| 2 | Transfection of mEpo gene to intestinal epithelium in vivo mediated by oral delivery of chitosan-DNA nanoparticles显示文摘AIM: To prepare the chitosan-pmEpo nanoparticles and to study their ability for transcellular and paracellular transport across intestinal epithelia by oral administration. METHODS: ICR mice were fed with recombinant plasmid AAV-tetO-CMV-mEpo (containing mEpo gene) or pCMVβ(containing LacZ gene), whether it was wrapped by chitosan or no. Its size and shape were observed by transmission electron microscopy. Agarose gel electrophoresis was used to assess the efficiency of encapsulation and stability against nuclease digestion. Before and after oral treatmant, blood samples were collected by retro-orbital puncture, and hematocrits were used to show the physiological effect of mEpo. RESULTS: Chitosan was able to successfully wrap the plasmid and to protect it from DNase degradation. Transmission electron microscopy showed that freshly prepared particles were approximately 70-150 nm in size and fairly spherical.Three days after fed the chitosan-pCMVβ complex was fed,the mice were killed and most of the stomach and 30% of the small intestine were stained. Hematocrit was not modified in naive and ‘naked' mEpo-fed mice, a rapid increase of hematocrit was observed during the first 4 days of treatment in chitosan-mEpo-fed animals, reaching 60.9±1.2% (P<0.01),and sustained for a week. The second feed (6 days after the first feed) was still able to promote a second hematocrit increase in chitosan-mEpo-fed animals, reaching 65.9±1.4%(P<0.01), while the second hematocrit increase did not appearin the ‘naked' mEpo-second-fed mice. | 3 | Protective effects of acupuncture and LGNHFD on expressions of vascular endothelial growth factor, basic fibroblast growth factor,and cluster of differentiation 34 in rats with cerebral ischemia-reperfusion injury显示文摘OBJECTIVE: To investigate the protective effects of electroacupuncture and LGNHFD on cerebral ischemia-reperfusion injury, and their effects on the expression of cluster of differentiation 34(CD 34), vascular endothelial growth factor(VEGF), and basic fibroblast growth factor(b FGF) in the peripheral tissue of the cerebral ischemic semi-dark zone in rats.The influence and mechanism of expression.METHODS: Healthy male Sprague-Dawley rats were randomly divided into the sham operation group, model group, Traditional Chinese Medicine (TCM) group, electroacupuncture group, and acupuncture group. The cerebral ischemia model was prepared by middle artery occlusion. The electroacupuncture and TCM groups received electroacupuncture at the 'Neiguan(PC6)', 'Baihui(GV20)', and 'Renzhong(GV26)' acupoints from 4 h after modeling, once daily for 14 d. After modeling,the acupuncture and TCM groups were administered LGNHFD once daily for 14 d. After treatment,the infarct volume and regional cerebral blood flow(r CBF) were measured in each group, the expression of CD34 in the surrounding ischemic penumbra was determined using immunohistochemistry,and the expression of angiogenesis-related factors in the ischemic hemisphere was detected by western blot.RESULTS: Compared with the sham operation group, r CBF was significantly decreased(P < 0.01)and cerebral infarction volume was significantly increased(P < 0.01) in the model group at four time points(5 min and 3, 7, and 14 d after modeling). At three postoperative time points(3, 7, and 14 d), the number of CD34 + cells in the model group were significantly increased(P < 0.01), and the relative expression levels of VEGF, b FGF, and CD34+ in the ischemic hemisphere were significantly increased(P < 0.01, P < 0.05). Compared with the model group, the r CBF of the acupuncture group was significantly increased at 3, 7, and 14 d after intervention(P < 0.01), while the r CBF of the electroacupuncture and TCM groups was significantly increased at 7 and 14 d after intervention(P < 0.01).After 3, 7, and 14 d of intervention, infarct volume was significantly reduced(P < 0.01), the number of CD34 + cells was significantly increased(P < 0.01,P < 0.05), and the relative expression levels of VEGF,b FGF, and CD34 in the ischemic hemisphere were significantly increased(P < 0.01, P < 0.05). Compared with the electroacupuncture and TCM groups, the r CBF of the acupuncture group was significantly increased(P < 0.01), the infarct volume was significantly reduced(P < 0.01), and the relative expression levels of VEGF, b FGF, and CD34 were significantly increased(P < 0.05, P < 0.01) after 3, 7,and 14 d of intervention.CONCLUSION: Acupuncture therapy is superior to simple drug and electroacupuncture therapy for reducing infarct volume, increasing r CBF and CD34 expression in cells, and promoting capillary regeneration in rats with cerebral ischemia. | 4 | Pioglitazone ameliorates neuronal damage after traumatic brain injury via the PPARg/NF-kB/IL-6 signaling pathway显示文摘Traumatic brain injury(TBI)is the major cause of high mortality and disability rates worldwide.Pioglitazone is an activator of peroxisome proliferator-activated receptor-gamma(PPARγ)that can reduce inflammation following TBI.Clinically,neuroinflammation after TBI lacks effective treatment.Although there are many studies on PPARγin TBI animals,only few could be converted into clinical,since TBI mechanisms in humans and animals are not completely consistent.The present study,provided a potential theoretical basis and therapeutic target for neuroinflammation treatment after TBI.First,we detected interleukin-6(IL-6),nitric oxide(NO)and Caspase-3 in TBI clinical specimens,confirming a presence of a high expression of inflammatory factors.Western blot(WB),quantitative real-time PCR(qRTPCR)and immunohistochemistry(IHC)were used to detect PPARγ,IL-6,and p-NF-kB to identify the mechanisms of neuroinflammation.Then,in the rat TBI model,neurobehavioral and cerebral edema levels were investigated after intervention with pioglitazone(PPARγactivator)or T0070907(PPARγinhibitor),and PPARγ,IL-6 and p-NF-kB were detected again by qRT-PCR,WB and immunofluorescence(IF).The obtained results revealed that:1)increased expression of IL-6,NO and Caspase-3 in serum and cerebrospinal fluid in patients after TBI,and decreased PPARγin brain tissue;2)pioglitazone could improve neurobehavioral and reduce brain edema in rats after TBI;3)the protective effect of pioglitazone was achieved by activating PPARγand reducing NF-kB and IL-6.The neuroprotective effect of pioglitazone on TBI was mediated through the PPARγ/NF-kB/IL-6 pathway. | Yongbing Deng Xue Jiang Xiaoyan Deng Hong Chen Jie Xu Zhaosi Zhang Geli Liu Zhu Yong Chengfu Yuan Xiaochuan Sun Changdong Wang | 2020 | Genes & Diseases2020,7,2: | 6 |

| 7 | Toll-like receptor 4 polymorphisms and bacterial infections in patients with cirrhosis and ascites显示文摘AIM To assess the relationship between the presence of toll-like receptor 4(TLR4) polymorphisms and bacterial infections in cirrhotic patients with ascites. METHODS We prospectively included consecutive patients with cirrhosis and ascites hospitalized during a 6-year period. Patients with human immunodeficiency virus(HIV) infection or any other immunodeficiency, patients with advanced hepatocellular carcinoma(beyond Milan's criteria) or any other condition determining poor short-term prognosis, and patients with a permanent urinary catheter were excluded. The presence of D299 G and/or T399 I TLR4 polymorphisms was determined by sequencing and related to the incidence and probability of bacterial infections, other complications of cirrhosis, hepatocellular carcinoma, and mortality during follow-up. A multivariate analysis to identify predictive variables of mortality in the whole series was performed. RESULTS We included 258 patients: 28(10.8%) were carriers of D299G and/or T399I TLR4 polymorphisms(polymorphism group) and 230 patients were not(wildtype group). The probability of developing any bacterial infection at one-year follow-up was 78% in the polymorphism group and 69% in the wild-type group(P = 0.54). The one-year probability of presenting infections caused by gram-negative bacilli(51% vs 44%, P = 0.68), infections caused by gram-positive cocci(49% vs 40%, P = 0.53), and spontaneous bacterial peritonitis(29% vs 34%, respectively, P = 0.99) did not differ between the two groups. The oneyear probability of transplant-free survival was 55% in the polymorphism group and 66% in the wild-type group(P = 0.15). Multivariate analysis confirmed that age, Child-Pugh score, active alcohol intake, previous hepatic encephalopathy, hepatocellular carcinoma and serum creatinine were associated with a higher risk of death during follow-up. | 8 | Spatial and temporal variation characteristics of ocean waves in the South China Sea during the boreal winter显示文摘The spatial and temporal variation characteristics of the waves in the South China Sea(SCS) in the boreal winter during the period of 1979/1980–2011/2012 have been investigated based on the European Centre for Medium-range Weather Forecasts interim(ERA-Interim) reanalysis dataset. The results show that the leading mode of significant wave height anomalies(SWHA) in the SCS exhibits significant interannual variation and a decadal shift around the mid-1990 s, and features a basin-wide pattern in the entire SCS with a center located in the west of the Luzon Strait. The decadal change from a weak regime to a strong regime is mainly associated with the enhancement of winter monsoon modulated by the Pacific decadal oscillation(PDO). The interannual variation of the SWHA has a significant negative correlation with the El Ni?o Southern Oscillation(ENSO) in the same season and the preceding autumn. For a better understanding of the physical mechanism between the SCS ocean waves and ENSO, further investigation is made by analyzing atmospheric circulation. The impact of the ENSO on the SWHA over the SCS is bridged by the East Asian winter monsoon and Pacific-East Asian teleconnection in the lower troposphere.
