4Review of Rapid Transformation of Floodplain Wetlands in Northeast China: Roles of Human Development and Global Environmental Change显示文摘Northeast China is the region with the largest area of wetlands in China. The Sanjiang Plain and the Songnen Plain are large freshwater marsh distribution regions that are affected by climate warming and by the increasing frequency and density of extreme weather and are the regions most subject to disturbances by human activities in Northeast China. The wetlands of the Sanjiang Plain and the Songnen Plain have shrunk severely in the past 60 years, and wetland functions have been reduced substantially because of climate change, unreasonable land use, fire episodes, engineering and construction works and urbanization. Large-scale agricultural development started in the 1950 s has been the most important driving factor for wetland loss and degradation in the Sanjiang Plain. Water shortage has been the most important factor for degradation and fragmentation of wetlands in the Songnen Plain. To mitigate wetland degradation and better protect wetlands, special regulations, long-term mechanisms and technical support of wetland protection should be established. A wetland compensation program should be implemented, and technologies for increasing the adaptive capacity of wetlands should be developed. 5Inhibition of ASCT2 induces hepatic stellate cell senescence with modified proinflammatory secretome through an IL-1α/NF-κB feedback pathway to inhibit liver fibrosis显示文摘Senescence of activated hepatic stellate cells(aHSCs) is a stable growth arrest that is implicated in liver fibrosis regression.Senescent cells often accompanied by a multi-faceted senescence-associated secretory phenotype(SASP).But little is known about how alanine-serine-cysteine transporter type-2(ASCT2),a high affinity glutamine transporter,affects HSC senescence and SASP during liver fibrosis.Here,we identified ASCT2 is mainly elevated in aHSCs and positively correlated with liver fibrosis in human and mouse fibrotic livers.We first discovered ASCT2 inhibition induced HSCs to senescence in vitro and in vivo.The proinflammatory SASP were restricted by ASCT2 inhibition at senescence initiation to prevent paracrine migration.Mechanically,ASCT2 was a direct target of glutaminolysisdependent proinflammatory SASP,interfering IL-1α/NF-κB feedback loop via interacting with precursor IL-1α at Lys82.From a translational perspective,atractylenolide Ⅲ is identified as ASCT2 inhibitor through directly bound to Asn230 of ASCT2.The presence of -OH group in atractylenolide Ⅲ is suggested to be favorable for the inhibition of ASCT2.Importantly,atractylenolide Ⅲ could be utilized to treat liver fibrosis mice.Taken together,ASCT2 controlled HSC senescence while modifying the proinflammatory SASP.Targeting ASCT2 by atractylenolide Ⅲ could be a therapeutic candidate for liver fibrosis.Feixia Wang Zhanghao Li Li Chen Ting Yang Baoyu Liang Zili Zhang Jiangjuan Shao Xuefen Xu Guoping Yin Shijun Wang Hai Ding Feng Zhang Shizhong Zheng 2022Acta Pharmaceutica Sinica B2022,12,9:2

8Efficient circular gene knockout system for Burkholderiales strain DSM 7029 and Mycobacterium smegmatis mc^2 155显示文摘Multiple gene knockouts are often employed in studies of microbial physiology and genetics. However, the selective markers that confer antibiotic resistance are generally limited, so it is necessary to remove these resistance genes before the next round of using, which is time consuming and labor intensive. Here, we created a universal circular gene knockout system for both the gram-negative bacterial Burkholderiales strain DSM 7029 and the gram-positive bacterial Mycobacterium smegmatis mc^2 155, by combining the homologous recombination with multiple serine integrase-meditated site-specific recombination systems. In this system, a resistance gene and an integrase gene were constructed within the two attachment sites corresponding to a second, different integrase to form a cassette for gene disruption, which could be easily removed by the second integrase during the subsequent round of gene knockout. The sacB gene was also employed for negative selection. As the integrase-mediated deletion of the resistance/integrase gene cassette was highly efficient and concurrent with the following knockout round, the cyclic use of three cassettes could achieve multiple gene knockout in a sequential manner. Following the modularity concept in synthetic biology, common components of the knockout plasmids were retained as BioBricks, accelerating the knockout plasmids construction process. 19Utility of the Surgical Apgar Score in pancreatic cancer and modification显示文摘Objectives:The Surgical Apgar Score(SAS)can predict the incidence of complications in different surgical fields.However,it is rarely studied in pancreatic cancer.The aim of the present study was to assess the predictive value of the SAS in pancreatic ductal adenocarcinoma(PDAC),and then propose a modified SAS which was more suitable for pancreatic cancer patients.Materials and methods:A prospective cohort study of 160 PDAC patients was concluded.The primary endpoint was 30-day major complications.The SAS was calculated as described.The overall discriminatory power of the score was analyzed using receiver operating characteristic curves and the area under the curve(AUC)with respect to major complications or death.Results:It showed a significant predictive value of SAS in major complications or death in PDAC(p=0.020,AUC=0.606),especially in complication of pneumonia(p=0.022)and pleural effusion(p=0.023).In addition,the SAS exert significant predictive value in distal pancreatectomy group,but it has a weak predictive value for pancreaticoduodenectomy group.On multivariable analyses,occurrence of major postoperative complications was associated with lowest mean arterial pressure,estimated blood loss and operative time.Interestingly,as a characteristic of SAS,lowest heart rate was not involved.The modified SAS we proposed including lowest mean arterial pressure,estimated blood loss and operative time increased AUC from 0.606 to 0.743.Conclusions:The SAS can be a simple,rapid scoring system that effectively predicts major postoperative complications.Besides,the modified SAS we proposed in this study,which included lowest mean arterial pressure,estimated blood loss and operative time,exert a better predictive value in PDAC patients.Guoping Ding Liangjing Zhou Wenchao Chen Zhengrong Wu Tao Shen Liping Cao 2019Laparoscopic, Endoscopic and Robotic Surgery2019,2,4:0
