| 2 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 3 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 4 | The High Energy X-ray telescope (HE) onboard the Insight-HXMT astronomy satellite显示文摘The Insight-Hard X-ray Modulation Telescope(Insight-HXMT) is a broadband X-ray and γ-ray(1-3000 ke V) astronomy satellite. One of its three main telescopes is the High Energy X-ray telescope(HE). The main detector plane of HE comprises 18 Na I(Tl)/Cs I(Na) phoswich detectors, where Na I(Tl) is used as the primary detector to measure ~ 20-250 ke V photons incident from the field of view(FOV) defined by collimators, and Cs I(Na) is used as the active shielding detector to Na I(Tl) by pulse shape discrimination. Additionally, Cs I(Na) is used as an omnidirectional γ-ray monitor. The HE collimators have a diverse FOV,i.e. 1.1°×5.7°(15 units), 5.7°×5.7°(2 units), and blocked(1 unit). Therefore, the combined FOV of HE is approximately5.7°×5.7°. Each HE detector has a diameter of 190 mm resulting in a total geometrical area of approximately 5100 cm2, and the energy resolution is ~15% at 60 ke V. For each recorded X-ray event by HE, the timing accuracy is less than 10 μs and the deadtime is less than 10 μs. HE is used for observing spectra and temporal variability of X-ray sources in the 20-250 ke V band either by pointing observations for known sources or scanning observations to unveil new sources. Additionally, HE is used for monitoring the γ-ray burst in 0.2-3 Me V band. | 7 | The Low Energy X-ray telescope (LE) onboard the Insight-HXMT astronomy satellite显示文摘The Low Energy X-ray telescope(LE) is one of the three main instruments of the Insight-Hard X-ray Modulation Telescope(Insight-HXMT). It is equipped with Swept Charge Device(SCD) sensor arrays with a total geometrical area of 384 cm^2 and an energy band from 0.7 to 13 ke V. In order to evaluate the particle induced X-ray background and the cosmic X-ray background simultaneously, LE adopts collimators to define four types of Field Of Views(FOVs), i.e., 1.6°×6°, 4°×6°, 50°-60°×2°-6 oand the blocked ones which block the X-ray by an aluminum cover. LE is constituted of three detector boxes(LEDs) and an electric control box(LEB) and achieves a good energy resolution of 140 e V@5.9 ke V, an excellent time resolution of 0.98 ms, as well as an extremely low pileup(<1%@18000 cts/s). | 9 | Insight-HXMT:China's First X-Ray Astronomical Satellite显示文摘The Hard X-ray Modulation Telescope(HXMT) satellite is the first X-ray astronomical satellite in China. Utilizing the 'direct demodulation image reconstruction method', HXMT will perform broad band(1-250 keV) survey of large sky areas in scanning mode, which can result in source detection characterized with high sensitivity and spatial resolution. Point observation is another key working mode of HXMT, and is specified for observations of galactic bright sources like X-Ray Binary(XRB) and others. Additionally, HXMT also has a powerful capability developed for monitoring gamma-ray bursts, which is quite unique in the world due to its energy coverage of 200 keV-3 MeV and largest detection area. The HXMT satellite was launched by a Long March 4 B(LM-4 B) rocket from the Jiuquan Satellite Launch Center on June 15, 2017. | 10 | Creating a novel herbicide-tolerance OsALS allele using CRISPR/Cas9-mediated gene editing显示文摘Weeds and weedy rice plague commercial rice fields in many countries. Developingherbicide-tolerance rice is the most efficient strategy to control weed proliferation. CRISPR/Cas9-mediated gene editing, which generates small InDels and nucleotide substitutions atand around target sites using error-prone non-homologous end joining DNA repairing, hasbeen widely adopted for generation of novel crop germplasm with a wide range of geneticvariation in important agronomic traits. We created a novel herbicide-tolerance allele inrice by targeting the acetolactate synthase (OsALS) gene using CRISPR/Cas9-mediated geneediting. The novel allele (G628W) arose from a G-to-T transversion at position 1882 of OsALSand conferred a high level of herbicide tolerance. Transgene-free progeny carryinghomozygous G628W allele were identified and showed agronomic performance similar tothat of wild-type plants, suggesting that the G628W allele is a valuable resource fordeveloping elite rice varieties with strong herbicide tolerance. To promote use of the G628Wallele and to accelerate introgression and/or pyramiding of the G628W allele with other elitealleles, we developed a DNA marker for the G628W allele that accurately and robustlydistinguished homozygous from heterozygous segregants. | 11 | Production of Two Elite Glutinous Rice Varieties by Editing Wx Gene显示文摘The waxy gene(Wx) in rice, which encodes the granule bound starch synthase enzyme, is responsible for amylose synthesis. Glutinous(sticky) rice has little or no amylose that can be used in various applications, such as brewing. In this study, knockout of the Wx gene with CRISPR/Cas9 technology was conducted in two elite japonica rice lines, Huaidao 5(HD5) and Suken 118(SK118), aiming to develop elite sticky rice varieties. We achieved six homozygous T_0 plants with more than 200 bp deletion in the Wx gene, as well as 36 wx-HD5 and 18 wx-SK118 homozygous transgene-free plants in the T_1 generation. The seeds of all the mutants were white and opaque, similar to those of sticky rice, and contained only 2.6%–3.2% amylose. Results of scanning electron microscopy showed that the quality of rice did not change. | 12 | Application of neural networks for permanent magnet synchronous motor direct torque control显示文摘Neural networks require a lot of training to understand the model of a plant or a process. Issues such as learning speed, stability, and weight convergence remain as areas of research and comparison of many training algorithms. The application of neural networks to control interior permanent magnet synchronous motor using direct torque control (DTC) is discussed. A neural network is used to emulate the state selector of the DTC. The neural networks used are the back-propagation and radial basis function. To reduce the training patterns and increase the execution speed of the training process, the inputs of switching table are converted to digital signals, i.e., one bit represent the flux error, one bit the torque error, and three bits the region of stator flux. Computer simulations of the motor and neural-network system using the two approaches are presented and compared. Discussions about the back-propagation and radial basis function as the most promising training techniques are presented, giving its advantages and disadvantages. | 13 | The Medium Energy X-ray telescope (ME) onboard the Insight-HXMT astronomy satellite显示文摘The Medium Energy X-ray telescope(ME) is one of the three main telescopes on board the Insight hard X-ray modulation telescope(Insight-HXMT) astronomy satellite. ME contains 1728 pixels of Si-PIN detectors sensitive in 5-30 ke V with a total geometrical area of 952 cm^2. The application specific integrated circuit(ASIC) chip, VA32TA6, is used to achieve low power consumption and low readout noise. The collimators define three kinds of field of views(FOVs) for the telescope, 1°×4°, 4°×4°,and blocked ones. Combination of such FOVs can be used to estimate the in-orbit X-ray and particle background components.The energy resolution of ME is ~3 ke V at 17.8 ke V(FWHM) and the time resolution is 255 μs. | 14 | Hard X-Ray Modulation Telescope(HXMT) Mission显示文摘The HXMT mission concept consists of a slat-collimated hard X-ray detector assembly sensitive in 20~250 keV with a collection area of about 5000 cm2. Based on the reconstruction technique by direct demodulation developed in recent years, HXMT is mainly devoted to performing a hard X-ray all-sky imaging survey with both high sensitivity and high spatial resolution. It can also be used to make pointed observations of X-ray sources to study their spectroscopic and temporal properties in details. The main detector of HXMT consists of 18 individual cylindrical NaI(T1)/CsI(Na) phoswich modules, each with anarea of 283.5 cm2 and a field of view of 5.7°× 1.1° (FWHM). Its spatial resolution and position accuracy are 5′ and 1′ by using the direct demodulation in 1994, and in 2000 its feasibility and technical demonstration study was selected as a project under the Major State Basic Research Program of China. In October 2005, this project entered the full design phase and was listed as a candidate for the first dedicated astronomy satellite around 2010. | 15 | A strategy with label-free quantification of the targeted peptides for quantitative peptidome analysis of human serum显示文摘Peptidomics draws more and more attention in discovering useful biomarkers for early diagnosis of disease. However, there is lack of efficient quantification strategy in peptidome analysis. In this study, a strategy with label-free quantification of the targeted endogenous peptides based on peak intensity using μUPLC-Q-TOF-MS/MS was developed for quantitative peptidome analysis of human serum. Different amounts of standard BSA tryptic digesting peptides were added into the same serum extracts for evaluation of the developed strategy, and it was observed that the average relative error of the targeted peptides was 6.42%, which was superior to the result obtained directly by commercially available software PLGS. It was also demonstrated that this quantification strategy could obviously increase the detection sensitivity of the peptide by DDA analysis. Then, this strategy was applied to comparatively analyze the peptides extracted from the serum of HCC or breast cancer patients and healthy individuals, respectively. Peptides with charge states up to 5 and molecular weight over 4000 can be reliably identified and quantified. This quantitative analysis method based on μUPLC-Q-TOF-MS/MS exhibited superior sensitivity than that by MALDI-TOF-MS commonly used in peptidome analysis. The rice blast resistant protein Pi-ta, differs from its susceptible counterpart, pi-ta, by a single amino acid in exon 2. To create new materials resistant to the rice blast disease, we inserted a genomic fragment containing the exon 2 and 3′ untranslated region(3′ UTR) of Pi-ta into intron 1 of pi-ta in rice materials susceptible to rice blast using the intron-targeted insertion strategy. The gene insertion frequency was3.8%. Several novel transgene-free progeny with stably inherited homozygous insert were identified in the T_1 generation, which have been crossed to rice germplasm bearing other resistance gene(R gene) for pyramiding of R genes. In this review, we focus on the design and application of multicolor fluorescent probes based on small organic molecules: single fluorophores with multiple reaction sites, multiple fluorophores with single reaction sites, or multiple fluorophores with multiple reaction sites. Moreover, a design strategy for multicolor fluorescent probes and its application in biological imaging are also summarized, providing a systematic plan for future research on fluorescent probes functionalized by small organic molecules. | 19 | Laser Remelting of Plasma Sprayed NiCrAlY and NiCrAlY-Al_2O_3 Coatings显示文摘Two types of plasma sprayed coatings (NiCrAlY and NiCrAlY-A12O3) were remelted by a 5 kW cw CO2 laser. With increasing laser power and decreasing traverse speed in the ranges of 200-700 W and 5-30 mm/s respectively, the melted track grew in width and depth. In the optimum range of laser parameters, a homogeneous remelted layer without voids, cavities, unmelted particles and microcracks was formed. On the surface of remelted layers, Al203 and YAIO3 were detected. As a result of isothermal oxidation tests, weight gains of laser remelted coatings were obviously lower than that only plasma sprayed, especially laser remelted NiCrAlY-Al2O3 coatings.
