1Distribution of chlorpyrifos in rice paddy environment and its potential dietary risk显示文摘Chlorpyrifos is one of the most extensively used insecticides in China. The distribution and residues of chlorpyrifos in a paddy environment were characterized under field and laboratory conditions. The half-lives of chlorpyrifos in the two conditions were 0.9–3.8 days(field) and 2.8–10.3 days(laboratory), respectively. The initial distribution of chlorpyrifos followed the increasing order of water < straw < soil, and soil was characterized as the major absorber. The ultimate residues in rice grain were below the maximum residue limit(MRL) with a harvest interval of 14 days. The chronic exposure for chlorpyrifos was rather low compared to the acceptable daily intake(ADI = 0.01 mg/kg bw) due to rice consumption. The chronic exposure risk from chlorpyrifos in rice grain was 5.90% and 1.30% ADI from field and laboratory results respectively. Concerning the acute dietary exposure,intake estimated for the highest chlorpyrifos level did not exceed the acute reference dose(ARf D = 0.1 mg/kg bw). The estimated short-term intakes(ESTIs) were 0.78% and 0.25% of the ARf D for chlorpyrifos. 2Impacts of land conversion and management measures on net primary productivity in semi-arid grassland显示文摘Ecological restoration measures implemented in China have profoundly impacted vegetation NPP.This study aimed to estimate the effects of the land conversion and management measures on the grassland ecosystem in semi-arid regions.Land use data were employed from 2000 to 2015 to compare land conversion and coverage changes in Xilingol grassland.Then,the contributions of land conversion and management policies were quantified by assessing the difference between actual NPP and climate-induced NPP changes.The results indicated that the grassland area had a net loss of 534.42 km^(2),and the net area of increased vegetation coverage was 74,683.05 km^(2).Furthermore,the total NPP increased by 8,010.73 Gg C·yr^(−1)(1 Gg=10^(9)g),of which the human activities,including grazing management measures(+6,809.40 Gg C·yr^(−1))and land conversion(45.72 Gg C·yr^(−1))contributed to 85.58%of the increase in NPP.Transformation from desert and farmland dominated grassland expansion and NPP increase,while urbanization and desertification caused large grassland reduction and NPP loss.The grazing management increased vegetation NPP in most regions except for some regions in the desert steppe and the farming-pastoral zone.Related policies should be further adjusted to strengthen the management of the desert steppe and farming-pastoral regions.Feifei Cao Jiaxun Li Xiao Fu Gang Wu 2020Ecosystem Health and Sustainability2020,6,1:4

6Fotemustine-based therapy in combination with rituximab as a first-line induction chemotherapy followed by WBRT for newly diagnosed primary central nervous system lymphoma: a prospective phase II trial显示文摘Objective:This study aimed to evaluate the safety,efficacy,and feasibility of the rituximab,fotemustine,pemetrexed,and dexamethasone(R-FPD)regimen followed by whole-brain radiotherapy(WBRT)for patients with primary central nervous system lymphoma(PCNSL).Methods:A prospective,single-center phase II clinical trial was conducted.Patients with PCNSL newly diagnosed at the First Affiliated Hospital of Zhengzhou University between July 2018 and July 2020 were studied.The R-FPD regimen consisted of rituximab(375 mg/m2 i.v.on D0),fotemustine(100 mg/m2 i.v.on D1),pemetrexed(600 mg/m2 i.v.on D1),and dexamethasone(40 mg i.v.on D1-5).Patients 60 years or younger who showed a complete response(CR)were treated with 23.4 Gy of WBRT after the end of chemotherapy;those older than 60 years with CR were treated with a wait-and-see approach;and those who did not show CR after the 4th cycle of chemotherapy were given salvage WBRT 30 Gy+local tumor field irradiation up to 45 Gy,regardless of age.Results:A total of 30 patients were included.After 2 cycles,the objective response rate(ORR)was 96.5%(28/29,1 CR,27 PR,0 SD,and 1 PD).After 4 cycles,the ORR was 73.1%(19/26,11 CR,8 PR,4 SD,and 3 PD).After WBRT,the ORR was 90.9%(10/11,7 CR,3 PR,and 1 SD).The grade III and IV toxicity responses were mainly leukopenia(20.0%),thrombocytopenia(23.3%),and anemia(10.0%).Conclusions:Fotemustine-based therapy in combination with rituximab chemotherapy followed by WBRT can improve outcomes,providing ORR benefits and favorable tolerability in patients newly diagnosed with PCNSL.Jingjing Wu Fenghua Gao Wenhua Wang Xudong Zhang Meng Dong Lei Zhang Xin Li Ling Li Zhenchang Sun Xinhua Wang Xiaorui Fu Linan Zhu Mengjie Ding Songtao Niu Zhaoming Li Yu Chang Feifei Nan Jiaqian Yan Hui Yu Xiaolong Wu Zhiyuan Zhou Jieming Zhang Mingzhi Zhang 2022Cancer Biology & Medicine2022,19,7:1

12SENP5 deteriorates traumatic brain injury via SUMO2- dependent suppression of E2F1 SUMOylation显示文摘Traumatic brain injury(TBI)represents a main public health concern during the past decade,attracting considerable interest because of its rising prevalence,wide-ranging risk factors and lifelong familial and societal influence.SUMO2 can conjugate to substrates upon various cellular stresses.Nevertheless,whether and how SUMO2-specific proteases partake in TBI is less understood.The aim of this study is to dissect the effects of SUMO-specific peptidase 5(SENP5)on accentuating TBI in rats in an effort to unveil its underlying mechanism.SENP5 is overexpressed in hippocampal tissues of TBI rats,and inhibition of SENP5 reduces neurological function scores,decreases brain water content,inhibits apoptosis in hippocampal tissues,and attenuates brain injury caused in rats.Moreover,SENP5 inhibits the SUMOylation level of E2F transcription factor 1(E2F1)and increases the protein expression of E2F1.Silencing of E2F1 blocks the p53 signaling pathway.Overexpression of E2F1 partially reverses the protective effect of sh-SENP5 on TBI in rats.These findings reveal an essential role of SENP5 and the SUMOylation status of E2F1 in the TBI development.Yanfu Ding Feifei Chen Weitao Yang Xiaobin Fu Yuanyuan Xie 2023Acta Biochimica et Biophysica Sinica2023,55,8:0
13Negative refraction mediated by bound states in the continuum显示文摘Negative refraction might occur at the interface between a two-dimensional photonic crystal(Ph C)slab and a homogeneous medium,where the guiding of the electromagnetic wave along the third dimension is governed by total internal reflection.Herein,we report on the observation of negative refraction in the Ph C slab where the vertical guiding is enabled by a bound state in the continuum and essentially beyond the light cone.Such abnormal refraction and guiding mechanism are based on the synchronous crafting of spatial dispersion and the radiative lifetime of Bloch modes within the radiative continuum.Microwave experiments are provided to further validate the numerical proposal in an all-dielectric Ph C platform.It is envisioned that the negative refraction observed beyond the light cone might facilitate the development of optical devices in integrated optics,such as couplers,multiplexers,and demultiplexers.ZHANYUAN ZHANG FEIFEI QIN YI XU SONGNIAN FU YUNCAI WANG YUWEN QIN 2021Photonics Research2021,9,8:0
14SYP-5 regulates meiotic thermotolerance in Caenorhabditis elegans显示文摘Meiosis produces the haploid gametes required by all sexually reproducing organisms,occurring in specific temperature ranges in different organisms.However,how meiotic thermotolerance is regulated remains largely unknown.Using the model organism Caenorhabditis elegans,here,we identified the synaptonemal complex(SC)protein SYP-5 as a critical regulator of meiotic thermotolerance.syp-5-null mutants maintained a high percentage of viable progeny at 20℃ but produced significantly fewer viable progeny at 25℃,a permissive temperature in wild-type worms.Cytological analysis of meiotic events in the mutants revealed that while SC assembly and disassembly,as well as DNA double-strand break repair kinetics,were not affected by the elevated temperature,crossover designation,and bivalent formation were significantly affected.More severe homolog segregation errors were also observed at elevated temperature.A temperature switching assay revealed that late meiotic prophase events were not temperature-sensitive and that meiotic defects during pachytene stage were responsible for the reduced viability of syp-5 mutants at the elevated temperature.Moreover,SC polycomplex formation and hexanediol sensitivity analysis suggested that SYP-5 was required for the normal properties of the SC,and charge-interacting elements in SC components were involved in regulating meiotic thermotolerance.Together,these findings provide a novel molecular mechanism for meiotic thermotolerance regulation.Yuanyuan Liu Qiuchen Zhao Hui Nie Fengguo Zhang Tingting Fu Zhenguo Zhang Feifei Qi Ruoxi Wang Jun Zhou Jinmin Gao 2021Journal of Molecular Cell Biology2021,13,9:0
15HBXIP blocks myosin-ⅡA assembly by phosphorylating and interacting with NMHC-ⅡA in breast cancer metastasis显示文摘Tumor metastasis depends on the dynamic balance of the actomyosin cytoskeleton.As a key component of actomyosin filaments,non-muscle myosin-ⅡA disassembly contributes to tumor cell spreading and migration.However,its regulatory mechanism in tumor migration and invasion is poorly understood.Here,we found that oncoprotein hepatitis B X-interacting protein(HBXIP) blocked the myosin-ⅡA assemble state promoting breast cancer cell migration.Mechanistically,mass spectrometry analysis,co-immunoprecipitation assay and GST-pull down assay proved that HBXIP directly interacted with the assembly-competent domain(ACD) of non-muscle heavy chain myosin-ⅡA(NMHC-ⅡA).The interaction was enhanced by NMHC-ⅡA S1916 phosphorylation via HBXIP-recruited protein kinase PKCβⅡ.Moreover,HBXIP induced the transcription of PRKCB,encoding PKCβⅡ,by coactivating Sp1,and triggered PKCβⅡ kinase activity.Interestingly,RNA sequencing and mouse metastasis model indicated that the anti-hyperlipidemic drug bezafibrate(BZF) suppressed breast cancer metastasis via inhibiting PKCβⅡ-mediated NMHC-ⅡA phosphorylation in vitro and in vivo.We reveal a novel mechanism by which HBXIP promotes myosin-ⅡA disassembly via interacting and phosphorylating NMHC-ⅡA,and BZF can serve as an effective anti-metastatic drug in breast cancer.Lu Zhang Xiaolei Zhou Bowen Liu Xuhe Shi Xianmeng Li Feifei Xu Xueli Fu Xue Wang Kai Ye Tianzhi Jin Huimin Sun Qianqian Li Weiying Zhang Lihong Ye 2023Acta Pharmaceutica Sinica B2023,13,3:0

17cGAS regulates the DNA damage response to maintain proliferative signaling in gastric cancer cells显示文摘The activation of some oncogenes promote cancer cell proliferation and growth,facilitate cancer progression and metastasis by induce DNA replication stress,even genome instability.Activation of the cyclic GMP-AMP synthase(cGAS)mediates classical DNA sensing,is involved in genome instability,and is linked to various tumor development or therapy.However,the function of cGAS in gastric cancer remains elusive.In this study,the TCGA database and retrospective immunohistochemical analyses revealed substantially high cGAS expression in gastric cancer tissues and cell lines.By employing cGAS high-expression gastric cancer cell lines,including AGS and MKN45,ectopic silencing of cGAS caused a significant reduction in the proliferation of the cells,tumor growth,and mass in xenograft mice.Mechanistically,database analysis predicted a possible involvement of cGAS in the DNA damage response(DDR),further data through cells revealed protein interactions of the cGAS and MRE11-RAD50-NBN(MRN)complex,which activated cell cycle checkpoints,even increased genome instability in gastric cancer cells,thereby contributing to gastric cancer progression and sensitivity to treatment with DNA damaging agents.Furthermore,the upregulation of cGAS significantly exacerbated the prognosis of gastric cancer patients while improving radiotherapeutic outcomes.Therefore,we concluded that cGAS is involved in gastric cancer progression by fueling genome instability,implying that intervening in the cGAS pathway could be a practicable therapeutic approach for gastric cancer.BIN LIU HAIPENG LIU FEIFEI REN HANGFAN LIU IHTISHAM BUKHARI YUMING FU WANQINGWU MINGHAI ZHAO SHAOGONG ZHU HUI MO FAZHAN LI MICHAEL B.ZHENG YOUCAI TANG PENGYUAN ZHENG YANG MI 2021Oncology Research2021,29,2:0
18Hierarchically Structured Nb_(2)O_5 Microflowers with Enhanced Capacity and Fast-Charging Capability for Flexible Planar Sodium Ion Micro-Supercapacitors显示文摘Planar Na ion micro-supercapacitors(NIMSCs) that offer both high energy density and power density are deemed to a promising class of miniaturized power sources for wearable and portable microelectron-ics. Nevertheless, the development of NIMSCs are hugely impeded by the low capacity and sluggish Na ion kinetics in the negative electrode.Herein, we demonstrate a novel carbon-coated Nb_(2)O_5 microflower with a hierarchical structure composed of vertically intercrossed and porous nanosheets, boosting Na ion storage performance. The unique structural merits, including uniform carbon coating, ultrathin nanosheets and abun-dant pores, endow the Nb_(2)O_5 microflower with highly reversible Na ion storage capacity of 245 mAh g^(-1) at 0.25 C and excellent rate capability.Benefiting from high capacity and fast charging of Nb_(2)O_5 microflower, the planar NIMSCs consisted of Nb_(2)O_5 negative electrode and activated car-bon positive electrode deliver high areal energy density of 60.7 μWh cm^(-2),considerable voltage window of 3.5 V and extraordinary cyclability.
