3MiR146a-loaded engineered exosomes released from silk fibroin patch promote diabetic wound healing by targeting IRAK1显示文摘Unhealable diabetic wounds need to be addressed with the help of newer,more efficacious strategies.Exosomes combined with biomaterials for sustained delivery of therapeutic agents are expected to bring new hope for chronic wound treatment.Here,the engineered exosomes modified for efficiently loading miR146a and attaching to silk fibroin patch(SFP)were demonstrated to promote diabetic wound healing.Silk fibroin binding peptide(SFBP)was screened through phage display,and SFBP-Gluc-MS2(SGM)and pac-miR146a-pac fusion protein were constructed.The designed exosomes(SGM-Exos,miR146a-Exos,and SGM-miR146a-Exos)were isolated from the engineered placental mesenchymal stem cells(PMSCs)transduced with SGM or/and pac-miR146a-pac protein.Gluc signals indicated SGM-Exo@SFP markedly increased the binding rate and the stability of SGM-Exo.Moreover,the loading efficiency of miR146a in SGM-miR146a-Exos was ten-fold higher than that in miR146a-Exos.Superior to untreated,SGM-miR146a-Exo-only treated,and SFP-only treated groups,SGM-miR146a-Exo@SFP drived wound healing associated with less inflammation,collagen deposition,and neovascularization.The transcriptomics analysis suggested anti-inflammatory and regenerative effects with SGM-miR146a-Exo@SFP treatment.Here,we show efficient exosome@biomaterial-based miRNA delivery systems for regenerative medicine and tissue engineering.Qiankun Li Wenzhi Hu Qilin Huang Jie Yang Bingmin Li Kui Ma Qian Wei Yaxi Wang Jianlong Su Mengli Sun Shengnan Cui Rungong Yang Haihong Li Xiaobing Fu Cuiping Zhang 2023Signal Transduction and Targeted Therapy2023,8,3:2
4Analysis of Hydrocarbon Mixture Performance of a Dual-Channel Swirl Engine显示文摘In order to understand and improve the oil and gas mixing performance of a dual-channel vortex chamber diesel engine,the BH175F dual-channel vortex chamber combustion system was used as the research foundation,and the oil-gas mixture process of the combustion system was numerically analyzed.By analyzing the cylinder temperature,cylinder pressure,mixing process and combustion process of the combustion system,the mixture performance of the combustion system was studied.Results indicated that:The mixture of the compression Top Dead Center(TDC)started to enter the main combustion chamber through the start-up hole;when the piston reached 4°After Top Dead Center(ATDC),the mixture started to enter the main combustion chamber through the connecting channels A and B,and the high-concentration mixture entered the main combustion from the start-up hole;when the piston continued running down to 20°and 25°ATDC,it could be seen that the main combustion chamber mixture was already relatively uniform;besides,when the equivalence ratio was between 0.8 and 1,the air-ftiel mixture was unevenly distributed in the main combustion chamber.It provides guidance for further improvement of the combustion system.YUAN Wenhua HUANG Qilin FU Jun LIAO Jingjing LI Yu HE Yong ZHANG Zengfeng MA Yi 2020Journal of Thermal Science2020,29,6:1
5Sericin microparticles enveloped with metal-organic networks as a pulmonary targeting delivery system for intra-tracheally treating metastatic lung cancer显示文摘Chemotherapy is one of the major approaches for the treatment of metastatic lung cancer.However,systemic chemotherapy is limited by poor therapeutic efficiency and severe toxic side effects,due to the extremely low delivery efficacy and non-specificity of anticancer drugs.Herein,we report a sericin microparticles enveloped with metal-organic networks as a pulmonary delivery system for treating lung metastasis of breast cancer in an animal model.The sericin microparticles(SMPs)were prepared using water in oil(w/o)emulsification method.After doxorubicin(DOX)loading,tannic acid(TA)/ferric irons(Fe3+)based metal organic networks(MON)were coated on the particles to obtain DOX-loaded microparticles(DOX@SMPs-MON).The SMPs-MON with good biocompatibility could effectively encapsulate DOX and sustainably unload cargos in a pH-dependent manner.The DOX-loaded microparticles could be uptaken by 4T1 cells,and effectively kill the cancer cells.In vivo,DOX@SMPs-MON was deposited in the lungs and remained for over 5 days after pulmonary administration.In contrast to conventional DOX treatment that did not show significantly inhibitory effects on lung metastatic tumor,DOX@SMPs-MON markedly decreased the number and size of metastatic nodules in lungs,and the lung weight and appearance were similar to those of healthy mice.In summary,the sericin microparticles with MON wrapping might be a promising pulmonary delivery system for treating lung metastatic cancer.Jia Liu Yan Deng Daan Fu Ye Yuan Qilin Li Lin Shi Guobin Wang Zheng Wang Lin Wang 2021Bioactive Materials2021,6,1:1

7Novel neutrophil extracellular trap-related mechanisms in diabetic wounds inspire a promising treatment strategy with hypoxia-challenged small extracellular vesicles显示文摘Neutrophil extracellular traps(NETs)have been considered a significant unfavorable factor for wound healing in diabetes,but the mechanisms remain unclear.The therapeutic application of small extracellular vesicles(sEVs)derived from mesenchymal stem cells(MSCs)has received considerable attention for their properties.Hypoxic preconditioning is reported to enhance the therapeutic potential of MSC-derived sEVs in regenerative medicine.Therefore,the aim of this study is to illustrate the detailed mechanism of NETs in impairment of diabetic wound healing and develop a promising NET-targeting treatment based on hypoxic pretreated MSC-derived sEVs(Hypo-sEVs).Excessive NETs were found in diabetic wounds and in high glucose(HG)-induced neutrophils.Further research showed that high concentration of NETs impaired the function of fibroblasts through activating endoplasmic reticulum(ER)stress.Hypo-sEVs efficiently promoted diabetic wound healing and reduced the excessive NET formation by transferring miR-17-5p.Bioinformatic analysis and RNA interference experiment revealed that miR-17-5p in Hypo-sEVs obstructed the NET formation by targeting TLR4/ROS/MAPK pathway.Additionally,miR-17-5p overexpression decreased NET formation and overcame NET-induced impairment in fibroblasts,similar to the effects of Hypo-sEVs.Overall,we identify a previously unrecognized NET-related mechanism in diabetic wounds and provide a promising NET-targeting strategy for wound treatment.Ziqiang Chu Qilin Huang Kui Ma Xi Liu Wenhua Zhang Shengnan Cui Qian Wei Huanhuan Gao Wenzhi Hu Zihao Wang Sheng Meng Lige Tian Haihong Li Xiaobing Fu Cuiping Zhang 2023Bioactive Materials2023,,9:1

13A review on treatment of disinfection byproduct precursors by biological activated carbon process显示文摘Disinfection by-products(DBPs)in water systems have attracted increasing attention due to their toxic effects.Removal of precursors(mainly natural organic matter(NOM))prior to the disinfection process has been recognized as the ideal strategy to control the DBP levels.Currently,biological activated carbon(BAC)process is a highly recommended and prevalent process for treatment of DBP precursors in advanced water treatment.This paper first introduces the fundamental knowledge of BAC process,including the history,basic principles,typical process flow,and basic operational parameters.Then,the selection of BAC process for treatment of DBP precursors is explained in detail based on the comparative analysis of dominant water treatment technologies from the aspects of mechanisms for NOM removal as well as the treatability of different groups of DBP precursors.Next,a thorough overview is presented to summarize the recent developments and breakthroughs in the removal of DBP precursors using BAC process,and the contents involved include effect of pre-BAC ozonation,removal performance of various DBP precursors,toxicity risk reduction,fractional analysis of NOM,effect of empty bed contact time(EBCT)and engineered biofiltration.Finally,some recommendations are made to strengthen current research and address the knowledge gaps,including the issues of microbial mechanisms,toxicity evaluation,degradation kinetics and microbial products.Jie Fu Ching-Hua Huang Chenyuan Dang Qilin Wang 2022Chinese Chemical Letters2022,33,10:0
