1Osteoarthritis:toward a comprehensive understanding of pathological mechanism显示文摘Osteoarthritis(OA) is the most common degenerative joint disease and a major cause of pain and disability in adult individuals. The etiology of OA includes joint injury, obesity, aging, and heredity. However, the detailed molecular mechanisms of OA initiation and progression remain poorly understood and, currently,there are no interventions available to restore degraded cartilage or decelerate disease progression. The diathrodial joint is a complicated organ and its function is to bear weight, perform physical activity and exhibit a joint-specific range of motion during movement. During OA development, the entire joint organ is affected, including articular cartilage, subchondral bone, synovial tissue and meniscus. 2A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 3Anisotropic coding metamaterials and their powerful manipulation of differently polarized terahertz waves显示文摘Metamaterials based on effective media can be used to produce a number of unusual physical properties(for example,negative refraction and invisibility cloaking)because they can be tailored with effective medium parameters that do not occur in nature.Recently,the use of coding metamaterials has been suggested for the control of electromagnetic waves through the design of coding sequences using digital elements‘0’and‘1,'which possess opposite phase responses.Here we propose the concept of an anisotropic coding metamaterial in which the coding behaviors in different directions are dependent on the polarization status of the electromagnetic waves.We experimentally demonstrate an ultrathin and flexible polarization-controlled anisotropic coding metasurface that functions in the terahertz regime using specially designed coding elements.By encoding the elements with elaborately designed coding sequences(both 1-bit and 2-bit sequences),the x-and y-polarized waves can be anomalously reflected or independently diffused in three dimensions.The simulated far-field scattering patterns and near-field distributions are presented to illustrate the dual-functional performance of the encoded metasurface,and the results are consistent with the measured results.We further demonstrate the ability of the anisotropic coding metasurfaces to generate a beam splitter and realize simultaneous anomalous reflections and polarization conversions,thus providing powerful control of differently polarized electromagnetic waves.The proposed method enables versatile beam behaviors under orthogonal polarizations using a single metasurface and has the potential for use in the development of interesting terahertz devices.Shuo Liu Tie Jun Cui Quan Xu Di Bao Liangliang Du Xiang Wan Wen Xuan Tang Chunmei Ouyang Xiao Yang Zhou Hao Yuan Hui Feng Ma Wei Xiang Jiang Jiaguang Han Weili Zhang Qiang Cheng 2016Light(Science & Applications)2016,5,1:36
4Global leaf nitrogen and phosphorus stoichiometry and their scaling exponent显示文摘Leaf nitrogen(N) and phosphorus(P) concentrations constrain photosynthetic and metabolic processes,growth and the productivity of plants. Their stoichiometry and scaling relationships regulate the allocation of N and P from subcellular to organism, and even ecosystem levels, and are crucial to the modelling of plant growth and nutrient cycles in terrestrial ecosystems. Prior work has revealed a general biogeographic pattern of leaf N and P stoichiometric relationships and shown that leaf N scales roughly as two-thirds the power of P. However, determining whether and how leaf N and P stoichiometries, especially their scaling exponents, change with functional groups and environmental conditions requires further verification. In this study, we compiled a global data set and documented the global leaf N and P concentrations and the N:P ratios by functional group, climate zone and continent. The global overall mean leaf N and P concentrations were 18.9 mg g^(-1) and 1.2 mg g^(-1), respectively, with significantly higher concentrations in herbaceous than woody plants(21.72 mg g^(-1) vs. 18.22 mg g^(-1) for N; and 1.64 mg g^(-1) vs. 1.10 mg g^(-1) for P). Both leaf N and P showed higher concentrations at high latitudes than low latitudes. Among six continents, Europe had the highest N and P concentrations(20.79 and 1.54 mg g^(-1)) and Oceania had the smallest values(10.01 and 0.46 mg g^(-1)). These numerical values may be used as a basis for the comparison of other individual studies. Further, we found that the scaling exponent varied significantly across different functional groups,latitudinal zones, ecoregions and sites. The exponents of herbaceous and woody plants were 0.659 and0.705, respectively, with significant latitudinal patterns decreasing from tropical to temperate to boreal zones. 6Molecular Epidemiological Characteristics of Streptococcus pyogenes Strains Involved in an Outbreak of Scarlet Fever in China,2011显示文摘Objective To investigate molecular characterization of streptococcus pyogenes isolates involved in an outbreak of scarlet fever in China in 2011.Methods Seventy-four Streptococcal pyogenes involved in an outbreak of scarlet fever were isolated from pediatric patients in the areas with high incidence in China from May to August of 2011.Emm genotyping,pulsed-field gel electrophoresis (PFGE),superantigen (SAg) genes and antimicrobial susceptibility profiling were analyzed for these isolates.Results A total of 4 different emm types were identified.Emm12 was the most prevalent type which contained four predominating PFGE patterns corresponding to four different virulence and superantigen profiles.Emm12 (79.7%) and emml (14.9%) accounted for approximately 94%of all the isolates.The speA gene was all negative in emm12 isolates and positive in emml isolates.All strains were resistant to erythromycin,and 89.4%of them were resistant to erythromycin,tracycline,and clindamycin simultaneously.Conclusion Several highly diversified clones with a high macrolide resistance rate comprise a predominant proportion of circulating strains,though no new emm type was found in this outbreak.The data provide a baseline for further surveillance of scarlet fever,which may contribute to the explanation of the outbreak and development of a GAS vaccine in China.YOU Yuan Hai SONG Yan Yan YAN Xiao Mei WANG Hai Bin ZHANG Meng Han TAO Xiao Xia Li Lei Lei ZHANG Yu Xin JIANG Xi Hong ZHANG Bing Hua ZHOU Hao XIAO Di JIN Lian Mei FENG Zi Jian LUO Feng Ji ZHANG Jian Zhong 2013Biomedical and Environmental Sciences2013,26,11:15

8Protective effect of recombinant human IL-1Ra on CCl_4-induced acute liver injury in mice显示文摘AIM: To evaluate the effects of positive regulation of recombinant human interleukin 1 receptor antagonist (rhIL-1Ra) on hepatic tissue recovery in acute liver injury in mice induced by carbon tetrachloride (CCl 4 ). METHODS: Acute liver damage was induced by injecting 8-wk-old mice with CCl 4 1 mL/kg (1:3 dilution in corn oil) intraperitoneally (ip). Survival after liver failure was assessed by injecting 8-wk-old mice with a lethal dose of CCl 4 2.6 mL/kg (1:1 dilution in corn oil) ip. Mice were subcutaneously injected with 1 mg/kg recombinant human IL-1Ra twice a day after CCl 4 treatment for 5 d. Serum alanine amino transferase (ALT) and aspartate aminotransferase (AST) levels were determined with a commercial assay kit. Serum IL-1β, IL-1Ra levels were measured by enzyme-linked immunosorbent assay kit. Quantitative real-time polymerase chain reaction was used to determine liver IL-1β, IL-1Ra and IL-6 expression during CCl 4-induced acute liver injury. Liver sections were stained with hematoxylin-eosin. A histology-injury grading system was used to evaluate the degree of necrosis after acute liver injury. Proliferating cell nuclear antigen (PCNA) staining was used to evaluate the role of rhIL-1Ra in promoting hepatocyte proliferation. RESULTS: Quantitative analysis showed a higher level of IL-6 mRNA expression and reduced serum AST and ALT levels in the livers of the rhIL-1Ra-treated group at the early phase of CCl 4-induced acute liver injury. Histological examination indicated a decrease in centrilobular necrotic areas in mice treated with rhIL-1Ra, and a novel role of rhIL-1Ra in promoting hepatocyte proliferation was also supported by an increase of PCNA staining. All these results, accompanied by a strong survival benefit in rhIL-1Ra-treated vs PBS-treated groups, demonstrated that rhIL-1Ra administration ameliorated the histological damage and accelerated the regeneration and recovery process of the liver. 10A phase I study of different doses and frequencies of pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) in patients with standard-dose chemotherapy-induced neutropenia显示文摘Objective: The recommended dose of prophylactic pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) is 100 μg/kg once per cycle for patients receiving intense-dose chemotherapy.However, few data are available on the proper dose for patients receiving less-intense chemotherapy. The aim of this phase I study is to explore the proper dose and administration schedule of PEG rhG-CSF for patients receiving standard-dose chemotherapy.Methods:Eligible patients received 3-cycle chemotherapy every 3 weeks.No PEG rhG-CSF was given in the first cycle.Patients experienced grade 3 or 4 neutropenia would then enter the cycle 2 and 3.In cycle 2,patients received a single subcutaneous injection of prophylactic PEG rhG-CSF on d 3,and received half-dose subcutaneous injection in cycle 3 on d 3 and d 5,respectively.Escalating doses(30,60,100 and 200μg/kg)of PEG rhG-CSF were investigated.Results:A total of 26 patients were enrolled and received chemotherapy,in which 24 and 18 patients entered cycle 2 and cycle 3 treatment,respectively.In cycle 2,the incidence of grade 3 or 4 neutropenia for patients receiving single-dose PEG rhG-CSF of 30,60,100 and 200 μg/kg was 66.67%,33.33%,22.22% and 0,respectively,with a median duration less than 1(0–2)d.No grade 3 or higher neutropenia was noted in cycle 3 in all dose cohorts.Conclusions:The pharmacokinetic and pharmacodynamic profiles of PEG rhG-CSF used in cancer patients were similar to those reported,as well as the safety.Double half dose administration model showed better efficacy result than a single dose model in terms of grade 3 neutropenia and above.The single dose of 60 μg/kg,100 μg/kg and double half dose of 30 μg/kg were recommended to the phase Ⅱ study,hoping to find a preferable method for neutropenia treatment.Yan Qin Xiaohong Han Lin Wang Ping Du Jiarui Yao Di Wu Yuanyuan Song Shuxiang Zhang Le Tang Yuankai Shi 2017Chinese Journal of Cancer Research2017,29,5:11

12The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 15Effects of multidrug resistance, antisense RNA on the chemosensitivity of hepatocellular carcinoma cells显示文摘BACKGROUND: Multidrug resistance is a major obstacle in cancer chemotherapy. We examined whether the antisense RNA of multidrug resistance gene 1 (mdr1) could reverse multidrug resistance in the human hepatocellular carcinoma (HCC) cell line SMMC7721/ADM. METHODS: The recombinant adenoviruses pAdEasy- GFP-ASmdr1 product was produced by the adenoviral vector AdEasy system, which can express antisense RNA against the mdr1 gene. Following that, the recombinant adenovirus was transfected into the P-glycoprotein- producing multidrug resistance cell line, SMMC7721/ADM human HCC cells resistant to adriamycin (ADM) and daunorubicin (DNR). In order to investigate the reversal of multidrug resistance phenotype, we measured the expression of mdr1 mRNA by RT-PCR and the production of P-glycoprotein by flow cytometry. The sensitivities for ADM and DNR SMMC7721/ADM cells were examined by [3-(4, 5-dimethylthi-azol-2-yl)-2,5 diphenyl-terazolium bromide] (MTT) analysis. RESULTS: The low-level expression of mdr1 mRNA and P-glycoprotein production were observed in parental sensitive cells SMMC/7721 in addition to the overexpressionof mdr1 mRNA and P-glycoprotein in SMMC7721/ADM cells. The transfection of antisense-RNA into SMMC7721/ ADM cells resulted in decreases of mdr1 mRNA and P-glycoprotein, but increase of drug sensitivities. The sensitivities of transfected SMMC7721/ADM cells to ADM and DNR in IC50 reduced by 31.25% and 62.96% respectively. CONCLUSIONS: Mdr1 antisense RNA can increase the sensitivities of SMMC7721/ADM cells to anticancer drug by decreasing the expression of the mdr1 gene and inhibiting P-glycoprotein expression. 16Future changes in coverage of 1.5℃ and 2℃warming thresholds显示文摘The areas covered by 1.5 ℃ and 2.0 ℃ warming thresholds under RCP2.6, RCP4.5, and RCP8.5 were analyzed based on 22 CMIP5 models. More than 90% of the model runs are in agreement that by the end of the 21 st century, near-surface air temperature changes over ~5%(~2%), ~40%(~18%), and ~92%(~86%)of the globe will cross the 1.5 ℃(2.0 ℃) threshold under RCP2.6, RCP4.5, and RCP8.5, respectively. Under RCP8.5, nearly the whole of North America, Europe + Russia, Africa, and Asia–Russia will cross the 1.5 ℃(2.0 ℃) threshold in ~2050(~2060), while the coverage rates over South America and Oceania are ~80%(~75%) and ~50%(~30%), respectively. The threshold-onset time(TOT) for 2 ℃ warming is earliest over Europe + Russia and North America, followed by Africa, Asia–Russia, South America, and finally Oceania under the RCP4.5 and RCP8.5 scenarios. 17Fabrication and characterization of laminated Ti-(TiB + La_2O_3)/Ti composite显示文摘The incorporation of ceramic particulate reinforcements into titanium alloys can improve the specific strength and specific stiffness,while inevitably reduce the plasticity and ductility.In this study,in situ synthesized multilayer Ti-(TiB +La_2O_3)/Ti composite was designed by learning from the microstructure of nature biological materials with excellent mechanical properties.The Ti-(TiB + La_2O_3)/Ti composite with unique characteristic of laminated structure was prepared by combined powder metallurgy and hot rolling.The method has the synthesize advantages with in-situ reaction of Ti and LaB_6 at high temperature and controllability of reinforcements size and constituent phases in composites.The result shows that the pores in the as sintered laminated structure composite completely disappeared after hot rolling at 1050℃.The agglomerated reinforcement particles were well dispersed and distributed uniformly along the rolling direction.The thickness of pure Ti layer and(TiB+La_2O_3)/Ti composite layer decreased from 1 mm to about 200 μm.Meanwhile,the grains size was refined obviously after rolling deformation.The room temperature tensile test indicates that the elongation of the laminated Ti-(TiB + La_2O_3)/Ti composite improved from 13%to 17%in comparison with the uniform(TiB + La_2O_3)/Ti composite,while the tensile strength had little change.It provides theoretical and experimental basis for fabricating the novel high performance laminated Ti-(TiB + La_2O_3)/Ti composites.Yuanfei Han Hongqiang Duan Weijie Lu Liqiang Wang Di Zhang 2015Progress in Natural Science:Materials International2015,25,5:8
18Improving Damping Properties and Thermal Stability of Epoxy/Polyurethane Grafted Copolymer by Adding Glycidyl POSS显示文摘Modified castor oil-based epoxy resin (EP)/polyurethane (PU) grafted copolymer by glycidyl polyhedral oligomeric silsesquioxane (glycidyl POSS) was synthesized. The damping properties, thermal stability, mechanical properties and morphology of the grafted copolymer modified by glycidyl POSS were studied systematically. The results revealed that the incorporation of glycidyl POSS improved the damping performance evidently and broadened damping temperature range, especially when the glycidyl POSS content was0.2%–1%. At the same time, there was a slight increase in thermal stability with the increase of POSS content. The tensile properties changed with the change of the copolymer's Tg, decreased at low POSS contents and increased at high POSS contents. 19A cyclodextrin-based nanoformulation achieves co-delivery of ginsenoside Rg3 and quercetin for chemo-immunotherapy in colorectal cancer显示文摘The immune checkpoint blockade therapy has profoundly revolutionized the field of cancer immunotherapy. However, despite great promise for a variety of cancers, the efficacy of immune checkpoint inhibitors is still low in colorectal cancer(CRC). This is mainly due to the immunosuppressive feature of the tumor microenvironment(TME). Emerging evidence reveals that certain chemotherapeutic drugs induce immunogenic cell death(ICD), demonstrating great potential for remodeling the immunosuppressive TME. In this study, the potential of ginsenoside Rg3(Rg3) as an ICD inducer against CRC cells was confirmed using in vitro and in vivo experimental approaches. The ICD efficacy of Rg3 could be significantly enhanced by quercetin(QTN) that elicited reactive oxygen species(ROS). To amelioratein vivo delivery barriers associated with chemotherapeutic drugs, a folate(FA)-targeted polyethylene glycol(PEG)-modified amphiphilic cyclodextrin nanoparticle(NP) was developed for co-encapsulation of Rg3 and QTN. The resultant nanoformulation(CD-PEG-FA.Rg3.QTN) significantly prolonged blood circulation and enhanced tumor targeting in an orthotopic CRC mouse model, resulting in the conversion of immunosuppressive TME. Furthermore, the CD-PEG-FA.Rg3.QTN achieved significantly longer survival of animals in combination with Anti-PD-L1. 20Green synthesis of fluorescent carbon quantum dots and carbon spheres from pericarp显示文摘An economical idea was developed to synthesize fluorescent carbon quantum dots(CQDs) directly from the refluxing extraction of orange pericarp via a hydrothermal technique. Hydrothermal temperatures and times were adjusted to control the particle sizes and the quantum yields of the obtained CQDs. The as-prepared carbon quantum dots showed narrow particle size distribution, good water solubility, and acceptable fluorescence lifetimes. Due to their high stability, these obtained carbon quantum dots have great application potential in nano-biotechnology.
