| 1 | Mitochondrial inhibitor as a new class of insulin sensitizer显示文摘Insulin resistance is a major risk factor for type 2 diabetes.AMP-activated protein kinase(AMPK)is a drug target in the improvement of insulin sensitivity.Several insulin-sensitizing medicines are able to activate AMPK through inhibition of mitochondrial functions.These drugs,such as metformin and STZ,inhibit ATP synthesis in mitochondria to raise AMP/ATP ratio in the.process of A MPK activation.However,chemicals that activate AMPK directly or by activating its upstream kinases have not been approved for treatment of type 2 diabetes in humans.In an early study,we reported that berberine inhibited oxygen consumption in mitochondria,and increased AMP/ATP ratio in cells.The observation suggests an indirect mechanism for AMPK activation by berberine.Berberine stimulates glycolysis for ATP production that offsets the cell toxicity after mitochondria inhibition.The study suggests that mitochondrial inhibition is an approach for AMPK activation.In this review article,literature is critically reviewed to interpret the role of mitochondria function in the mechanism of insulin resistance,which supports that mitochondria inhibitors represent a new class of AMPK activator.The inhibitors are promising candidates for insulin sensitizers.This review provides a guideline in search for small molecule AMPK activators in the drug discovery for type 2 diabetes. | Yong Zhang Jianping Ye | 2012 | Acta Pharmaceutica Sinica B2012,2,4: | 15 |
| 2 | Necking of Q&P Steel during Uniaxial Tensile Test with the Aid of DIC Technique显示文摘A lot of research has been focused on the necking process during the plastic deformation of sheet metals, but the localized necking is rarely distinguished form diffused necking by experiments, due to the limit of measurement equipment and method. Quenching and Partitioning (Q&P) steel is a 3rd generation advanced high strength steel (AHSS). Its good combination of high strength and ductility ensures potential application in automobile industry. Uniaxial tensile tests of QP980 steel sheet at five strain rates are performed to investigate the necking process and the effect of strain rate on necking behavior of Q&P steel. Digital image correlation (DIC) method is applied during tensile tests, and evolutions of major strain, minor strain and normal strain distributions along gauge section of the tensile specimens are obtained. The diffused and localized necking strains are determined according to SWIFT necking theory and HILL necking theory respectively. The test results indicate that with the increasing of strain rate in the investigated range, the diffused necking strain decreases from 0.152 to 0.120 and localized necking strain decreases from 0.245 to 0.137. Meanwhile, the difference of the two strains decreases form 0.096 to 0.017. Thus it can be concluded that strain rate has an influence on both necking strains during the deformation of QP980 steel sheet. | 3 | Mechanisms of insulin resistance in obesity显示文摘Obesity increases the risk for type 2 diabetes through induction of insulin resistance.Treatment of type 2 diabetes has been limited by little translational knowledge of insulin resistance although there have been several well-documented hypotheses for insulin resistance.In those hypotheses,inflammation,mitochondrial dysfunction,hyperinsulinemia and lipotoxicity have been the major concepts and have received a lot of attention.Oxidative stress,endoplasmic reticulum(ER)stress,genetic background,aging,fatty liver,hypoxia and lipodystrophy are active subjects in the study of these concepts.However,none of those concepts or views has led to an effective therapy for type 2 diabetes.The reason is that there has been no consensus for a unifying mechanism of insulin resistance.In this review article,literature is critically analyzed and reinterpreted for a new energy-based concept of insulin resistance,in which insulin resistance is a result of energy surplus in cells.The energy surplus signal is mediated by ATP and sensed by adenosine monophosphate-activated protein kinase(AMPK)signaling pathway.Decreasing ATP level by suppression of production or stimulation of utilization is a promising approach in the treatment of insulin resistance.In support,many of existing insulin sensitizing medicines inhibit ATP production in mitochondria.The effective therapies such as weight loss,exercise,and caloric restriction all reduce ATP in insulin sensitive cells.This new concept provides a unifying cellular and molecular mechanism of insulin resistance in obesity,which may apply to insulin resistance in aging and lipodystrophy. | Jianping Ye | 2013 | Frontiers of Medicine2013,7,1: | 11 |
| 4 | Mechanical and dielectric properties of porous and wave-transparent Si3N4–Si3N4 composite ceramics fabricated by 3D printing combined with chemical vapor infiltration显示文摘Porous Si3N4–Si3N4 composite ceramics were fabricated by 3D printing combined with low-pressure chemical vapor infiltration(CVI).This technique could effectively improve the designability of porous Si3N4 ceramics and optimize the mechanical and dielectric properties.The effects of process parameters including the deposition time and heat treatment on the microstructure and properties of porous Si3N4–Si3N4 composite ceramics were studied.The study highlights following:When CVI processing time was increased from 0 to 12 h,the porosity decreased from68.65%to 26.07%and the density increased from 0.99 to 2.02 g/cm3.At the same time,the dielectric constant gradually increased from 1.72 to 3.60;however,the dielectric loss always remained less than0.01,indicating the excellent electromagnetic(EM)wave-transparent performance of porous Si3N4–Si3N4 composite ceramics.The maximum flexural strength of 47±2 MPa was achieved when the deposition time attained 6 h.After heat treatment,the porosity increased from 26.07%to 36.02%and the dielectric constant got a slight increase from 3.60 to 3.70 with the dielectric loss still maintaining lower than 0.01.It has been demonstrated that the porous Si3N4–Si3N4 composite ceramics are a promising structural and EM wave-transparent material suitable for high temperature service. | Zanlin CHENG Fang YE Yongsheng LIU Tianlu QIAO Jianping LI Hailong QIN Laifei CHENG Litong ZHANG | 2019 | Journal of Advanced Ceramics2019,8,3: | 10 |

| 7 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. | 9 | Construction,Characterization,and Chromosomal Mapping of a Fosmid Library of the White-Cheeked Gibbon (Nomascus leucogenys)显示文摘Gibbons have experienced extensive karyotype rearrangements during evolution and represent an ideal model for studying the underlying molecular mechanism of evolutionary chromosomal rearrangements. It is anticipated that the cloning and sequence characterization of evolutionary chromosomal breakpoints will provide vital insights into the molecular force that has driven such a radical karyotype reshuffle in gibbons. We constructed and characterized a high-quality fosmid li-brary of the white-cheeked gibbon (Nomascus leucogenys) containing 192,000 non-redundant clones with an average insert size of 38 kb and 2.5-fold genome coverage. By end sequencing of 100 randomly selected fosmid clones,we generated 196 se-quence tags for the library. These end-sequenced fosmid clones were then mapped onto the chromosomes of the white-cheeked gibbon by fluorescence in situ hy-bridization,and no spurious chimeric clone was detected. BLAST search against the human genome showed a good correlation between the number of hit clones and the number of chromosomes,an indication of unbiased chromosomal distribu-tion of the fosmid library. The chromosomal distribution of the mapped clones is also consistent with the BLAST search result against human and white-cheeked gibbon genomes. | 10 | Regulation of microbiota–GLP1 axis by sennoside A in diet-induced obese mice显示文摘Sennoside A(SA) is a bioactive component of Chinese herbal medicines with an activity of irritant laxative, which is often used in the treatment of constipation and obesity. However, its activity remains unknown in the regulation of insulin sensitivity. In this study, the impact of SA on insulin sensitivity was tested in high fat diet(HFD)-induced obese mice through dietary supplementation. At a dosage of 30 mg/kg/day, SA improved insulin sensitivity in the mice after 8-week treatment as indicated by HOMA-IR(homeostatic model assessment for insulin resistance) and glucose tolerance test(GTT). SA restored plasma level of glucagon-like peptide 1(GLP1) by 90% and mRNA expression of Glp1 by 80% in the large intestine of HFD mice. In the mechanism, SA restored the gut microbiota profile, short chain fatty acids(SCFAs), and mucosal structure in the colon. A mitochondrial stress was observed in the enterocytes of HFD mice with ATP elevation, structural damage, and complex dysfunction. The mitochondrial response was induced in enterocytes by the dietary fat as the same responses were induced by palmitic acid in the cell culture. The mitochondrial response was inhibited in HFD mice by SA treatment. | 12 | Valence state manipulation of Sm3+ ions via a phase-shaped femtosecond laser field显示文摘The ability to manipulate the valence state conversion of rare-earth ions is crucial for their applications in color displays, optoelectronic devices, laser sources, and optical memory. The conventional femtosecond laser pulse has been shown to be a well-established tool for realizing the valence state conversion of rare-earth ions, although the valence state conversion efficiency is relatively low. Here, we first propose a femtosecond laser pulse shaping technique for improving the valence state conversion efficiency of rare-earth ions. Our experimental results demonstrate that the photoreduction efficiency from Sm^(3+) to Sm^(2+) in Sm^(3+)-doped sodium aluminoborate glass using a π phase step modulation can be comparable to that using a transform-limited femtosecond laser field, while the peak laser intensity is decreased by about 63%, which is very beneficial for improving the valence state conversion efficiency under the laser-induced damage threshold of the glass sample. | 16 | Mechanism of insulin resistance in obesity: a role of ATP显示文摘Obesity increases the risk of type 2 diabetes through the induction of insulin resistance.The mechanism of insulin resistance has been extensively investigated for more than 60 years,but the essential pathogenic signal remains missing.Existing hypotheses include inflammation,mitochondrial dysfunction,hyperinsulinemia,hyperglucagonemia,glucotoxicity,and lipotoxicity.Drug discoveries based on these hypotheses are unsuccessful in the development of new medicines.In this review,multidisciplinary literature is integrated to evaluate ATP as a primary signal for insulin resistance.The ATP production is elevated in insulin-sensitive cells under obese conditions independent of energy demand,which we have named“mitochondrial overheating.”Overheating occurs because of substrate oversupply to mitochondria,leading to extra ATP production.The ATP overproduction contributes to the systemic insulin resistance through several mechanisms,such as inhibition of AMPK,induction of mTOR,hyperinsulinemia,hyperglucagonemia,and mitochondrial dysfunction.Insulin resistance represents a feedback regulation of energy oversupply in cells to control mitochondrial overloading by substrates.Insulin resistance cuts down the substrate uptake to attenuate mitochondrial overloading.The downregulation of the mitochondrial overloading by medicines,bypass surgeries,calorie restriction,and physical exercise leads to insulin sensitization in patients.Therefore,ATP may represent the primary signal of insulin resistance in the cellular protective response to the substrate oversupply.The prevention of ATP overproduction represents a key strategy for insulin sensitization. | Jianping Ye | 2021 | Frontiers of Medicine2021,15,3: | 4 |
| 17 | A New Species of Genus Microhyla(Amphibia: Anura: Microhylidae) from Zhejiang Province, China显示文摘We described a new species, Microhyla beilunensis sp. nov., from Zhejiang Province of China. Phylogenetic analyses based on the mitochondrial 12 S, 16 S and CO1 gene sequences suggested that the new taxon was distinctly separated from its congeners and closed to M. mixtura and M. okinavensis. Morphologically, the new species could be identified from its congeners except M. mixtura by several characters:(1) rudimentary webs on toe base;(2) absence of disks and dorsal median longitudinal grooves on finger tips;(3) presence of disks and dorsal median longitudinal grooves on toe tips. | 18 | Comparative evaluation of the physicochemical properties of nano-hydroxyapatite/collagen and natural bone ceramic/collagen scaffolds and their osteogenesis-promoting effect on MC3T3-E1 cells显示文摘The use of various types of calcium phosphate has been reported in the preparation of repairing materials for bone defects.However,the physicochemical and biological properties among them might be vastly different.In this study,we prepared two types of calcium phosphates,nano-hydroxyapatite(nHA)and natural bone ceramic(NBC),into 3D scaffolds by mixing with type I collagen(CoL),resulting in the nHA/CoL and NBC/CoL scaffolds.We then evaluated and compared the physicochemical and biological properties of these two calcium phosphates and their composite scaffold with CoL.Scanning electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),Fourier-transform infrared spectroscopy(FTIR),X-ray diffraction spectroscopy(XRD)and compressive tests were used to,respectively,characterize the morphology,composition,distribution and the effect of nHA and NBC to collagen.Next,we examined the biological properties of the scaffolds using cytotoxicity testing,flow cytometry,immunofluorescence staining,biocompatibility testing,CCK-8 assays and RT-PCR.The results reflected that the Ca2t released from nHA and NBC could bind chemically with collagen and affect its physicochemical properties,including the infrared absorption spectrum and compression modulus,among others.Furthermore,the two kinds of scaffolds could promote the expression of osteo-relative genes,but showed different gene induction properties.In short,NBC/CoL could promote the expression of early osteogenic genes,while nHA/CoL could upregulate late osteogenic genes.Conclusively,these two composite scaffolds could provide MC3T3-E1 cells with a biomimetic surface for adhesion,proliferation and the formation of mineralized extracellular matrices.Moreover,nHA/CoL and NBC/CoL had different effects on the period and extent ofMC3T3-E1 cell mineralization. | Xiongxin Lei Jianping Gao Fangyu Xing Yang Zhang Ye Ma Guifeng Zhang | 2019 | Regenerative Biomaterials2019,6,6: | 4 |

| 20 | Live cell imaging and proteomic profiling of endogenous NEAT1 lncRNA by CRISPR/ Cas9-mediated knock-in显示文摘In mammalian cells,long noncoding RNAs(lncRNAs)form complexes with proteins to execute various biological functions such as gene transcription,RNA processing and other signaling activities.However,methods to track endogenous lncRNA dynamics in live cells and screen for lncRNA interacting proteins are limited.Here,we report the development of CERTIS(CRISPR-mediated Endogenous lncRNA Tracking and Immunoprecipitation System)to visualize and isolate endogenous lncRNA,by precisely inserting a 24-repeat MS2 tag into the distal end of lncRNA locus through the CRISPR7Cas9 technology.In this study,we show that CERTIS effectively labeled the paraspeckle lncRNA NEAT1 without disturbing its physiological properties and could monitor the endogenous expression variation of NEAT1.In addition,CERTIS displayed superior performance on both short-and long-term tracking of NEAT1 dynamics in live cells.We found that NEAT1 and paraspeckles were sensitive to topoisomerase I specific inhibitors.Moreover,RNA Immunoprecipitation(RIP)of the MS2-tagged NEAT1 lncRNA successfully revealed several new protein components of paraspeckle.Our results support CERTIS as a tool suitable to track both spatial and temporal lncRNA regulation in live cells as well as study the lncRNA-protein interactomes. | Bohong Chen Shengcheng Deng Tianyu Ge Miaoman Ye Jianping Yu Song Lin Wenbin Ma Zhou Songyang | 2020 | Protein & Cell2020,11,9: | 4 |
