| 2 | The relationship between platelet endothelial cell adhesion molecule-1 and paraquat-induced lung injury in rabbits显示文摘BACKGROUND:Platelet endothelial cell adhesion molecule-1(PECAM-1),also known as CD31,is mainly distributed in vascular endothelial cells.Studies have shown that PECAM-1 is a very significant indicator of angiogenesis,and has been used as an indicator for vascular endothelial cells.The present study aimed to explore the relationship between the expression of PECAM-1 and the degree of acute lung injury(ALI) and fibrosis in paraquat(PQ) induced lung injury in rabbits.METHODS:Thirty-six adult New Zealand rabbits were randomly divided into three groups(12rabbits in each group) according to PQ dosage:8 mg/kg(group A),16 mg/kg(group B),and 32 mg/kg(group C).After PQ infusion,the rabbits were monitored for 7 days and then euthanized.The lungs were removed for histological evaluation.Masson staining was used to determine the degree of lung fibrosis(LF),and semi-quantitative immune-histochemistry analysis to determine the expression of PECAM-1.Pearson's product-moment correlation analysis was performed to evaluate the relationship between the expression of PECAM-1 and the extent of lung injuries expressed by ALI score and degree of LF.RESULTS:Rabbits in the three groups showed apparent poisoning.The rabbits survived longer in group A than in groups B and C(6.47±0.99 days vs.6.09±1.04 days vs.4.77±2.04 days)(P<0.05).ALI score was lower in group A than in groups B and C(8.33±1.03 vs.9.83±1.17 vs.11.50±1.38)(P<0.05),and there was statistically significant difference between group B and group C(P=0.03).LF was slighter in group A than in groups B and C(31.09%±2.05%vs.34.37%±1.62%vs.36.54%±0.44%)(P<0.05),and there was statistically significant difference between group B and group C(P=0.026).The PEACAM-1 expression was higher in group A than in groups B and C(20.31%±0.70%vs.19.34%±0.68%vs.18.37%±0.46%)(P<0.05),and there was statistically significant difference between group B and group C(P=0.017).Pearson's correlation analysis showed that the expression of PECAM-1 was negatively correlated to both ALI score(Coe=-0.732,P=0.001)and degree of LF(Coe=-0.779,P<0.001).CONCLUSIONS:The PECAM-1 expression significantly decreases in New Zealand rabbits after PQ poisoning,and the decrease is dose-dependent.The PECAM-1 expression is negatively correlated with ALI score and LF,showing a significant role in the development of lung injuries induced by PQ. | Jing Shi Chun-lin Hu Yu-feng Gao Xiao-xing Liao Hope Xu | 2012 | World Journal of Emergency Medicine2012，3，1： | 7 |
| 3 | 晚期Luminal(HER2阴性)型乳腺癌患者的MDT诊疗显示文摘本文介绍1例激素受体阳性、人表皮生长因子受体-2(human epidermal growth factor receptor-2,HER2)阴性晚期乳腺癌多线内分泌治疗的多学科诊疗过程。该病例有较长的无瘤生存期(10年),初始复发转移肿瘤分布范围广,涉及两肺多发、多处骨骼及颈部锁骨上等区域多发淋巴结转移,经锁骨上转移淋巴结穿刺病理证实,免疫组织化学检测仍为雌激素受体(estrogen receptor,ER)强阳性/HER2阴性,但是孕激素受体(progesterone receptor,PR)仅小区阳性。经过多学科讨论后患者依次一线接受高剂量氟维司群(500 mg,肌注,28 d/次,首月第14天负荷剂量)内分泌治疗,二线选择哌柏西利(125 mg/d,服21 d,休7 d)+来曲唑(2. 5 mg/d),三线接受哌柏西利+氟维司群(同前),其2次进展仅表现为新发淋巴结转移或胸腔积液,并未发现内脏新发病灶或者病灶增大。其一、二线内分泌治疗的无进展生存期(progression free survival,PFS)均为12个月,三线内分泌治疗保留哌柏西利,换用一线的内分泌治疗药物(氟维司群)。临床上,还有全身化疗和局部放疗等选择。由于患者自发病以来一直拒绝接受化疗,术后辅助仅选择阿那曲唑(1 mg/d)辅助内分泌治疗(5年),解救治疗期间同样要求仅选择内分泌治疗。鉴于患者的意愿和肿瘤特征与演变、分子检测与监测,结合MDT专家意见,连续选择内分泌治疗作为解救治疗,取得较长的疾病控制实践,患者维持较好的生活质量。 | 曹文明 邓雪英 陈波 杜向慧 陈占红 郑亚兵 Hope Rugo Geraldine M.Jacobson 郑鸿钧 王晓稼 | 2018 | 实用肿瘤杂志2018，33，6： | 6 |
| 4 | Failed biliary cannulation: Clinical and technical outcomes after tertiary referral endoscopic retrograde cholangiopancreatography显示文摘AIM: Prospective evaluation of repeat endoscopic retrograde cholangiopancreatography (ERCP) for failed Schutz grade 1 biliary cannulation in a high-volume center. METHODS: Prospective intention-to-treat analysis of patients referred for biliary cannulation following recent unsuccessful ERCP. RESULTS: Fifty-one patients (35 female; mean age: 62.5 years; age range: 40-87 years) with previous failed biliary cannulation were referred for repeat ERCP. The indication for ERCP was primarily choledocholithiasis (45%) or pancreatic malignancy (18%). Successful biliary can- nulation was 100%. The precut needle knife sphincterotomy (NKS) rate was 27.4%. Complications occurred in 3.9% (post-ERCP pancreatitis). An identif iable reason for initial unsuccessful biliary cannulation was present in 55% of cases. Compared to a cohort of 940 nave pa-pilla patients (female 61%; mean age: 59.9 years; age range: 18-94 years) who required sphincterotomy over the same time period, there was no statistical difference in the cannulation success rate (100% vs 98%) or postERCP pancreatitis (3.1% vs 3.9%). Precut NKS use was more frequent (27.4% vs 12.7%) (P = 0.017). After briefly reviewing the standard agency cost arguments, in this article I emphasize the heterogeneity of concentrated ownership and in particular focus on the roles of families, institutions, governments, and employee ownership. I also discuss the role of large shareholders in private(i.e., unlisted) firms, where ownership tends to be more concentrated than in publicly traded firms. Unlike the almost ubiquitous GPX-1, GPX-GI is mainly expressed in the epithelium of gastrointestinal tract. GPX-GI contributes to at least fifty percent of GPX activity in rodent small intestmal epithelium. The total GPX activity consists of at least 70% of selenium-dependent GPX activity in this compartment.By analyzing a panel of mouse mterspecies DNA from the Jackson Laboratory’s backcross resource,we mapped Gpx2 gene to mouse chromosome 12 between D12Mit4 and D12Mit5, near the Ccs1 locus which contains a colon cancer susceptibility gene. A pseudogene, Gpx2-ps is mapped to mouse chromosome 7.Comparison of Gpx2 gene expression in three pairs of C57BL/6Ha and ICR/Ha mice which are respectively resistant and sensitive to dimethylhydrazine-induced colon cancer, we found a higher Gpx2 mRNA level in C57BL/6Ha colon than ICR/Ha colon. Interestingly, a lower level of GPX activity is found in the resistant strain of mice. | 15 | Immune profiling and cancer post transplantation显示文摘Half of all long-term(> 10 year) australian kidney transplant recipients(KTR) will develop squamous cell carcinoma(SCC) or solid organ cancer(SOC), making cancer the leading cause of death with a functioning graft. At least 30% of KTR with a history of SCC or SOC will develop a subsequent SCC orSOC lesion. Pharmacological immunosuppression is a major contributor of the increased risk of cancer for KTR, with the cancer lesions themselves further adding to systemic immunosuppression and could explain, in part, these phenomena. Immune profiling includes; measuring immunosuppressive drug levels and pharmacokinetics, enumerating leucocytes and leucocyte subsets as well as testing leucocyte function in either an antigen specific or non-specific manner. Outputs can vary from assay to assay according to methods used. In this review we define the rationale behind post-transplant immune monitoring assays and focus on assays that associate and/or have the ability to predict cancer and rejection in the KTR. We find that immune monitoring can identify those KTR of developing multiple SCC lesions and provide evidence they may benefit from pharmacological immunosuppressive drug dose reductions.
