1UGT1A1 gene polymorphism: Impact on toxicity and efficacy of irinotecan-based regimens in metastatic colorectal cancer显示文摘AIM: To investigate the correlation between uridine diphosphate glucuronosyl transferase 1A1 (UGT1A1) gene polymorphisms and irinotecan-associated side effects and parameters of drug efficacy in patients with metastatic colorectal cancer (mCRC) receiving a low-dose weekly irinotecan chemotherapeutic regimen. METHODS: Genotypes were retrospectively evaluated by gene scan analysis on the ABI 310 sequencer of the TATAA box in the promoter region of the UGT1A1 gene in blood samples from 105 patients who had received 1st line irinotecan-based chemotherapy for mCRC. RESULTS: The distribution of the genotypes was as follows: wild type genotype (WT) (6/6 ) 39.0%, heterozygous genotype (6/7) 49.5%, and homozygous genotype (7/7) 9.5%. The overall response rate (OR) was similar between patients carrying the (6/7, 7/7) or the WT genotype (6/6) (44.3% vs 43.2%, P = 0.75). Neither time to progression [(TTP) 8.1 vs 8.2 mo, P = 0.97] nor overall survival [(OS) 21.2 vs 18.9 mo, P = 0.73] differed significantly in patients who carried the(6/6) when compared to the (6/7, 7/7) genotype. No significant differences in toxicity were observed: Grade 3 and 4 delayed diarrhoea [(6/7, 7/7) vs (6/6); 13.0% vs 6.2%, P =0.08], treatment delays [(6/7, 7/7) vs (6/6); 25.1% vs 19.3%, P = 0.24] or dose reductions [(6/7, 7/7) vs (6/6); 21.5% vs 27.2%, P = 0.07].CONCLUSION: This analysis demonstrates the non-significant influence of the UGT1A1 gene polymorphism on efficacy and rate of irinotecan-associated toxicity in mCRC patients receiving low-dose irinotecan based chemotherapy.Christoph Schulz Volker Heinemann Andreas Schalhorn Nikolas Moosmann Thomas Zwingers Stefan Boeck Clemens Giessen Hans-Joachim Stemmler 2009World Journal of Gastroenterology2009,15,40:31

3Ultrathin monolithic 3D printed optical coherence tomography endoscopy for preclinical and clinical use显示文摘Preclinical and clinical diagnostics increasingly rely on techniques to visualize internal organs at high resolution via endoscopes.Miniaturized endoscopic probes are necessary for imaging small luminal or delicate organs without causing trauma to tissue.However,current fabrication methods limit the imaging performance of highly miniaturized probes,restricting their widespread application.To overcome this limitation,we developed a novel ultrathin probe fabrication technique that utilizes 3D microprinting to reliably create side-facing freeform micro-optics(<130μm diameter)on single-mode fibers.Using this technique,we built a fully functional ultrathin aberration-corrected optical coherence tomography probe.This is the smallest freeform 3D imaging probe yet reported,with a diameter of 0.457 mm,including the catheter sheath.We demonstrated image quality and mechanical flexibility by imaging atherosclerotic human and mouse arteries.The ability to provide microstructural information with the smallest optical coherence tomography catheter opens a gateway for novel minimally invasive applications in disease.Jiawen Li Simon Thiele Bryden C.Quirk Rodney W.Kirk Johan W.Verjans Emma Akers Christina A.Bursill Stephen J.Nicholls Alois M.Herkommer Harald Giessen Robert A.McLaughlin 2020Light(Science & Applications)2020,9,1:6

6Tunable and switchable polarization rotation with non-reciprocal plasmonic thin films at designated wavelengths显示文摘We experimentally demonstrate an ultra-thin plasmonic optical rotator in the visible regime that induces a polarization rotation that is continuously tunable and switchable by an external magnetic field.The rotator is a magneto-plasmonic hybrid structure consisting of a magneto-optical EuSe slab and a one-dimensional plasmonic gold grating.At low temperatures,EuSe possesses a large Verdet constant and exhibits Faraday rotation,which does not saturate over a regime of several Tesla.By combining these properties with plasmonic Faraday rotation enhancement,a large tuning range of the polarization rotation of up to 8.4° for a film thickness of 220 nm is achieved.Furthermore,through experiments and simulations,we demonstrate that the unique dispersion properties of the structure enable us to tailor the wavelengths of the tunable polarization rotation to arbitrary spectral positions within the transparency window of the magneto-optical slab.The demonstrated concept might lead to important,highly integrated,non-reciprocal,photonic devices for light modulation,optical isolation,and magnetic field optical sensing.The simple fabrication of EuSe nanostructures by physical vapor deposition opens the way for many potentially interesting magneto-plasmonic systems and three-dimensional magneto-optical metamaterials.Dominik Floess Jessie Y Chin Akihito Kawatani Daniel Dregely Hanns-Ulrich Habermeier Thomas Weiss Harald Giessen 2015Light(Science & Applications)2015,4,1:4
7Important progress on the use of isotope techniques and methods in catchment hydrology显示文摘The use of isotope techniques and methods in catchment hydrology in the last 50 years has generated two major types of progress: (1) Assessment of the temporal variations of the major stocks and flows of water in catchments, from which the estimation of wa-ter residence times is introduced in this paper. (2) Assessment of catchment hydrologic processes, in which the interactions be-tween different waters, hydrographical separation, and bio-geochemical process are described by using isotopes tracers. Future progress on isotope techniques and methods in hydrology is toward the understanding of the hydrological process in large river basins. 9Indentation of elastically soft and plastically compressible solids显示文摘The effect of soft elasticity, i.e., a relatively small value of the ratio of Young's modulus to yield strength and plastic compressibility on the indentation of isotropically hardening elastic–viscoplastic solids is investigated. Calculations are carried out for indentation of a perfectly sticking rigid sharp indenter into a cylinder modeling indentation of a half space. The material is characterized by a finit strain elastic–viscoplastic constitutive relation that allows for plastic as well as elastic compressibility. Both soft elasticity and plastic compressibility significantl reduce the ratio of nominal indentation hardness to yield strength. A linear relation is found between the nominal indentation hardness and the logarithm of the ratio of Young's modulus to yield strength, but with a different coefficien than reported in previous studies.The nominal indentation hardness decreases rapidly for small deviations from plastic incompressibility and then decreases rather slowly for values of the plastic Poisson's ratio less than0.25. 123D-printed miniature spectrometer for the visible range with a 100×100μm^(2) footprint显示文摘The miniaturisation of spectroscopic measurement devices opens novel information channels for size critical applications such as endoscopy or consumer electronics.Computational spectrometers in the micrometre size range have been demonstrated,however,these are calibration sensitive and based on complex reconstruction algorithms.Herein we present an angle-insensitive 3D-printed miniature spectrometer with a direct separated spatial-spectral response.The spectrometer was fabricated via two-photon direct laser writing combined with a super-fine inkjet process.It has a volume of less than 100×100×300μm^(3).Its tailored and chirped high-frequency grating enables strongly dispersive behaviour.The miniature spectrometer features a wavelength range of 200 nm in the visible range from 490 nm to 690 nm.It has a spectral resolution of 9.2±1.1 nm at 532 nm and 17.8±1.7 nm at a wavelength of 633 nm.Printing this spectrometer directly onto camera sensors is feasible and can be replicated for use as a macro-pixel of a snapshot hyperspectral camera.Andrea Toulouse Johannes Drozella Simon Thiele Harald Giessen Alois Herkommer 2021Light(Advanced Manufacturing)2021,2,1:2
