| 2 | Combined prednisone and levothyroxine improve treatment of severe thrombocytopenia in hepatitis B with compensatory cirrhosis accompanied by subclinical and overt hypothyroidism显示文摘The aim of the present study was to investigate the relationship between hypothyroidism and thrombocytopenia in hepatitis Brelated compensatory liver cirrhosis and to determine whether treatment with levothyroxine and prednisone is superior in a multicenter, open-label, observational study in China. In total, 125 consecutive hepatitis B-related compensated liver cirrhosis patients with severe thrombocytopenia accompanied by hypothyroidism were included. The patients were divided into four groups according to treatment strategy: a control group(n=29), a prednisone group(n=25), a levothyroxine group(n=32) and a prednisone plus levothyroxine group(n=39). Severe thrombocytopenia was more prevalent in hepatitis B-associated compensatory liver cirrhosis patients with hypothyroidism than in euthyroid patients(29.6% vs. 14.7%, P<0.05). Combination treatment with prednisone and levothyroxine decreased the risk of bleeding and improved platelet recovery compared to control treatment and treatment with either prednisone or levothyroxine alone. The platelet count before therapy, serum thyroid stimulating hormone and combination treatment with prednisone and levothyroxine were associated with bleeding events. Therefore, the present study suggests that hypothyroidism is associated with severe thrombocytopenia in hepatitis B-associated compensatory liver cirrhosis. | 3 | Arsenic trioxide alleviates acute graft-versus-host disease by modulating macrophage polarization显示文摘This study aimed to explore macrophage polarization in acute graft-versus-host disease after hematopoietic stem cell transplantation, and investigated if arsenic trioxide(ATO) could correct this imbalance. In the colon of GVHD mice, we found that the number of F4/80+iNOS+ cells as well as the expression intensity of TNF-α and IL-1β was greater in the GVHD group than in the BM group, whereas the number of F4/80+CD206+ cells and the expression intensity of IL-10 and TGF-β was greater in the BM group than in the GVHD group. We investigated the effect of ATO on GVHD mice, and found that ATO treatment clearly improved the survival of the mice and reduced the severity of GVHD. In addition, ATO reduced the number of F4/80+iNOS+ cells, and increased the number of F4/80+CD206+ cells in the colon of GVHD mice. Furthermore, ATO sharply decreased CD86 and CD80 expression, and increased CD163 and CD206 expression in macrophages induced from aGVHD patients. Therefore,ATO can modulate the M1 and M2 phenotype in GVHD mice or in macrophages from aGVHD patients. | 5 | Reduced β2-GPI is associated with increased platelet aggregation and activation in patients with prolonged isolated thrombocytopenia after allo-HSCT显示文摘We aimed to measure platelet function and its relationship with β2-GPI in prolonged isolated thrombocytopenia(PT) after allogeneic hematopoietic stem cell transplantation(allo-HSCT).Fifty-six patients with PT and 60 allo-HSCT recipients without PT(non-PT controls) were enrolled.Platelet aggregation and activation,β2-GPI and anti-β2-GPI antibody levels,vWF antigen,and vWF activity were analyzed.The effect of β2-GPI on platelet aggregation was also measured ex vivo.Results showed that ADP-induced platelet aggregation significantly increased(39%±7.5% vs.23%±8.5%,P=0.032),and the platelet expression of both CD62 p(33.6%±11.6% vs.8.5%±3.5%,P<0.001) and PAC-1(42.4%±7.6% vs.6.8%±2.2%,P<0.001) was significantly higher in patients with PT than in those without PT.Significantly lower β2-GPI levels(164.2±12 μg m L^-1 vs.234.2±16 μg mL^-1,P<0.001),higher anti-β2-GPI IgG levels(1.78±0.46 U mL^-1 vs.0.94±0.39 U mL^-1,P<0.001),and increased vWF activity(133.06%±30.50% vs.102.17%±25.90%,P<0.001) were observed in patients with PT than in those without PT.Both ADPinduced platelet aggregation(n=116,r^2=-0.5042,P<0.001) and vWF activity(n=116,r^2=-0.2872,P<0.001) were negatively correlated with β2-GPI levels.In summary,our data suggested that platelet aggregation and activation were significantly higher in patients with PT than in those without PT,which might be associated with reduced β2-GPI levels.The reduced β2-GPI levels might be due to the existence of anti-β2-GPI IgG. | Haixia Fu Jingzhong Zhao Lanping Xu Kaiyan Liu Yu Wang Huan Chen Wei Han Jingzhi Wang Fengrong Wang Xiaojun Huang Xiaohui Zhang | 2019 | Science China(Life Sciences)2019,62,7: | 4 |

Methods: Forty SD rats were randomly assigned to five groups: control group (group N), asthma model group (group A), two pretreated groups of different concentrations of ketamine (group K1, K2) and dexamethasone group(group D) with eight rats in each group.The rats in group A were sensitized by injection of ovalbumin (OA) together with aluminum hydroxide and bordetella pertussis as adjuvants. Two weeks after the sensitization,aerosolized OA was used to cause asthma. The rats in group K1 and K2 were sensitized with OA as group A , and then exposed to aerosol of ketamine ,with the concentration of 25 g/L and 50 g/L respectively.Before using aerosolized OA,the rats in group D were exposed to aerosol of 0.01% dexamethasone .The level of NO2-/NO3- in lung tissues, inducible nitric oxide synthetase(iNOS) and constitute nitric oxide synthetase(cNOS) was measured in all groups. Results: The level of NO2-/NO3- and the activity of iNOS in lung tissues in group A were signiticantly higher than those in the other groups. The iNOS activity and the level of NO2-/NO3- in lung tissues were highly positively correlated. Conclusion: NO can induce airway hyperreactivity that may worsen asthma. | 16 | Rumen microbiota-host transcriptome interaction mediates the protective effects of trans-10,cis-12 CLA on facilitating weaning transition of lambs显示文摘Developing alternatives to antibiotics for prevention of gastrointestinal dysbiosis in early-weaning farmed animals is urgently needed.This study was to explore the potential effects of trans-10,cis-12 conjugated linoleic acid(CLA)on maintaining ruminal homeostasis of young ruminants during the weaning transition period.Thirty neonatal lambs were selected(6 lambs per group)and euthanized for rumen microbial and epithelial analysis.The lambs were weaned at 28 d and experienced the following 5 treatments:euthanized on d 28 as the pre-weaning control(CON0),fed starter feed for 5(CON5)or 21(CON21)d,fed starter feed with 1%of CLA supplemented for 5(CLA5)or 21(CLA21)d.Results showed that the average daily weight gain and dry matter intake were significantly higher in CLA5 than CON5 group.As compared with the CON5 and CON21 group,the relative abundances of volatile fatty acid(VFA)producing bacteria including Bacteroides,Treponema,Parabacteroides and Anaerovibrio,as well as the concentrations of acetate,butyrate and total VFA were significantly increased in CLA5 and CLA21 group,respectively.Integrating microbial profiling and epithelial transcriptome results showed that 7 downregulated inflammatory signaling-related host genes IL2RA,CXCL9,CD4,CCR4,LTB,SPP1,and BCL2A1 with CLA supplementation were significantly negatively correlated with both VFA concentration and VFA producing bacteria,while 3(GPX2,SLC27A2 and ALDH3A1)and 2(GSTM3 and GSTA1)upregulated metabolism-related genes,significantly positively correlated with either VFA concentration or VFA producing bacteria,respectively.To confirm the effects of CLA on epithelial signal transduction,in vitro experiment was further conducted by treating rumen epithelial cells without or with IL-17A+TNF-αfor 12 h after pretreatment of 100μM CLA or not(6 replicates per treatment).The results demonstrated the anti-inflammatory effect of CLA via suppressing the protein expression of NF-кB p-p65/p65 with the activation of peroxisome proliferator-activated receptor gamma(PPARγ).In conclusion,CLA supplementation enhanced the ruminal microbiota-driven transcriptional regulation in healthy rumen epithelial development via rumen VFA production,and CLA may therefore serve as an alternative way to alleviate early-weaning stress and improve physiological and metabolic conditions of young ruminants. | Chunlei Yang Xiangfei Deng Peter Lund Haixia Liu Xingwang Ding Zhengwei Fu Naifeng Zhang Jinjun Li Lifeng Dong | 2023 | Animal Nutrition2023,,1: | 0 |

| 18 | Bending strain effects on the optical and optoelectric properties of GaN nanowires显示文摘Elastic strain has been an important method to regulate the electronic structures and physical properties of nanoscale semiconductors due to the promising potentials in improving the performance of their optoelectronic devices.Here,we report the investigation of bending strain effects on the optical and optoelectric properties of individual gallium nitride(GaN)nanowires(NWs).By charactering the near-band emission spectrum of individual GaN NWs at different bending strains with low temperature cathodoluminescence(CL),we reveal that the near-band emission splits into two peaks,where the low energy peak displays a linear redshift with increasing the bending strain while the high energy one shows a slight blueshift.Further localized ultraviolet(UV)photoresponse measurements illustrate that the photoresponse of the GaN NWs shows a linear increase with the bending train,and the maximum enhancement is more than two orders of magnitude.The experimental observations are well interpreted by theoretical calculations on the strain modulation on the electronic band structure of GaN combined with analysis of carrier dynamics and optical waveguide effect in the bending strain field.Our results not only shed light on the bending strain effects on the optical and optoelectric properties of semiconductors,but also hold potential to help the future design of high performance nano-optoelectric devices. | Xuewen Fu Haixia Nie Zepeng Sun Min Feng Xiang Chen Can Liu Fang Liu Dapeng Yu Zhimin Liao | 2022 | Nano Research2022,15,5: | 0 |
