1Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. 2Chemical fingerprint analysis of Gentianae Radix et Rhizoma by high-performance liquid chromatography显示文摘Gentianae Radix et Rhizoma(also called 'Longdan' in Chinese)is commonly used for eliminating damp-heat and quenching the fire of liver and gall bladder in traditional Chinese medicine.In this study,a novel and reliable method using high-performance liquid chromatography(HPLC)was developed both for quantitative analysis of four bioactive compounds(loganic acid,swertiamarin,gentiopicroside and sweroside)and chemical fingerprint analysis of 'Longdan'.In quantitative analysis,four compounds showed good regressions(R^(2)>40.9987)within the test ranges and the recovery of the method was in the range 97.61-102.49%.In fingerprint analysis,ten characteristic peaks were selected to evaluate the similarities of the crude drugs,and the HPLC chromatograms of twenty samples from different regions of China showed similar patterns.The results demonstrated that the combination of the quantitative and chromatographic fingerprint analyses offered an efficient way to evaluate the quality consistency of Gentianae Radix et Rhizoma.Baozhong Duan Jianyong Hu Linfang Huang Xianying Yang Fuying Chen 2012Acta Pharmaceutica Sinica B2012,2,1:8
3Excessive gestational weight gain in early pregnancy and insufficient gestational weight gain in middle pregnancy increased risk of gestational diabetes mellitus显示文摘Background:Gestational weight gain(GWG)is associated with the risk of gestational diabetes mellitus(GDM).However,the effect of weight gain in different trimesters on the risk of GDM is unclear.This study aimed to evaluate the effect of GWG on GDM during different trimesters.Methods:A birth cohort study was conducted from 2017 to 2020 in Shenzhen,China.In total,51,205 participants were included comprising two models(early pregnancy model and middle pregnancy model).Gestational weight(kg)was measured at each prenatal clinical visit using a standardized weight scale.Logistic regression analysis was used to assess the risk of GDM.Interaction analysis and mediation effect analysis were performed in the middle pregnancy model.Results:In the early pregnancy model,the risk of GDM was 0.858 times lower(95%confidence interval[CI]:0.786,0.937)with insufficient GWG(iGWG)and 1.201 times higher(95%CI:1.097,1.316)with excessive GWG after adjustment.In the middle pregnancy model,the risk of GDM associated with iGWG increased 1.595 times(95%CI:1.418,1.794)after adjustment;for excessive GWG,no significant difference was found(P=0.223).Interaction analysis showed no interaction between GWG in early pregnancy(GWG-E)and GWG in middle pregnancy(GWG-M)(F=1.268;P=0.280).The mediation effect analysis indicated that GWG-M plays a partial mediating role,with an effect proportion of 14.9%.Conclusions:eGWG-E and iGWG-M are associated with an increased risk of GDM.Strict control of weight gain in early pregnancy is needed,and sufficient nutrition should be provided in middle pregnancy.Aiqi Yin Fuying Tian Xiaoxia Wu Yixuan Chen Kan Liu Jianing Tong Xiaonian Guan Huafan Zhang Linlin Wu Jianmin Niu 2022Chinese Medical Journal2022,,9:5
4Innate immune checkpoint Siglec10 in cancers:mining of comprehensive omics data and validation in patient samples显示文摘Sialic acid binding Ig-like lectin 10(Siglec10)is a member of innate immune checkpoints that inhibits the activation of immune cells through the interaction with its ligand CD24 on tumor cells.Here,by analyzing public databases containing 64517 patients of 33 cancer types,we found that the expression of Siglec10 was altered in 18 types of cancers and was associated with the clinical outcomes of 11 cancer types.In particular,Siglec10 was upregulated in patients with kidney renal clear cell carcinoma(KIRC)and was inversely associated with the prognosis of the patients.In 131 KIRC patients of our settings,Siglec10 was elevated in the tumor tissues of 83(63.4%)patients compared with that in their counterpart normal kidney tissues.Moreover,higher level of Siglec10 was associated with advanced disease(stages III and IV)and worse prognosis.Silencing of CD24 in KIRC cells significantly increased the number of Siglec10-expressing macrophages phagocytosing KIRC cells.In addition,luciferase activity assays suggested that Siglec10 was a potential target of the transcription factors c-FOS and GATA1,which were identified by data mining.These results demonstrate that Siglec10 may have important oncogenic functions in KIRC,and represents a novel target for the development of immunotherapies.Chen Zhang Jiandong Zhang Fan Liang Han Guo Sanhui Gao Fuying Yang Hua Guo Guizhen Wang Wei Wang Guangbiao Zhou 2022Frontiers of Medicine2022,16,4:2
5Bullet-like vanadium-based MOFs as a highly active catalyst for promoting the hydrogen storage property in MgH_(2)显示文摘The practical application of magnesium hydride(MgH_(2))was seriously limited by its high desorption temperature and slow desorp-tion kinetics.In this study,a bullet-like catalyst based on vanadium related MOFs(MOFs-V)was successfully synthesized and doped with MgH_(2) by ball milling to improve its hydrogen storage performance.Microstructure analysis demonstrated that the as-synthesized MOFs was consisted of V_(2)O_(3) with a bullet-like structure.After adding 7wt%MOFs-V,the initial desorption temperature of MgH_(2) was reduced from 340.0 to 190.6℃.Besides,the MgH_(2)+7wt%MOFs-V composite released 6.4wt%H_(2) within 5 min at 300℃.Hydrogen uptake was started at 60℃under 3200 kPa hydrogen pressure for the 7wt%MOFs-V containing sample.The desorption and absorption apparent activity energies of the MgH_(2)+7wt%MOFs-V composite were calculated to be(98.4±2.9)and(30.3±2.1)kJ·mol^(-1),much lower than(157.5±3.3)and(78.2±3.4)kJ·mol^(−1) for the as-prepared MgH_(2).The MgH_(2)+7wt%MOFs-V composite exhibited superior cyclic property.During the 20 cycles isothermal dehydrogenation and hydrogenation experiments,the hydrogen storage capacity stayed almost unchanged.X-ray diffraction(XRD)and X-ray photoelectron spectrometer(XPS)measurements confirmed the presence of metallic vanadium in the MgH_(2)+7wt%MOFs-V composite,which served as catalytic unit to markedly improve the hydrogen storage properties of Mg/MgH_(2) system.Zhiyu Lu Jiahuan He Mengchen Song Yan Zhang Fuying Wu Jiaguang Zheng Liuting Zhang Lixin Chen 2023International Journal of Minerals,Metallurgy and Materials2023,30,1:1

10Recent advances of magnesium hydride as an energy storage material显示文摘Energy storage is the key for large-scale application of renewable energy,however,massive efficient en-ergy storage is very challenging.Magnesium hydride(MgH_(2))offers a wide range of potential applications as an energy carrier due to its advantages of low cost,abundant supplies,and high energy storage capac-ity.However,the practical application of MgH_(2) for energy storage is still impeded by its sluggish kinetics,poor cycling stability,etc.Herein,we provide an overview of recent advances of MgH_(2) for enhancing the hydrogen storage,lithium-ion storage,and heat storage performances.For hydrogen storage,a particular emphasis was put on altering the kinetics and thermodynamics of MgH_(2) via catalyzing,alloying,nano-sizing,and compositing.Modifications to MgH_(2) as a battery anode material mainly focus on the effect of additives,electrolytes,and structure configurations.Prototype heat storage apparatus was proposed based on MgH_(2) and coupled with other materials or systems to improve the heat storage and economic efficiency.Besides,prospects of MgH_(2) in these fields are discussed.This review would stimulate more insightful and pioneering research for the design and preparation of MgH_(2) with excellent energy storage performances.Mengchen Song Liuting Zhang Fuying Wu Haoyu Zhang Hu Zhao Lixin Chen Hong Li 2023Journal of Materials Science & Technology2023,,18:0
11Significant expansion and red-shifting of fluorescent protein chromophore determined through computational design and genetic code expansion显示文摘Fluorescent proteins (FPs)with emission wavelengths in the far-red and infrared regions of the spectrum provide powerful tools for deep-tissue and super-resolution imaging.The development of red-shifted FPs has evoked widespread interest and continuous engineering efforts.In this article, based on a computational design and genetic code expansion,we report a rational approach to significantly expand and red-shift the chromophore of green fluorescent protein (GFP).We applied computational calculations to predict the excitation and emission wavelengths of a FP chromophore harboring unnatural amino acids (UAA)and identify in silico an appropriate UAA,2-amino-3-(6- hydroxynaphthalen-2-yl)propanoic acid (naphthol-Ala).Our methodology allowed us to formulate a GFP variant (cpsfGFP-66-Naphthol-Ala)with red-shifted absorbance and emission spectral maxima exceeding 60 and 130 nm,respectively,compared to those of GFP.The GFP chromophore is formed through autocatalytic post-translational modification to generate a planar 4-(p-hydroxybenzylidene)-5- imidazolinone chromophore.We solved the crystal structure of cpsfGFP-66-naphthol-Ala at 1.3 ■ resolution and demonstrated the formation of a much larger conjugated n-system when the phenol group is replaced by naphthol.These results explain the significant red-shifting of the excitation and emission spectra of cpsfGFP-66-naphthol-Ala.Li Wang Xian Chen Xuzhen GUO liasong Li Qi Liu Fuying Kang Xudong Wang Cheng HU Haiping Liu Weimin Gong Wei Zhuang Xiaohong Liu Jiangyun Wang 2018Biophysics Reports2018,4,5:0

13Metal organic framework supported niobium pentoxide nanoparticles with exceptional catalytic effect on hydrogen storage behavior of MgH_(2)显示文摘Nb_(2)O_(5)nanoparticles with an average particle size of 10 nm supported on a rhombic dodecahedral metal organic framework(MOF)were successfully synthesized by a facile one-pot hydrothermal reaction and subsequent calcination process.Experimental results demonstrated that the prepared catalyst drastically improved the hydrogen storage behavior of MgH_(2).7 wt%Nb_(2)O_(5)@MOF doped MgH_(2)started to desorb hydrogen at 181.9℃and 6.2 wt%hydrogen could be released within 2.6 min and 6.3 min at 275℃and 250℃,respectively.The fully dehydrogenated composite also displayed excellent hydrogenation by decreasing the onset absorption temperature to 25℃and taking up4.9 wt%and 6.5 wt%hydrogen within 6 min at 1750C and 1500C,respectively.Moreover,the corresponding activation energy was calculated to be 75.57±4.16 kJ mol^(-1)for desorption reaction and 51.38±1.09 kJ mol^(-1)for absorption reaction.After 20 cycles,0.5 wt%hydrogen capacity was lost for the MgH_(2)+7 wt%Nb_(2)O_(5)@MOF composite,much lower than 1.5 wt%of the MgH_(2)+7 wt%Nb_(2)O_(5)composite.However,the addition of Nb_(2)O_(5)@MOF had limited effect on reducing the dehydrogenation enthalpy of MgH_(2).Microstructure analysis revealed that Nb_(2)O_(5)particles were uniformly distributed on surface of the MgH_(2)matrix and synergistically improved the hydrogen storage property of MgH_(2)with MOF.Liuting Zhang Farai Michael Nyahuma Haoyu Zhang Changshan Cheng Jiaguang Zheng Fuying Wu Lixin Chen 2023Green Energy & Environment2023,8,2:0
14Co-expression of Clostridium perfringens α and ε Toxins and Their Immunogenicity显示文摘The gene CPA and ETX encoding α and ε toxins were amplified from the standard strain C60-2 of type D Clostridium perfingens by PCR,and then they were inserted into pet-Duet-1 vecto,respectively for the construction of co-expression plasmid p ETDuet-1-CPA-ETX. The co-expression plasmid was transformed into BL21( DE3) competent cells. α and ε toxins proteins were induced by IPTG before analysis by SDS-PAGE. As a result,both of α and ε toxins were detected at 43 and 34 KU by western-blot. Mice immunized with the co-expressed α and ε toxins proteins produced high titers of neutralizing antibodies in the serum,which protected the mice against the attack of type D C. perfringens culture filtrate. In addition,mice immunized with the produced co-expressed α and ε toxins proteins showed significantly higher surviving rate than with ε toxins protein alone when infected with culture filtrate of type D C. perfringens.
