1Helium Isotope Geochemistry of Ultrahigh-Pressure Metamorphic Eclogites From the Dabie-Sulu Terrane in East China显示文摘The 3He/4He ratios of most eclogites from the Dabie-Sulu terrane range from 0.056 to 0.67 Ra; the data points fall into the mixing part of the crust and the mantle in the 3He-4He diagram. The 3He/4He ratios of eclogites are obviously correlated with the types of their surrounding rocks. The helium isotope composition of the eclogites from the Bixiling complex possesses characters of mantle-derived rocks with the 3He/4He ratio being 5.6 Ra. The 4He concentration of the eclogites exhibits visible inverse correlation with the δ18O value of the quartz in the eclogites from the Sulu area. The δ18O values of the eclogites change synchronously with those of the country rocks. Those results suggest that protoliths of the eclogites were basic-ultrabasic rock bodies or veins intruding into the continental crust in the early stage; strong exchange and hybridization between the basic-ultrabasic rocks and continental rocks and the atmospheric water during the intrusion led to abrupt increase of the 3He/4He ratios,LI Yanhe LI Jincheng SONG Hebin LIU Xiaochun 2000Acta Geologica Sinica(English Edition)2000,74,1:12
2Characteristics of authigenic pyrite and its sulfur isotopes influenced by methane seep at Core A, Site 79 of the middle Okinawa Trough显示文摘The anaerobic oxidation of methane(AOM) has strongly developed at Core A, Site 79 of the middle Okinawa Trough, East China Sea, and a large amount of authigenic pyrite is preserved in the surface sediment. In this study, we analyze the characteristics of the authigenic pyrite and its sulfur isotopic values. The authigenic pyrite is stripy and tubular, and there were foraminifera compartments filled with pyrite. The pyrite is extracted using chromium reduction, and the values of ? 34 S are found to lie between ?41.20‰ and 8.92‰ V-CDT. The bulk pyrite tends to be more enriched in 34 S with increasing depth. Particularly, the ? 34 S value of the pyrite lies between ?32.73‰ and ?41.20‰ V-CDT above 278 cmbsf, but it quickly increases below this depth(?21.49‰–8.92‰ V-CDT). At the same time, the total sulfur content of the pyrite shows an abrupt increase above 100 cmbsf but is otherwise stable between 1.04% and 0.55% below 100 cmbsf. The stable and negative values of ? 34 S and the decreasing values of total sulfur above 278 cmbsf indicate reduced AOM activities in 17.18–5.3 ka. In addition, the increasing ? 34 S and pyrite content indicate strong AOM development and methane seep below 278 cmbsf in 18.8–17.18 ka. In particular, the highest positive value of ? 34 S occurring in 18.78 ka indicates the most intense AOM activity. 3Geochemical constraints on the methane seep activity in western slope of the middle Okinawa Trough, the East China Sea显示文摘Anaerobic oxidation of methane and sulfate reduction was studied in the pore waters of four cores at two stations of the middle Okinawa Trough. Pore water vertical distributions of sulfate, methane, sulfide, total alkalinity, ammonium, and phosphate were determined in this study. Our results show strong linear sulfate concentration gradients of 6.83 mmol/L m?1 in Core A and 5.96 mmol/L m?1 in Core C, which were collected from two stations. Concurrent variations of methane, total alkalinity and hydrogen sulfide all exhibit steep increases with depth at both cores, which indicate active methane seep activities around two stations. Pore water ammonium and phosphate concentrations reveal minor influences of organic matter degradation on sulfate reduction at two stations. Sulfate methane interface(SMI) was extrapolated from linear sulfate profiles in methane seep cores. 4Extreme Enrichment of Tellurium in Deep-Sea Sediments显示文摘Tellurium is a sort of scattered rare element on the earth. Its concentration is very low in earth’s crust, only 1.0 ng/g. However, it has extremely high abundance in Co-rich crusts, marine polymetallic nodules, deep-sea sediments and aerolites. To find out the origin of tellurium enrichment in deep-sea sediments, we analyzed and compared tellurium concentrations and helium isotope compositions in the magnetic parts and those in the bulk parts of deep-sea sediments. The result indicates that the helium content, 3He/4He ratio and tellurium concentration are obviously higher in the magnetic parts than those in the bulk parts. The 3He abundance varies synchronously with the tellurium concentration. 3He and Te have a distinct positive correlation with each other. It is the first time that the paper brings forward that the extreme enrichment of tellurium in deep-sea sediments, like helium isotope anomalies, probably results from the input of interplanetary dust particles (IDPs). 6Overexpression and characterization of a thermostable β-agarase producing neoagarotetraose from a marine isolate Microbulbifer sp.AG1显示文摘An agarase gene containing 1 302 bp was cloned from Microbulbifer sp. AG1. It encoded a mature protein of 413 amino acids plus a 20-residue signal peptide. The recombinant enzyme without the signal peptide was expressed and purified from Escherichia coli BL21(DE3). When agarose was used as a substrate, the optimal temperature and pH for the enzyme were 60℃ and 7.5, respectively. The recombinant agarase showed excellent thermostability with 67% and 19% of residual activities after incubation at 50℃ and 60℃ for 1 h, respectively.Except SDS, the recombinant agarase had a relatively good resistance against the detected inhibitors, detergents and urea denaturant. Thin layer chromatography analysis and enzyme assay using p-nitrophenyl-α/β-Dgalactopyranoside revealed that the recombinant agarase was a β-agarase that degraded agarose into neoagarotetraose as the main end product. 8A continuous simulation of Holocene effective moisture change represented by variability of virtual lake level in East and Central Asia显示文摘The fluctuation of a single lake level is a comprehensive reflection of water balance within the basin, while the regional consistent fluctuations of lake level can indicate the change of regional effective moisture. Previous researches were mainly focused on reconstructing effective moisture by multiproxy analyses of lake sediments. We carried out a series of experiments, including a transient climate evolution model, a lake energy balance model and a lake water balance model to simulate continuous Holocene effective moisture change represented by variability of virtual lake level in East and Central Asia.The virtual lake level, area, water depth and salinity are not equivalent to actual values, but we estimated relative changes of the regional effective moisture. We also explored the driving mechanisms of effective moisture change in different geographical regions. Our results indicated that gradually falling effective moisture during the Holocene in northern China was due to the combined effects of high lake evaporation caused by longwave and shortwave radiation, and low precipitation caused by reductions of summer solar insolation. A decline in effective moisture through the Holocene in the Tibetan Plateau and southern Central Asia resulted from decreased precipitation because of the weakening of the Asian summer monsoon. 11Cloning and characterization of a thermostable carboxylesterase from inshore hot spring thermophile Geobacillus sp.ZH1显示文摘The gene(741 bp) encoding carboxylesterase from the thermophilic bacterium Geobacillus sp.ZHl was cloned and overexpressed in Escherichia coli.The purified recombinant protein presented a molecular mass of about 40 kDa by SDS-PAGE analysis.Enzyme assays using p-nitrophenyl esters with different acyl chain lengths as the substrates confirmed its esterase activity,yielding highest specific activity with p-nitrophenyl acetate.Among the p-nitrophenyl esters tested,the carboxylesterase presented preference for p-nitrophenyl caprylate,but hydrolyzed p-nitrophenyl butyrate more efficiently.When p-nitrophenyl butyrate was used as a substrate,the recombinant carboxylesterase exhibited highest activity at pH 8.0 and 60℃.Almost no decrease in esterase activity was observed at 60℃for 3 h,and over 40% of activity was still maintained after incubation at 90℃for 3 h.These results indicate that Geobacillus sp.ZH1 recombinant esterase was thermostable.The enzymatic activity was inhibited by the addition of phenylmethylsulfonyl fluoride,indicating that it contains serine residue,which plays a key role in the catalytic mechanism.Except SDS and xylene,this esterase showed stability toward other tested detergents and organic solvents.Cloning,expression,and biochemical characterization of Geobacillus sp.ZH1 carboxylesterase lay a good foundation for its structural characterization and industrial application.ZHU Yanbing LIU Guangming LI Hebin LIU Jingwen BAI Xiaoming CAI Huinong 2012Acta Oceanologica Sinica2012,31,6:1
12Characterization of a thermostable manganese-containing superoxide dismutase from inshore hot spring thermophile Thermus sp.JM1显示文摘A thermostable superoxide dismutase (SOD) from the inshore thermophile Thermus sp. JM1 was purified to homogeneity by steps of fractional ammonium sulfate precipitation, DEAE-Sepharose chromatography and Phenyl-Sepharose chromatography. The specific activity of the purified native enzyme was 1 656 U/mg. A sod gene from this strain was cloned and overexpressed in Escherichia coli (E. coli). The prepared apo-enzyme of the purified recombinant SOD (rSOD) was reconstituted with either Fe or Mn by means of incubation with appropriate metal salts. As a result, only Mn 2+ - reconstituted rSOD (Mn-rSOD) exhibited the specific activity of 1 598 U/mg. SOD from Thermus sp. JM1 was Mn-SOD, judging by the specific activities analysis of Fe or Mn reconstituted rSODs and the insensitivity of the native SOD to both cyanide and H 2 O 2 . Both the native SOD and Mn- rSOD were determined to be homotetramers with monomeric molecular mass of 26 kDa and 27.5 kDa, respectively. 13Purification and biochemical characterization of manganesecontaining superoxide dismutase from deep-sea thermophile Geobacillus sp. EPT3显示文摘Thermostable SOD is a promising enzyme in biotechnological applications. In the present study, thermophile Geobacillus sp. EPT3 was isolated from a deep-sea hydrothermal field in the East Pacific. A thermostable superoxide dismutase(SOD) from this strain was purified to homogeneity by steps of fractional ammonium sulfate precipitation, DEAE-Sepharose chromatography, and Phenyl-Sepharose chromatography. SOD was purified 13.4 fold to homogeneity with a specific activity of 3 354 U/mg and 11.1% recovery. SOD from Geobacillus sp. EPT3 was of the Mn-SOD type, judged by the insensitivity of the enzyme to both KCN and H2O2. SOD was determined to be a homodimer with monomeric molecular mass of 26.0 k Da. It had high thermostability at 50°C and 60°C. At tested conditions, SOD was relatively stable in the presence of some inhibitors and denaturants, such as β-mercaptoethanol(β-ME), dithiothreitol(DTT), phenylmethylsulfonyl fluoride(PMSF), urea, and guanidine hydrochloride. Geobacillus sp. EPT3 SOD showed striking stability across a wide p H range from 5.0 to 11.0. 14ADAP restraint of STAT1 signaling regulates macrophage phagocytosis in immune thrombocytopenia显示文摘Heightened platelet phagocytosis by macrophages accompanied by an increase in IFN-γplay key roles in the etiology of immune thrombocytopenia(ITP);however,it remains elusive how macrophage-mediated platelet clearance is regulated in ITP.Here,we report that adhesion and degranulation-protein adaptor protein(ADAP)restrains platelet phagocytosis by macrophages in ITP via modulation of signal transducer and activator of transcription 1(STAT1)-FcγR signaling.We show that ITP was associated with the underexpression of ADAP in splenic macrophages.Furthermore,macrophages from Adap^(−/−)mice exhibited elevated platelet phagocytosis and upregulated proinflammatory signaling,and thrombocytopenia in Adap^(−/−)mice was mitigated by the depletion of macrophages.Mechanistically,ADAP interacted and competed with STAT1 binding to importinα5.ADAP deficiency potentiated STAT1 nuclear entry,leading to a selective enhancement of FcγRI/IV transcription in macrophages.Moreover,pharmacological inhibition of STAT1 or disruption of the STAT1-importinα5 interaction relieved thrombocytopenia in Adap^(−/−)mice.Thus,our findings not only reveal a critical role for ADAP as an intracellular immune checkpoint for shaping macrophage phagocytosis in ITP but also identify the ADAP-STAT1-importinα5 module as a promising therapeutic target in the treatment of ITP.Yiwei Xiong Yanli Li Xinxing Cui Lifeng Zhang Xiaodong Yang Hebin Liu 2022Cellular & Molecular Immunology2022,19,8:0

17Substitution of His260 residue alters the thermostability of Pseudoalteromonas carrageenovora arylsulfatase显示文摘This study aimed to improve the thermostability of arylsulfatase from Pseudoalteromonas carrageenovora. A library of P. carrageenovora arylsulfatase mutants was constructed by introducing random mutagenesis using error-prone PCR. After screening, two mutants of H260L and D84A/H260L showed enhanced thermal stability than the wild-type predecessor (WT). Site-directed mutagenesis demonstrated that only amino acid residue at Position 260 plays an important role in the thermostability of P. carrageenovora arylsulfatase. Thermal inactivation analysis showed that the half-life (t1/2) values at 55°C for H260L, H260I, H260Q, H260F and H260R were 40.6, 48.4, 30.9, 29.1 and 34.5 min, respectively, while that of WT was 9.1 min. 18Laparoscopic duodenum-preserving pancreatic head resection:a narrative review显示文摘Pancreatic surgery is one of the most complex and challenging fields in abdominal surgery associated with extensive surgical trauma,damage to adjacent organs,a long operation time and a high incidence of postoperative complications.Since the early 1990s,laparoscopic techniques have been applied to a growing number of pancreas surgeries,and great progress has been achieved in laparoscopic pancreaticoduodenectomy.As surgeons become proficient in laparoscopic pancreaticoduodenectomy techniques,laparoscopic techniques are gradually used in other pancreatic surgeries,such as laparoscopic distal pancreatectomies and laparoscopic duodenum-preserving pancreatic head resection(LDPPHR),which may benefit patients by reducing postoperative pain and hospital stays and providing a quick recovery to normal activity.Recently,a great number of literature have introduced LDPPHR.It is a good surgical method for benign and low-grade malignant tumors of the pancreatic head.Although LDPPHR is technically feasible,it is not yet generally practicable and limited to highly skilled endoscopic surgeons,and the long-term results after LDPPHR are still not well defined.This article aims to provide a literature review of LDPPHR to assess its feasibility,safety,postoperative recovery,and future outlook according to early experiences of this technique.Nuerabula Wujimaimaiti Yi Wu Jingxiong Yuan Jikuan Jin Hebin Wang Shizhen Li Hang Zhang Min Wang Renyi Qin 2021Journal of Pancreatology2021,4,4:0
