| 2 | Engineering a New Chloroplastic Photorespiratory Bypass to In crease Photosynthetic Efficiency and Productivity in Rice显示文摘Over the past few years, three photorespiratory bypasses have been introduced into plants, two of which led to observable in creases in photos yn thesis and biomass yield. However, most of the experiments were carried out using Arabidopsis under controlled environmental conditions, and the increases were only observed under low-light and short-day conditions. In this study, we designed a new photorespiratory bypass (called GOC bypass), characterized by no reducing equivalents being produced during a complete oxidation of glycolate into CO2 catalyzed by three rice-self-originating enzymes, i.e., glycolate oxidase, oxalate oxidase, and catalase. We successfully established this bypass in rice chloroplasts using a multi-gene assembly and transformation system. Transgenic rice plants carrying GOC bypass (GOC plants) showed significant increases in photosynthesis efficiency, biomass yield, and nitrogen content, as well as several other CO2-enriched phe no types under both greenhouse and field conditions .Grain yield of GOC plants varied depending on seeding season and was increased significantly in the spring. We further demonstrated that GOC plants had significant advantages under high-light conditions and that the improvements in GOC plants resulted primarily from a photosynthetic CO2-concentrating effect rather than from improved energy balance. | 3 | Clinical and experimental study on regional administration of phosphorus32 glassmicrospheres in treating hepatic carcinoma显示文摘AIM To study the therapeutical effectiveness, dosage range and toxic adverse effects of domestic phosphorus 32 glass microsphere and evaluate its clinical significance. METHODS Ⅰ. Fifty two BALB/*!c tumor bearing male nude mice were allocated into treatment group( n =38) and control group( n =14). In the former group different doses of 32 P GMS were injected into the tumor mass, while in the latter 31 P GMS or no treatment was given. The experimental animals were sacrificed in batches, and then the tumors and their nearby tissues were examined by light and electron microscopy. Ⅱ. Through selective catheterization of hepatic artery, 32 P GMS was infused to 5 healthy domestic pigs in a dosage equivalent to the therapeutic dose for human being, and 31 P GMS was infused to another 5 healthy domestic pigs. Two pigs infused with contrast medium served as whole course blank controls. One pig from each group was surrendered to euthanasia at week 1, 4, 8 and 16 respectively. The ultrastructural histopath ological changes in liver tissues taken from different sites were evaluated semiquan titatively. Ⅲ. One hundred and twenty seven times of 32 P GMS intrahepatic artery interventional therapies were performed on 93 patients with hepatic carcinoma, including 79 cases of primary hepatic carcinoma and 14 cases of secondary hepatic carcinoma. 32 P GMS ( n =30), and group B, 32 P GMS and half dose of trans hepatic artery embolization (TAE) ( n =49) , and 18 patients with HCC by TAE only as control group C. Fourteen patients with secondary hepatic carcinoma were treated in the same way as group B or C. RESULTS Ⅰ. Comparing with the control group, the treatment group of tumor bearing nude mice attained the tumor inhibition rates of 59 7%-93 7% ( F =579 62, P <0 01) at 14*!d . At an absorbed dose of 7320Gy, the tumor cells were completely destroyed. When the absorbed doses ranged from 1830Gy to 3660Gy, most of the tumor cells showed the evidences of injury or necrosis, but there appeared some well differentiated tumor cells and enhanced effect of the autoimmunocytes. At an absorbed dose of 366Gy or less, some tumor cells still remained active proliferative ability. The definite anticancer effect appeared as early as 3d after intratumoral injection of 32 P GMS. Ⅱ. The cumulative amount of 32 P GMS in the target tissue after trans hepatic artery instillation attained more than 90% of the total dose administrated. Semiquantitative analysis of ultrastructral morphology in the experimental group showed no statistical difference between the nuclear abnormality (N abn ) and mitochondrial variability (M var ) at week 1 or 2, but revealed prominent difference (χ 2=6 70-9 68, P <0 01 , χ 2=65 09-115 09, P <0 001 ) as compared with those in the other groups. In the experimental group the N abn in tissues showed no significant difference between week 8 and week 16. No apparent changes were found in the stomach, spleen, kidney and lung tissues of the experimental pigs. Ⅲ. The therapeutical results of HCC patients in group A were closely approximated to those of group C, no hematological toxic side effects were noted, and the systemic reaction was mild. In some patients 2*!mos - 3*!mos after treatment some secondary foci appeared around the periphery of the primary lesion. In general better effectiveness was obtained in patients with small lesion. After analyzing by RIDIT method, the therapeutic result in group B was significantly better than that in group C, and secondary foci around the original lesion were rarely seen at 3*!mos after treatment. In group C the collateral circulation was reestablished along the periphery of primary foci and the secondary foci appeared more frequently, and required to undergo several courses of treatment. And the phase difference correction algorithm based on the discrete spectrum and the HSCW is given. The HSCW has a low peak side lobe level, a high side lobe roll-off rate, and a simple spectrum representation. Hence, leakage errors and harmonic interferences can be considerably reduced by weighting samples with the HSCW, the parameter estimation by the HSCW-based phase difference correction algorithm is free of solving high order equations, and the overall method can be easily implemented in embedded systems. | 11 | Effects of Chinese herbal medicine Yiqi Huaju Formula on hypertensive patients with metabolic syndrome:a randomized,placebo-controlled trial显示文摘BACKGROUND:Patients with hypertension coupled with metabolic syndrome(MetS) are among the high risk population in cardiovascular and cerebrovascular diseases.To reduce the prevalence of cardiovascular and cerebrovascular diseases,it is essential to appropriately control blood pressure together with other cardiovascular risk factors. OBJECTIVE:The current study was designed to investigate the therapeutic effects on blood pressure,blood pressure variability and other cardiovascular risk factors by giving Yiqi Huaju Formula,a compound traditional Chinese herbal medicine,in addition to routine treatment to hypertensive patients coupled with MetS. DESIGN,SETTING,PARTICIPANTS AND INTERVENTIONS:A total of 43 patients with hypertension coupled with MetS were recruited into this study.The enrolled patients were randomly divided into the Chinese herbal formula group(anti-hypertensive drugs plus Yiqi Huaju Formula,CHF) and the control group(anti-hypertensive drugs plus placebo).The CHF group enrolled 22 patients while the control group received 21 cases.Treatments were given for 12 weeks in both groups. MAIN OUTCOME MEASURES:Parameters examined include 24-hour ambulatory blood pressure monitoring,body mass index,waist circumference,waist-to-hip ratio,homeostatic model assessment for insulin resistance(HOMA-IR),fasting glycosylated hemoglobin(HbA1c),fasting plasma glucose,2-hour postprandial plasma glucose(PPG),fasting plasma insulin,serum lipid,etc. RESULTS:Compared with the control group,the CHF group had significant improvement (PO.01) in anthropometric parameters,FPG,HOMA-IR,blood pressure amplitude,blood pressure variability and blood pressure load. CONCLUSION:This study showed that integrated traditional Chinese and Western medicine treatment can achieve better results in controlling blood pressure as well as other cardiovascular risk factors.The mechanism of controlling of blood pressure may be associated with the improvement of insulin sensitivity due to the Yiqi Huaju intervention. The causal link between disruptive PTEN/Akt signaling and chondrocyte senescence and the underlying mechanism are unclear. In this study, we found activated Akt signaling in human OA cartilage as well as in a mouse OA model with surgical destabilization of the medial meniscus.Genetic mouse models mimicking sustained Akt signaling in articular chondrocytes via PTEN deficiency driven by either Col2a1-Cre or Col2a1-Cre^(ERT2) developed OA, whereas restriction of Akt signaling reversed the OA phenotypes in PTEN-deficient mice.Mechanistically, prolonged activation of Akt signaling caused an accumulation of reactive oxygen species and triggered chondrocyte senescence as well as a senescence-associated secretory phenotype, whereas chronic administration of the antioxidant N-acetylcysteine suppressed chondrocyte senescence and mitigated OA progression in PTEN-deficient mice. Therefore,inhibition of Akt signaling by PTEN is required for the maintenance of articular cartilage. | 16 | Effects of Chinese herbal medicine Yiqi Huaju Qingli Formula in metabolic syndrome patients with microalbuminuria:a randomized placebo-controlled trial显示文摘BACKGROUND:Microalbuminuria(MAU) is a key component of metabolic syndrome(MetS) and is an early sign of diabetic nephropathy as well.Although routine Western medicine treatments are given to MetS patients to control high blood pressure,hyperglycemia and dyslipidemia,some patients still experience progressive renal lesions and it is necessary to modify and improve the treatment strategy for MetS patients. OBJECTIVE:To investigate the efficacy of Yiqi Huaju Qingli Herb Formula,a compound traditional Chinese herbal medicine,in MetS patients with MAU when it is combined with routine Western medicine treatment. DESIGN,SETTING,PARTICIPANTS AND INTERVENTIONS:Sixty patients with MetS were randomized into the Chinese herbal formula group(CHF,Yiqi Huaju Qingli formula treatment in combination with Western medicine) and control group(placebo in combination with Western medicine).All treatments were administered for 12 weeks. MAIN OUTCOME MEASURES:Urinary microalbumin(MA),urinary albumin-to-creatinine ratio (UACR),24-hour total urine protein(24-hTP),body mass index(BMI),waist circumference (WC),waist-to-hip ratio(WHR),fasting plasma glucose(FPG),2-hour postprandial plasma glucose(2-hPPG),glycosylated hemoglobin(HbA1c),homeostasis model assessment for insulin resistance(HOMA-IR),blood lipid profile and blood pressure were observed. RESULTS:Compared with the control group,CHF treatment significantly decreased BMI (P<0.05),WC(P<0.01) and WHR(P<0.01).Both groups had significant decreases in FPG, 2-hPPG,HbA1c,HOMA-IR,MA,and UACR,with CHF treatment showing better effects on these parameters compared with the control treatment(P<0.05).Both treatments significantly reduced the levels of total cholesterol,low-density lipoprotein cholesterol and triacylglycerol(TAG),and a greater reduction in TAG was observed with CHF treatment(P<0.05).The level of high-density lipoprotein cholesterol did not change in the control group after treatment(P>0.05),whereas it significantly increased with CHF treatment(P<0.01).Compared with before the treatment,significant decreases in systolic blood pressure,diastolic blood pressure and mean arterial blood pressure were observed in both groups(P<0.01).However,there was no significant difference between the two groups(P>0.05). CONCLUSION:Combined treatment of Yiqi Huaju Qingli Formula and Western medicine significantly alleviated MAU,which may correlate with the improvement of insulin sensitivity and glucose and lipid metabolism. | 17 | Association study of the single nucleotide polymorphisms in adiponectin-associated genes with type 2 diabetes in Han Chinese显示文摘Single-nucleotide polymorphisms (SNPs) of ADIPOQ, ADIPOR1, and ADIPOR2 have been associated with type 2 diabetes mellitus (T2DM), but there are many conflicting results especially in Chinese populations. To investigate the contribution of the adiponectin genes and their receptors to T2DM, a case-control study was performed and 11 SNPs of ADIPOQ, ADIPOR1, and ADIPOR2 were genotyped in 985 T2DM and 1,050 control subjects. rs16861194 (-11426 A>G) in the putative promoter of ADIPOQ was associated with T2DM (P = 0.007; OR = 1.29, 95% CI 1.08-1.55). None of the other 10 SNPs were associated with T2DM in this study, although rs2241766 and rs1501299 were reported to be associated with T2DM in previous Chinese studies. There was also no significant difference found from the ADIPOQ haplotype analysis, which contains rs16861194. In addition, we also assessed potential gene-gene interactions in three genes and no interactions were found. | 18 | Effect of Silicon on Growth, Physiology, and Cadmium Translocation of Tobacco (Nicotiana tabacum L.) in Cadmium-Contaminated Soil显示文摘Silicon(Si) offers beneficial effect on plants under cadmium(Cd) stress such as promoting plant growth and increasing resistance to heavy metal toxicity. In this study, a pot experiment was performed to study the role of Si in alleviating Cd toxicity in tobacco(Nicotiana tabacum L.) plants on a yellow soil taken from Guiyang, China. Nine treatments consisting of three concentrations of Cd(0, 1, and 5 mg kg^(-1)) together with three Si levels(0, 1, and 4 g kg^(-1)) were established. Plant growth parameters, Cd concentration,and the malondialdehyde(MDA), chlorophyll, and carotenoid contents were determined 100 d after transplanting of tobacco seedlings.Application of exogenous Si enhanced the growth of tobacco plants under Cd stress. When 5 mg kg^(-1) Cd was added, Si addition at 1 and 4 g kg^(-1) increased root, stem, and leaf biomass by 26.1%–43.3%, 33.7%–43.8%, and 50.8%–69.9%, respectively, compared to Si addition at 0 g kg^(-1). With Si application, the transfer factor of Cd in tobacco from root to shoot under both 1 and 5 mg kg^(-1) Cd treatments decreased by 21%. The MDA contents in the Si-treated tobacco plants declined by 5.5%–17.1% compared to those in the non-Si-treated plants, indicating a higher Cd tolerance. Silicon application also increased the chlorophyll and carotenoid contents by 33.9%–41% and 25.8%–47.3% compared to the Cd only treatments. | 19 | Protective effect of Saccharomyces boulardii on intestinal mucosal barrier of dextran sodium sulfate-induced colitis in mice显示文摘Background: The effect and mechanism of Saccharomyces boulardii (Sb) in inflammatory bowel disease are unclear. The objective of the study was to evaluate the impact of Sb on intestinal mucosal barrier and intestinal flora in a colitis mouse model. Methods: Forty C57BL/6J male mice were randomly assigned to five groups: normal control group (A), pathologic control group (B), Sb treatment group (C), mesalazine treatment group (D), and Sb combined with mesalazine treatment group (E). Colitis was induced by the addition of 2.5%(wt/vol) dextran sodium sulfate (DSS) in the drinking water ad libitum for 7 days. The general condition, weight change, stool property, and bloody stool level of mice were observed to evaluate the disease activity index. The expression of zona occludens-1 (ZO-1) and occludin in intestinal tissue were measured by immunohistochemistry. The level of tumor necrosis factor-α(TNF-α) and interleukin (IL)-8 in plasma was measured by enzyme linked immunosorbent assay. Inter-cellular tight junctions were observed by transmission electron microscopy. The feces and intestinal contents were collected sterilely, and intestinal flora was analyzed by 16S rRNA sequencing. Results: Compared with group B, Sb reduced the disease activity index and histological score of group C (disease activity index: group B 2.708 ± 0.628, group C 1.542 ± 0.616, PBC = 0.005;histological score: group B 9.875 ± 3.271, group C 4.750 ± 1.832, PBC = 0.005) in DSS-induced colitis in mice. Sb exerted a protect effect on the expression of ZO-1 (group B 2.075 ± 1.176, group C 4.225 ± 1.316, PBC = 0.019) and occludin (group B 2.200 ± 0.968, group C 3.525 ± 1.047, PBC = 0.023). Compared with group B, Sb decreased the level of TNF-α and IL-8 of group C (TNF-α: group B 716.323 ± 44.691 ng/L, group C 521.740 ± 90.121 ng/L, PBC = 0.001;IL-8: group B 128.992 ± 11.475 pg/mL, group C 106.283 ± 15.906 pg/mL, PBC = 0.012). Treatment with Sb preserved the tight junctions and ameliorated microvilli and inter-cellular space. Treatment with Sb also showed its own characteristics: a higher percentage of Bacteroidetes and a lower percentage of Firmicutes, with significant differences or a significant trend. The proportion of the S24-7 family was increased significantly in the Sb treatment group. Conclusions: Sb shows an anti-inflammatory effect and has a protective effect on the intestinal mucosal mechanical barrier. | 20 | Comparison of Artisan iris-claw intraocular lens implantation and posterior chamber intraocular lens sulcus fixation for aphakic eyes显示文摘AIM:To compare the efficacy and complications of Artisan iris-claw intraocular lens(IOL)implantation and posterior chamber IOL sulcus fixation for the treatment of aphakic eyes without capsular support after vitrectomy.METHODS:A prospective study of 45 cases was conducted.Forty-five eyes without sufficient lens capsule support following pars plana vitrectomy(PPV)combined lens extraction were divided into two groups.Group A:25 eyes received Artisan iris-claw IOL implantation.Group B:20 eyes received posterior chamber IOL sulcus fixation.The corrected distance visual acuity(CDVA)and intraocular pressure(IOP),corneal endothelial cell loss rate,surgical time and complications were compared between the two groups.Pigment changes of trabecular meshwork and anterior chamber depths were measured at each time point in Artisan group.RESULTS:The mean surgical time of Artisan group was significantly shorter(P<0.05).No statistically significant difference in endothelial cell loss rate was noted between two groups at any time point(P>0.05).CDVA of Artian group was better than that of the sulcus fixation group 1d after surgery(P<0.05)and there was no statistically significant difference 1 and 3mo after surgery(P>0.05).Mean IOP showed no significant differences between groups before and after surgery.The postoperative complications of Artisan group were anterior uveitis,iris depigmentation,pupillary distortion and spontaneous lens dislocation.The complications of sulcus fixation group include choroidal detachment,intraocular haemorrhage,tilt of IOL optic part and retinal detachment.CONCLUSION:Secondary Artisan IOL implantation canbe performed less invasively and in a shorter surgical time period with earlier visual recovery after surgery compared to transscleral suturing fixation of an IOL.This technique is an effective and safe procedure.It is a promising option for the treatment of aphakic eyes without capsular support after vitrectomy. | He Teng Hong Zhang | 2014 | International Journal of Ophthalmology(English edition)2014,7,2: | 7 |
