1Hes1 Knockdown Exacerbates Ischemic Stroke Following tMCAO by Increasing ER Stress-Dependent Apoptosis via the PERK/ eIF2a/ATF4/CHOP Signaling Pathway显示文摘Apoptosis induced by endoplasmic reticulum(ER)stress plays a crucial role in mediating brain damage after ischemic stroke.Recently,Hes1(hairy and enhancer of split 1)has been implicated in the regulation of ER stress,but whether it plays a functional role after ischemic stroke and the underlying mechanism remain unclear.In this study,using a mouse model of ischemic stroke via transient middle cerebral artery occlusion(tMCAO),we found that Hes1 was induced following brain injury,and that siRNA-mediated knockdown of Hes1 increased the cerebral infarction and worsened the neurological outcome,suggesting that Hes1 knockdown exacerbates ischemic stroke.In addition,mechanistically,Hes1 knockdown promoted apoptosis and activated the PERK/eIF2a/ATF4/CHOP signaling pathway after tMCAO.These results suggest that Hes1 knockdown promotes ER stress-induced apoptosis.Furthermore,inhibition of PERK with the specific inhibitor GSK2606414 markedly attenuated the Hes1 knockdown-induced apoptosis and the increased cerebral infarction as well as the worsened neurological outcome following tMCAO,implying that the protection of Hes1 against ischemic stroke is associated with the amelioration of ER stress via modulating the PERK/eIF2a/ATF4/CHOP signaling pathway.Taken together,these results unveil the detrimental role of Hes1 knockdown after ischemic stroke and further relate it to the regulation of ER stress-induced apoptosis,thus highlighting the importance of targeting ER stress in the treatment of ischemic stroke.Yueyong Li Yingjun Zhang Huangde Fu Huadong Huang Qifeng Lu Houji Qin Yingning Wu Huatuo Huang Guizhen Mao Zhongheng Wei Pinhu Liao 2020Neuroscience Bulletin2020,36,2:12
2Shuganjieyu capsule increases neurotrophic factor expression in a rat model of depression显示文摘Shuganjieyu capsule has been approved for clinical treatment by the State Food and Drug Administration of China since 2008. In the clinic, Shuganjieyu capsule is often used to treat mild to moderate depression. In the rat model of depression established in this study, Shuganjieyu capsule was administered intragastrically daily before stress. Behavioral results confirmed that depressive symptoms lessened after treatment with high-dose(150 mg/kg) Shuganjieyu capsule. Immunohistochemistry results showed that high-dose Shuganjieyu capsule significantly increased phosphorylation levels of phosphorylation cyclic adenosine monophosphate response element binding protein and brain-derived neurotrophic factor expression in the medial prefrontal cortex and hippocampal CA3 area. 3Platelet transfusion refractoriness after T-cell-replete haploidentical transplantation is associated with inferior clinical outcomes显示文摘Haploidentical stem cell transplantation (haplo-SCT) has been an alternative source of bone marrow for patients without human leukocyte antigen (HLA)-matched donors. The aim of this study was to investigate the relationships between platelet transfusion refractoriness (PTR) and clinical outcomes in the setting of haplo-SCT. Between May 2012 and March 2014, 345 patients who underwent unmanipulated haplo-SCT were retrospectively enrolled. PTR occurred in 20.6% of all patients. Patients in the PTR group experienced higher transplant-related mortality (TRM, 43.7% vs. 13.5%, P<0.001), lower overall survival (OS, 47.9%vs. 76.3%, P<0.001) and lower leukemia-free survival (LFS, 47.9% vs. 72.3%, P<0.001) compared to patients in the non-PTR group. The multivariate analysis showed that PTR was associated with TRM (P=0.002), LFS (P<0.001), and OS (P<0.001).The cumulative incidences of PTR in patients receiving >12 platelet (PLT) transfusions (third quartile of PLT transfusions) were higher than in patients receiving either >6 (second quartile) or >3 (first quartile) PLT transfusions (56.1% vs. 41.6% vs. 28.2%,respectively; P<0.001). The multivariate analysis also showed that PTR was associated with the number of PLT transfusions(P<0.001). 6Protist Interactions and Seasonal Dynamics in the Coast of Yantai, Northern Yellow Sea of China as Revealed by Metabarcoding显示文摘Facilitated by the high-throughput sequencing(HTS)technique,the importance of protists to aquatic systems has been widely acknowledged in the last decade.However,information of protistan biotic interactions and seasonal dynamics is much less known in the coast ecosystem with intensive anthropic disturbance.In this study,year-round changes of protist community composition and diversity in the coastal water of Yantai,a city along the northern Yellow Sea in China,were investigated using HTS for the V4 region of 18S rDNA.The interactions among protist groups were also analyzed using the co-occurrence network.Data analyses showed that Alveolata,Chlorophyta,and Stramenopiles are the most dominant phytoplanktonic protists in the investigated coastal area.The community composition displayed strong seasonal variation.The abundant families Dino-Group-I-Clade-1 and Ulotrichales_X had higher proportions in spring and summer,while Bathycoccaceae exhibited higher ratios in autumn and winter.Alpha diversities(Shannon and Simpson)were the highest in autumn and the lowest in spring(ANOVA test,P<0.05).Nutrients(SiO42−,PO43−),total organic carbon(TOC),and pH seemed to drive the variation of alpha diversity,while temperature,PO43−and TON were the most significant factors influencing the whole protist community.Co-variance network analyses reveal frequent co-occurrence events among ciliates,chlorophytes and dinoflagellate,suggesting biotic interactions have been induced by predation,parasitism and mixotrophy.FU Yingjun ZHENG Pengfei ZHANG Xiaoli ZHANG Qianqian JI Daode 2020Journal of Ocean University of China2020,19,4:3
7Application of Preoperative Visits during the Perioperative Period of Ophthalmic Surgery显示文摘Purpose:.To evaluate the effect of preoperative visits on patients′ psychology, physiology, and behavior during the perioperative period of eye surgery under local anesthesia, with the aim of enhancing patients′ cooperation with the surgery and improving their degree of satisfaction.Methods:.A total of 240 patients scheduled to undergo eye surgery between August and October 2013 were randomly divided into an observation(n = 120) and a control(n = 120)group..Patients in the observation group attended preoperative visits with nurses and received conventional nursing care. The control group received only conventional nursing.Results:.The Zung self-rating anxiety scale(SAS) scores were significantly lower in patients from the observation group than in the control group(P<0.05). Surgeons operating on the observation group were more satisfied with their patients′ cooperation with the surgery than were surgeons operating on the controls(P<0.01). 8Segmented Au/PtCo heterojunction nanowires for efficient formic acid oxidation catalysis显示文摘Exploring a new strategy for the removal of adsorbed CO (CO^(*)) on a Pt surface at a low potential is the key to achieving enhanced catalysis for the formic acid oxidation reaction (FAOR);however, the development of such a strategy remains a significant challenge. Herein, we report a class of Au/PtCo heterojunction nanowires (HNWs) as efficient electrocatalysts for accelerating the FAOR. This heterojunction structure and the induced Co alloying effects can facilitate formic acid adsorption/activation on Pt with high CO tolerance, generating the FAOR pathway from dehydration to dehydrogenation. The optimized Au_(23)/Pt_(63)Co_(14) HNWs showed the highest specific and mass activities of 11.7 mA cm^(−2)Pt and 6.42 A mg^(−1)Pt reported to date, respectively, which are considerably higher than those of commercial Pt/C. 10Modification of adipose mesenchymal stem cells-derived small extracellular vesicles with fibrin-targeting peptide CREKA for enhanced bone repair显示文摘The process of bone repair is highly regulated by a large number of bioactive factors.Thus,a“cocktail”of bioactive factors supplemented to the defect sites is desirable for bone repair.In this regard,small extracellular vesicles(sEVs)derived from mesenchymal stem cells hold great potential in tissue repair.Nevertheless,the poor homing and retention of sEVs greatly limited their possible clinical application.In the present work,DMPE-PEG-CREKA was inserted into the membrane of sEVs released from adipose-derived mesenchymal stem cells to obtain CREKA functionalized sEVs(CREKA-sEVs),which could target fibrin to accumulate and retain in bone defects.Our results showed that CREKA-sEVs,like sEVs,promoted the osteogenic differentiation of BMSCs,the angiogenic property of HUVECs,and modulated the polarization of macrophages in vitro.Furthermore,due to the improved fibrin-binding and retention capacity of CREKA-sEVs,they enhanced the bone repair substantially in the rat femoral defect model.This study provided a new strategy to improve the therapeutic efficiency of sEVs and showed that CREKA-sEVs had great application value in bone tissue repair.Qi Wu Xiaoling Fu Xian Li Jing Li Weiju Han Yingjun Wang 2023Bioactive Materials2023,,2:1

12Switching between eltrombopag and recombinant human thrombopoietin in patients with immune thrombocytopenia: an observational study显示文摘Background:Recombinant human thrombopoietin(rh-TPO)and eltrombopag are two distinct TPO receptor agonists(TPO-RAs)with different mechanisms.During the pandemic,when immunosuppressive medications are controversial,switching to another TPO-RA may be worth exploring in patients who do not benefit from their first TPO-RA.We investigated the outcomes of switching from rh-TPO to eltrombopag or vice versa in immune thrombocytopenia(ITP)patients.Methods:This prospective,open-label,observational investigation included 96 adult ITP patients who needed to switch between rh-TPO and eltrombopag between January 2020 and January 2021 at Peking University People’s Hospital in China.The study evaluated response rates and platelet counts at different time points after the switch,bleeding events,time to response,duration of response,and adverse events.Results:At 6 weeks after switching,response was observed in 21/49 patients(43%)who switched for inefficacy and 34/47 patients(72%)who switched for non-efficacy-related issues.In the inefficacy group,9/27 patients(33%)responded to eltrombopag,and 12/22 patients(55%)responded to rh-TPO.In the non-efficacy-related group,21/26(81%)and 13/21(62%)patients in the eltrombopag and rh-TPO groups maintained their response rates at 6 weeks after switching,respectively.Response at 6 months was achieved in 24/49 patients(49%)switching for inefficacy and 37/47 patients(79%)switching for non-efficacy issues.In the inefficacy group,13/27 patients(48%)responded to eltrombopag,and 11/22 patients(50%)responded to rh-TPO.In the non-efficacy-related group,22/26 patients(85%)and 15/21 patients(71%)in the eltrombopag and rh-TPO groups maintained their response rates at 6 months after switching,respectively.Both eltrombopag and rh-TPO were well tolerated.Conclusions:Our study confirmed the safety and effectiveness of switching between rh-TPO and eltrombopag for ITP patients who had no response to or experienced adverse events with their first TPO-RA.When the switch was motivated by other reasons,including patient preference and platelet count fluctuations,the probability of response was high.Registration:ClinicalTrials.gov,NCT04214951.Xuan Cai Xiaixia Fu Xiangyu Zhao Jin Lu Qian Jiang Yingjun Chang Xiaojun Huang Xiaohui Zhang 2022Chinese Medical Journal2022,135,19:0
