D5Mit308 on chro-mosome 5, about 42.19 cM from the centromere. In the same way, W-2Bao and W-3Bao are mapped nearby W-1Bao, and Wbct is located on chromosome 1, about 41.6 cM from the centro-mere. After searching for the mouse genome database (MGD) and performing a one-by-one study of all genes located on chromosome subregion, it is believed that the kit gene is an excellent candidate for the white spot mutations of W-1Bao, W-2Bao and W-3Bao.WU Baojin1,2, MAO Huihua1, SHAO Yixiang1, XUE Zhengfeng1 & LI Houda1 1. 2Isolation and characterization of facultative mixotrophic ammonia-oxidizing bacteria from constructed wetlands显示文摘Autotrophic ammonia-oxidizing bacteria (AOB) have been widely studied in constructed wetlands systems,while mixotrophic AOB have been less thoroughly examined.Heterotrophic bacteria were isolated from wastewater and rhizospheres of macrophytes of constructed wetlands,and then cultivated in a mixotrophic medium containing ammonium and acetic acid.A molecular characterization was accomplished using ITS-PCR amplification,and phylogenetic analysis based on 16S rRNA gene sequences.Results showed the presence of 35 bacteria,among 400 initially heterotrophic isolates,that were able to remove ammonia.These 35 isolates were classified into 10 genetically different groups based on ITS pattern.Then,a collection of 10 isolates were selected because of their relatively high ammonia removal efficiencies (ARE 80%) and their phylogenetic diversity.In conditions of mixotrophy,these strains were shown to be able to grow (increase of optical density OD 660 during incubation with assimilation of nitrogen into cellular biomass) and to oxidize ammonia (important ammonia oxidation efficiencies,AOE between 79% and 87%).Among these facultative mixotrophic AOB,four isolates were genetically related to Firmicutes (Bacillus and Exiguobacterium),three isolates were affiliated to Actinobacteria (Arthrobacter) and three other isolates were associated with Proteobacteria (Pseudomonas,Ochrobactrum and Bordetella).Soulwène Kouki Neila Saidi Fadhel M'hiri Houda Nasr Hanène Cherif Hadda Ouzari Abdennaceur Hassen 2011Journal of Environmental Sciences2011,23,10:9
3A New Sliding Function for Discrete Predictive Sliding Mode Control of Time Delay Systems显示文摘The control of time delay systems is still an open area for research. This paper proposes an enhanced model predictive discrete-time sliding mode control with a new sliding function for a linear system with state delay. Firstly, a new sliding function including a present value and a past value of the state, called dynamic surface, is designed by means of linear matrix inequalities (LMIs). Then, using this dynamic function and the rolling optimization method in the predictive control strategy, a discrete predictive sliding mode controller is synthesized. This new strategy is proposed to eliminate the undesirable effect of the delay term in the closed loop system. Also, the designed control strategy is more robust, and has a chattering reduction property and a faster convergence of the system s state. 4褐铁矿处理水中六价铬显示文摘Cr(Ⅵ)是土壤和水体中常见的有毒污染物,将Cr(Ⅵ )转化为毒性较低的Cr(Ⅲ)是常用而有效的治理方法.本文采用褐铁矿对水中Cr(Ⅵ )进行处理研究,考察了褐铁矿投加量、Cr(Ⅵ)初始浓度、pH值、反应温度、铁矿石粒径大小以及添加不同物质等因素对褐铁矿去除效果的影响.结果表明,褐铁矿对Cr(Ⅵ)有很好的还原去除效果,Cr(Ⅵ)去除效率随褐铁矿用量的增加而增大,随Cr(Ⅵ )初始浓度的增加而减小,pH值越低越有利于反应的进行.铁矿石颗粒的直径越小,比表面积越大,也对反应越有利,而温度对Cr(Ⅵ)的去除影响不大.100目的褐铁矿颗粒去除Cr(Ⅵ )的能力为7.77mgCr(VI)/g.使用褐铁矿不仅可以将Cr(Ⅵ)还原为Cr(Ⅲ),还能将Cr(Ⅵ )彻底从水中去除.吴燕君 王倩 Zaidi Houda 张珍 徐新华 2008浙江大学学报(理学版)2008,35,6:6
5Cytokine and apoptosis gene polymorphisms influence the outcome of hepatitis C virus infection显示文摘BACKGROUND:Hepatitis C virus (HCV) infection is thought to be chronic and the factors leading to viral clearance or persistence are poorly understood.This study was undertaken to investigate the possibility of a significant relationship between the spontaneous clearance or the persistence of hepatitis C virus (HCV) infection and cytokine and apoptosis gene polymorphisms in Tunisian patients on hemodialysis.METHODS:Polymorphisms of the genes IL-1 (-889 IL-1α,-511 and +3954 IL-1β,IL-1Ra),IL-18 (-137 and-607),IL-12 (-1188) and Apo1/Fas (-670) were determined by PCR-RFLP,PCR-SSP and PCR-VNTR in 100 healthy blood donors and 100 patients infected with HCV and undergoing hemodialysis.The patients were classified into two groups:G1 consisted of 76 active chronic hepatitis patients (positive for HCV RNA) and G2 consisted of 24 hemodialysed patients who spontaneously eliminated the virus (negative for HCV RNA).RESULTS:The frequency of genotype association [-137GC/-607CA] IL-18 was higher in G2 (41.7%) than in G1 (15.8%) (P=0.008;OR=0.26;95% CI,0.10-0.73).We also found a higher frequency of the AA genotype of the Apo1/Fas gene in G2 (41.6%) than in G1 (17.5%) (P=0.026;OR=3.49;95% CI,1.13-10.69).Adjustment for known covariate factors (age,gender and genotype) confirmed these univariate findings and revealed that the genotype association GC-CA of the (-137 and-607) IL-18 gene and the AA genotype of the Apo1/Fas gene were associated with the clearance of HCV (P=0.041 and 0.017,respectively).CONCLUSION:The two genotypes GC-CA of the (-137 and-607) IL-18 polymorphism and the AA genotype of the Apo1/Fas gene influence the outcome of HCV infection in Tunisian patients on hemodialysis.Leila Ksiaa Cheikhrouhou Imen Sfar Hajer Aounallah-Skhiri Houda Aouadi Salwa Jendoubi-Ayed Taieb Ben Abdallah Khaled Ayed Yousr Lakhoua-Gorgi 2011Hepatobiliary & Pancreatic Diseases International2011,10,3:4
6Analysis of the Influence of Viscosity and Thermal Conductivity on Heat Transfer by Al2O3-Water Nanofluid显示文摘The addition of nanoparticles into liquid,even at low concentrations,leads to an increase in both,dynamic viscosity and thermal conductivity.Furthermore,the increase in temperature causes an increase in thermal conductivity and a decrease in the nanofluid viscosity.In this context,a numerical investigation of the competition between viscosity and thermal conductivity about their effects on heat transfer by Al_(2)O_(3)-water nanofluid was conducted.A numerical study of heat transfer in a square cavity,filled with Al_(2)O_(3)-water nanofluid and heated from the left side,was presented in this paper.Continuity,momentum,and thermal energy equations are solved by the finite volume method.Regarding the pressure-velocity coupling,the SIMPLER algorithm was used.The working conditions,allowing the increase of heat transfer,are established.In addition,two correlations for viscosity and thermal conductivity of Al_(2)O_(3)-water nanofluid as functions of the concentration and diameter size based on experimental measurement are proposed.These correlations were more precisely compared to those given by the theoretical models.Moreover,other models for viscosity and conductivity depending on temperature are used and discussed.The results reveal that heat transfer by Al_(2)O_(3)-water nanofluid is enhanced only when the temperature exceeds 40℃ and the diameter size does not exceed a certain limit of the order of 45-50 nm depending on temperature.Houda Jalali Hassan Abbassi 2020Fluid Dynamics & Materials Processing2020,16,2:3
7Solid-state characterization and impurities determination of fluconazol generic products marketed in Morocco显示文摘In this paper, we report the results of quality control based in physicochemical characterization and impurities determination of three samples of fluconazole drug substances marketed in Morocco. These samples were supplied by different pharmaceuticals companies. The sample A, as the discovered product, was supplied by Pfizer, while samples B and C (generics), were manufactured by two different Indian industries. Solid-state characterization of the three samples was realized with different physicochemical methods as: X-ray powder diffraction, Fourier-transformation infrared spectroscopy, differential scanning calorimetry. High performance liquid chromatography was used to quantify the impurities in the different samples. The results from the physicochemical methods cited above, showed difference in polymorph structure of the three drug substances. Sample A consisted in pure polymorph Ⅲ, sample B consisted in pure polymorph Ⅱ, sample C consisted in a mixture of fluconazole Form Ⅲ, form Ⅱ and the monohydrate. This result was confirmed by differential scanning calorimetry. Also it was demonstrated that solvents used during the re-crystallization step were among the origins of these differences in the structure form. On the other hand, the result of the stability study under humidity and temperature showed that fluconazole polymorphic transformation could be owed to the no compliance with the conditions of storage. 8Antioxidant Machinery Related to Decreased MDA Generation by Thymus Algeriensis Essential Oil-induced Liver and Kidney Regeneration显示文摘Objective This study was conducted to determine the histopathological and biochemical effects of Thymus algeriensis essential oil(TEO) on hydrogen peroxide(H_2O_2)-induced oxidative stress in liver and kidney tissues of rats. Methods Rats were treated in six groups and were exposed for 2 weeks to low(LD; 100 μmol/L) and high doses(HD;1 mmol/L) of H_2O_2 in the presence or absence of TEO(180 mg/kg). Liver and kidney atrophy was measured by using biochemical and histopathological assays. Results Our study demonstrated that H_2O_2 induced liver and kidney atrophy, as evidenced by the significant elevation of serum aminotransferase, urea, and creatinine levels compared with those in the control rats. Urea levels were estimated by evaluating the activity of serum urease that hydrolyzes urea into CO_2 and ammonia. However, TEO treatment significantly alleviated oxidative stress in the H_2O_2-induced liver and kidney toxicity model by reducing the levels of malondialdehyde concomitantly with marked elevations in superoxide dismutase, catalase, glutathione peroxidase, and glutathione S-transferase, as well as decrease in glutathione activity. 9Association of Fas/Apo1 gene promoter (-670 A/G) polymorphism in Tunisian patients with IBD显示文摘AIM: To detect a possible association between the polymorphism of the (-670 A/G) Fas/Apo1 gene promoter and susceptibility to Crohn's disease (CD) and ulcerative colitis (UC) in the Tunisian population. METHODS: The (-670 A/G) Fas polymorphism was analyzed in 105 patients with CD, 59 patients with UC, and 100 controls using the polymerase chain reaction restriction fragment length polymorphism method. RESULTS: Significantly lower frequencies of the Fas -670 A allele and A/A homozygous individuals were observed in CD and UC patients when compared with controls. Analysis of (-670 A/G) Fas polymorphism with respect to sex in CD and UC showed a significant difference in A/A genotypes between female patients and controls (P corrected = 0.004 in CD patients and P corrected = 0.02 in UC patients, respectively). Analysis also showed a statistically significant association between genotype AA of the (-670 A/G) polymorphism and the ileum localization of the lesions (P corrected = 0.048) and between genotype GG and the colon localization (Pcorrected = 0.009). The analysis ofinflammatory bowel disease patients according to clinical behavior revealed no difference. 11Aggressive primary hepatic epithelioid hemangioendothelioma:a case report and literature review显示文摘A new case of epithelioid hemangioendothelioma is reported to have occurred to a 67-year-old patient who consulted for rightsided chest pain. The work-up showed multiple right pulmonary lesions associated with bilateral moderate pleural effusion and left-sided pleural thickening and three hypodense nodules in the right lobe of the liver, peritoneal thickening, ascites, and multiple vertebral lytic lesions. The diagnosis of an epithelioid hemangioendothelioma was concluded through a histological examination of a computed tomography scan guided biopsy of the liver. The patient received a primary mono-chemotherapy with Adriamycin(75 mg/m^2 every three weeks) and intravenous bisphosphonates without response and general status impairment. 12Second Order Sliding Mode Control for Discrete Decouplable Multivariable Systems via Input-output Models显示文摘The problem of the chattering phenomenon is still the main drawback of the classical sliding mode control. To resolve this problem, a discrete second order sliding mode control via input-output model is proposed in this paper. The proposed control law is synthesized for decouplable multivariable systems. A robustness analysis of the proposed discrete second order sliding mode control is carried out. 15Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants,are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. 17Effects of Barium Stress in Brassica juncea and Cakile maritima:The Indicator Role of Some Antioxidant Enzymes and Secondary Metabolites显示文摘Soil contamination by toxic trace metal elements,like barium(Ba),may stimulate various undesirable changes in the metabolic activity of plants.The plant responses are fast and with,direct or indirect,generation of reactive oxygen species(ROS).To cope with the stress imposed by the ROS production,plants developed a dual cellular system composed of enzymatic and non-enzymatic players that convert ROS,and their by-products,into stable nontoxic molecules.To assess the Ba stress response of two Brassicaceae species(Brassica juncea,a glycophyte,and Cakile maritime,a halophyte),plants were exposure to different Ba concentrations(0,100,200,300 and 500µM).The plants response was evaluated through their morphology and development,the determination of plant leaves antioxidant enzymatic activities and by the production of plants secondary metabolites.Results indicated that the two Brassicaceae species have the ability to survive in an environment containing Ba(even at 500µM).The biomass production of C.maritima was slightly affected whereas an increase in biomass B.juncea was noticed.The stress imposed by Ba activated the antioxidant defense system in the two species,noticed by the changes in the leaves activity of catalase(CAT),ascorbate peroxidase(APX)and guaicol peroxidase(GPX),and of the secondary metabolites,through the production of total phenols and flavonoids.The enzymatic response was not similar within the two plant species:CAT and APX seem to have a more important role against the oxidative stress in C.maritima while in B.juncea is GPX.Overall,total phenols and flavonoids production was more significant in the plants aerial part than in the roots,of the both species.Although the two Brassicaceae species response was different,in both plants catalytic and non-catalytic transformation of ROS occurs,and both were able to overcome the Ba toxicity and prevent the cell damage.Houda Bouslimi Renata Ferreira Nesrine Dridi Pedro Brito Susete Martins-Dias Isabel Caçador Noomene Sleimi 2021Phyton-International Journal of Experimental Botany2021,90,1:2
18Natural evolution of hepatitis C virus infection in hemodialysis Tunisian patients and CTLA-4 SNP's显示文摘AIM: To analyze the polymorphisms of CTLA-4 gene involved in the response against hepatitis C virus(HCV) infection.METHODS: We recruited 500 hemodialysed patients from several hemodialysis centers, all HCV-antibody positive, spread over different regions of Tunisia, as part of a national survey in 2008 conducted in the laboratory of immunology at the Charles Nicolle hospital Tunisia, classified into two groups G1(PCR+) and G2(PCR-) according to the presence or absence of viral RNA. Of these patients, 307 were followed prospectively on a viral molecular level over a period from 2002 to 2008, divided into two groups based on the persistence and viral clearance. PCR-RFLP was performed for the analysis of SNPs(+49) A/G and(+6230) G/A CTLA-4 for these 500 patients and 358 healthy controls.RESULTS: Analysis of clinical and virological charac-teristics of our cohort suggests a nosocomial infection in our hemodialysed patients with transfusion history as a primary risk factor and a predominance of genotype 1b. The haplotype analysis revealed an increase of frequencies of GG(+49)/(CT60) CTLA-4 in the entire patients group compared to controls(P = 0.0036 and OR = 1.42; 95%CI: 1.12-1.79, respectively). This haplotype is therefore associated with susceptibility to HCV infection.
