| 2 | Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. | 13 | A Study on Super Speed Forming of Metal Sheet by Laser Shock Waves显示文摘Metal sheet plastic deformation or forming is gener at ed through a mechanical pressure or a thermal variation. These pressure variatio ns or thermal variations can be created by a variety of means such as press form ing, hydroforming, imploding detonation and so on. According to the magnitude of the strain rates all these forming methods can be divided into quasi-static fo rming and dynamical forming. Up to now there are no reports of forming methods w ith the strain rates above 10 5sec -1, even though the exploding forming. In this article, we work on a dynamic super-speed forming method driven by lase r shock waves and advanced a novel concept of laser shock forming. The initial o bservation of the laser shock forming is done through a bugle testing with speci mens of SUS430 sheet metal, using a neodymium-glass laser of pulse energy 10J~ 3 0J and duration of 20 ns (FWHM). The investigation revealed that the plastic de formation during the laser shock forming is characterized as ultrahigh strain ra te up to 10 7sec -1. We indicate that plastic deformation increases nonlin early when the energy density of the laser varies. By investigating the hardness and residual stress of the surfaces, we conclude that laser shock forming is a combination technique of laser shock strengthening and metal forming for introdu cing a strain harden and a compressive residual stress on the surface of the wor k-piece, and the treated surface by laser shock forming has good properties in fatigue and corrosion resistance. | 16 | Research on acoustic holography method for identification of sound sources显示文摘This paper presents the method named acoustic holography which can be used to identify noise sources. A new formula of holography reconstruction is obtained, based on the Kirchhoff integral formula. Some simulating tests are carried out using the new formula. The comparison with other reconstruction formulas proves that the new formula is more effective. By using acoustic holography method, some interesting results about the noise of a vehicle are shown. | 17 | Magnetic and Structural Properties in Co/Cu/Co Sandwiches with Ni and Cr Buffer Layers显示文摘The magnetic and structural properties in Co/Cu/Co sandwiches with Ni and Cr buffer layers were investigated. It was found that the coercivity in Ni layer buffered samples decreases with increasing Ni layer thickness, while that in Cr layer buffered ones increases with increasing Cr layer thickness, leading to a large difference in field sensitivity of their giant magnetoresistance (GMR) properties. X-ray diffraction and high resolution transmission electron microscope images exhibited that there is a strong fcc (111) texture in the samples with Ni buffer layer. But there are only randomly oriented polycrystalline grains in Cr buffered sandwiches. According to atomic force microscope topography, the surface roughness of Cr buffered sandwiches is smaller than that of Ni buffered ones.
