| 2 | Isolation and characterisation of human gingival margin-derived STRO-1/MACS^+ and MACS^-cell populations显示文摘Recently,gingival margin-derived stem/progenitor cells isolated via STRO-1/magnetic activated cell sorting(MACS) showed remarkable periodontal regenerative potential in vivo.As a second-stage investigation,the present study's aim was to perform in vitro characterisation and comparison of the stem/progenitor cell characteristics of sorted STRO-1-positive(MACS^+) and STRO-1-negative(MACS^-) cell populations from the human free gingival margin.Cells were isolated from the free gingiva using a minimally invasive technique and were magnetically sorted using anti-STRO-1 antibodies.Subsequently,the MACS^+ and MACS^- cell fractions were characterized by flow cytometry for expression of CD14,CD34,CD45,CD73,CD90,CD105,CD146/MUC18 and STRO-1.Colony-forming unit(CFU) and multilineage differentiation potential were assayed for both cell fractions.Mineralisation marker expression was examined using real-time polymerase chain reaction(PCR).MACS^+ and MACS- cell fractions showed plastic adherence.MACS^+ cells,in contrast to MACS- cells,showed all of the predefined mesenchymal stem/progenitor cell characteristics and a significantly higher number of CFUs(P<0.01).More than 95%of MACS^+ cells expressed CD105,CD90 and CD73;lacked the haematopoietic markers CD45,CD34 and CD14,and expressed STRO-1 and CD146/MUC18.MACS- cells showed a different surface marker expression profile,with almost no expression of CD14 or STRO-1,and more than 95%of these cells expressed CD73,CD90 and CD146/MUC18,as well as the haematopoietic markers CD34 and CD45 and CD105.MACS^+ cells could be differentiated along osteoblastic,adipocytic and chondroblastic lineages.In contrast,MACS- cells demonstrated slight osteogenic potential.Unstimulated MACS^+ cells showed significantly higher expression of collagen I(P<0.05) and collagen III(P<0.01),whereas MACS^- cells demonstrated higher expression of osteonectin(P<0.05;MannWhitney).The present study is the first to compare gingival MACS^+ and MACS- cell populations demonstrating that MACS^+ cells,in contrast to MACS- cells,harbour stem/progenitor cell characteristics.This study also validates the effectiveness of the STRO-l/MACS^+technique for the isolation of gingival stem/progenitor cells.Human free gingival margin-derived STRO-1/MACS^+ cells are a unique renewable source of multipotent stem/progenitor cells. | Karim M Fawzy El-Sayed Sebastian Paris Christian Graetz Neemat Kassem Mohamed Mekhemar Hendrick Ungefroren Fred Fndrich Christof Drfer | 2015 | International Journal of Oral Science2015,7,2: | 6 |
| 3 | Clinical significance of serum expression of GROβ in esophageal squamous cell carcinoma显示文摘AIM:To determine the association between serum levels of growth-related gene product β(GROβ) and clinical parameters in esophageal squamous cell carcinoma(ESCC).METHODS:Using enzyme-linked immunosorbent assay,serum GROβ levels were measured in ESCC patients(n = 72) and healthy volunteers(n = 83).The association between serum levels of GROβ and clinical parameters of ESCC was analyzed statistically.RESULTS:The serum GROβ levels were much higher in ESCC patients than in healthy controls(median:645 ng/L vs 269 ng/L,P < 0.05).Serum GROβ levels were correlated positively with tumor size,lymph node metastasis,and tumor-node-metastasis(TNM) staging,but not with gender or the histological grade of tumors in ESCC patients.The sensitivity and specificity of the assay for serum GROβ were 73.61% and 56.63%,respectively.CONCLUSION:GROβ may function as an oncogene product and contribute to tumorigenesis and metastasis of ESCC. | Qiao-Mei Dong Jin-Qiang Zhang Qian Li Jacqueline C Bracher Denver T Hendricks Xiao-Hang Zhao | 2011 | World Journal of Gastroenterology2011,17,21: | 5 |

| 5 | RECENT ADVANCES IN RESEARCH AND FORECASTING OF TROPICAL CYCLONE TRACK, INTENSITY, AND STRUCTURE AT LANDFALL显示文摘This review prepared for the fourth International Workshop on Tropical Cyclone Landfall Processes(IWTCLP-4) summarizes the most recent(2015-2017) theoretical and practical knowledge in the field of tropical cyclone(TC) track, intensity, and structure rapid changes at or near landfall. Although the focus of IWTCLPIV was on landfall, this summary necessarily embraces the characteristics of storms during their course over the ocean prior to and leading up to landfall. In the past few years, extremely valuable observational datasets have been collected for TC forecasting guidance and research studies using both aircraft reconnaissance and new geostationary or low-earth orbiting satellites at high temporal and spatial resolution. Track deflections for systems near complex topography such as that of Taiwan and La Réunion have been further investigated, and advanced numerical models with high spatial resolution necessary to predict the interaction of the TC circulation with steep island topography have been developed. An analog technique has been designed to meet the need for longer range landfall intensity forecast guidance that will provide more time for emergency preparedness. Probabilistic track and intensity forecasts have also been developed to better communicate on forecast uncertainty. Operational practices of several TC forecast centers are described herein and some challenges regarding forecasts and warnings for TCs making landfall are identified. | 6 | The Spatial and Temporal Variability of Tropospheric NO_(2) during 2005–14 over China Observed by the OMI显示文摘As improved and accumulated satellite records become available,it is significant to provide up-to-date perspectives on the spatiotemporal signatures of tropospheric nitrogen dioxide(NO2)over China,the knowledge of which is helpful for air pollution control.In this study,the Ozone Monitoring Instrument NO2 dataset for the last 10 years(2005–14)was retrieved to examine multiple aspects of NO2 columns,including distributions,trends,and seasonal cycle.The pattern of average NO2suggests five hotspots with column density higher than 20×1015 molec cm-2:Jing-Jin-Tang;combined southern Hebei and northern Henan;Jinan;the Yangtze River Delta;and the Pearl River Delta.Furthermore,substantial and widespread NO2 growths are distributed over the North China Plain.By contrast,downward trends in NO2 amounts prevail in the megacities of Beijing,Shanghai,and Guangzhou,despite generally high loading levels.Except for the Pearl River Delta,there appears to be temporally consistent behaviors across all regions considered,where NO2 had an abrupt decline during 2008 to 2009,then a drastic increase up to 2013,before beginning to reduce again after 2013.However,the NO2 over the Pearl River Delta is not coevolving with the rest,having experienced a moderate rise from 2005 to 2007,followed by a reduction thereafter.A marked seasonality is apparent,with a maximum in winter and a minimum in summer,regardless of the region.The annual amplitude of NO2 is less pronounced over the Pearl River Delta,whereas the largest range is observed over the combined Southern Hebei and Northern Henan region,induced by enhanced NO2emission in wintertime due to intense domestic heating. | WANG Ting WANG Pu-Cai Francois HENDRICK YU Huan Michel VAN ROOZENDAEL | 2015 | Atmospheric and Oceanic Science Letters2015,8,6: | 2 |
