| 1 | Effect of Silicon on Growth, Physiology, and Cadmium Translocation of Tobacco (Nicotiana tabacum L.) in Cadmium-Contaminated Soil显示文摘Silicon(Si) offers beneficial effect on plants under cadmium(Cd) stress such as promoting plant growth and increasing resistance to heavy metal toxicity. In this study, a pot experiment was performed to study the role of Si in alleviating Cd toxicity in tobacco(Nicotiana tabacum L.) plants on a yellow soil taken from Guiyang, China. Nine treatments consisting of three concentrations of Cd(0, 1, and 5 mg kg^(-1)) together with three Si levels(0, 1, and 4 g kg^(-1)) were established. Plant growth parameters, Cd concentration,and the malondialdehyde(MDA), chlorophyll, and carotenoid contents were determined 100 d after transplanting of tobacco seedlings.Application of exogenous Si enhanced the growth of tobacco plants under Cd stress. When 5 mg kg^(-1) Cd was added, Si addition at 1 and 4 g kg^(-1) increased root, stem, and leaf biomass by 26.1%–43.3%, 33.7%–43.8%, and 50.8%–69.9%, respectively, compared to Si addition at 0 g kg^(-1). With Si application, the transfer factor of Cd in tobacco from root to shoot under both 1 and 5 mg kg^(-1) Cd treatments decreased by 21%. The MDA contents in the Si-treated tobacco plants declined by 5.5%–17.1% compared to those in the non-Si-treated plants, indicating a higher Cd tolerance. Silicon application also increased the chlorophyll and carotenoid contents by 33.9%–41% and 25.8%–47.3% compared to the Cd only treatments. | 2 | SARS-CoV-2 promotes RIPK1 activation to facilitate viral propagation显示文摘Coronavirus disease 2019(COVID-19),caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),is the ongoing global pandemic that poses substantial challenges to public health worldwide.A subset of COVID-19 patients experience systemic inflammatory response,known as cytokine storm,which may lead to death.Receptor-interacting serine/threonine-protein kinase 1(RIPK1)is an important mediator of inflammation and cell death.Here,we examined the interaction of RIPK1-mediated innate immunity with SARS-CoV-2 infection.We found evidence of RIPK1 activation in human COVID-19 lung pathological samples,and cultured human lung organoids and ACE2 transgenic mice infected by SARS-CoV-2.Inhibition of RIPK1 using multiple smallmolecule inhibitors reduced the viral load of SARS-CoV-2 in human lung organoids.Furthermore,therapeutic dosing of the RIPK1 inhibitor Nec-1s reduced mortality and lung viral load,and blocked the CNS manifestation of SARS-CoV-2 in ACE2 transgenic mice.Mechanistically,we found that the RNA-dependent RNA polymerase of SARS-CoV-2,NSP12,a highly conserved central component of coronaviral replication and transcription machinery,promoted the activation of RIPK1.Furthermore,NSP12323L variant,encoded by the SARS-CoV-2 C14408T variant first detected in Lombardy,Italy,that carries a Pro323Leu amino acid substitution in NSP12,showed increased ability to activate RIPK1.Inhibition of RIPK1 downregulated the transcriptional induction of proinflammatory cytokines and host factors including ACE2 and EGFR that promote viral entry into cells.Our results suggest that SARS-CoV-2 may have an unexpected and unusual ability to hijack the RIPK1-mediated host defense response to promote its own propagation and that inhibition of RIPK1 may provide a therapeutic option for the treatment of COVID-19. | Gang Xu Ying Li Shengyuan Zhang Haoran Peng Yunyun Wang Dekang Li Taijie Jin Zhuohao He Yilun Tong Chunting Qi Guowei Wu Kangyun Dong Jizhou Gou Yang Liu Tongyang Xiao Jing Qu Liang Li Liang Liu Ping Zhao Zheng Zhang Junying Yuan | 2021 | Cell Research2021,31,12: | 6 |

| 6 | Efficacy and safety of Liangxue Jiedu decoction for the treatment of progressive psoriasis vulgaris: a multicenter, randomized, controlled study显示文摘OBJECTIVE: To determine the clinical efficacy and safety of Liangxue Jiedu decoction(LJD) for the treatment of progressive psoriasis vulgaris and to provide the basis for the development of a standardized treatment protocol for psoriasis vulgaris.METHODS: In this multicenter, randomized, controlled study, patients with blood-heat type psoriasis were randomly assigned to receive either Chinese herbal medicine(LJD;treatment group) or Western Medicine(cetirizine hydrochloride, vitamin C, and vitamin B complex;control group). Psoriasis Area and Severity Index(PASI) scores were calculated in addition to the number of patients who achieved ≥ 50% or ≥ 75% improvement in PASI score from baseline. The change in symptoms of Chinese medicine(color of rash, burning sensation,itchiness, severity of irritation, and anger) was evaluated and safety was assessed as adverse events and laboratory analysis. t test, independent sample non-parametric test and χ2 test were used to analyze the results.RESULTS: A total of 238 participants were included in the study [treatment group, n = 122(PP analysis117);control group, n = 116(PP analysis 104)]. LJD treatment was associated with a significant improvement in skin lesions and symptoms com-pared with Western Medicine treatment. At the end of the 8-week treatment period, 23.77%(PP analysis 24.79%) of patients in the treatment group had achieved PASI75 and 57.38%(PP analysis 58.97%)had achieved PASI50;the corresponding figures in the control group were 9.48%(9.62%) and 25.00%(25.00%), respectively. | 7 | Facial Beauty Prediction Based on Lighted Deep Convolution Neural Network with Feature Extraction Strengthened显示文摘Convolutionneural network(CNN) has significantly pushed forward machine vision,which has achieved very significant results in face recognition, image classification and objection detection,and provides a new method for facial beauty prediction(FBP). Although the approach is widely applied in FBP,the research progress in FBP is relatively slow compared with face recognition.The first one is that there is less public database for FBP,and experiments for FBP are tested on small-scale database.The second one is that evaluation of facial beauty is subjective and lack of criterion,and CNN model is hard to train. In view of the problems of FBP, we expand Largescale database of Asian women's face database(LSAFBD)with data augmentation. A lighted deep convolution neural network(LDCNN) for FBP including 5650 K parameters is constructed by both Inception model of Google Net and Max-Feature-Max activation layer, which can extract multi-scale features of an image,get compacted presentation and reduce parameters. | 13 | Prediction of Cancer-Associated piRNA–mRNA and piRNA–lncRNA Interactions by Integrated Analysis of Expression and Sequence Data显示文摘piwi-interactingRNAs(piRNAs) are valuable biomarkers, but functional studies are still very limited.Recent research shows that piRNA-mediated cleavage acts on Transposable Elements(TEs), messengerRNAs(mRNAs), and long non-codingRNAs(lncRNAs). This study aimed to predict cancer-associated piRNA-mRNA and piRNA-lncRNA interactions as well as piRNA regulatory functions. Four cancer types(BRCA, HNSC, KIRC,and LUAD) were investigated. Interactions were identified by integrated analysis of the expression and sequence data. For the expression analysis, only piRNA–mRNA and piRNA–lncRNA pairs with expression profiles that were significantly inversely correlated were retained to reduce false-positive rates during the prediction. For the sequence analysis, miRanda was used for the target prediction. We identified 198 piRNA–mRNA and 10 piRNA–lncRNA pairs. Unlike mRNA and lncRNA expressions, the piRNA expression was relatively consistent across the cancer types. Furthermore, the identified piRNAs were consistent with previously published cancer biomarkers, such as piRNA-36741, piR-21032, and piRNA-57125.
