1Helium Isotope Geochemistry of Ultrahigh-Pressure Metamorphic Eclogites From the Dabie-Sulu Terrane in East China显示文摘The 3He/4He ratios of most eclogites from the Dabie-Sulu terrane range from 0.056 to 0.67 Ra; the data points fall into the mixing part of the crust and the mantle in the 3He-4He diagram. The 3He/4He ratios of eclogites are obviously correlated with the types of their surrounding rocks. The helium isotope composition of the eclogites from the Bixiling complex possesses characters of mantle-derived rocks with the 3He/4He ratio being 5.6 Ra. The 4He concentration of the eclogites exhibits visible inverse correlation with the δ18O value of the quartz in the eclogites from the Sulu area. The δ18O values of the eclogites change synchronously with those of the country rocks. Those results suggest that protoliths of the eclogites were basic-ultrabasic rock bodies or veins intruding into the continental crust in the early stage; strong exchange and hybridization between the basic-ultrabasic rocks and continental rocks and the atmospheric water during the intrusion led to abrupt increase of the 3He/4He ratios,LI Yanhe LI Jincheng SONG Hebin LIU Xiaochun 2000Acta Geologica Sinica(English Edition)2000,74,1:12
2Plasmacytoid dendritic cell deficiency in neonates enhances allergic airway inflammation via reduced production of IFN-α显示文摘Allergic asthma,a chronic inflammatory airway disease associated with type 2 cytokines,often originates in early life.Immune responses at an early age exhibit a Th2 cell bias,but the precise mechanisms remain elusive.Plasmacytoid dendritic cells(pDCs),which play a regulatory role in allergic asthma,were shown to be deficient in neonatal mice.We report here that this pDC deficiency renders neonatal mice more susceptible to severe allergic airway inflammation than adult mice in an OVA-induced experimental asthma model.Adoptive transfer of pDCs or administration of IFN-αto neonatal mice prevented the development of allergic inflammation in wild type but not in IFNAR1−/−mice.Similarly,adult mice developed more severe allergic inflammation when pDCs were depleted.The protective effects of pDCs were mediated by the pDC-/IFN-α-mediated negative regulation of the secretion of epithelial cell-derived CCL20,GM-CSF,and IL-33,which in turn impaired the recruitment of cDC2 and ILC2 cells to the airway.In asthmatic patients,the percentage of pDCs and the level of IFN-αwere lower in children than in adults.These results indicate that impairment of pDC-epithelial cell crosstalk in neonates is a susceptibility factor for the development of allergeninduced allergic airway inflammation.Min Wu Liuchuang Gao Miao He Hangyu Liu Han Jiang Ketai Shi Runshi Shang Bing Liu Shan Gao Hebin Chen Feili Gong Erwin WGelfand Yafei Huang Junyan Han 2020Cellular & Molecular Immunology2020,17,5:3

4A continuous simulation of Holocene effective moisture change represented by variability of virtual lake level in East and Central Asia显示文摘The fluctuation of a single lake level is a comprehensive reflection of water balance within the basin, while the regional consistent fluctuations of lake level can indicate the change of regional effective moisture. Previous researches were mainly focused on reconstructing effective moisture by multiproxy analyses of lake sediments. We carried out a series of experiments, including a transient climate evolution model, a lake energy balance model and a lake water balance model to simulate continuous Holocene effective moisture change represented by variability of virtual lake level in East and Central Asia.The virtual lake level, area, water depth and salinity are not equivalent to actual values, but we estimated relative changes of the regional effective moisture. We also explored the driving mechanisms of effective moisture change in different geographical regions. Our results indicated that gradually falling effective moisture during the Holocene in northern China was due to the combined effects of high lake evaporation caused by longwave and shortwave radiation, and low precipitation caused by reductions of summer solar insolation. A decline in effective moisture through the Holocene in the Tibetan Plateau and southern Central Asia resulted from decreased precipitation because of the weakening of the Asian summer monsoon. 6A 3D bio-printed spheroids based perfusion in vitro liver on chip for drug toxicity assays显示文摘Liver is the foremost organ of human being for drug metabolism,and it played a significant role in toxicity evaluation of drugs.Establishing a liver model in vitro can accelerate the process of the drug screening and new drug research and development.We provide a 3D printing based hepatic sinusoid-on-a-chip microdevice that reconstitutes organ-level liver functions to create a drug screening model of toxicity evaluation on chip.The microfluidic device,which recapitulates the hepatic sinusoid microenvironment,consists of PET polyporous membranes which mimic the perisinusoidal space,and experience fluid flow to mimic the hepatic arterial capillaries.The PET membrane was used to separate the hepatocyte and endotheliocyte.The endotheliocyte was cultured on the downside of the membrane and the hepatocyte were 3D seeded on the membrane via the 3D printer.This device was used to reproduce the in vitro liver model for drug toxicity assays.The expression of several biomarkers of liver was compared with the monoculture and 2D cultured conditions,and the results reveal that this organ-on-a-chip microdevice mimics the drug hepatoxicity that has not been possible by 2D cell-based and animal models,providing a useful platform for screening the drugs and developing an effective therapy in hepatopathy.Tian Tian Yuhan Ho Chen Chen Hebin Sun Janan Hui Panhui Yang Yuqing Ge Ting Liu Jian Yang Hongju Mao 2022Chinese Chemical Letters2022,33,6:1
7Cloning and characterization of a thermostable carboxylesterase from inshore hot spring thermophile Geobacillus sp.ZH1显示文摘The gene(741 bp) encoding carboxylesterase from the thermophilic bacterium Geobacillus sp.ZHl was cloned and overexpressed in Escherichia coli.The purified recombinant protein presented a molecular mass of about 40 kDa by SDS-PAGE analysis.Enzyme assays using p-nitrophenyl esters with different acyl chain lengths as the substrates confirmed its esterase activity,yielding highest specific activity with p-nitrophenyl acetate.Among the p-nitrophenyl esters tested,the carboxylesterase presented preference for p-nitrophenyl caprylate,but hydrolyzed p-nitrophenyl butyrate more efficiently.When p-nitrophenyl butyrate was used as a substrate,the recombinant carboxylesterase exhibited highest activity at pH 8.0 and 60℃.Almost no decrease in esterase activity was observed at 60℃for 3 h,and over 40% of activity was still maintained after incubation at 90℃for 3 h.These results indicate that Geobacillus sp.ZH1 recombinant esterase was thermostable.The enzymatic activity was inhibited by the addition of phenylmethylsulfonyl fluoride,indicating that it contains serine residue,which plays a key role in the catalytic mechanism.Except SDS and xylene,this esterase showed stability toward other tested detergents and organic solvents.Cloning,expression,and biochemical characterization of Geobacillus sp.ZH1 carboxylesterase lay a good foundation for its structural characterization and industrial application.ZHU Yanbing LIU Guangming LI Hebin LIU Jingwen BAI Xiaoming CAI Huinong 2012Acta Oceanologica Sinica2012,31,6:1

9Characterization of a thermostable manganese-containing superoxide dismutase from inshore hot spring thermophile Thermus sp.JM1显示文摘A thermostable superoxide dismutase (SOD) from the inshore thermophile Thermus sp. JM1 was purified to homogeneity by steps of fractional ammonium sulfate precipitation, DEAE-Sepharose chromatography and Phenyl-Sepharose chromatography. The specific activity of the purified native enzyme was 1 656 U/mg. A sod gene from this strain was cloned and overexpressed in Escherichia coli (E. coli). The prepared apo-enzyme of the purified recombinant SOD (rSOD) was reconstituted with either Fe or Mn by means of incubation with appropriate metal salts. As a result, only Mn 2+ - reconstituted rSOD (Mn-rSOD) exhibited the specific activity of 1 598 U/mg. SOD from Thermus sp. JM1 was Mn-SOD, judging by the specific activities analysis of Fe or Mn reconstituted rSODs and the insensitivity of the native SOD to both cyanide and H 2 O 2 . Both the native SOD and Mn- rSOD were determined to be homotetramers with monomeric molecular mass of 26 kDa and 27.5 kDa, respectively. 10Purification and biochemical characterization of manganesecontaining superoxide dismutase from deep-sea thermophile Geobacillus sp. EPT3显示文摘Thermostable SOD is a promising enzyme in biotechnological applications. In the present study, thermophile Geobacillus sp. EPT3 was isolated from a deep-sea hydrothermal field in the East Pacific. A thermostable superoxide dismutase(SOD) from this strain was purified to homogeneity by steps of fractional ammonium sulfate precipitation, DEAE-Sepharose chromatography, and Phenyl-Sepharose chromatography. SOD was purified 13.4 fold to homogeneity with a specific activity of 3 354 U/mg and 11.1% recovery. SOD from Geobacillus sp. EPT3 was of the Mn-SOD type, judged by the insensitivity of the enzyme to both KCN and H2O2. SOD was determined to be a homodimer with monomeric molecular mass of 26.0 k Da. It had high thermostability at 50°C and 60°C. At tested conditions, SOD was relatively stable in the presence of some inhibitors and denaturants, such as β-mercaptoethanol(β-ME), dithiothreitol(DTT), phenylmethylsulfonyl fluoride(PMSF), urea, and guanidine hydrochloride. Geobacillus sp. EPT3 SOD showed striking stability across a wide p H range from 5.0 to 11.0. 11ADAP restraint of STAT1 signaling regulates macrophage phagocytosis in immune thrombocytopenia显示文摘Heightened platelet phagocytosis by macrophages accompanied by an increase in IFN-γplay key roles in the etiology of immune thrombocytopenia(ITP);however,it remains elusive how macrophage-mediated platelet clearance is regulated in ITP.Here,we report that adhesion and degranulation-protein adaptor protein(ADAP)restrains platelet phagocytosis by macrophages in ITP via modulation of signal transducer and activator of transcription 1(STAT1)-FcγR signaling.We show that ITP was associated with the underexpression of ADAP in splenic macrophages.Furthermore,macrophages from Adap^(−/−)mice exhibited elevated platelet phagocytosis and upregulated proinflammatory signaling,and thrombocytopenia in Adap^(−/−)mice was mitigated by the depletion of macrophages.Mechanistically,ADAP interacted and competed with STAT1 binding to importinα5.ADAP deficiency potentiated STAT1 nuclear entry,leading to a selective enhancement of FcγRI/IV transcription in macrophages.Moreover,pharmacological inhibition of STAT1 or disruption of the STAT1-importinα5 interaction relieved thrombocytopenia in Adap^(−/−)mice.Thus,our findings not only reveal a critical role for ADAP as an intracellular immune checkpoint for shaping macrophage phagocytosis in ITP but also identify the ADAP-STAT1-importinα5 module as a promising therapeutic target in the treatment of ITP.Yiwei Xiong Yanli Li Xinxing Cui Lifeng Zhang Xiaodong Yang Hebin Liu 2022Cellular & Molecular Immunology2022,19,8:0
12Substitution of His260 residue alters the thermostability of Pseudoalteromonas carrageenovora arylsulfatase显示文摘This study aimed to improve the thermostability of arylsulfatase from Pseudoalteromonas carrageenovora. A library of P. carrageenovora arylsulfatase mutants was constructed by introducing random mutagenesis using error-prone PCR. After screening, two mutants of H260L and D84A/H260L showed enhanced thermal stability than the wild-type predecessor (WT). Site-directed mutagenesis demonstrated that only amino acid residue at Position 260 plays an important role in the thermostability of P. carrageenovora arylsulfatase. Thermal inactivation analysis showed that the half-life (t1/2) values at 55°C for H260L, H260I, H260Q, H260F and H260R were 40.6, 48.4, 30.9, 29.1 and 34.5 min, respectively, while that of WT was 9.1 min.
