| 1 | TNF-αTNFR1 Signaling is Required for the Full Expression of Acute and Chronic Itch in Mice via Peripheral and Central Mechanisms显示文摘Increasing evidence suggests that cytokines and chemokines play crucial roles in chronic itch. In the present study, we evaluated the roles of tumor necrosis factor-alpha(TNF-a) and its receptors TNF receptor subtype-1(TNFR1) and TNFR2 in acute and chronic itch in mice.Compared to wild-type(WT) mice, TNFR1-knockout(TNFR1-KO) and TNFR1/R2 double-KO(DKO), but not TNFR2-KO mice, exhibited reduced acute itch induced by compound 48/80 and chloroquine(CQ). Application of the TNF-synthesis inhibitor thalidomide and the TNF-αantagonist etanercept dose-dependently suppressed acute itch. Intradermal injection of TNF-α was not sufficient to evoke scratching, but potentiated itch induced by compound 48/80, but not CQ. In addition, compound 48/80 induced TNF-α mRNA expression in the skin, while CQ induced its expression in the dorsal root ganglia(DRG) and spinal cord. Furthermore, chronic itch induced by dry skin was reduced by administration of thalidomide and etanercept and in TNFR1/R2 DKO mice. Dry skin induced TNF-α expression in the skin, DRG, and spinal cord and TNFR1 expression only in the spinal cord. | 2 | Loss of Lkb1 impairs Treg function and stability to aggravate graft-versus-host disease after bone marrow transplantation显示文摘Accumulating evidence suggests that a reduction in the number of Foxp3^(+) regulatory T cells(Tregs)contributes to the pathogenesis of acute graft-versus-host disease(aGVHD),which is a major adverse complication that can occur after allogeneic hematopoietic stem cell transplantation(allo-HSCT).However,the precise features and mechanism underlying the defects in Tregs remain largely unknown.In this study,we demonstrated that Tregs were more dramatically decreased in bone marrow compared with those in peripheral blood from aGVHD patients and that bone marrow Treg defects were negatively associated with hematopoietic reconstitution.Tregs from aGVHD patients exhibited multiple defects,including the instability of Foxp3 expression,especially in response to IL-12,impaired suppressor function,decreased migratory capacity,and increased apoptosis.Transcriptional profiling revealed the downregulation of Lkb1,a previously identified critical regulator of murine Treg identity and metabolism,and murine Lkb1-regulated genes in Tregs from aGVHD patients.Foxp3 expression in human Tregs could be decreased and increased by the knockdown and overexpression of the Lkb1 gene,respectively.Furthermore,a loss-of-function assay in an aGVHD murine model confirmed that Lkb1 deficiency could impair Tregs and aggravate disease severity.These findings reveal that Lkb1 downregulation contributes to multiple defects in Tregs in human aGVHD and highlight the Lkb1-related pathways that could serve as therapeutic targets that may potentially be manipulated to mitigate aGVHD. | Xiuhua Su Qianqian Wang Wei Guo Xiaolei Pei Qing Niu Maolan Liu Yuanyuan Liu Song Chen Sizhou Feng Yi He Donglin Yang Rongli Zhang Qiaoling Ma Weihua Zhai Aiming Pang Jialin Wei Yong Huang Yuechen Luo Mingzhe Han Xiaoming Feng Erlie Jiang | 2020 | Cellular & Molecular Immunology2020,17,5: | 3 |
| 3 | Effects of dry dietary protein on digestibility, nitrogen-balance and growth performance of young male mink显示文摘The experiment was to study the nutrient digestibility and metabolism performance of male minks,which were fed different protein level diets during growth period. Effects of protein quantity on growth and development of minks and feed conversion ratio(FCR) were also investigated. Sixty healthy male minks of 45 d were randomly allocated into six groups with ten replicates, which was one sable for each replicate. The minks in six groups were fed diets in which protein levels were 28, 30,32, 34, 36 and 38%,respectively. The six groups were denoted as P28, P30, P32, P34, P36 and P38. After 2 wk, all minks were weighed, average daily gains(ADG) were calculated, and the digestibility values of nutrients were determined. The results indicated that digestibility of calcium, nitrogen of fence, nitrogen deposition, net protein utilization(NPU), and biological value of protein(BV) were similar(P > 0.05), however, nitrogen intake greatly varied among groups(P < 0.01). Compared with group P28, groups of P34, P36 and P38 showed significant difference(P < 0.01) in ADG and FCR. This paper derives the image signature of the ground moving target after the processing of Range-Doppler (RD) algorithm, the SAR signatures of ground moving targets are analyzed, including the geometry correction, the offsets and defocusing in both range and azimuth direction. Finally, computer simulation results validate its effectiveness. | 6 | Effects of dietary methionine supplementation on growth performance of cubs,nutrient digestibility, nitrogen metabolism and serum biochemical indicators of female blue foxes(Alopex lagopus)显示文摘The objective of the present study was to investigate the effects of methionine(Met) supplementation on growth performance of cubs, nutrient digestibility, nitrogen metabolism and serum biochemical parameters of female blue foxes. One hundred primiparous female blue foxes that were similar in breeding date, pedigree, age, and weight were selected for the trial. The foxes were randomly assigned to four groups(n = 25 each group) and fed diets supplemented with Met at 2(Met2), 4(Met4), 6(Met6)and 8 g/kg(Met8), respectively, for 40 days. Our data showed that body weights at 20 and 40 d were significantly higher in the Met4 group than in the Met2 group(P < 0.05). The Met4 group also had the highest apparent digestibility of dry matter and crude protein compared with either the Met2, Met6, or Met8 group(P< 0.05). The serum Met and isoleucine(Ile) concentrations were significantly higher in the Met4 group than in the Met6 or Met8 group(P < 0.05). In summary, these data indicate that supplementary Met improves growth performance of cubs likely due to increased crude protein and dry matter and increased nitrogen retention of female blue foxes. | 12 | Effects of degenerative autologous nucleus pulposus on the lumbar sympathetic trunk: Sympathetic inflammation in rats显示文摘Objective: We have reported the presence of discogenic visceral pain secondary to anterior herniation of the lumbar disc. The aim of this study was to observe the inflammatory response of the sympathetic trunk to an autologous degenerative nucleus pulposus(NP) injection under fluoroscopy. Methods: A total of 72 rats were used. In 24 rats,the autologous NP suspension was injected into the right sympathetic trunk. Next,food intake and body weight of each rat were monitored for 14 days. Fourteen days after the injection,the right lumbar sympathetic trunk was harvested for histological assessment,and protein levels of IL-1 β,IL-6,and TNF-α were quantified via ELISA. Results: In the NP-treated group,endoneural hyperemia and intensive infiltration of inflammatory cells could be identified in sections of the sympathetic trunk by the hematoxylin and eosin(H&E) stain. Meanwhile,elevated concentrations of IL-1 β,IL-6,and TNF-α were determined in the sympathetic trunk of the NP group as compared to that of the na?ve and control groups,which indicated the development of an inflammatory response. Furthermore,food intake and body weight of rats in the NP group decreased significantly. | 13 | Arc Erosion Behavior of Cu–0.23Be–0.84Co Alloy after Heat Treatment: An Experimental Study显示文摘The arc erosion behavior of Cu–0.23Be–0.84 Co alloy after heat treatment was investigated experimentally by a JF04 C electric contact test system. The arc duration, arc energy, contact resistance and contact pressure of Cu–0.23Be–0.84 Co alloy after solution treatment and aging treatment were analyzed. The arc erosion morphologies were contrastively observed by a three-dimensional measuring system and scanning electron microscopy. For the Cu–0.23Be–0.84 Co alloy in solution state and aging state, the maximum values of arc duration are 90 and 110 ms, and the arc energies are 15,000 and18,000 m J, respectively. The maximum value of the contact resistance of Cu–0.23Be–0.84 Co alloy in different states is about 33 m X. The contact pressure of Cu–0.23Be–0.84 Co alloy in solution state generally changes between 50 and 60 c N during whole make-and-break contacts, while in aging state, it has a larger fluctuation range. Moreover, the quality of moving contact(anode) decreases, while static contact(cathode) increases. The materials transfer from anode to cathode during make-and-break contacts. The total mass losses of Cu–0.23Be–0.84 Co alloy in solution state and aging state are 3and 1.2 mg, respectively. In addition, a number of discrete corrosion pits, molten droplet, porosity and cavity distribute on the surface of moving contact and static contact. The arc erosion model of Cu–0.23Be–0.84 Co alloy in make-and-break contact was built. The arc erosion resistance of Cu–0.23Be–0.84 Co alloy after heat treatment is closely related to the microstructure and the properties of contact materials.
