| 2 | Construct hepatic analog by cell-matrix controlled assembly technology显示文摘A mixture of hepatic cells and chito- san/gelatin solution was deposited to construct a hepatic analog by way of layer-by-layer deposition technique using a home-made devise. The size and cell concentration of the analogs can be controlled freely. Approximately 90% of the hepatic cells re- mained viable under 0.2 Mpa extrusion pressure. Cultured in vitro 8 weeks before animal test, hepatic cells in structure maintained their phenotype and kept proliferating, and albumin and other secretion of the cells increased. Cords and hepaton-like structures were observed after culture for 20 d. These results indicate that hepatic cells could be assembled di- rectly into a 3D viable structure and expanded to form a hepatic organoid. | 8 | Association of therapeutic occasion, gross motor function grading and developmental level with gross motor functional recovery in children with cerebral palsy显示文摘BACKGROUND: The abnormal posture and motor pattern have not stabilized in children with cerebral palsy at early period, thus timely treatment can establish normal postural reflex and motor pattern, and prevent complications of muscle contracture, ankylosis, skeletal deformity, etc. The clinical factors affecting the rehabilitative efficacy of gross motor function in children with cerebral palsy should be observed. OBJECTIVE: To observe the effects of therapeutic occasion, grading of gross motor function and developmental level on the rehabilitative efficacy in children with cerebral palsy. DESIGN: A case-controlled analysis. SETTING: Qilu Children's Hospital of Shandong University. PARTICIPANTS: Totally 138 children with cerebral palsy, who were hospitalized for 12 months in the Rehabilitation Center of Qilu Children's Hospital, Shandong University, were selected from April 2004 to September 2006, and all the children were diagnosed to be accorded with the standard set by the national seminar on cerebral palsy in 2004. There were 97 males and 41 females, including 55 cases of 0-2 years old, 47 cases of 2-4 years and 36 cases of 4-6 years. Informed contents were obtained from relatives of all the children. METHODS: ① Comprehensive rehabilitation treatment: Vojta method was to induce the children to turn over the body and crawl by stimulating reflective turn over and crawling on belly. Bobath method including trainings of head control, turning over body, keeping sitting position, keeping balance, crawling, keeping standing position, and walking, etc.; The children were massaged by using the maneuvers of push, press, rub, pull, wave, etc. according to the sites and types of palsy. Acupuncture was performed mainly at bilateral motor areas, the needle was retained for 1 hour per time, 6 days continuously every week, and followed by a 1-day interval. ② Prognosis assessment: The gross motor functional grading of the children with cerebral palsy at admission was assessed using the Chinese version of gross motor function classification system (GMFCS). The gross motor functions of the children with cerebral palsy were assessed before rehabilitation treatment and 12 months after rehabilitation treatment by using gross motor function measure (GMFM). The improved GMFM total score was calculated. Developmental quotient was evaluated using Gesell developmental schedules (GDS) after 12-month rehabilitation treatment. Univariate analysis of variance was performed using the improved GMFM total score with age, GMFCS grade and developmental level (developmental quotient). MAIN OUTCOME MEASURES: Effects of age started to accept rehabilitation treatment, GMFCS grade and developmental level on the recovery of gross motor function in children with cerebral palsy. RESULTS: All the 138 children with cerebral palsy were involved in the analysis of results. The improved GMFM total scores were significantly different among the children with cerebral palsy of different ages started to accept treatment, GMFCS grades and developmental levels (F=13.464, 37.283, 30.814, P < 0.01). The younger the children started to accept treatment, the lower the their GMFCS grades and the higher their developmental levels, then the higher their improved GMFM total scores. | 10 | Comparisons of corneal biomechanical and tomographic parameters among thin normal cornea,forme fruste keratoconus,and mild keratoconus显示文摘Background:To compare the dynamic corneal response(DCR)and tomographic parameters of thin normal cornea(TNC)with thinnest corneal thickness(TCT)(≤500μm),forme fruste keratoconus(FFKC)and mild keratoconus(MKC)had their central corneal thickness(CCT)matched by Scheimpflug imaging(Pentacam)and corneal visualization Scheimpflug technology(Corvis ST).Methods:CCT were matched in 50 eyes with FFKC,50 eyes with MKC,and 53 TNC eyes with TCT≤500μm.The differences in DCR and tomographic parameters among the three groups were compared.The receiver operating characteristic(ROC)curve was used to analyze the diagnostic significance of these parameters.Back propagation(BP)neural network was used to establish the keratoconus diagnosis model.Results:Fifty CCT-matched FFKC eyes,50 MKC eyes and 50 TNC eyes were included.The age and biomechanically corrected intraocular pressure(bIOP)did not differ significantly among the three groups(all P>0.05).The index of height asymmetry(IHA)and height decentration(IHD)differed significantly among the three groups(all P<0.05).IHD also had sufficient strength(area under the ROC curves(AUC)>0.80)to differentiate FFKC and MKC from TNC eyes.Partial DCR parameters showed significant differences between the MKC and TNC groups,and the deflection amplitude of the first applanation(A1DA)showed a good potential to differentiate(AUC>0.70)FFKC and MKC from TNC eyes.Diagnosis model by BP neural network showed an accurate diagnostic efficiency of about 91%.Conclusions:The majority of the tomographic and DCR parameters differed among the three groups.The IHD and partial DCR parameters assessed by Corvis ST distinguished FFKC and MKC from TNC when controlled for CCT. | Lei Tian Di Zhang Lili Guo Xiao Qin Hui Zhang Haixia Zhang Ying Jie Lin Li | 2021 | Eye and Vision2021,8,1: | 1 |

| 17 | MdWRKY11 improves copper tolerance by directly promoting the expression of the copper transporter gene MdHMA5显示文摘Overuse of fungicides and fertilizers has resulted in copper(Cu)contamination of soils and toxic levels of Cu in apple fruits.To breed Cu-resistant apple(Malus domestica)cultivars,the underlying molecular mechanisms and key genes involved in Cu resistance must be identified.Here,we show that MdWRKY11 increases Cu tolerance by directly promoting the transcription of MdHMA5.MdHMA5 is a Cu transporter that may function in the storage of excess Cu in root cell walls and stems for Cu tolerance in apple.The transcription factor MdWRKY11 is highly induced by excess Cu.MdWRKY11 overexpression in transgenic apple enhanced Cu tolerance and decreased Cu accumulation.Apple calli transformed with an MdWRKY11-RNAi construct exhibited the opposite phenotype.Both an in vivo chromatin immunoprecipitation assay and an in vitro electrophoretic mobility shift assay indicated that MdWRKY11 binds to the promoter of MdHMA5.Furthermore,MdWRKY11 promoted MdHMA5 expression in transgenic apple plants,as revealed by quantitative PCR.Moreover,inhibition of MdWRKY11 expression by RNA interference led to a significant decrease in MdHMA5 transcription.Thus,MdWRKY11 directly regulates MdHMA5 transcription.Our work resulted in the identification of a novel MdWRKY11-MdHMA5 pathway that mediates Cu resistance in apple. | Kun Shi Xuan Liu Yunpeng Zhu Yixue Bai Dongqian Shan Xiaodong Zheng Lin Wang Haixia Zhang Chanyu Wang Tianci Yan Fangfang Zhou Zehui Hu Yanzhao Sun Yan Guo Jin Kong | 2020 | Horticulture Research2020,7,1: | 1 |
