| 1 | An in vivo study with an MRI tracer method reveals the biophysical properties of interstitial fluid in the rat brain显示文摘The nature of brain interstitial fluid (ISF) has long been a subject of controversy. Most of the previous studies on brain ISF were carried out in vitro. In the present study, a novel method was developed to characterize ISF in the living rat brain by magnetic resonance (MR) imaging using gadolinium-diethylenetriaminepentaacetic acid (Gd-DTPA) as a tracer. Sprague Dawley rats (n=8) were subjected to MR scanning before and after the introduction of Gd-DTPA into the caudate nucleus. A one-way drainage of brain ISF was demonstrated on the dynamic MR images. According to the traditional diffusion model, the diffusion and clearance rate constants of the tracer within brain extracellular space (ECS) were derived as (3.38±1.07)×10-4 mm 2 s-1 and (7.60±4.18)×10-5 s-1 . Both diffusion and bulk flow contributed to the drainage of ISF from the caudate nucleus, which demonstrated an ISF-cerebrospinal fluid confluence in the subarachnoid space at the lateral and ventral surface of the brain cortex at 3 h after the injection. | 4 | PhaseⅡstudy of induction chemotherapy followed by concurrent chemoradiotherapy with raltitrexed and cisplatin in locally advanced nasopharyngeal carcinoma显示文摘Objective:For locally advanced nasopharyngeal carcinoma(LA-NPC)patients,high incidences of distant metastases and severe treatment related toxicities are the main obstacles needed to be overcome.Raltitrexed,a specific thymidylate synthase inhibitor with a convenient administration schedule,has an acceptable and manageable toxicity,and possesses radio-sensitizing properties.To investigate the efficacy and safety of raltitrexed and cisplatin induction chemotherapy and concurrent chemoradiotherapy(IC+CCRT)in patients with LA-NPC,a phaseⅡclinical study was conducted.Methods:Sixty eligible patients with LA-NPC were enrolled into this study.A raltitrexed-cisplatin combination was used as part of an IC+CCRT regimen.Raltitrexed-cisplatin IC was given once every 3 weeks(q3 w)for two cycles,followed by raltitrexed-cisplatin based CCRT q3 w for two cycles.Intensity-modulated radiotherapy(IMRT)was given for all enrolled patients.Results:All patients were included in survival analysis according to the intent-to-treat principle.The objective response rate(ORR)3 months after treatment was 98%.The 2-year overall survival(OS)rate was 92%.The median relapse-free survival(RFS)time was 30.5[95%confidence interval(95%CI),28.4-32.3]months.The 2-year RFS rate was 85%.The 2-year local failure-free survival(LFFS)rate was 97%and the 2-year distant metastasis-free survival(DMFS)rate was 88%.Acute toxicities were mostly grade 2 and 3 reactions in bone marrow suppression,gastrointestinal side effect and oropharyngeal mucositis.Only two patients occurred grade 4 acute toxicities,one was bone marrow suppression and the other was dermatitis radiation.Conclusions:The combination of raltitrexed and cisplatin has a comparable efficacy to those in standard firstline therapy. | Yuan Wu Xueyan Wei Zilong Yuan Hongbin Xu Yanping Li Ying Li Liu Hu Guang Han Yu Qian Desheng Hu | 2020 | Chinese Journal of Cancer Research2020,32,5: | 4 |
| 5 | SARS-CoV-2 ORF10 suppresses the antiviral innate immune response by degrading MAVS through mitophagy显示文摘The global coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused severe morbidity and mortality in humans.It is urgent to understand the function of viral genes.However,the function of open reading frame 10(ORF10),which is uniquely expressed by SARS-CoV-2,remains unclear.In this study,we showed that overexpression of ORF10 markedly suppressed the expression of type I interferon(IFN-I)genes and IFN-stimulated genes.Then,mitochondrial antiviral signaling protein(MAVS)was identified as the target via which ORF10 suppresses the IFN-I signaling pathway,and MAVS was found to be degraded through the ORF10-induced autophagy pathway.Furthermore,overexpression of ORF10 promoted the accumulation of LC3 in mitochondria and induced mitophagy.Mechanistically,ORF10 was translocated to mitochondria by interacting with the mitophagy receptor Nip3-like protein X(NIX)and induced mitophagy through its interaction with both NIX and LC3B.Moreover,knockdown of NIX expression blocked mitophagy activation,MAVS degradation,and IFN-I signaling pathway inhibition by ORF10.Consistent with our observations,in the context of SARS-CoV-2 infection,ORF10 inhibited MAVS expression and facilitated viral replication.In brief,our results reveal a novel mechanism by which SARS-CoV-2 inhibits the innate immune response;that is,ORF10 induces mitophagy-mediated MAVS degradation by binding to NIX. | Xingyu Li Peili Hou Wenqing Ma Xuefeng Wang Hongmei Wang Zhangping Yu Huasong Chang Tiecheng Wang Song Jin Xue Wang Wenqi Wang Yudong Zhao Yong Zhao Chunqing Xu Xiaomei Ma Yuwei Gao Hongbin He | 2022 | Cellular & Molecular Immunology2022,19,1: | 4 |

Among 118 putative miRNAs,47% and 53% of the predicted miRNAs from the red flour beetle are harbored by known protein-coding genes (intronic) and genes located outside (intergenic miRNA),respectively. | 8 | IAP's Solar-Powered Unmanned Surface Vehicle Actively Passes through the Center of typhoon Sinlaku(2020)显示文摘The solar-powered marine unmanned surface vehicle(USV) developed by the USV team of the Institute of Atmospheric Physics is a rugged, long-duration, and autonomous navigation vessel designed for the collection of longrange, continuous, real-time, meteorological and oceanographic measurements, especially under extreme sea conditions(sea state 6–7). These solar-powered USVs completed a long-term continuous navigation observation test over 26 days.During this time, they coordinated double-USV observations and actively navigated into the path of Typhoon Sinlaku(2020) before collecting data very close to its center during the 2020 USV South China Sea Typhoon Observation Experiment. Detailed high temporal resolution(1 min) real-time observations collected by the USV on the typhoon were used for operational typhoon forecasting and warning for the first time. | 10 | Self-lubricate and anisotropic wear behavior of AZ91D magnesium alloy reinforced with ternary Ti_2AlC MAX phases显示文摘The dry sliding wear behavior of Ti_2AlC reinforced AZ91 magnesium composites was investigated at sliding velocity of 0.5 m/s under loads of 10, 20, 40 and 80 N using pin-on-disk configuration against a Cr15 steel disc. Wear rates and friction coefficients were registered during wear tests. Worn tracks and wear debris were examined by scanning electron microscopy, energy dispersive X-ray spectrometry and transmission electron microscopy in order to obtain the wear mechanisms of the studied materials. The main mechanisms were characterized as the magnesium matrix oxidation and self-lubrication of Ti_2AlC MAX phase. In all conditions, the composites exhibit superior wear resistance and self-lubricated ability than the AZ91 Mg alloy. | 11 | Effects of ethanol extracts of scorpion on hippocampal apoptosis and caspase-3 expression in lithium chloride-pilocarpine-induced status epilepticus rats显示文摘BACKGROUND: Previous studies have demonstrated that scorpion venom in the scorpion can inhibit epilepsy and apoptosis.However,it remains unclear whether ethanol extracts of scorpion (EES) exhibit similar effects.OBJECTIVE: To investigate the effects of EES on hippocampal apoptosis and caspase-3 expression,and to compare the effects on sodium valproate (positive control drug) in a rat model of status epilepticus induced by lithium chloride-pilocarpine.DESIGN,TIME AND SETTING: This randomized,controlled study was conducted at the Drug Research and Development Center,Kanghong Pharmaceuticals Group,and the Department of Pathology,Sichuan Academy of Medical Sciences & Sichuan Provincial People’s Hospital,China from May 2007 to April 2008.MATERIALS: EES were prepared by Huashen Pharmaceutical,China.Sodium valproate (Hunan Xiangzhong Pharmaceutical,China) and lithium chloride-pilocarpine (Sigma,USA) were also used in the present study.METHODS: From a total of 156 rats,six served as normal controls.The remaining rats were intraperitoneally injected with lithium chloride-pilocarpine to establish status epilepticus models,and then assigned to five groups (n = 30,respectively).Animals in each group were administered drugs at 15 minutes after epileptic seizure by gavage.i.e.in the normal control and model groups,rats were treated with 1 mL/0.1 kg saline.The sodium valproate group was administered 120 mg/kg/d sodium valproate.The low-,moderate-,and high-dose EES groups received treatments of 290,580,and 1 160 mg/kg/d EES.The dispensed concentration was 1 mL/0.1 kg.Rat seizure behavior was observed.If status epilepticus did not terminated after 1 hour,the rats were intraperitoneally administered atropine (1 mg/kg) and diazepam (10 mg/kg) to terminate seizure.These rats were continuously observed for 6 hours to ensure seizure termination.Then rats were treated with the above-mentioned drugs at 8: 00 am each day until sacrifice,which took place 4 hours after drug administration.MAIN OUTCOME MEASURES: Terminal dUTP nick end labeling (TUNEL)-positive cells and caspase-3 expression were,respectively,determined by TUNEL and immunohistochemistry at 6,24,48,and 72 hours,as well as 7 days,after status epilepticus.Behavioral changes were also measured.RESULTS: A few caspase-3-positive cells were observed.TUNEL-and caspase-3-positive cells were mainly visible in the hippocampal CA1 and CA3 regions 6 hours following status epilepticus in the model and drug intervention groups.The number of TUNEL-positive cells reached a peak at 48 hours following status epilepticus in the sodium valproate group,as well as the moderate-and high-dose EES groups,and number of TUNEL-positive cells reached a peak at 72 hours in the model and low-dose EES groups.The number of caspase-3-positive cells reached a peak at 48 hours in each group.Following treatment of sodium valproate and EES,the number of TUNEL-and caspase-3-positive cells significantly decreased compared with the model group at various time points (P < 0.05).The number of TUNEL-and caspase-3-positive cells was greatest in the low-dose EES group,followed by the moderate-and high-dose EES groups.The number of TUNEL-and caspase-3-positive cells was similar between the sodium valproate and high-dose EES groups.Epileptic seizure was significantly improved in the sodium valproate group,as well as the moderate-and high-dose EES groups,compared with the model group (P < 0.05 or P < 0.01).Treatment with sodium valproate and high-dose EES resulted in the best outcome,although the results were similar (P > 0.05).CONCLUSION: A dose of 1 160 mg/kg/d EES significantly inhibited status epilepticus.This outcome corresponded to a decreased number of apoptotic cells and caspase-3-positive cells,which was similar to sodium valproate.These results suggest that it is not necessary to extract a component from the scorpion for the treatment of epilepsy.The high dose of EES significantly inhibited epilepsy,which correlated with decreased hippocampal caspase-3 expression. | Liang Yu Hongbin Sun Yi Liang Yan Xie Baoming He Fei Xu | 2010 | Neural Regeneration Research2010,5,2: | 2 |
| 12 | Scorpion ethanol extract and valproic acid effects on hippocampal glial fibrillary acidic protein expression in a rat model of chronic-kindling epilepsy induced by lithium chloride-pilocarpine显示文摘The present study analyzed the effects of ethanol extracts of scorpion on epilepsy prevention and hippocampal expression of glial fibrillary acidic protein in a lithium chloride-pilocarpine epileptic rat model. Results were subsequently compared with valproic acid. Results showed gradually- increased hippocampal glial fibrillary acidic protein expression following model establishment; glial fibrillary acidic protein mRNA expression was significantly increased at 3 days, reached a peak at 7 days, and then gradually decreased thereafter. Ethanol extracts of scorpion doses of 580 and 1 160 mg/kg, as well as 120 mg/kg valproic acid, led to a decreased number of glial fibrillary acidic protein-positive cells and glial fibrillary acidic protein mRNA expression, as well as decreased seizure grades and frequency of spontaneously recurrent seizures. The effects of 1 160 mg/kg ethanol extracts of scorpion were equal to those of 120 mg/kg valproic acid. | 13 | Effects of topiramate versus other antiepileptic drugs on the cognitive function of patients with epilepsy显示文摘BACKGROUND: Only very large dose of topiramate has neurotoxicity, indicating that topiramate has low neurotoxicity and high safety. The residual rate of topiramate is affected by many cognitive-related adverse effects. Patients who take topiramate often accompany with thought slowness, difficulty in finding words, dyscalculia, blunt reaction, attention decreasing, memory deterioration, etc. OBJECTIVE: To compare the effects of topiramate with traditional anti-epileptic drugs (including carbamazepine and Valproic acid (VPA) on cognitive function of patients with epilepsy. DESIGN: Observational experiment, self-control and intergroup comparison. SETTING: Sichuan Academy of Medical Science. PARTICIPANTS: Eighty-seven inpatients and outpatients with newly diagnosed epilepsy who received preliminary diagnosis and follow-up in the Department of Neurology, Sichuan People's Hospital between January 2004 and June 2006 were involved in this survey. They were diagnosed according to disease history and electroencephalogram (EEG). The onset type was diagnosed following the definition of epilepsy and epileptic syndrome in 1989 International Anti-epileptic League. The involved patients and their relatives were informed of detection and therapeutic regimen. The patients were assigned into two groups according to table of random digit: traditional antiepileptic drugs group (AEDs group, n =44) and topiramate (TPM) group (n =43). METHODS: ①Among the patients in AEDs group, carbamazepine was the first choice for 21 patients with partial seizures or partial secondarily generalized seizures, and VPA for 23 patients with generalized seizures. The initial dose of carbamazepine was 300 mg/d, and that of VPA was 500 mg/d. Patients in the TPM group took TPM with the initial dose of 25 mg/d, increased by 25 mg/d each week to target dose 150 mg/d within 8 weeks. ② Curative effect was graded into 4 degrees: markedly effective, effective, ineffective and aggravated. Total effective rate was calculated. ③ Cognitive function of patients was tested before and 6 months after administration by using Wechsler Adult Intelligence Scale(WAIS) or Wechsler Intelligence Scale for Children (WISC, Chinese edition), (Higher scores indicated better cognitive function), Stroop color word interference, test of memory of past numbers, test of telling the names of fruits and vegetables within 1 minute (Shorter time for reading word, telling color and memory of past numbers demonstrated better cognitive function. Less errors in reading words, telling colors and memory of past numbers, numbering and telling the names of fruits and vegetables within 1 minute indicated better cognitive function), etc. totally 22 items. ④ t test and paired t test were used for measurement data. MAIN OUTCOME MEASURES: Clinical curative effects and adverse reactions as well as neurological tests. RESULTS: Eighty-four pationts praticipated final analysis and 3 dropped out. ① Inthe AEDS group and TPM group, total effective rate was 86% and 99%, respectively. ② In the AEDs group, there were no significant changes in the scores of each test of WIS before and after treatment (P > 0.05). In the TPM group, total IQ, word scores, verbal IQ and digit span scores were significantly decreased (t =2.097–4.423,P < 0.05–0.01).Following treatment, the time for reading word and telling color for patients in the AEDs group was prolonged in Stroop color interference test(t =–2.304,–2.454,P < 0.05), and time for reading word and memory of past numbers for patients in the topiramate group was significantly prolonged (t =–3.054, 2.272,P < 0.01,0.05). ③There were no significant differences in scores of WIS before and after treatment in AEDs group and TPM group (P > 0.05). Following treatment, verbal IQ, word scores, total IQ, digit span of patients in the TPM group were significantly lower than those in the AEDs group(t =2.052–3.297,P < 0.05–0.01).There were no significant differences in Stroop color word interference, memory of past numbers and telling the names of fruits and vegetables within 1 minute before and after treatment in AEDsgroup and TPM group (P > 0.05).
