1Evolutionary conservation of Dmrt gene family in amphibi-ans, reptiles and birds显示文摘Sex determining gene Mab-3 of C. elegans and doublesex of Drosophila contain a common DNA binding motif called a DM domain, both of which regulate similar aspects of sexual development. Human doublesex-related gene DMRT1 has been identified, which also contains the conserved DM-related DNA-binding domain and plays an essential role in gonadal differentiation. We present the amplification of a broad spectrum of DM domain sequences from phylogenetic diverse vertebrates (Cynops orientalis, Chrysemys scripta elegans and Coturnix coturnix) using degenerate PCR. 3Non-fragile switching tracking control for a flexible air-breathing hypersonic vehicle based on polytopic LPV model显示文摘This article proposes a linear parameter varying (LPV) switching tracking control scheme for a flexible air-breathing hypersonic vehicle (FAHV). First, a polytopic LPV model is constructed to represent the complex nonlinear longitudinal model of the FAHV by using Jacobian linearization and tensor-product (T-P) model transformation approach. Second, for less conservative controller design purpose, the flight envelope is divided into four sub-regions and a non-fragile LPV controller is designed for each parameter sub-region. These non-fragile LPV controllers are then switched in order to guarantee the closed-loop FAHV system to be asymptotically stable and satisfy a specified performance criterion. The desired non-fragile LPV switching controller is found by solving a convex constraint problem which can be efficiently solved using available linear matrix inequality (LMI) techniques, and robust stability analysis of the closed-loop FAHV system is verified based on multiple Lypapunov functions (MLFs). 4Integrative lipidomic and transcriptomic study unravels the therapeutic effects of saikosaponins A and D on non-alcoholic fatty liver disease显示文摘Nonalcoholic fatty liver disease(NAFLD)has become one of the most prominent causes of chronic liver diseases and malignancies.However,few therapy has been approved.Radix Bupleuri(RB)is the most frequently used herbal medicine for the treatment of liver diseases.In the current study,we aim to systemically evaluate the therapeutic effects of saikosaponin A(SSa)and saikosaponin D(SSd),the major bioactive monomers in RB,against NAFLD and to investigate the underlying mechanisms.Our results demonstrated that both SSa and SSd improved diet-induced NAFLD.Integrative lipidomic and transcriptomic analysis revealed that SSa and SSd modulated glycerolipid metabolism by regulating related genes,like Lipe and Lipg.SSd profoundly suppressed the fatty acid biosynthesis by downregulating Fasn and Acaca expression and promoted fatty acid degradation by inducing Acox1 and Cpt1 a expression.Bioinformatic analysis further predicted the implication of master transcription factors,including peroxisome proliferator-activated receptor alpha(PPARα),in the protective effects of SSa and SSd.These results were further confirmed in vitro in mouse primary hepatocytes.In summary,our study uncoded the complicated mechanisms underlying the promising anti-steatosis activities of saikosaponins(SSs),and provided critical evidence inspiring the discovery of innovative therapies based on SSa and SSd for the treatment of NAFLD and related complications.Xiaojiaoyang Li Junde Ge Yajing Li Yajie Cai Qi Zheng Nana Huang Yiqing Gu Qi Han Yunqian Li Rong Sun Runping Liu 2021Acta Pharmaceutica Sinica B2021,11,11:15
5Association of KIR Genotypes and Haplotypes with Susceptibility to Chronic Hepatitis B Virus Infection in Chinese Han Population显示文摘Killer immunoglobulin-like receptor (KIR) genes can regulate the activation of NK and T cells upon interaction with HLA class I molecules. Hepatitis B virus (HBV) infection has been regarded as a multi-factorial disorder disease. Previous studies revealed that KIRs were involved in HCV and HIV infection or clearance. The aim of this study was to explore the possibility of the inheritance of KIR genotypes and haplotypes as a candidate for susceptibility to persistent HBV infection or HBV clearance. The sequence specific primer polymerase chain reaction (SSP-PCR) was employed to identify the KIR genes and pseudogenes in 150 chronic hepatitis B (CHB) patients, 251 spontaneously recovered (SR) controls, and 412 healthy controls. The frequencies of genotype G, M, FZ1 increased in CHB patients compared with healthy control subjects. The frequency of genotype AH was higher in SR controls than that in both CHB patients and healthy controls. The carriage frequencies of genotype G and AH were higher; while, the frequencies of AF and AJ were lower in SR controls than those in healthy control subjects. The frequency of A haplotype was lower, whereas, the frequency of B haplotype was higher in CHB patients and SR controls than those in healthy controls. In healthy controls, haplotype 4 was found lower compared with that in CHB patients and SR controls and the frequency of haplotype 5 was higher in SR controls than that in other two groups. Based on these findings, it seems that the genotypes M and FZ1 are HBV susceptive genotypes; AH, on the other hand, may be protective genotypes that facilitate the clearance of HBV. 7Polytopic LPV modeling and gain-scheduled switching control for a flexible air-breathing hypersonic vehicle显示文摘A novel gain-scheduled switching control method for the longitudinal motion of a flexible air-breathing hypersonic vehicle (FAHV) is proposed.Firstly,velocity and altitude are selected as scheduling variables,a polytopic linear parameter varying(LPV) model is developed to represent the complex nonlinear longitudinal dynamics of the FAHV.Secondly,based on the obtained polytopic LPV model,the flight envelope is divided into four smaller subregions, and four gain-scheduled controllers are designed for these parameter subregions.Then,by the defined switching characteristic function,these gain-scheduled controllers are switched in order to guarantee the closed-loop FAHV system to be asymptotically stable and satisfy a given tracking error performance criterion.The condition of gain-scheduled switching controller synthesis is given in terms of linear matrix inequalities(LMIs) which can be easily solved by using standard software packages.Finally,simulation results show the effectiveness of the presented method.Yiqing Huang Changyin Sun Chengshan Qian Jingmei Zhang Li Wang 2013Journal of Systems Engineering and Electronics2013,24,1:10
8Primarily screening and analyzing ESTs differentially expressed in rats' primary liver cancer显示文摘Objective: To screen and analyze key express sequence tags (ESTs) which were differentially displayed in every period of SD rats' primary hepatic carcinoma and reveal the molecular mechanism of carcinogenesis. Methods: Using diethylnitrosamine (DENA) as a cancerigenic agent, animal models with different phases of primary hepatic cancer were constructed in SD rats. Rats were respectively sacrificed at d 14, d 28, d 56, d 77, d 105 and d 112 after the rats received DENA by gavage, then the livers were harvested. One part of the livers was classified according to their pathological changes, while the other was reserved for molecular mechanism studies on hepatocarcinogenesis. The differentially expressed genes were isolated from both normal and morbid tissues by mRNA differential display technique (DDRT-PCR). After the fragments were sequenced, bioinformatics were used to analyze the results. Results: Twelve differentially expressed cDNA fragments were obtained. Nine fragments had the homology with known cDNA clones, especially EST-7 was similar to BN/SsNHsdMCW mitochondrion gene and the identity was 100% which suggested EST-7 may be the part of BN/SsNHsdMCW mitochondrion gene. In contrast, other three fragments (EST-1, EST-3 and EST-5) had extremely low identity to any genes registered in GENBANK databases. Conclusions: BN/SsNHsdMCW mitochondrion gene was expressed in different periods of hepatocarcinogenesis. 11Distributed Brillouin frequency shift extraction via a convolutional neural network显示文摘Distributed optical fiber Brillouin sensors detect the temperature and strain along a fiber according to the local Brillouin frequency shift(BFS),which is usually calculated by the measured Brillouin spectrum using Lorentzian curve fitting.In addition,cross-correlation,principal component analysis,and machine learning methods have been proposed for the more efficient extraction of BFS.However,existing methods only process the Brillouin spectrum individually,ignoring the correlation in the time domain,indicating that there is still room for improvement.Here,we propose and experimentally demonstrate a BFS extraction convolutional neural network(BFSCNN)to retrieve the distributed BFS directly from the measured two-dimensional data.Simulated ideal Brillouin spectra with various parameters are used to train the BFSCNN.Both the simulation and experimental results show that the extraction accuracy of the BFSCNN is better than that of the traditional curve fitting algorithm with a much shorter processing time.The BFSCNN has good universality and robustness and can effectively improve the performances of existing Brillouin sensors.YIQING CHANG HAO WU CAN ZHAO LI SHEN SONGNIAN FU MING TANG 2020Photonics Research2020,8,5:6
12Thrombolysis with alteplase 3-4.5 hours after acute ischaemic stroke:the first multicentre,phase III trial in China显示文摘background and purpose Data on the efficacy and safety of alteplase for acute ischaemic stroke(AIS)administered 3-4.5 hours after the onset of stroke symptoms in Chinese patients is limited.We sought to determine whether AIS patients would benefit from thrombolysis with alteplase between 3 and 4.5 hours after the onset of stroke symptoms in a prospective,multicentre,single-arm trial in China.Materials and methods Eligible AIS patients were given 0.9 mg/kg alteplase intravenously.The primary efficacy endpoint was a favourable outcome at 3 months,defined as a score of 0 or 1 on the modified Rankin Scale.Thresholds for the primary efficacy endpoint were determined to be 40%based on the literature review.The primary safety endpoint was symptomatic intracranial haemorrhage(sICH)according to the European Cooperative Acute Stroke Study III(ECASS III)trial definition.Post hoc analysis between this study and the ECASS III trial were compared using the propensity score matching(PSM)method.results A total of 120 eligible AIS patients from 11 sites in China received thrombolysis therapy in this study.The median time from onset of symptoms to needle was 3 hours 54 min.The percentage of patients with a favourable outcome was 63.3%(95%CI 54.4 to 71.4),significantly higher than the predefined threshold(p<0.0001).Three patients(2.5%,95%CI 0.5 to 7.1)had sICH,including two fatal sICH.Six patients died within 3 months after treatment.The post hoc PSM analysis showed a numerically higher rate of the primary efficacy endpoint in this study(63.3%)than the matched placebo arm(56.7%)in the ECASS III trial.Conclusions Intravenous alteplase with a standard dose administered between 3 and 4.5 hours after onset of symptoms is effective and safe for Chinese AIS patients.Huaguang Zheng Yi Yang Huisheng Chen Chuanling Li Yangkun Chen Fu-Dong Shi Li Yang Xiaoping Cui Zuneng Lu Yanling Liang Songbiao Cui Anding Xu Yiqing Wu Yaqing Sun Yongjun Wang 2020Stroke & Vascular Neurology2020,5,3:4
13Virtual and augmented reality technologies for emergency management in the built environments: A state-of-the-art review显示文摘With the rapid technological advancements in recent decades,virtual reality(VR)and augmented reality(AR)technologies have been increasingly adopted to address various challenges in emergency management in the built environments.This paper presents a review of state-of-the-art applications in this rapidly evolving area.A total of 84 relevant articles are identified based on searching in the Web of Science Core Collection and snowballing.These papers are then organized based on a taxonomy developed in this study.Next,a range of VR/AR appli-cations presented in these papers that are aimed to enhance various processes associated with pre-emergency preparedness,responses during emergency and post-emergency recovery are reviewed in detail.The existing VR/AR applications are also described from a human-computer interaction perspective.Finally,current research trends,knowledge gaps and directions for future research are discussed.The findings presented in this paper are expected to provide a synthetic and critical review of state-of-the-art VR/AR applications for emergency management in the built environment and facilitate further advancements in both research and practice in this area.Yiqing Zhu Nan Li 2021Journal of Safety Science and Resilience2021,2,1:4

15into the Role of Additives in Catalytic Synthesis of Insight Cyclohexylamine from Nitrobenzene显示文摘Cyclohexylamine is a versatile intermediate in various chemical industries,which can be expediently synthesized via hydrogenation of aniline or nitrobenzene.However,such processes always suffer from side reactions.Many reports have found that specific additives can suppress the formation of side products,but the mechanism is still not clear.In this work,results suggest that it is the hydroxide ion of alkali metal hydroxides,rather than the cations,which plays a key role in suppressing the side reactions on supported Ru-based catalysts.With the assistance of LiOH,the selectivity toward cy- clohexylamine increased from 85.4% to 100%.Side products,such as dicyciohexylamine,cyclohexanol and N-isopropyl cyclohexylamine,could no longer be detected.Theoretical calculations further disclosed that addition of alkali metal hydroxides inhibited the dissociation of enamine and decreased the adsorption energy of cyclohexylamine,which might be the reasons for a better selectivity.However,the addition of alkali metal hydroxides reduced the activity of nitrobenzene hydrogenation by unfolding the condensation reaction route.To recover or even further enhance the catalytic performance,a second metal component was introduced and the resultant RuNi/AC exhibited a significant improvement in activity compared with Ru/AC.Xuefeng Li Zhe Wang Shanjun Mao Yiqing chen Minghui Tang Haoran Li Yong Wang 2018Chinese Journal of Chemistry2018,36,12:4
16Heteroatom-doped porous carbon from methyl orange dye wastewater for oxygen reduction显示文摘Banana peel-derived porous carbon(BPPC) was prepared from banana peel and used as an adsorbent for methyl orange(MO) wastewater removal. BPPC-MO50 is a N,S-doped BPPC obtained via secondary carbonization. The BPPC-MO50 exhibited a high specific surface area of1774.3 m^2/g. Heteroatom-doped porous carbon(PC) was successfully synthesized from the BPPC absorbed MO at high temperature and used for oxygen reduction. The BPPC-MO50 displayed the highest ORR onset potential among all carbon-based electrocatalysts, i.e., 0.93 V vs.reversible hydrogen electrode(RHE). This is the first report to describe porous carbon-activated materials from agriculture and forestry waste that is used for adsorption of dyes from wastewater via an enhanced heteroatom(N,S) content. 17The genome of Magnolia biondii Pamp. provides insights into the evolution of Magnoliales and biosynthesis of terpenoids显示文摘Magnolia biondii Pamp.(Magnoliaceae,magnoliids)is a phylogenetically,economically,and medicinally important ornamental tree species widely grown and cultivated in the north-temperate regions of China.Determining the genome sequence of M.biondii would help resolve the phylogenetic uncertainty of magnoliids and improve the understanding of individual trait evolution within the Magnolia genus.We assembled a chromosome-level reference genome of M.biondii using~67,~175,and~154Gb of raw DNA sequences generated via Pacific Biosciences single-molecule real-time sequencing,10X Genomics Chromium,and Hi-C scaffolding strategies,respectively.The final genome assembly was~2.22Gb,with a contig N50 value of 269.11 kb and a BUSCO complete gene percentage of 91.90%.Approximately 89.17%of the genome was organized into 19 chromosomes,resulting in a scaffold N50 of 92.86Mb.The genome contained 47,547 protein-coding genes,accounting for 23.47%of the genome length,whereas 66.48%of the genome length consisted of repetitive elements.We confirmed a WGD event that occurred very close to the time of the split between the Magnoliales and Laurales.Functional enrichment of the Magnolia-specific and expanded gene families highlighted genes involved in the biosynthesis of secondary metabolites,plant–pathogen interactions,and responses to stimuli,which may improve the ecological fitness and biological adaptability of the lineage.Phylogenomic analyses revealed a sister relationship of magnoliids and Chloranthaceae,which are sister to a clade comprising monocots and eudicots.The genome sequence of M.biondii could lead to trait improvement,germplasm conservation,and evolutionary studies on the rapid radiation of early angiosperms.Shanshan Dong Min Liu Yang Liu Fei Chen Ting Yang Lu Chen Xingtan Zhang Xing Guo Dongming Fang Linzhou Li Tian Deng Zhangxiu Yao Xiaoan Lang Yiqing Gong Ernest Wu Yaling Wang Yamei Shen Xun Gong Huan Liu Shouzhou Zhang 2021Horticulture Research2021,8,1:4

19Platelet-mediated adhesion facilitates leukocyte sequestration in hypoxia-reoxygenated microvessels显示文摘Leukocyte transendothelial migration and sequestration are two distinct outcomes following leukocyte adhesion to endothelium during ischemia-reperfusion injury, in which platelets may play a pivotal role. In the present study, we established an in vitro hypoxia-reoxygenation model to mimic ischemia-reperfusion injury and found platelet pre-incubation significantly increased leukocyte adhesion to endothelial cells after hyoxia-reoxygenation(over 67%). Blockade of endothelial-cell-expressed adhesion molecules inhibited leukocyte direct adhesion to endothelial cells, while platelet-mediated leukocyte adhesion was suppressed by blockade of platelet-expressed adhesion molecules. Further experiments revealed platelets acted as a bridge to mediate leukocyte adhesion, and platelet-mediated adhesion was the predominant pattern in the presence of platelets. However, platelet pre-incubation significantly suppressed leukocyte transendothelial migration after hypoxia-reoxygenation(over 31%), which could be aggravated by blockade of endothelial-cell-expressed adhesion molecules, but alleviated by blockade of platelet-expressed adhesion molecules. This would indicate that platelet-mediated adhesion disrupted leukocyte transendothelial migration. An in vivo mesenteric ischemia-reperfusion model demonstrated leukocyte transfusion alone caused mild leukocyte adhesion to reperfused vessels and subsequent leukocyte infiltration, while simultaneous leukocyte and platelet transfusion led to massive leukocyte adhesion and sequestration within reperfused microvessels. Our studies revealed platelets enhanced leukocyte adhesion to endothelial cells, but suppressed leukocyte transendothelial migration. 20History,progress and future challenges of artificial blood vessels:a narrative review显示文摘Cardiovascular disease serves as the leading cause of death worldwide,with stenosis,occlusion,or severe dysfunction of blood vessels being its pathophysiological mechanism.Vascular replacement is the preferred surgical option for treating obstructed vascular structures.Due to the limited availability of healthy autologous vessels as well as the incidence of postoperative complications,there is an increasing demand for artificial blood vessels.From synthetic to natural,or a mixture of these components,numerous materials have been used to prepare artificial vascular grafts.Although synthetic grafts are more appropriate for use in medium to large-diameter vessels,they fail when replacing small-diameter vessels.Tissue-engineered vascular grafts are very likely to be an ideal alternative to autologous grafts in small-diameter vessels and are worthy of further investigation.However,a multitude of problems remain that must be resolved before they can be used in biomedical applications.Accordingly,this review attempts to describe these problems and provide a discussion of the generation of artificial blood vessels.In addition,we deliberate on current state-of-the-art technologies for creating artificial blood vessels,including advances in materials,fabrication techniques,various methods of surface modification,as well as preclinical and clinical applications.Furthermore,the evaluation of grafts both in vivo and in vitro,mechanical properties,challenges,and directions for further research are also discussed.Ke Hu Yuxuan Li Zunxiang Ke Hongjun Yang Chanjun Lu Yiqing Li Yi Guo Weici Wang 2022Biomaterials Translational2022,3,1:3
